Recent genomic studies have demonstrated that fungi can possess gene clusters encoding for the production of previously unobserved secondary metabolites. Activation of these attenuated or silenced genes to obtain either improved titers of known compounds or new ones altogether has been a subject of considerable interest. In our efforts to discover new chemotypes that are effective against infectious diseases, including malaria and methicillin-resistant Staphylococcus aureus (MRSA), we have isolated a strain of marine fungus, Leucostoma persoonii, that produces bioactive cytosporones. Epigenetic modifiers employed to activate secondary metabolite genes resulted in enhanced production of known cytosporones B (1, 360%), C (2, 580%) and E (3, 890%), as well as the production of the previously undescribed cytosporone R (4). Cytosporone E was the most bioactive, displaying an IC(90) of 13 μM toward Plasmodium falciparum, with A549 cytotoxicity IC(90) of 437 μM, representing a 90% inhibition therapeutic index (TI(90) = IC(90) A459/IC(90)P. falciparum) of 33. In addition, cytosporone E was active against MRSA with a minimal inhibitory concentration (MIC) of 72 μM and inhibition of MRSA biofilm at roughly half that value (minimum biofilm eradication counts, MBEC90, was found to be 39 μM).  相似文献

The objective of this study was to determine the association between space allowance and in-transit loss of finishing pigs going to select abattoirs in Ontario during summer weather conditions. The study included data from 2- or 3-tiered trailers transporting ≥ 130 pigs in June, July, and August 2003 to 3 packers that processed 76% of Ontario market pigs. Daily in-transit loss data were merged with packer data to determine the number of pigs on each trailer. Space allowance (in square meters per pig) was estimated from the percentage of each trailer's capacity that was filled by the load size. Actual pig weights were not available. Hourly temperature and relative humidity were obtained from 2 local Ontario weather stations. In-transit loss increased with environmental temperature, by 6.6 times at temperatures between 28°C and 34.2°C compared with < 17°C. At space allowances between 0.44 and 0.43 m(2)/pig compared with ≥ 0.515 m(2)/pig, in-transit losses increased 2.12 times when environmental temperatures were < 21°C. Temperature is likely a more important determinant of in-transit loss than space allowance. However, in-transit losses in hot weather are likely to be reduced by increasing space allowance or by adding a cooling device.  相似文献

In large-scale disasters, it is not always possible to identify every potential toxic agent to which SAR dogs may be exposed. However, an understanding of the basic means by which dogs may be exposed to toxic agents can aid veterinarians in determining basic risks for particular SAR sites and allow veterinarians to institute general preventive measures (eg, frequent eye washes) to minimize exposure. Discussions with public health and other authorities on-site may aid in identifying site-specific risks for SAR dogs. Finally, ensuring that SAR dog handlers are aware of basic risks, precautions, and decontamination measures is essential, as handlers are the first line of defense in preventing illness or injury to SAR dogs as they work a disaster area.  相似文献

In-transit losses and stage of transport when deaths occurred were determined for 4 760 213 market-weight pigs produced in 2001 by 4159 Ontario producers and marketed through 117 transport companies to 33 packers located in Canada (96%) and the United States. Approximately 73% and 21% of producers marketed < 2000 pigs and < 500 pigs, respectively. In-transit loss was 0.017%, with 75% of producers losing ≤ 5 pigs annually. Approximately half of in-transit losses occurred on the truck, with 14% of the other deaths occurring at the assembly yards, 4% on the producers’ trucks, and 24% at the abattoir. Fifteen percent of in-transit deaths, representing 1212 pigs, occurred in pigs that were previously identified as abnormal by the transporter or personnel working at the assembly yard or abattoir. Average losses were higher for producers marketing < 2000 pigs, and in-transit loss ratio (ITLR) was highest among those marketing < 100 pigs. Pigs from small farms traveled greater distances than those from larger operations. In-transit losses increased sharply between 590 and 720 km traveled, and decreased at distances > 980 km. Environmental temperatures reached ≥ 31°C for 4.2% of pigs shipped in June, July, and August, with median and mean temperatures of 20.6°C and 20.3°C, respectively, for these months. Twenty percent of all in-transit losses (1617 pigs) occurred in August.  相似文献

ABSTRACT

The aim of this work was to evaluate the effects of isochoric freezing on the quality of tilapia fillet. Isochoric freezing was compared to chilling, super-chilling, and freezing. Isochoric freezing showed muscle color alterations similar to the other preservation methods. All preservation methods resulted in softer fillets, with the isochoric frozen fillet having the most similar texture to that of the fresh sample. Thiobarbituric acid reactive substances (TBARS) for isochoric samples were similar to those of fresh samples. However, there was a 53%, 55%, and 34% increase in TBARS for chilled, super-chilled, and frozen samples, respectively. Total volatile basic nitrogen (TVB-N) content was 1.4 times higher for isochoric samples than for fresh samples. For chilled, super-chilled, and frozen samples, TVB-N content was 3.0, 1.9, and 1.3, respectively, times higher than for fresh samples. Microstructural analysis indicated that isochoric samples showed less cell damage compared to those using other methods. Subfreezing temperatures in conjunction with no ice formation during isochoric freezing contributed to improved quality of tilapia fillet. This study may find application in the commercial preservation of fish to increase shelf life and allow for expanded distribution of raw fish. This study might also be a potential solution to “food desert” areas, where residents have low access to fresh healthy foods.  相似文献

The genomes of individuals from the same species vary in sequence as a result of different evolutionary processes. To examine the patterns of, and the forces shaping, sequence variation in Arabidopsis thaliana, we performed high-density array resequencing of 20 diverse strains (accessions). More than 1 million nonredundant single-nucleotide polymorphisms (SNPs) were identified at moderate false discovery rates (FDRs), and approximately 4% of the genome was identified as being highly dissimilar or deleted relative to the reference genome sequence. Patterns of polymorphism are highly nonrandom among gene families, with genes mediating interaction with the biotic environment having exceptional polymorphism levels. At the chromosomal scale, regional variation in polymorphism was readily apparent. A scan for recent selective sweeps revealed several candidate regions, including a notable example in which almost all variation was removed in a 500-kilobase window. Analyzing the polymorphisms we describe in larger sets of accessions will enable a detailed understanding of forces shaping population-wide sequence variation in A. thaliana.  相似文献
