Beech (Fagus sylvatica L.) seedlings were cultivated from seeds sown in pots or directly in the ground in outdoor chambers that were transparent to solar radiation, and provided either ambient air or CO(2)-enriched air (ambient + 350 &mgr;mol mol(-1)). The rooting volume was high in all experiments. In the short-term experiment, potted plants were assigned to a factorial CO(2) x nutrient treatment (optimal nutrient supply and severe nutrient shortage) for 1 year. In the long-term experiment, plants were grown directly in the ground and received an optimal supply of water and nutrients in both CO(2) treatments for 3 years. Nutrient stress caused carboxylation capacity (V(m)) to decrease in the potted seedlings exposed to CO(2)-enriched air during their first growing season. In the long-term experiment with optimal nutrient supply, CO(2)-enriched air did not affect V(m), but caused an upward acclimation of maximum electron transport rate (J(m)). Consequently, there was a 14% increase in the J(m)/V(m) ratio, indicating nitrogen reallocation to maintain an equilibrium between RuBP consumption and RuBP regeneration. Both V(m) and J(m) decreased during the growing season in both CO(2) treatments. Although upward acclimation of J(m) was no longer apparent at the end of the third growing season, plants in CO(2)-enriched air maintained a higher J(m)/V(m) ratio than plants in ambient air, indicating that photosynthetic acclimation always occurred. Second flush leaves appeared during each growing season. When expressed on the basis of foliar nitrogen concentration, their photosynthetic characteristics (V(m) and J(m)) were enhanced compared with other leaves. Because the number of second flush leaves was also increased in the elevated CO(2) treatment, this response should be taken into account when modeling the effects of elevated CO(2) concentration on canopy photosynthesis. Stomatal conductance decreased in response to atmospheric CO(2) enrichment; however, the stomatal response to irradiance followed a single relationship based on two stomatal conductance models.  相似文献

Residue quality is a key factor governing biodegradation and the fate of C in soil. Most investigations of relationships existing between crop residue quality and soil decomposition have been based on determining the relative proportions of soluble, cellulose, hemicellulose and lignin components. However, cell wall cohesion is increased by tight interconnections between polysaccharides and lignin that involve cross-linking agents (phenolic acids). The aim of this study was to determine the role of lignin composition and phenolic acids on short- to medium-term decomposition of maize roots in soil. Sixteen maize genotypes, presenting a range of chemical characteristics related to root lignin and phenolic acids, were used. The main components were characterized by Van Soest (VS) extraction and cell wall acid hydrolysis, and the non-condensed Syringyl and Guaicyl lignin monomers, esterified phenolic acids and etherified phenolic acids were determined. Maize roots were then incubated in soil under controlled conditions (15 °C, −80 kPa moisture) for 796 days. Results showed that VS extraction over-estimated the structural hemicellulose content and that VS lignin was more recalcitrant than Klason lignin. The tremendous effect of cell wall chemical characteristics was shown by marked variations (almost two-fold differences in C mineralization), between the 16 maize roots. Decomposition was controlled by soluble residue components in the short term whereas lignin and the interconnections between cell wall polymers were important in the long-term. Notably the cell wall domain rich in non-condensed lignin and esterified phenolic acids was prone to decomposition whereas the presence of etherified ferulic acids seemed to hamper cell wall decomposition.  相似文献

Understanding and monitoring deformation and water content changes in meat during cooking is of prime importance. We show the possibilities offered by nuclear magnetic resonance imaging (MRI) for the in situ dynamic measurement of deformation fields and water content mapping during beef heating from 20 to 75 °C. MRIs were acquired during heating, and image registration was used to calculate the deformation field. The temperature distribution in the sample was simulated numerically to link structural modifications and water transfer to temperature values. During heating, proton density decreases because of a magnetic susceptibility drop with temperature and water expulsion due to muscle contraction. A positive relationship was found between local cumulative deformation and water content. This new approach makes it possible to identify the deformation field and water transfer simultaneously and to trace thermal history to build heuristic models linking these parameters.  相似文献

