Mycoplasma pneumonia of swine (MPS) is responsible for significant economic losses in the swine industry. We selected Landrace pigs for reduced MPS pulmonary lesions over five generations, and measured concentrations of the following cytokines: interleukin (IL)‐10, IL‐13, IL‐17, tumor necrosis factor (TNF)‐α and interferon (IFN)‐γ to estimate their correlation with MPS lesions. Sheep red blood cells (SRBC) were injected twice intramuscularly at 70 and 95 kg body weight. Blood serum samples were collected after 1 week of secondary SRBC inoculation and cytokine concentrations were analyzed by ELISA. Genetic parameters and breeding values were estimated. The heritability estimates of IL‐10, IL‐13, IL‐17, TNF‐α and IFN‐γ were 0.20 ± 0.06, 0.12 ± 0.06, 0.27 ± 0.07, 0.20 ± 0.10 and 0.05 ± 0.03, respectively. Genetic correlations of IL‐17 and TNF‐α with pulmonary MPS lesions were high (‐0.86 ± 0.13 and 0.69 ± 0.29, respectively) and those of IFN‐γ and IL‐13 with MPS lesions were moderately negative (‐0.45). Through selection, the breeding values of IL‐17 and IFN‐γ increased substantially and those of TNF‐α decreased. These results suggest that innate and cellular immunity are more important for the suppression of pulmonary lesions in MPS than humoral‐mediated immunity, such as antibody response.  相似文献

We investigated potential relationships between rumen microbiota and milk production in dairy cows during the transition period. Twelve dairy cows were divided into a low‐yield (LY) or high‐yield (HY) group based on their milk yield. Rumen samples were taken from dairy cows at 3 weeks before parturition, and at 4, 8, and 12 weeks after parturition. 16S rDNA‐based metagenomic analysis showed that diversities of rumen microbiota in both groups were similar and the number of operational taxonomic units (OTUs) was lower in the postpartum than prepartum period in both groups. The abundance of Bacteroidetes and ratio of Bacteroidetes:Firmicutes was higher in the HY than the LY group. OTUs assigned to Prevotella bryantii, Fibrobacter succinogenes, Ruminococcus albus, Butyrivibrio fibrisolvens, and Succinivibrio sp. were abundant in the HY group. These OTUs were significantly related to the propionate molar proportion of rumen fluids in the HY group. OTUs assigned to Lachnospiraceae, Bifidobacterium sp. and Saccharofermentans were dominant in the LY group. Predictive functional profiling revealed that abundance of gene families involved in amino acid and vitamin metabolism was higher in the HY than the LY group. These results suggest that the community structure and fermentation products of rumen microbiota could be associated with milk production of dairy cows.  相似文献

硫氧还蛋白家族是约12 kD的小分子蛋白质,含有高度保守的活性反应位点WCG/PPC,催化硫醇一二硫键相互交换,调节细胞内的氧化还原作用.在本研究中,通过RT-PCR法获得目的基因拟南芥硫氧还蛋白M1型基因(AtTRX m1,GenBank Accession No.At1g03680).通过Northern杂交分析拟南芥悬浮细胞系在100 mmol/L NaCl、10 mmol/L NaHCO3、50 μmol/L ABA、10 mmol/L H2O2等逆境处理下mRNA表达水平的变化和硫氧还蛋白M1型转基因植株对NaCl、NaHCO3盐及ABA逆境的抗性分析,表明硫氧还蛋白M1型基因与生物环境胁迫有一定的相关性,尤其是氧化胁迫,能够增强植物对逆境的耐受力.从而为进一步研究硫氧还蛋白基因家族与逆境之间的相关作用奠定了基础.  相似文献

ABSTRACT

Enhanced phosphorus (P) uptake from the soil and increased plant growth related to arbuscular mycorrhizal (AM) fungi in pot culture, using sterilized soil, are well-known phenomena. However, these enhancements are not widely observed under field conditions because field sterilization is difficult. The aim of this study was to investigate the effects of AM fungi on P uptake and the growth of Allium fistulosum in non-fumigated and fumigated fields, under different levels of P availability. Plants were inoculated with the AM fungus Glomus R-10 and grown in fumigated soil. For the uninoculated treatment, a sterilized inoculum was applied directly. The field was fumigated using dazomet. Superphosphate was applied to the field at the rates of 0 (P0) or 500 (P500) kg P2O5 ha?1. The inoculated and uninoculated plants were transplanted into the fields and sampled three times to measure AM fungal colonization, shoot P concentration, and shoot dry weight of the plants. At the transplanting stage, AM fungal colonization was observed in the inoculated plants (>70%) but not in the uninoculated plants. At the third sampling, irrespective of P treatment, AM fungal colonization was observed both in the uninoculated and inoculated plants in the non-fumigated field, and there was no difference in shoot P content and shoot dry weight between the inoculated and uninoculated plants. AM fungal colonization in the fumigated field was higher in the inoculated than uninoculated plants, irrespective of P treatment; shoot P content and shoot dry weight were both higher in the inoculated plants than in the uninoculated plants with P0. These results suggest that the responses of A. fistulosum to AM fungal inoculation under the low-P and fumigated conditions are similar to those observed in sterilized pot culture conditions.  相似文献

