Summary The effect of fungicides and the antagonistic fungusTrichoderma harzianum (Th) on the potato leak pathogenPythium aphanidermatum (Pa) was investigated in vitro. Rot was reduced by 73% to 89% when tubers inoculated withPa were treated by immersion in solutions (0.5% w/v) of the fungicides Ridomil MZ58. Dithane M45. Manebe 80 and Tachigaren 360. Complete protection was achieved by dusting inoculated tubers with Ridomil MZ58 diluted in kaolin at a final concentration of 0.1% (w/w). The biological treatments of dipping inoculated tubers in a conidial water suspension ofTh at 108 conidia/ml or of dusting with a mixture of a barley culture ofTh and kaolin to a final concentration of 2×109 conidia/g were as effective as the chemical treatments. Control was still effective when inoculated tubers were treated with Ridomil MZ58. Tachigaren 360 or the conidial suspension ofTh after 24h incubation at 25°C.  相似文献

Summary Soft rot caused by Erwinia carotovora ssp. atroseptica is a major disease of stored potatoes. Since varietal resistance can contribute to control, the work reported was designed to find new sources of resistance among related tuber-bearing Solanum spp. True seeds were imported from two international collections and families were screened for resistance to tuber soft rot. Forty-eight resistant clones were found in 21 out of 100 accessions. These clones will be used in breeding programmes at the diploid or tetraploid level.  相似文献

Fibrillar collagens are the more abundant extracellular proteins. They form a metazoan-specific family, and are highly conserved from sponge to human. Their structural and physiological properties have been successfully used in the food, cosmetic, and pharmaceutical industries. On the other hand, the increase of jellyfish has led us to consider this marine animal as a natural product for food and medicine. Here, we have tested different Mediterranean jellyfish species in order to investigate the economic potential of their collagens. We have studied different methods of collagen purification (tissues and experimental procedures). The best collagen yield was obtained using Rhizostoma pulmo oral arms and the pepsin extraction method (2-10 mg collagen/g of wet tissue). Although a significant yield was obtained with Cotylorhiza tuberculata (0.45 mg/g), R. pulmo was used for further experiments, this jellyfish being considered as harmless to humans and being an abundant source of material. Then, we compared the biological properties of R. pulmo collagen with mammalian fibrillar collagens in cell cytotoxicity assays and cell adhesion. There was no statistical difference in cytotoxicity (p > 0.05) between R. pulmo collagen and rat type I collagen. However, since heparin inhibits cell adhesion to jellyfish-native collagen by 55%, the main difference is that heparan sulfate proteoglycans could be preferentially involved in fibroblast and osteoblast adhesion to jellyfish collagens. Our data confirm the broad harmlessness of jellyfish collagens, and their biological effect on human cells that are similar to that of mammalian type I collagen. Given the bioavailability of jellyfish collagen and its biological properties, this marine material is thus a good candidate for replacing bovine or human collagens in selected biomedical applications.  相似文献

Some legume species of the Crotalaria genus are specifically nodulated by methylotrophic bacteria belonging to the Methylobacterium nodulans species. The feature of this symbiotic bacterium is its ability to oxidize methanol, a property based on the presence of a methanol dehydrogenase enzyme. Despite a good knowledge of this property and its implication in symbiosis, the molecular dialogue between M. nodulans and crotalaria podocarpa leading to symbiosis is largely unknown, except the presence of a nodA nodulation gene in the genome of M. nodulans ORS 2060. To investigate if M. nodulans ORS 2060 produces Nod factors, molecules considered as the major bacteria-to-plant signals essential for the establishment of rhizobia–legume symbiosis, we identified and sequenced a nodDABCUIJHQ cluster from a genomic library of ORS 2060. Phylogenetic analyses of nod genes revealed that M. nodulans ORS 2060 form a branch together with Burkholderia tuberum STM678 and a strain of Methylobacterium sp. (4-46) isolated from Lotononis, and distinct from all the other rhizobia. To analyse the regulation of ORS 2060 nod genes, we constructed a nodA–LacZ promoter fusion to monitor the nod gene expression with various flavonoids. The flavone apigenin was found to be the strongest inducer of nod gene expression in M. nodulans ORS 2060. This latter flavonoid was used to induce ORS 2060, and Nod factors were purified by high-performance liquid chromatography (HPLC) and further characterized by mass spectrometry. One major Nod factor structure was identified as a pentamer of chitin substituted by C18:1 or C16:0 acyl chains on the non-reducing end and 6-O-sulphated on the other end, suggesting a classic symbiotic dialogue between M. nodulans and C. podocarpa.  相似文献

