We investigated reproduction of the clonal tree Robinia pseudoacacia in a Pinus thunbergii windbreak. Microsatellite polymorphism analysis revealed that asexual reproduction through horizontal root elongation and ramet formation was the primary mode of reproduction in this population. Germination experiments indicated that the inability of established genets to produce viable seeds inhibited sexual reproduction. The boundaries between genet areas were relatively distinct, indicating that ramets within genets spatially excluded one another. Analyses of the number of annual rings and thickness of excavated horizontal roots provided new insight regarding photosynthate transfer through horizontal roots. The number of annual rings and thickness were highest for horizontal roots near the base of ramets and decreased along portions far from ramets. This result indicates that the new horizontal root is supplied with photosynthate from the mother ramet for a few years only, and that resource translocation between neighboring ramets within the same clone ceases after several years. Hypotheses about horizontal root expansion and cessation were considered to explain the exclusive distribution pattern of the horizontal root between genets.  相似文献

To initiate defense responses against invasion of pathogenic organisms, animals and plants must recognize microbe-associated molecular patterns (MAMPs). In this study, the elicitor activity of bacterial DNA on the model plant Arabidopsis thaliana was examined. EcoRI-digested plasmid DNA induced defense responses such as generation of reactive oxygen species and deposition of callose, whereas SmaI- and HapII-digested plasmid DNA and EcoRI-digested herring DNA did not remarkably induce these responses. Further, methylation of the CpG sequence of plasmid DNA and Escherichia coli DNA reduced the level of the defense responses. The endocytosis inhibitors wortmannin and amantadine significantly inhibited DNA-induced defense responses. These results suggest that non-methylated CpG DNA, as a MAMP, induced defense responses in Arabidopsis and that non-methylated DNA seems to be translocated into the cytoplasm by endocytosis.  相似文献

We examined the age and growth of the marbled sole Pseudopleuronectes yokohamae collected from Tokyo Bay, Japan, during two study periods between 2002 and 2008, when the stock size was consistently low. The von Bertalanffy growth equations were found to differ significantly between sexes, and females attained a larger standard length than males of the same age. These results were then compared with those reported for 1974–1983 and 1986–1988, two study periods when the stock size was high. The growth of both sexes of marbled sole in Tokyo Bay during the 2000s was found to be significantly higher than that in the 1970s and 1980s. To explore possible factors causing these changes in the growth, we examined bottom water temperature and population density. We found that the population density decreased and the bottom water temperature increased between the late 1970s and the late 2000s. However, despite evidence of changes in population densities and water temperatures, further study is needed to determine the main factors causing the observed changes in growth.  相似文献

ABSTRACT:   The Bioplorer (BP) (Matsushita Ecology Systems Co. Ltd, Kasugai, Aichi-ken, Japan) is an apparatus that consists of a light-emitting diode and optical and image analysis systems. This instrument has been developed and used for the semi-automatic counting of total microbes, mainly in food, cosmetics and industrial products. In this study, the applicability of BP to the detection and enumeration of eukaryotic and prokaryotic cells, after the fluorescent in situ hybridization (FISH) procedure, was examined. The cells of a yeast ( Candida albicans ) and a bacterium ( Escherichia coli ) were specifically labeled using an oligonucleotide probe with fluorochrome and then counterstained with 4', 6-diamidino-2-phenylindole (DAPI). The numbers of cells after FISH treatment and DAPI staining agreed well, indicating that the BP optical device and image analysis system had enough sensitivity to detect and quantify eukaryotic and prokaryotic cells. Because of its simplicity and reliability, BP can be a new tool for quantification of specifically labeled target microorganisms, both in industrial products and natural samples.  相似文献

ABSTRACT:   Samples of river waters containing high concentrations of zinc and other heavy metals but low concentrations of other anthropogenic contaminants were collected to investigate the relationship between toxicity of heavy metals and naturally present organic matters or hardness, as well as the effects of heavy metals on aquatic organisms. Acute toxicity tests were conducted for the water samples using Daphnia magna and medaka Oryzias latipes . Almost all the D. magna died in river waters containing high concentrations of zinc, but O. latipes in the same waters were hardly affected. Since the test organisms were not only exposed to zinc but also other heavy metals in the river waters, we examined the toxicity using toxic units composed of zinc, copper, lead, and cadmium. The results of a bioassay with the river waters showed that the mortality of D. magna did not depend solely on the total number of toxic units of heavy metals. The organic matters and the hardness of the river waters could decrease the acute toxicity of zinc and other heavy metals to D. magna .  相似文献

