Chicken pathogenic Escherichia coli strains were found to autoagglutinate in a static culture of trypticase soy broth (TSB). One strain, designated PDI-386, was further studied for its autoagglutinating property. Acidity in the cultured medium caused by glucose degradation induced the autoagglutination. The bacterial cells grown in a glucose-free L-broth could be aggregated by adding acid, which suggests a potentiality of autoagglutination of the strain grown in the L-broth. The autoagglutinating parent (Agg) formed small colonies with irregular edges like rough colonies on the TS agar, whereas its non-autoagglutinating variant (Nag) formed larger smooth colonies with a perfectly round edge. The Nag colony was easily generated from the Agg colony on the TS agar. The autoagglutinating property was very unstable when the bacteria was passed in the TSB, but rather stable in the L-broth. Under electron microscope, the Agg were found to possess pili of more than 20 microns in length. However, the phenotypic expression of autoagglutination did not correlate with that of mannose-sensitive hemagglutination against guinea pig erythrocytes. Incubation of the Nag in the L-broth at room temperature for more than 10 days provoked the reversion of the autoagglutination. There was no difference between the Agg and the Nag in terms of surface hydrophobicity, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) patterns of membrane proteins and LPS, and plasmid profiles. The virulence of the Agg was higher than that of the Nag. The autoagglutination property is, however, so unstable that the pathogenicity of E. coli isolates from chickens should be carefully evaluated.  相似文献

Inhalt: An 81 Versuchs- und 92 Kontrolltieren der Rasse “Potnische Schwarzbunte” aus 3 Problembetrieben wurde ein Versuch zur Sterilitätsprophylaxe rnit der PRID-Spirale durchgeführt. Anhand klinischer Befunde und Milchprogesteronwerten wurde die Untergruppe aus Kühen mit Ovardystrophie (18 Versuchs- und 22 Kontrolltiere) gebildet und extra ausgewertet. Der Behandlungsbeginn lag zwischen 60 und 80 Tage post partum. Die Versuchstiere wurden mit der PRID-Spirale 12 Tage lang behandelt, die Kontrolltiere erhielten Injcktionen rnit Kochsalzlösung und solche rnit Ovardystrophie wurden zusätzlich mit einer Eierstocksmassage behandelt. Die Versuchstiere wurden 56 und 72 Stunden nach Entfernung der Spirale blind besamt. Die Brunstin-duktionsrate betrug insgesamt 90,1%, bei denen rnit Ovardystrophie 77,7%. Die An-wendung der PRID-Spirale führte zur Verbesserung der Fertilitätslage. Die Gesamt-trächtigkeitsrate lag mit 87,6% bei den Versuchskühenüber derjenigen der Kontrollkü-he mit 79,3%. Die Behandlung der Tiere rnit Ovardystrophie erbrachte eine Gesamt-trächtigkeitsrate von 88,8% bei den Versuchs- und 72,7% bei den Kontrolltieren. Die Zwischentragezeit betrug bei den Versuchstieren 101,2 Tage und bei den Kontrolltieren 113,3 Tage (p ≤ 0,05). Bei den azyklischen Tieren lag die Zwischentragezeit bei 104,6 Tage bzw. 134,7 Tage (p ≤ 0,05). In einem anderen Versuch wurde 20 Hybriden zwischen Wisent und Hausrind rwecks Zyklussynchronisation die PRID-Spirale verabreicht. Die Brunstinduktionsrate betrug 66,6%, das Erstbesamungsergebnis nach Doppelbesamung 30%, die Gesamtträchtig-keitsrate nach dem Decken der umrindernden Kreuzungstiere rnit einem Bullen 90%. Die Deckperiode konnte verkürzt werden. Contents: Investigations on the synchronization of estrus cycle in cattle from farms with fertility problems and in cross-breeds between bison and cattle with a PRID-de-vice In 81 experimental and 92 control animals of the “Polish Black and White” breed derived from 3 farms with fertility problems an investigation was carried out for sterility prophylaxis using the PRID-device. Based on clinical findings and milk progesterone values cattle with ovarian dystrophy (18 experimental and 22 control animals) from a special group were used. The begin of treatment was between 60 and 80 days post partum. All experimental animals had received a PRID-device for a duration of 12 days. Control animals were injected with physiological saline solution and animals with ovarian dystrophy were treated additionally with ovarian massage. Experimental animals were inseminated 56 and 72 hrs after removal of the device. All animals taken, estrus was induced in 90.1% of the animals, while only 77.7% of animals with ovarian dystrophy came to estrus. Application of the PRID-device led to an improvement of fertility. Pregnancy rates in experimental animals were 87.6%, while only 79.3% of the control animals were pregnant. Treatment of animals with ovarian dystrophy resulted in 88.8% pregnancies for treated and 72.7% for control animals. Experimental animals were non-pregnant for a duration of 101.2 days, while control animals remained non-pregnant for 113.3 days (p < 0.05). Acyclic cattle had a non-pregnant period of 104.6 days and 134.7 days (p < 0.05) respectively. In a second experiment 20 cross-breeds between bison and domestic cattle were treated with a PRID-device for synchronization of the estrus cycle. The rate of estrus induction was 66.6%, the fertility after double insemination 30%, the total pregnancy rate improved after mating with a bull of animals that had not conceived to 90%.  相似文献

