We investigated the effects of beta‐glucans (Saccharomyces cerevisiae) ingestion on metabolic parameters of Wistar rats receiving high‐fat diet. The experimental period was divided into two stages: in the first one, the animals were divided into two groups containing 12 animals each. The first group received commercial feed and the second received high‐fat diet containing 20% of pork fat during 60 days. At the end of this period, body weight, blood glucose and Lee index were assessed. In the second stage, those 24 animals were redivided into four groups: (C) – control diet; (CB) – control diet and treated with Beta‐glucan (BG); (O) – obese animals and (OB) – obese animals treated with BG. Animals from groups CB and OB received 30 mg/kg of BG dissolved in saline solution by gavage. Animals from groups C and O received only saline solution for 28 days. The design used was totally randomized in 2 × 2 factorial scheme. Data were submitted to analysis of variance (anova ). Animals from OB group showed inferior levels (p < 0.05) of total cholesterol (13.33%), triacylglycerols (16.77%) and blood glucose (23.97%) when compared to the animals from group O. The use of BG has provided smaller increase in Lee index (p < 0.05), without promoting alteration in feed and water consumption, organs weight, HDL‐C, LDL+VLDL‐C, carcass composition, villus/crypt ratio, and pancreas, kidney and stomach histology. BG from S. cerevisiae promoted beneficial metabolic effects in rats receiving high‐fat diet. 相似文献

Grapevine Pinot gris Virus (GPGV) is a single stranded RNA of the genus Trichovirus infecting grapevine (Vitis vinifera) and associated with stunting, chlorotic mottling and leaf deformation symptoms. During a monitoring of GPGV infection in vineyards of the Trentino region in Italy, we have detected the virus in the herbaceous plants Silene latifolia subsp. Alba (Mill.) (bladder campion) and Chenopodium album L. (white goosefoot), which showed symptoms of viral infection. The full-length GPGV RNA genome, amplified from these infected hosts, was sequenced and a phylogenetic analysis revealed that its closest relative is the strain SK13, recently isolated in Slovakia. Our results indicate that herbaceous plants can be considered as a reservoir for the GPGV virus. This finding is important for studying the epidemiological aspects of GPGV disease and to formulate appropriate control measures. 相似文献

We here isolate fulvic acids from vermicompost to prepare and characterize novel fulvic acid-coated magnetite nanoparticles. UV-A irradiation of suspensions of the nanoparticles under different experimental conditions led to photo-reduction of Cr(VI). In anoxic conditions in the presence of formic acid, after 60 min of irradiation ca. 100% of Cr(VI) was reduced. Under these conditions, the carbon dioxide radical anions, CO2 .- , mediated the photo-reduction of Cr(VI). However, the high reduction potential of Cr(VI) and the variety of reactive species generated upon UV-A irradiation make this nanomaterial also suitable for Cr(VI) photo-reduction also under aerobic conditions in the presence of formic acid or under anoxic conditions without the addition of formic acid. The possible photodegradation routes involved are discussed in detail. 相似文献