Since the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, we have established an archive system of livestock and wild animals from the surrounding ex-evacuation zone. Wildlife within the alert zone have been exposed to low-dose-rate (LDR) radiation for a long continuous time. In this study, we analysed the morphological characteristics of the testes and in vitro fertilization (IVF) capacity of cryopreserved sperm of racoons from the ex-evacuation zone of the FDNPP accident. The radioactivity of caesium-137 (137Cs) was measured by gamma-ray spectrometry, and the measured radioactivity concentration was 300–6,630 Bq/kg in the Fukushima raccoons. Notably, normal spermatogenesis was observed in the seminiferous tubules of the testes, with the germinal epithelium composed of a spermatogenic cell lineage with no evident ultrastructural alterations; freeze-thawing sperm penetration ability was confirmed using the interspecific zona pellucida-free mouse oocytes IVF assays. This study revealed that the chronic and LDR radiation exposure associated with the FDNPP accident had no adverse effect on the reproductive characteristics and functions of male raccoons.  相似文献

Arterial blood gas analysis is an important diagnostic and monitoring tool for respiratory abnormalities. In human medicine, lung complications often occur as a result of liver disease. Although pulmonary complications of liver disease have not been reported in dogs, we have frequently encountered hypoxemia in dogs with liver disorders, especially extrahepatic biliary obstruction. In addition, respiratory disorders account for 20% of perioperative fatalities in dogs. Therefore, in this study, we evaluated the respiratory status in dogs with hepatobiliary disease by arterial blood gas analysis. PaO2 and PaCO2 were measured. Alveolar-arterial oxygen difference (AaDO2), the indicator of gas exchange efficiency, was calculated. Compared to healthy dogs (control group), hepatobiliary disease dogs had significantly lower PaO2 and higher AaDO2. Hypoxemia (PaO2 of ≤80 mmHg) was observed in 28/71 dogs with hepatobiliary disease. AaDO2 was higher (≥30 mmHg) than the control group range (11.6 to 26.4 mmHg) in 32/71 hepatobiliary disease dogs. By classifying type of hepatobiliary disease, dogs with extrahepatic biliary obstruction and chronic hepatitis showed significantly lower PaO2 and higher AaDO2 than in a control group. Dogs with chronic hepatitis also had significantly lower PaCO2. The present study shows that dogs with hepatobiliary disease have respiratory abnormalities more than healthy dogs. Preanesthetic or routine arterial blood gas analysis is likely beneficial to detect the respiratory abnormalities in dogs with hepatobiliary disease, especially extrahepatic biliary obstruction and chronic hepatitis.  相似文献

Wheat grain size and shape are associated not only with yield but also with product and milling quality. A subspecies of cultivated tetraploid wheat, Triticum turgidum ssp. polonicum, is characterized by elongated glumes. To elucidate morphological effects of the subspecies differentiation-related gene, we conducted QTL analysis for grain and spikelet shape using a mapping population between two tetraploid wheat subspecies, polonicum and durum. P1, the gene controlling the elongated glumes, was located on chromosome 7A, and the polonicum-type allele acted in an incomplete dominance manner to express the elongated glume phenotype. The polonicum allele of the P1 locus significantly affected not only glume length but also grain shape, spike shape, awn length and seed fertility in tetraploid wheat. The elongated glume phenotype was correlated with an increase in spike length, grain length and grain weight, and with a decrease in fertility, grain number and awn length. Thus, the subspecies differentiation-related gene in subspecies polonicum dramatically affects grain shape accompanied by alteration of spikelet shape in tetraploid wheat.  相似文献

Second malignancies are frequent complications in human patients with chronic lymphocytic leukemia (CLL). However, the clinical details and outcome of this phenomenon were unclear in their canine counterparts. Here, we report a dog with high-grade lymphoma concurrent with T-cell CLL. A 10-year-old male golden retriever presented with lymphadenopathies. The lymph nodes contained large-sized lymphocytes, raising suspicion of high-grade lymphoma. Meanwhile, small lymphocytic lymphocytosis in the peripheral blood was consistent with CLL. Interestingly, molecular biological analyses revealed that CLL cells were of the T-cell type, whereas lymphoma cells were of the B-cell type. Chemotherapy using the L-VCA short protocol was effective for 155 days, but the dog died on day 194 after diagnosis, despite rescue therapies.  相似文献

The intake, digestion, and ruminal fermentation properties; the antioxidative activity; and the physical effectiveness of pineapple residue (PR) from the cut-fruit industry as a feed for cattle were investigated. Four nonlactating Holstein cows were fed a basal diet and diets with 1.4- and 3.6-kg dry matter (DM)/day of PR (low-PR and high-PR diets, respectively). The DM digestibility and total digestible nutrients were 71% and 70%, respectively. Ruminal pH, volatile fatty acids, and ammonia-N did not differ among the diets. The numbers of total viable, amylolytic, cellulolytic, and methanogenic bacteria and protozoa, and microbial protein synthesis in the rumen also did not differ significantly. Blood triglyceride, nonesterified fatty acids, urea nitrogen, and aspartate transaminase were lower in the high-PR diet. Feeding PR did not affect blood malondialdehyde, although PR has a higher antioxidative value than other commonly used feeds. The particle distribution of PR satisfied the recommended range for haylage. The chewing time for ingestion and rumination did not change with PR content, and the roughage value index of PR was 57-min/kg DM. These results suggest that PR has good intake and digestion properties and satisfactory physical effectiveness. Even a high-PR-content feed unlikely induces the risk of ruminal acidosis.  相似文献