Abstract. A two year field experiment was carried out in a semiarid Mediterranean area in order to evaluate, the effect on soil erosion of adding different urban organic wastes: a stabilized municipal waste (compost), an unstabilized municipal waste, and an aerobic sewage sludge. All the treatments significantly reduced soil erosion, compared to the control soil. The soil amended with compost was the most effective treatment, reducing soil loss by 94% and runoff by 54%.  相似文献

Salivary or secreted carbonic anhydrase (CA), which constitutes a new class of CA, designated CA-VI, was isolated. Swine CA-VI purified from swine saliva by inhibitor-affinity chromatography and ion exchange chromatography had a specific activity of 5,468 units/mg. The molecular weight was 250,000, as determined by gel filtration under non-denaturing conditions, and the subunit molecular weight was found to be 37,000 by sodium dodecyl sulfate polyacrylamide gel electrophoresis, indicating that swine CA-VI consists of 7 subunits. The treatment of the enzyme with endo-N-acetylglucosaminidase F reduced its subunit molecular weight from 37,000 to 35,000 and 32,000. We raised a rabbit antibody against purfied swine CA-VI. Double immunodiffusion showed that anti-swine CA-VI serum reacted with swine CA-VI and swine saliva, but not with hemolysate (containing CA-I and CA-Il) or muscle extracts (containing CA-III). The concentration of CA-VI in swine saliva, measured using single radial immunodiffusion, was 0.027 +/- 0.017 mg/mg total protein.  相似文献

The study describes plumage modifications and specific feather malformations, as related to the domestication process of different poultry species. The modifications include naked necks, leg feathering, frizzle feathering, silky feathering, fat quills, and feather abnormalities caused by behavioural hypertrophies. Most of these plumage modifications correspond to the breed standard for exhibition poultry fancy. However, they impair the normal function of these animals. The negative influences comprise disorders in social behaviour, loss of typical plumage functions and disabilities of normal mobility, as well as genetic defects and pathogenic predispositions.  相似文献

The anthelmintic efficacy of abamectin (avermectin B1) was evaluated against gastrointestinal nematodes, including Ostertagia ostertagi inhibited larvae and lungworm, in yearling crossbred beef heifers during late spring. The calves were grazed on contaminated pasture for 10 weeks and then held under conditions free of nematode infection for 3 weeks prior to allotment and treatment on 5 June. Thirteen calves were randomly assigned to two groups of six by restricted randomization on body weights; the extra lightest calf was assigned to the non-treated control group. Group 1 calves were treated with abamectin at 200 micrograms kg-1 body weight by s.c. injection and Group 2 calves were not treated; all were killed at 14 days after treatment. Ostertagia ostertagi was present in all controls; arithmetic mean numbers of adults, developing fourth stage larvae (L4) and inhibited EL4 were 7683, 605 and 36,102, respectively. Other nematode genera present in controls in sufficient numbers for the experiment were Haemonchus placei adults, Trichostrongylus axei adults, Cooperia spp. adults, Oesophagostomum radiatum adults, Bunostomum phlebotomum adults, Dictyocaulus viviparus adults and E5 (immature adults). Abamectin was highly effective (consistently greater than 99% efficacy and P less than 0.05) in removing all nematodes present in treated calves as represented in non-treated controls, including the primary target of Ostertagia ostertagi inhibited EL4. The lowest efficacy was 93.8%, against D. viviparus E5.  相似文献

A 20-year-old pony mare with persistent ventricular tachycardia (VT) was examined cardiopathologically. At necropsy, the heart was enlarged and rounded with both ventricular dilatation. In a longitudinal section of the ventricular septum, a large grayish white patchy lesion (5 x 25 mm) was detected in the relatively higher portion. Microscopically, the lesion was extensive myocardial fibrosis located in the vicinity of the proximal part of the left bundle branch. Partially the fibrotic lesion was in contact with the branch. Such a lesion might play an important role in creating a suitable background for the development of VT via automaticity or reentry mechanism.  相似文献

