The hydroxylation and acetylation of 0.5 mM sulphadimidine (SDD) was studied in primary cultures of hepatocytes from male and female rats, and from castrated male and sham operated male rats. In addition, SDD metabolism was investigated in hepatocytes from castrated male rats treated with testosterone, prior to liver cell isolation. In male rat hepatocytes a significantly higher hydroxylation activity was observed than in hepatocytes from female and castrated male rats. Acetylation activity was higher in females. Testosterone induced hydroxylation but did not affect acetylation. These results correlate well with data from previous in vivo studies, showing the relevance of this in vitro model.  相似文献

Trypanosoma evansi is the cause of surra, a camel disease which is the most important single cause of economic losses in camel rearing areas. Sheep and goats herded with camels are the most likely hosts for T evansi. Upon intravenous infections goats developed erratic parasitaemia, lost weight and their packed cell volume dropped significantly (P<0–001). Trypanosomes were demonstrated by direct microscopy in extravascular locations such as synovial, peritoneal and cerebrospinal fluids and also in lymph by subinoculations into mice. The carcases were emaciated and pale. Histologically there was lymphatic tissue hyperplasia, muscular atrophy and nephrotic changes. Two animals had necrotic foci in the liver, kidneys, lymph nodes, spleen and lungs and also bronchopneumonia. Histologically there was depopulation of lymphocytes in lymphatic tissues, destruction of hepatocytes in the liver with infiltration by inflammatory cells in the liver, lymph nodes, spleen and the kidneys.  相似文献

The tone of respiratory smooth muscle is largely determined by the input from autonomic nerves. The distribution of adrenergic and selected nonadrenergic, non-cholinergic (NANC) nerves in the normal equine trachealis muscle was investigated using immunohistochemistry. The smooth muscle of the trachealis was found to contain numerous nerves immunoreactive for an enzymatic marker of adrenergic nerves, as well as many nerves immunoreactive for a putative NANC neurotransmitter, peptide histidine isoleucine, a potent bronchodilator. The tissues surrounding the respiratory smooth muscle contained numerous nerves immunoreactive for the neuropeptides substance P and calcitonin gene-related peptide, which can cause marked vasodilation and bronchoconstriction. The complex innervation of the equine trachea should be kept in mind when interpreting the results of physiological experiments.  相似文献

The lack of protective action of oestrogens which appears during menopausal period may be the reason of serious metabolic disturbances including oxidative stress. The hypothesis was stated that ovariectomy may induce the variations of antioxidant/oxidant status which can be detected in rat liver. A total of 102 healthy Wistar female rats were included in the experiment and divided into control (CON; n = 6), sham-operated (SHO; n = 48) and ovariectomized (OVX; n = 48) groups. Animals from SHO (n = 6) and OVX (n = 6) groups were killed every week during 8 weeks of experiment to detect dynamic changes in examined parameters. Anti-oxidative enzyme activities [glutathione peroxidase (GSH-Px); superoxide dismutase (SOD)] as well as total antioxidant capacity (TAC) and the intensity of lipid peroxidation, measured by the concentration of N,N,diethyl-p-phenylene diamine (DEPPD) radical, were determined in liver homogenates by the use of spectrophotometric methods. Wave-like patterns of examined parameters within 8 weeks of experiment were detected. GSH-Px activity tended to be higher in OVX animals and was significantly lower at 8th week when compared with 1st week of experiment. SOD activity was higher in SHO animals and showed significant differences between 3rd, 4th, 7th and 1st week. TAC values were significantly higher in OVX when compared with SHO groups in 2nd, 4th, 5th week and significantly lower in 3rd, 6th and 7th week of the experiment. The concentration of DEPPD radicals tended to increase in OVX group. In conclusion, ovariectomy which leads to oestrogen insufficiency is reflected as well in variation of anti-oxidative/oxidative parameters in rat liver homogenates.  相似文献

The recently recognized potential of sodium chlorate as a possible preharvest food safety tool for pathogen reduction in meat animals has spurred interest in the pharmacokinetics of intraruminally dosed chlorate. Six Loala cattle were assigned (one heifer and one steer per treatment) to one of three intraruminal doses of radiolabeled sodium [36Cl]chlorate (21, 42, or 63 mg/kg body weight) administered in four equal aliquots over a 24-h period. Blood and serum were collected (29 samples in 48 h). Total radioactive residues were measured and the radioactive moieties were speciated. Chlorate appeared rapidly in blood and serum after dosing. For animals administered a dose of 42 or 63 mg/kg, the half-life of absorption was estimated at 0.6-0.9 h. Serum chlorate concentrations progressively increased with aliquot administration until peaking at 6-21 parts per million at 26 h. Between aliquot administrations, serum chlorate levels typically peaked in 3.5 h or less. The half-life of chlorate elimination ranged between 6.9 and 11 h, depending on the dose. Ultimately, absorption of chlorate removes it from its desired site of action, the lower gastrointestinal tract, thereby reducing its efficacy. Further research is needed to develop a chlorate formulation that will allow passage to the lower gastrointestinal tract.  相似文献

Bovine cysticercosis is an important food safety issue that is of economic concern. In Belgium, in the last years an increase in the number of cases, mostly light infections, was observed. The role of contact with contaminated surface water has been hypothesized as the main route of transmission. Based on abattoir records from 2001 till 2003 the distribution and risk factors of bovine cysticercosis among dairy and mixed farms were studied in four provinces, using Geographic Information Systems (GIS) and questionnaires. The risk factors were analysed using a case-control study design. The case group consisted of herds from which homebred cattle with cysticercosis had been detected at the abattoir; the control group was composed of herds where no cases had been detected. Case herds were distributed over the study area. A logistic regression analysis revealed that the location (province), the number of slaughtered cattle, the flooding of pastures, free access of cattle to surface water and the proximity of wastewater effluent were significant explanatory variables for bovine cysticercosis to be recorded in a herd.  相似文献