Phytoplasma-induced floral malformations such as virescence, phyllody, and proliferation were observed on hydrangeas in Gunma Prefecture, Japan. Phylogenetic analyses based on 16S rRNA, secY, groEL, and amp gene sequences indicated that the affected hydrangea plants were associated with phytoplasmas belonging to ‘Candidatus Phytoplasma asteris’, but not to ‘Ca. P. japonicum’, which occurs in hydrangeas showing phyllody in Japan. This is the first molecular evidence of an association of ‘Ca. P. asteris’ with hydrangea plants in Japan.  相似文献

A viroid was detected for the first time in symptomless petunia plants (Petunia spp.) and identified as Tomato chlorotic dwarf viroid (TCDVd) based on an analysis of the complete genomic sequence. These petunia plants are a likely source of inoculum for tomato or potato plants because TCDVd induces severe symptoms on these plants. The genomic sequence of this petunia isolate from Japan shared 100 % identity with petunia isolates from the Netherlands and United Kingdom and a tomato isolate from Japan. Phylogenetic analysis showed that all petunia isolates and the tomato isolate from Japan formed a monophyletic clade.  相似文献

Incorporation of newly-immobilized N into major soil organic matter fractions during a cropping period under paddy and upland cropping systems in the tropics was investigated in Jawa paddy fields with and without fish cultivation and a Sumatra cassava field in Indonesia. 15N-labelled urea (15N urea) was applied as basal fertilizer, and the soil samples were collected after harvest. The percentage of distribution of the residual N in soil from 15N urea into the humic acids, fulvic acid fraction, and humin were 13.1–13.9, 19.0–20.5, and 53.4–54.3%, respectively, for the Jawa paddy soils, and 14.9, 27.4, and 52.4%, respectively, for the Sumatra cassava soil. These values were comparable to the reported ones for other climatic zones. The percentage of distribution of 15N urea-derived N into humic acids was larger than that of total N into the same fraction in all the soils. The distribution into the fulvic acid fraction was also larger for 15N urea-derived N than for total N in the Jawa soils. Humic and non-humic substances in the fulvic acid fraction were separated using insoluble polyvinylpyrrolidone (PVP) into the adsorbed and non-adsorbed fractions, respectively. Less than 5% of the 15N urea-derived N in fulvic acid fraction was detected in the PVP-adsorbed fraction (generic fulvic acids). The proportion of non-hydrolyzable N remained after boiling with 6 M HCl in the 15N urea-derived N was 9.4–13.5%, 17.3–26.7%, and 8.4–16.6% for the humic acids, generic fulvic acids, and humin, respectively. The significantly low resistance to acid hydrolysis suggested that the 15N urea-derived N was less stable than the total N in soil regardless of the fractions of humus.  相似文献

Unprecedented large-scale algal blooms were observed during autumn 2021 around the Katsurakoi fishing port, Kushiro, eastern Hokkaido, Japan. Monitoring of shoreline water showed that chlorophyll a (Chl a) concentrations and the cell density of Karenia spp., dominated by Karenia selliformis, repeatedly increased synchronously between September and November 2021. These increases were associated with a southerly wind-driven current, which transported offshore water on the shelf towards the shoreline at the sea surface. The blooms were prolonged as a result of algal accumulation in the semi-closed fishing port. The maximum Chl a concentration and cell density exceeded 50 µg Chl a/L and 104 cells/mL, respectively. During the autumn bloom of Karenia spp., the nitrate?+?nitrite and phosphate concentrations in the water were lower than those in 2019 and 2020, and the silicate concentration was comparable. The ammonium concentration during the bloom was notably higher than before the bloom period, reaching 15 µM. Mass mortality of several fish species and echinoderms that were cultured using rearing water intake from the same shoreline occurred synchronously with the increase in Karenia spp.