Purpose
The use of humic substances (HS) in agriculture is beneficial and has positive environmental impacts. However, to optimize the use of HS possible links between their structural characteristics and bioactivity must be shown. The goal of this study is to evaluate the bioactivity of different humic fractions extracted from vermicompost (VC) in rice plants and to shed light to possible structure-function relationships.Materials and methods
Humic-like fractions were obtained from cattle manure vermicompost processed by African nightcrawlers (Eudrilus eugeniae spp.). Humic-like acid fraction using only water as extractor (HLAw), HLA fraction extracted following the International Humic Substances Society (IHSS) recommended method, and the solid residue (humified residual (HR)) after extraction of HLA were characterized using complementary chemical, physic, and spectroscopic technics (elemental composition, UV-Vis and Fourier transform infrared spectroscopy (FTIR) spectroscopies, 13C-CP MAS NMR, and MEV). Biological activity of the three HS was conducted in growth chambers and measured in roots using WinRhizo Arabidopsis software. Principal component analysis (PCA) was used to find a grouping pattern between the structural variables evaluated and the obtained root parameters.Results and discussion
Differences were found in elemental composition among HS with larger C/N ratio in HR than in HLA and HLAw. HLA and HLAw FTIR spectra showed carboxyl band at 1714.66 cm?1 better resolved than in HR. Bands at 1642 cm?1 (amide I) and 1510 cm?1 (lignin), were better resolved in HLA. 13C-NMR showed the following order of aromaticity: HLA > HLAw > HR. For HLAw bioactivity, the structures CAlkyl-H,R, CC=O, and CCOO-H,R correlated with the number and growth of smaller root. The aromatic CAr-H,R, CAr-O,N, and aliphatic CAlkyl-O,N, CAlkyl-O, and CAlkyl-di-O structures in HLA, correlated with larger roots growth. HR also stimulated root growth and development in rice plants.Conclusions
Aliphatic and oxygenated structures in HLAw showed a relation with induction of initial root emissions, whereas the presence of aromatic compounds in HLA was related with root growth stimulation activity. Higher concentration of HLAw was necessary to produce an equivalent stimulus compared with HLA; it could indicate that, although both fractions showed similar types of structures in their composition, differences in the predominant structures may be determining different effects on the root.135.
Purpose
Excitation–emission matrices spectroscopy (EEMS) of soil humic acids (HAs) contains large amount of information on their properties, as well as on the dynamics related to their intra- and inter-molecular interactions. The objective of this research was (i) to show that EEMS represents a useful tool to investigate the molecular and mechanistic aspects of HA adsorbing capacity towards atrazine (A) and paraquat (P) with or without Cu2+ ions and (ii) to evaluate if additional information on these mechanisms can be obtained by combining EEMS with principal component analysis (PCA).Materials and methods
HAs have been isolated from soil samples collected in a citrus field at three locations, within the plant rows (HAa); between the rows (HAb) and in a adjacent, control soil (HAc). Interaction products were obtained between each HA and A and P, with or without Cu ions. Elemental analysis and Fourier-transorm Infrared Spectroscopy were applied to support fluorescence data. Fluorescence spectra were recorded on aqueous solutions, and fluorescence intensity (FI) values were normalized using a quinine sulphate standard. PCA analysis was performed using the software STATGRAPHICS Centurion XV.I.Results and discussion
The EEM spectra of the three unreacted HAs are characterized by the presence of two fluorophores α and β, in the region of longer wavelengths pairs (EEWP). The EEM spectra of the HA-A interaction products are featured by the same two fluorophores, with FI values decreased of about 50%, whereas those of the HA-P interaction products show a unique peak, γ, at intermediate EEWP. Finally, the EEM spectra of HA-A-Cu2+ show, with respect to HA-A samples, a blue shift of the α peak with an additional decrease of FI values (about 60%) and the disappearance of the peaks β, whereas those of HA-P-Cu2+ feature, in comparison with those of HA-P, a small red-shift of the peak γ. PCA data suggest that Cu ions do not affect the interaction mechanism between HA and P, whereas it appears to exercise a strong influence on interaction between HA and A.Conclusions
The results obtained indicate that EEMS allows direct measurements of the adsorbing capacity of HA towards atrazine and paraquat. Additional information obtained by PCA analysis show that Cu ions behave like a good antagonist in preventing the formation of ionic bonds between HA and atrazine, whereas Cu is not able to affect the prevalent mechanism of HA interaction with paraquat, which is a charge-transfer bond.136.

Plants of the urban environment are exposed to a wide range of pollutants, including heavy metals. This research studies in situ the eco-physiological and antioxidant responses of holm oak (Q. ilex) leaves to Pb and Cd to assess the mechanisms of metal tolerance in this species, widely used as biomonitor. Leaves of plants grown at parks and roadsides were analyzed for photosynthetic activity, Pb and Cd concentration in tissues and cell-free extracts, thiol groups, D1 and Rubisco protein content, ascorbic acid (AsA) amount, and catalase (CAT) activity. The main results evidenced that Cd concentration was higher in leaves collected at the park out from the downtown; whereas Pb was most abundant in leaves sampled at the roadside nearby the highway. Pb in cell-free extracts was higher in park than in roadside leaves. Although Cd in the leaf tissues was twofold lower than Pb, it was more abundant than Pb in cellular extracts deprived of all particulate matter. Leaves responded to different concentration of Cd and Pb modulating some eco-physiological and biochemical traits, roadside leaves showed reduced leaf lamina, higher content of photosynthetic pigments, hydrogen peroxide, and AsA, as well as higher CAT activity compared to park leaves. In the roadside leaves, a stress condition for photosynthetic apparatus can be hypothesized on the basis of the decline of photochemical activity, the increase of NPQ, and the reduction of Rubisco and D1 protein content. The elevated presence of thiol groups in these leaves suggests a possible role of Pb and Cd in activation of antioxidant responses. 相似文献