Equine arteritis virus (EAV), a non-arthropod borne togavirus, has been shown to have a global distribution. To date, no major antigenic variation has been demonstrated between EAV isolates from different geographic origins. In this study, the genomic RNA of EAV isolates obtained from horses of different breeds in various countries around the world was oligonucleotide fingerprinted. Comparisons of these fingerprints were used to determine the extent of genomic variation among such isolates. Comparisons among isolates from North American horses revealed, for the most part, oligonucleotide homologies of less than 60%. Only 29 of the 98 comparisons revealed greater than 60% oligonucleotide homology. Nonetheless, several comparisons indicated a close epidemiologic relationship between isolates from horses of different breeds located in different states. Though all European isolates were of Standardbred origin and were from horses located in northern European countries, the majority had oligonucleotide homologies of less than 60%. Where oligonucleotide homology was apparent, it was, with one exception, greater than 70%. The two isolates from New Zealand had 93.2% oligonucleotide homology. This is indicative of an extremely close epidemiologic relationship. Comparisons between EAV isolates from around the world revealed oligonucleotide homologies between viruses from North America, Europe and New Zealand. In several instances, this homology was greater than 70% and in one case greater than 80%. No oligonucleotide homology was evident in comparisons involving the virus from South Africa. The high level of genomic conservation between certain EAV isolates of disparate geographic origins may reflect dissemination of the virus associated with the international movement of horses. The extent of genomic variation demonstrated between most of the EAV isolates used in this study confirms the need for further investigation of genomic heterogeneity among strains of this virus before techniques that rely upon nucleic acid hybridization can be effectively applied as diagnostic procedures.  相似文献

Abstract –  Foraging juvenile fish with relatively high food demands are usually vulnerable to various aquatic and avian predators. To compromise between foraging and antipredator activity, they need exact and reliable information about current predation risk. Among direct predator-induced cues, visual and olfactory signals are considered to be most important. Food intake rates and prey-size selectivity of laboratory-reared, naive young-of-the-year (YOY) perch, Perca fluviatilis , were studied in experiments with Daphnia magna of two size classes: 2.8 and 1.3 mm as prey and northern pike, Esox lucius , as predator. Neither total intake rate nor prey-size selectivity was modified by predator kairomones alone (water from an aquarium with a pike was pumped into the test aquaria) under daylight conditions. Visual presentation of pike reduced total food intake by perch. This effect was significantly more pronounced (synergistic) when visual and olfactory cues were presented simultaneously to foraging perch. Moreover, the combination of cues caused a significant shift in prey-size selection, expressed as a reduced proportion of large prey in the diet. Our observations demonstrate that predator-induced olfactory cues alone are less important modifiers of the feeding behaviour of naive YOY perch than visual cues under daylight conditions. However, pike odour acts as a modulatory stimulus enhancing the effects of visual cues, which trigger an innate response in perch.  相似文献

Abstract. The Pan‐European Soil Erosion Risk Assessment (pesera ) model was evaluated using existing soil erosion data collected under various types of climate, vegetation, landscape and soil conditions. The data used represent a variety of typical Mediterranean land uses such as winter wheat, vines, olives and bare, stony land prevailing in hilly areas. Using this data, the model was calibrated for sediment transport by overland flow and results compared to measured soil erosion values from runoff plots and a watershed on a monthly basis. The performance of the model was assessed statistically, showing that it can be satisfactorily used for predicting soil erosion rates under the conditions included in the study. The overall model estimate including all the available experimental data was 0.69 t ha?1 yr?1 with a maximum error of 1.49 t ha?1 yr?1. After validation, the model was applied to a small watershed (60 ha) of great ecological importance for the sea turtle Caretta caretta. For this purpose, soil and vegetation maps were compiled from all the necessary data for applying the model. The model was run for three years using daily data from an existing nearby meteorological station. The predicted and measured soil erosion rates for a 7‐month period were 0.31 t and 0.18 t, respectively. Application of the model to each mapping unit showed the over‐riding importance of land use for sediment generation under the given climatic conditions. Bare land, occupying 5.5% of the watershed area, generated up to 69% of the total sediments estimated for the watershed. It is concluded that the pesera model can be used as a regional diagnostic tool under a range of soil, topographic and climatic conditions for identifying the best land use type and vegetation cover to protect hilly areas from soil erosion. The calculated overall root mean square error for the model is 0.06 t ha?1 yr?1, compared to a soil erosion rate of 0.04 t ha?1 yr?1, which can be tolerated for protecting the area for the sea turtle.  相似文献
