Invasion of apple fruit by Erwinia amylovora from fruit-bearing twigs through the abscission layer at fruit maturation was examined. Erwinia amylovora (ca. 105 cfu) tagged with bioluminescence genes from Vibrio fischeri was deposited in artificial wounds on fruit-bearing twigs of apple trees grown in a containment greenhouse on September 22, 27, or October 5, 2004. On October 22, 176 apples were harvested and cut horizontally in half. The upper halves were stamped on plates of selective medium, and the lower halves were flooded with iodine solution to assess maturity. All fruit were symptomless and fully mature. The pathogen was recovered from 19 (10.8%) apples. The result showed that if at least ca. 105 cfu of E. amylovora are present in fruit-bearing twigs at the time of fruit maturation, the bacteria can pass through the abscission layer into the fruit, even though the mature fruit lack symptoms.  相似文献

Two neuropeptides, neuropeptide B (NPB) and prolactin-releasing peptide (PrRP), have been suggested to play important roles in control of the hypothalamic-pituitary-adrenal (HPA) axis in rodents. The aim of the present study was to clarify the central actions of NPB or PrRP in sheep. Ovariectomized ewes were surgically implanted with a cannula directed to the lateral ventricle. They received intracerebroventricular (icv) administration of 400 mul of artificial cerebrospinal fluid, NPB (0.05, 0.5 or 5 nmol), PrRP (0.5, 5 or 50 nmol) or corticotropin-releasing hormone (CRH, 0.5 or 5 nmol) through the cannula, and blood samples were taken 30 and 0 min prior to and 15, 30, 60 and 90 min after the injection. Cortisol concentrations in plasma were determined by enzyme immunoassay. Administration of 0.5 nmol NPB resulted in a significant increase in the cortisol concentration compared with the vehicle control, whereas the cortisol concentration after lower or higher doses of NPB did not differ from the control value. Thus, an icv injection of NPB produced a bell-shaped dose-response of cortisol concentration. Administration of PrRP had no significant effect on the cortisol concentrations at any dose examined. Icv injection of CRH dose-dependently increased plasma cortisol concentrations. These results demonstrate that central NPB stimulates cortisol secretion, suggesting that this neuropeptide plays some roles in control of the HPA axis in sheep. On the other hand, unlike its role in rodents, PrRP is unlikely to be involved in control of the HPA axis in this species.  相似文献

Sensitivity maps of atmospheric acid deposition in Japan have not been updated in 20 years. Here, we propose new relative risk maps of surface water acidification in forests based on a weighted overlay of cumulative potential acid deposition (CPAD) simulated for a 25-year period (1981–2005), including the sensitivities of soil and bedrock to acidification. We assumed that relative acidification risk is high in areas that exhibit high sensitivities of soil and bedrock to acid and have received a large amount of cumulative acid deposition over the past several decades. We aggregated fine soil and bedrock maps into a 20-km mesh for overlay onto an 80-km mesh map of CPAD by considering their spatial structures in Japan. Allocation of the weights among CPAD and soil and bedrock sensitivities was performed based on observational trends in river pH over the past 30 years. The resulting risk map for surface water acidification showed that large areas of western Japan, as well as smaller areas of Hokkaido, Tohoku, Kanto, and Kyushu, are at high risk of surface water acidification. Seventy-seven percent of all rivers for which a declining trend in pH was observed from 2001 to 2009 were also high-risk areas. Acid deposition might be one factor controlling surface water acidification in areas with high bedrock sensitivity, in addition to high CPAD and soil sensitivity, although the risk of soil acidification remains unclear.  相似文献

Heartwoods of 22 African tropical wood species were extracted with methanol and the contents of total phenolic compounds in these extracts were measured. Three bioassays were conducted to evaluate the antioxidant activity, tyrosinase inhibitory activity, and antifungal activity of the methanol extracts. The results indicated that the extracts from 13 species exhibited high antioxidant potential, and their inhibitory concentrations that caused 50% scavenging of the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical (IC50) were less than 10 μg/ml. The crude extract from Cylicodiscus gabunensis showed the highest antioxidant activity, and was even higher than that of (+)-catechin, which is known as a potent antioxidant. There was a good correlation between the antioxidant activity and the content of total phenolic compounds, indicating that phenolic compounds played a predominant role in the antioxidant property of the wood extracts. Among all 22 species, only 1 species, Milicia excelsa, contained extractives that showed very high tyrosinase inhibitory activity. The extracts from 9 species showed high antifungal activity. No consistent relationship was observed between the tyrosinase inhibitory activity or antifungal activity and the content of total phenolics in the extracts. Canine herpesvirus 1 (CHV-1) has a relatively narrow host cell range when compared to other alphaherpesviruses. The early events of CHV-1 infection in a permissive Madin-Darby canine kidney (MDCK) and non-permissive cell lines. In order to quantify attachment and penetration, were investigated quantitative competitive PCR (QCPCR) method was established for quantitation of CHV-1 DNA. In all non-permissive cells tested, no significant decrease in viral attachment was observed. When CHV-1 was treated with heparin, viral attachment to MDCK cells was reduced by 25% of the input CHV-1 attached to MDCK cells even in the presence of 50 microg/ml heparin. However, the attachment of CHV-1 to non-permissive cells was severely impaired by heparin treatment. In permissive MDCK cells, about 80% of attached CHV-1 penetrated into cells. However, only 4-10% of CHV-1 attached to non-permissive cells penetrated into cells. Our data indicated that CHV-1, like other herpesviruses, attached to permissive MDCK cells through two mechanisms: the first one is through the interaction mediated by heparan sulfate (HS) on the cell surface and the second involves unidentified viral component and the cellular receptor. In contrast, the non-permissive cells lacked the cellular receptor for the second attachment mechanism and the defect in viral penetration into non-permissive cell might be related to the lack of the cellular receptor.  相似文献

