Effect of miR-19a on lipid catabolism in hepatocyte LO2

AIM: To observe the effect of microRNA-19a (miR-19a) on the lipid catabolism of hepatocyte LO2, and to explore the potential mechanism. METHODS: miR-19a was over-expressed or silenced by transfection of miR-19a mimics or miR-19a inhibitor into LO2 cells, then the mRNA level of miR-19a was detected by real-time PCR. The potential target of miR-19a was found by the method of bioinformatics through internet website. The effect of miR-19a on the 3' UTR of peroxisome proliferator-activated receptor α (PPARα) was measured by dual luciferase reporter assay, and the protein level of PPARα and its 2 major downstream rate-limiting enzymes involved in lipid catabolism, acyl-coenzyme a dehydrogenase (ACADM) and carnitine palmitoyltransferase 1A (CPT1A), were detected by Western blotting. Meanwhile, the effect of miR-19a on the generation of ketone body was measured by beta-hydroxybutyric acid (β-OHB) detection assay. RESULTS: The mRNA level of miR-19a was dramatically elevated by the transfection of miR-19a mimics, and sharply decreased by the transfection of miR-19a inhibitor (P<0.05). PPARα was found as a potential target of miR-19a, and dual luciferase reporter assay and Western blotting confirmed the regulatory effect of miR-19a on the expression of PPARα, with the protein level changes of ACADM and CPT1A. miR-19a mimics down-regulated, while miR-19a inhibitor up-regulated the concentration of β-OHB in LO2 cells (P<0.05). CONCLUSION: miR-19a regulates the lipid catabolism of hepatocytes by targeting the PPARα and its 2 downstream rate-limiting enzymes.     

农村建设中太阳能路灯综合效益评价研究

我国新农村建设中太阳能利用取得了快速发展,本文简要介绍了太阳能路灯系统的工作原理,结合某市循环经济示范村太阳能路灯系统的使用现状,采用费用-效益评价指标从经济、环境和社会效益3个方面对太阳能路灯系统进行综合评价与分析,得到太阳能路灯与普通路灯相比较,在经济上是可行的;安装太阳能路灯系统减少了对环境的污染,提高了可再生能源的利用价值,对于该旅游区的环境保护具有重大意义;对新农村建设新能源的利用,起到了很好的引导作用,为太阳能路灯系统大力推广提供一定的实践依据。     

拷贝数变异在畜禽中的研究进展

拷贝数变异（copy number variation,CNV）具有多种形式的变异结构,在品种多样性、生物进化和疾病相关性等研究中起着重要作用,并具有片段长度大、覆盖范围广等特点。随着分子生物学的发展及DNA测序技术的日渐成熟,人们对遗传变异的研究不断向DNA分子水平深入,多态性标记在畜禽育种中已逐渐成为动物育种研究的趋势和主流。由于CNV对基因的调控和表达所造成的影响更为显著,因此,CNV在重要畜禽中的研究越来越多。目前,已检测出大量有关畜禽重要经济性状的基因序列变异,并有许多研究均表明CNV与动物的重要经济特征及疾病的发生有关。笔者主要通过参考国内外相关的研究报道,简述了CNV的相关研究背景、概念、突变机制,归纳总结了CNV对牛、羊、猪、鸡的经济性状、繁殖性状和疾病调控的影响,以期通过对这些重要畜禽的基因组学研究揭示其适应性遗传机理和表型性状差异的遗传基础,开发相应的分子遗传标记,为畜禽的标记辅助选育提供理论基础。     

河北省水果产业发展现状及对策研究

河北是农业大省,水果作为河北省的农业三大产业之一,自加入世贸组织以后,水果产业也有了一定的发展。该研究从供给、消费和出口3个方面出发,通过对河北省水果产业发展现状的分析,提出了促进河北省水果产业进一步发展的对策建议。     

土壤压砂保水处理含水率对西瓜生产潜力的影响

以西瓜为试材,针对压砂保墒问题,选择压砂时期和灌溉定额作为研究因素,采用对比试验方法研究了压砂保墒效果及其种植西瓜的生产潜力。结果表明:压砂覆盖能够有效保持土壤水分,上年雨季前压砂与雨季后压砂相比较,播种初期前者处理土壤质量含水率可达10.8%以上,而后者仅为8.9%;补水条件下前者比后者处理西瓜纵径大2.8cm,西瓜横径大3.1cm,未补水条件下前者比后者处理西瓜纵径大3.9cm,西瓜横径大2.7cm;补水条件下,前者处理西瓜产量为12 301.0kg·hm~(-2),后者处理西瓜产量为7 549.1kg·hm~(-2),未补水条件下,前者处理西瓜产量为6 723.3kg·hm~(~(-2)),后者处理西瓜产量为3 829.5kg·hm~(-2),可见,压砂保水能够明显提高西瓜生产潜力。     

苹果果实表皮总蛋白提取及双向电泳方法的优化

以苹果果实表皮为试材,采用3种方法提取果实表皮总蛋白,并对双向电泳(2-DE)操作体系进行优化。结果表明:改良酚提法适于苹果果实表皮总蛋白的提取,优化后的2-DE分离体系为预制IPG胶条18cm pH 4~7,上样量600μg,改进后的等电聚焦程序,浓度16%的分离胶,适于苹果果实表皮总蛋白的2-DE分离。     

弱光胁迫对七个辣椒组合的生长发育及产量的影响

以驻马店市农业科学院园艺所近年来培育的7个辣椒组合(品种)为试材,利用遮阳网模拟弱光胁迫,调查了在弱光胁迫下辣椒植物学性状及产量性状,以株高、茎粗作为参考指标,以叶面积、比叶干质量作为重要指标,以产量、叶绿素a/b值作为主要指标,对其弱光耐受性进行了综合评价。结果表明:弱光逆境胁迫导致辣椒植株增高、茎秆变细、叶面积增大、比叶干质量降低、产量及叶绿素a/b值均降低;且叶绿素a/b值与产量的变化幅度基本一致,该方法可用来鉴定辣椒不同组合的耐弱光性能。     

利用SSR标记构建贵州山区玉米杂交种的指纹库

对贵州山区目前在生产上正在使用的西山系列玉米杂交种12份进行了SSR分析。从60对引物中筛选到稳定性好、多态性丰富的8对引物作为核心引物。用这8对引物的指纹组合构建了贵州山区西山系列玉米杂交种的DNA指纹数据库。结果表明,利用SSR技术进行玉米杂交种真实性的鉴定是可行、有效的。     

Nonlinear Characteristic Modeling of Servo Value on Hydraulic Bending Roll System''s Dynamic Characteristic

Nonlinear characteristic of servo value is the important factor influencing hydraulic system, the model of electro-hydraulic servo pressure control system is designed. In this model, flux-equation of servo valve is specially applied and some non-linear characteristics are considered. Furthermore, SIMULINK toolbox of MATLAB language software is applied to solve mathematic model and simulation curves of some variables are obtained. The calculation results are in good agreement with the experimental results.