Stylosanthes Sw. is considered one of the economically most important tropical forage legumes. The genus is essentially Neotropical, and Brazil, Venezuela and Mesoamerica are the main centres of species diversification. In view of its importance, an extensive review of Venezuelan Stylosanthes genetic resources was carried out. Firstly, the known natural distributions of all 11 species based on herbarium specimens were georeferenced and mapped and, by comparing them with information of germplasm (=seed) collection site distributions, for each species regions were identified which in terms of collecting and safeguarding genetic resources of Stylosanthes had not been explored yet. Secondly, the actual status of the Stylosanthes collection in Venezuela in terms of collected seed samples and available germplasm is discussed, as is the need for further explorations, including targeting ecologically important locations. Finally, the different Stylosanthes species in Venezuela are grouped according to their perceived agronomic value and potential as forage for animal production systems, and recommendations are given regarding research, collection and preservation needs. Overall, this study compiled baseline information for future considerations of an important genetic resource in Venezuela. The endangerment of the three endemic species, Stylosanthes falconensis Calles et Schultze-Kr., Stylosanthes sericeiceps S.F. Blake and Stylosanthes venezuelensis Calles et Schultze-Kr. should be a major concern and warrants priority considerations for preservation. 相似文献

Ixodid ticks require comparatively large bloodmeals for their development and survival. Blood-feeding elicits signaling events in the host leading to wound healing responses (hemostasis, inflammation, and tissue repair) and immunity. Bioactive molecules present in tick saliva sabotage these host responses at several levels. One of them is neutralization of cellular communication by binding of specific saliva molecules to cytokines that have important roles in innate and adaptive immunity. Chemokines are a subset of cytokines having chemotactic activities. We show anti-chemokine activities in salivary gland extracts (SGE) of adult Rhipicephalus appendiculatus ticks against human chemokines CXCL8, CCL2, CCL3, CCL5, and CCL11. At comparable protein concentrations, male Ixodes ricinus SGE showed activity against all the chemokines; SGE of female I. ricinus had comparatively lower levels of activity against all the chemokines but no detectable activity against CCL5 and CCL11. However, when the equivalent of a single pair of salivary glands was tested, male I. ricinus showed little or no activity against CCL3 and CCL5. No fundamental differences in activity were observed against mouse compared with human chemokines. A comparison with previously published data for Dermacentor reticulatus and Amblyomma variegatum indicates that the level of anti-cytokine activity depends on the species, developmental stage (adult or nymph), and amount of SGE used, as well as on the number of days the tick has been feeding. 相似文献

Magnetic resonance (MR) imaging has become an important diagnostic tool in the investigation of foot pain in horses. The aim of this study was to qualitatively compare ex-vivo MR images of the same equine feet obtained at three magnetic field strengths: 0.27, 1.5, and 3 tesla (T). Ten cadaver feet were used. All feet were imaged with two high-field systems (3T, 1.5T) and with a low-field (LF) system at 0.27 T designed for standing horses. Images were acquired using similar pulse sequences in all 3 MR units. MR images were subjectively evaluated by three independent experienced image analysts for image quality and clarity of visualization of individual anatomical structures using a four-point grading scale. The images from all of the examinations were considered to be of diagnostic value except for the hoof capsule where substantial artifacts were present in LF images with distortion and loss of signal at the dorsal/distal aspect of the hoof capsule in LF images. Anatomical structure scoring values of images obtained at 3T and 1.5T were significantly greater than scores of images obtained at 0.27T. Scores for images obtained at 3T were significantly higher than those for images obtained at 1.5T. Mean score differences between 1.5T and 3T were higher for cartilage of the distal interphalangeal joint and for the ungular cartilages. 相似文献