Two malting hulled varieties (Sebastian, Malz) and one nonmalting hull-less variety (AF Lucius) were used to assess vitamins C and E in the green biomass of young plants of spring barley ( Hordeum vulgare L.) in three stages of growth and development (BBCH 29, 31, 32-33). The samples from sampling I (BBCH 29) had statistically significantly higher vitamin C content and vitamin E activity compared to sampling I (BBCH 31). The highest average vitamin content was determined in the malting variety Sebastian (vitamin C, 520 mg 100 g(-1) DM; vitamin E, 73.06 mg kg(-1) DM) compared to the varieties Malz (501 mg 100 g(-1) DM; 61.84 mg kg(-1) DM) and AF Lucius (508 mg 100 g(-1) DM; 67.81 mg 100 g(-1) DM). The locality Krome?r?i?z? (Czech Republic, CR), with vitamin C and E contents of 524 mg 100 g(-1) DM and 68.74 mg kg(-1) DM, respectively, proved to be more suitable for growing green biomass compared to the locality Z?abc?ice (CR) (content of vitamins C and E, 477 mg 100 g(-1) DM and 66.39 mg kg(-1) DM, respectively). During the research period (2005-2007), it was determined that the green mass of young plants of spring barley was a significant source of vitamins C and E in the growth stage BBCH 29; in later samplings (BBCH 32-33) the vitamin levels dropped (by as much as 48%). These vitamins are important antioxidants for human health. Therefore, "green barley" can be recommended for the preparation of natural dietary supplements and is preferred to synthetic vitamin preparations.  相似文献

Average population growth in the African Sudanian belt is 3 % per year. This leads to a significant increase in cultivated areas at the expense of fallows and forests. For centuries, rural populations have been practicing agroforestry dominated by Vitellaria paradoxa parklands. We wanted to know whether agroforestry can improve local rainfall recycling as well as forest. We compared transpiration and its seasonal variations between Vitellaria paradoxa, the dominant species in fallows, and Isoberlinia doka, the dominant species in dry forests in the Sudanian belt. The fallow and dry forest we studied are located in northwestern Benin, where average annual rainfall is 1200 mm. Sap flow density (SFD) was measured by transient thermal dissipation, from which tree transpiration was deduced. Transpiration of five trees per species was estimated by taking into account the radial profile of SFD. The effect of the species and of the season on transpiration was tested with a generalized linear mixed model. Over the three-year study period, daily transpiration of the agroforestry trees, V. paradoxa (diameters 8–38 cm) ranged between 4.4 and 26.8 L day?1 while that of the forest trees, I. doka, (diameters 20–38 cm) ranged from 9.8 to 92.6 L day?1. Daily transpiration of V. paradoxa was significantly lower (15 %) in the dry season than in the rainy season, whereas daily transpiration by I. doka was significantly higher (13 %) in the dry season than in the rainy season. Our results indicate that the woody cover of agroforestry systems is less efficient in recycling local rainfall than forest cover, not only due to lower tree density but also to species composition.  相似文献

ABSTRACT: Scrapie in sheep and goats has been known for more than 250 years and belongs nowadays to the so-called prion diseases that also include e.g. bovine spongiform encephalopathy in cattle (BSE) and Creutzfeldt-Jakob disease in humans. According to the prion hypothesis, the pathological isoform (PrPSc) of the cellular prion protein (PrPc) comprises the essential, if not exclusive, component of the transmissible agent. Currently, two types of scrapie disease are known - classical and atypical/Nor98 scrapie. In the present study we examine 24 cases of classical and 25 cases of atypical/Nor98 scrapie with the sensitive PET blot method and validate the results with conventional immunohistochemistry. The sequential detection of PrPSc aggregates in the CNS of classical scrapie sheep implies that after neuroinvasion a spread from spinal cord and obex to the cerebellum, diencephalon and frontal cortex via the rostral brainstem takes place. We categorize the spread of PrPSc into four stages: the CNS entry stage, the brainstem stage, the cruciate sulcus stage and finally the basal ganglia stage. Such a sequential development of PrPSc was not detectable upon analysis of the present atypical/Nor98 scrapie cases. PrPSc distribution in one case of atypical/Nor98 scrapie in a presumably early disease phase suggests that the spread of PrPSc aggregates starts in the di- or telencephalon. In addition to the spontaneous generation of PrPSc, an uptake of the infectious agent into the brain, that bypasses the brainstem and starts its accumulation in the thalamus, needs to be taken into consideration for atypical/Nor98 scrapie.  相似文献