The aim of this study is to assess the premature mortality risks caused by exposure to particulate matter with aerodynamic diameter less than 2.5???m (PM2.5) and ozone elevated concentrations for the years?2000, 2005, and 2020 in East Asia. The spatial distributions and temporal variations of PM2.5 and ozone concentrations are simulated using the Models-3 Community Multiscale Air Quality Modeling System coupled with the Regional Emission Inventory in Asia. The premature mortality risks caused by exposure to PM2.5 and ozone are calculated based on a relative risk (RR) value of 1.04 (95?% confidence interval (CI): 1.01?C1.08) for PM2.5 concentrations above the annual mean limit of 10???g?m?3 taken from the World Health Organization?CAir Quality Guideline and based on a RR value of 1.003 (95?% CI: 1.001?C1.004) for ozone concentration above 35?ppb of the SOMO35 index (the sum of ozone daily maximum 8-h mean concentrations above 35?ppb). We demonstrate one of the implications of the policy making in the area of environmental atmospheric management in East Asia by highlighting the annual premature mortalities associated with exposure to PM2.5 concentrations that just meet an annual mean concentration of 10???g?m?3, as well as ozone concentrations that have a daily zero SOMO35 index in vulnerable places. Our results point to a growing health risk that may endanger human life in East Asia. We find that the effect of PM2.5 on human health is greater than the effect of ozone for the age group of 30?years and above. We estimate the corresponding premature mortality due to the effects of both ozone and PM2.5 in East Asia for the years?2000 and 2005 to be around 316,000 and 520,000 cases, respectively. For future scenarios of the year?2020, policy succeed case, reference, and policy failed case, the estimated annual premature mortality rates are 451,000, 649,000, and 1,035,000 respectively.  相似文献

To evaluate the characteristics of wood ash as fertilizer, composition of inorganic elements and the leaching behavior were studied. The leaching behavior was evaluated by three leaching methods using different solvents (water, hydrochloric acid, and acetic acid: Japanese leaching test Nos. 18 and 19 and modified toxicity characteristic leaching procedure prescribed by the US Environmental Protection Agency). The ash composition varied according to raw materials for fuel and ash collection systems. Concentration of Na, Al, Si, and V became higher in bark ashes while that of K became higher in wood ashes. Pb, Cd, Se, and Zn were more volatile and enriched in fly ashes. Of the nutrient elements, K showed high water solubility and its phytoavailability also appeared high, although the amount of water-soluble K was low in bark ash. Ca and Mg had intermediate solubility while P was less soluble in both water and acetic acid. The amount of hazardous substances having leached was low for most, but not all the ashes, in terms of Pb, Cr and As. Nevertheless, the phytoavailability of these hazardous substances after their application to the forest appeared low, due to their amphoteric leaching behavior and the acidity of Japanese forest soils.  相似文献

The process of solid-state fermentation was used to produce a cocktail enzyme of Trichoderma reesei ATCC 66587 and Aspergillus tubingensis KRCF 700-33. Wheat bran, corncob, and sugi pulp were supplemented with ammonium sulfate as an enzyme-producing medium using T. reesei and A. tubingensis. The corncob blend ratio, duration of incubation, and ammonium sulfate concentration were optimized for enhancing cellulase production from T. reesei using a Box-Behnken design. Filter paper degrading activity more than tripled when T. reesei was grown in the optimized medium, as compared with the initial medium. The highest activity of 4.03 FPU/ml (about 29 FPU/g of dry material) was obtained with a cocktail enzyme having a 25 % content of A. tubingensis and 75 % of T. reesei. The sugi pulp was then fermented to ethanol with the cocktail enzyme and thermotolerant yeast (Saccharomyces cerevisiae BA-11) under simultaneous saccharification and fermentation (SSF) at 40 °C. An ethanol concentration of 4.48 % (w/v) was achieved using the cocktail enzyme (4 FPU/g-pulp) that was produced on-site with a substrate loading level of 12.5 wt %, which achieved an ethanol yield of 76 % after 72 h.  相似文献

There is an urgent need for the international community to address not only the issues of preserving the global environment and reducing poverty in developing countries, but to also face up to new challenges, such as providing assistance to the reconstruction in Afghanistan. Furthermore, it is necessary to address these issues and challenges by means of mutual collaboration. This latter point has been the subject of lively discussion.In this paper, we describe a new direction in international cooperation taken by the Rural Development Bureau of the Ministry of Agriculture, Forestry and Fisheries, Japan in response to the trend of international discussions. This shoulb be achieved by utilizing Japans experience and expertise gained in the development of its own paddy rice agriculture.Specifically, based on the philosophy that sustainable agricultural and rural development can conserve a regional environment, the Bureau intends to undertake: (1) extension of the results from a cooperation project to other regions, by modifying it and converting it to multi-purpose use; (2) collaboration with aid-related entities in Japan and abroad, and dissemination of related information to them; and (3) promotion of cooperation focused on a specific topic.Furthermore, the Bureau will promote the following matters as the basis for international cooperation in each major task. (1) Contribution towards sustainable water use. (2) Extension of the participatory integrated rural development, and its modification for reconstruction assistance. (3) Extension of desertification control, and its modification and conversion for multi-purpose use. (4) Conversion of the technology for soil erosion control into multi-purpose use.  相似文献
