Phenotypic analysis for a cell line of canine epidermal keratinocytes   总被引:1,自引:0,他引:1  
Epidermal keratinocytes have the potential to produce inflammatory mediators that are considered to play an important role in skin diseases such as atopic dermatitis (AD). Thus, cell lines of canine epidermal keratinocytes are useful for studying the biological reactivity of keratinocytes in vitro. However, there has been no report on properly analyzing the phenotype of canine keratinocyte cell lines. In this work, we performed phenotypic analysis of CPEK, which was derived from the epidermis of an adult dog in order to examine the phenotypic similarity with epidermal keratinocytes. The present findings indicated that CPEK cells expressed markers for epidermal keratinocytes including cytokeratin 14, alpha6 integrin and PCNA. Our findings demonstrated that CPEK could be a useful cell line for investigating the central role of epidermal keratinocytes in the pathogenesis of AD in vitro.  相似文献

A microplate agglutination test (MAT) was compared with the tube agglutinin test (TAT), a standard test for the diagnosis of Brucella canis, in terms of the sensitivity and specificity. The results showed that MAT was more sensitive, simpler to perform and easier to read the results than TAT. On top of that the MAT allows us to handle a larger number of samples at once. Using this method we conducted sero-surveillance of the prevalence of B. canis in dogs kept in an Animal Shelter located in Kanagawa Prefecture. Twelve of 485 (2.5%) showed seropositive against B. canis. These results indicate that B. canis infection in dogs is still occurring in Japan.  相似文献

Twenty ferrets with histopathologically diagnosed lymphoma were classified cytomorphologically and immunohistochemically. According to site of origin, multicentric, gastrointestinal, mediastinal and cutaneous lymphomas accounted for 8 (40%), 9 (45%), 2 (10%) and 1 case (5%), respectively. According to the National Cancer Institute Working Formulation (NCI-WF), low-, high- and intermediate-grade lymphomas accounted for 4 (20%), 4 (20%) and 12 cases (60%), respectively. The 4 low-grade lymphomas showed no mitotic figures, whereas all 4 high-grade lymphomas exhibited > or = 3 mitotic figures (median,6). Higher grade thus appears to be associated with a higher number of mitotic figures. Immunohistochemical examination of 18 specimens, excluding 2 insufficient specimens, showed that 16 (88.9%) and 2 (11.1%) lymphomas were of T-cell origin and B-cell origin, respectively. According to the combination of the NCI-WF and immunophenotypes, all 4 low-grade lymphomas (2 multicentric, 1 gastrointestinal, and 1 cutaneous lymphoma) were classified as diffuse small lymphocytic lymphoma of T-cell origin. Of the 12 intermediate-grade lymphomas (6 multicentric, 4 gastrointestinal, and 2 mediastinal lymphomas), 11 were classified as diffuse mixed-cell lymphoma, and 1 as diffuse large cell lymphoma. Of these 11 lymphomas, 2 (both multicentric) were of B-cell origin, 7 (3 multicentric, 3 gastrointestinal, 1 mediastinal) were of T-cell origin, and 2 (1 multicentric, 1 mediastinal) were of unknown cell origin. The remaining 1 lymphoma (gastrointestinal) was of T-cell origin. All 4 high-grade lymphomas (gastrointestinal) were classified as diffuse immunoblastic lymphoma of T-cell origin.  相似文献

Climate change will have significant impacts on the rain-fed rice production ecosystem, and particularly on the ecosystem’s hydrology and water resources. Under rain-fed lowland conditions, substantial variations among fields in grain yield are commonly observed, but a method that can account for field-scale yield variability to produce regional-scale yield estimates is lacking, thereby limiting our ability to predict future rice production under changing climate and variable water resources. In this study, we developed a model for estimating regional yields of rain-fed lowland rice in Northeast Thailand, by combining a simple crop model with a crop calendar model. The crop model incorporates the effects of two important resources (water and nitrogen) on crop growth. The biomass accumulation is driven by water use, whereas the nitrogen supply determines canopy development and thereby constrains crop water use. Accounting for the wide range of planting dates and the strong photoperiod-sensitive characteristics of rice varieties through the calendar model is an essential component in determining regional yield estimates. The present model does not account for the effects of mid-season drought or flooding, but was nonetheless able to explain the spatial and temporal yield variations at the province level for the past 25 years. Thus, it can be used as a prototype for simulating regional yields of rain-fed lowland rice.  相似文献

Rain-fed lowlands are major agricultural ecosystems used for rice production in Northeast Thailand. Developing a tool to assess the effects of variable water conditions on the regional scale yield is also important to predict the effects of climate change on food supply. To estimate regional yields, we need a simple but accurate measure of the crop calendar (i.e., the distribution of planting dates), which has a strong influence on grain yield. In this article, we modeled the dependence of the crop calendar on rainfall patterns based on a survey of the region’s farmers as a part of an effort to provide a stronger basis for regional yield estimates. Our survey, conducted in 11 provinces for 2 years, confirmed the existence of large windows for sowing and transplanting versus narrow windows for heading and harvesting for rain-fed lowland rice culture in all the provinces. Variable water, soil, and toposequential conditions in the paddy fields were responsible for the large sowing and transplanting windows, whereas the use of photoperiod-sensitive varieties explained the narrow windows for heading and harvesting. The crop calendar was well expressed as a function of cumulative precipitation from June onward. When the crop calendar model was combined with a simple phenology-based model that uses growing degree-days adjusted by a day-length factor, we could estimate the rice crop calendar under rain-fed lowland conditions with acceptable accuracy. The model described in this article will be combined with a crop growth model to improve regional yield estimates for rain-fed lowland rice.  相似文献

A 9-year, 7-month-old female German shepherd weighing 26.6 kg was admitted to the hospital for pica and diarrhea. A large mass was found in the right ovary and removed, and cross section of the mass revealed a multilobular tumor consisting of several cystic cavities which contained tufts of dark hair in thick creamy-white sebaceous fluid. Histologically, the tumor consisted of adipose tissue, central nervous tissue, crystalline lens, cartilage and bone. In the central nervous tissue, lens and lesions like nonsuppurative inflammation comprizing of accumulation of glial cells and lymphocytic perivascular cuffing were observed. The tumor was diagnosed as a mature cystic teratoma.  相似文献

A novel colitis model using Syrian hamsters was developed. Colitis was induced by intracolonic administration of 1% acetic acid, and the ulcer area, tissue myeloperoxidase (MPO) activity, and luminal neutrophil elastase (NE) activity of the colon were determined at 1, 3, 8, 24 and 48 hr after colitis induction. The histopathological changes of the colon were also examined in this model. An increase of tissue MPO activity and NE activity was evident at 3 hr after induction of colitis, peaked at 24 hr, and decreased subsequently. The increase of luminal NE activity was well correlated with the colonic ulcer area. In histopathological examination, ulceration, erosion, crypt abscesses, neutrophil infiltration, hemorrhage, and edema were seen. The effects of prednisolone were examined to evaluate the adequacy of our colitis model. Syrian hamsters were treated orally with prednisolone at 18 and 1 hr before and at 6 hr after induction of colitis, and the ulcer area, tissue MPO activity, and luminal NE activity were evaluated at 24 hr after colitis induction. Prednisolone therapy had little effect on the tissue MPO activity. However, the NE activity of the prednisolone-treated group was significantly decreased. In addition, although prednisolone did not significantly decrease the ulcer area, a tendency toward decrease was noted. We conclude that this new model of experimental colitis in Syrian hamsters is useful for investigating the pathophysiology of colitis, especially useful for studying the relationship between colitis and NE activity.  相似文献

A total of 1,335 archived human sera collected in 1985 from an area in Japan where a tick-borne disease is endemic were examined by indirect immunofluorescence antibody test (IFAT) to estimate seroprevalence against three serologically distinct types of Babesia microti-like parasites; namely, Hobetsu, Kobe, and U.S. types. Eighteen sera (1.3%) were found to be IFAT-positive (titer 1:100-1:6,400), of which 14 and three were ascertained by Western blot analysis to be positive against the Hobetsu and Kobe types, respectively. In addition, four sera showed an IFAT titer of 1:100 against the U.S. type, but they appeared to be false-positive because they were cross-reactive against the Hobetsu and Kobe types, and also because a U.S.-type parasite has not been found in Japan. Our results suggest that human babesiosis in Japan occurred prior to the discovery of the index case in 1999 and that the infections were caused mainly by Hobetsu-type parasites.  相似文献