A new sesquiterpene lactone, 7alpha,10alpha-epoxy-1alpha(H),5alpha(H)-guaia-3,11(13)-dien-8alpha,12-olide, was isolated from the leaves of Hedyosmum arborescens. The structure of this unusual 7,10-epoxy-guaianolide was determined by spectroscopic methods, particularly one- and two-dimensional 1H and 13C NMR.  相似文献

As part of a study of the processes involved in litter biodegradation following sewage sludge (SS) addition, the variations over 14 months of phosphatase activities in a cork oak litter (Quercus suber L.) were investigated. A field experimental design was carried out using the litter-bag method on both a fertilized plot receiving SS applications (twice ) and a control plot. Acid (APH) and alkaline (BPH) phosphatases were measured, along with several biotic and abiotic variables potentially involved in the regulation of these enzymes. These included moisture, temperature, pH, water-extractable inorganic P (PW), culturable heterotrophic bacteria and fungi. Sludge addition had significant effects on all the variables measured. Indeed, sludge increased significantly BPH activities, available PW, microbial densities (i.e. bacteria and fungi) and pH in the fertilized plot. In contrast, APH activities decreased significantly following sludge addition. As a consequence, the BPH/APH ratio increased markedly and immediately in the fertilized plot, but only after the 1st amendment. Following the 1st preconditioning SS amendment, the 2nd fertilization had fewer effects on biological variables, because of summer dryness. The different properties examined varied significantly with incubation time, and most were significantly related to the seasonal patterns of litter moisture in this Mediterranean forest ecosystem. Hence, sewage sludge application modified the intensity of microbial responses to environmental factors, but biological patterns regulating P turn-over were maintained.  相似文献

Using specific radioimmunoassays for the two GnRH molecular forms present in the European eel, Anguilla anguilla, (mGnRH and cGnRH II), we compared their distributions in the pituitary and different parts of the brain of female silver eels, as well as the modifications of their levels in experimentally matured female eels (treated with carp pituitary extract). In control eels, mGnRH levels were higher than cGnRH II levels in the pituitary, olfactory lobes and telencephalon, di- and mesencephalon, while the opposite was found in the posterior part of the brain (met- and myelencephalon). Experimental sexual maturation of the gonads significantly increased mGnRH levels in the pituitary and anterior parts of the brain; such a positive effect was not observed on the low cGnRH II levels, which were, in contrast, reduced. These data indicate that the positive feedback of gonadal hormones on GnRH, that we previously demonstrated, would specifically affect the mGnRH form. The differential distribution and control of mGnRH and cGnRH II suggest that these two forms have different physiological roles in the eel. The large increase in mGnRH during sexual maturation suggests the prime implication of this form in the neuroendocrine control of reproduction.  相似文献

Field isolates of Mycosphaerella fijiensis, causing black Sigatoka of banana, were characterised for their sensitivity to different inhibitors of the cytochrome bc1 enzyme complex (Qo respiration inhibitors, strobilurin fungicides), using physiological, biochemical and molecular genetic methods. Strobilurin‐resistant isolates exhibited very high resistance factors both in mycelial growth inhibition and NADH consumption assays. Cross‐resistance was observed among all Qo inhibitors, including trifloxystrobin, azoxystrobin, famoxadone, strobilurin B and myxothiazol. However, the Qi and the cytochrome aa3 inhibitors, antimycin A and potassium cyanide, respectively, were not cross‐resistant to Qo inhibitors. In sensitive but not in resistant isolates, mixtures of Qo inhibitors and SHAM, an inhibitor of the alternative oxidase (AOX), were more active than the components alone, indicating that the alternative pathway is essential in metabolism, but not causal for resistance. In the cell‐free NADH‐consumption assay, the Qo inhibitors affected the sensitive but not the resistant isolates, suggesting that AOX was not active in sub‐mitochondrial particles. In whole cells, however, the AOX has a basic expression level and is probably not inducible by trifloxystrobin. Sequencing of the cytochrome b gene of sensitive and resistant M fijiensis isolates revealed a difference in the nucleotide sequence leading to a single amino acid change from glycine to alanine at position 143 in the resistant isolate. This change is known to occur also in the naturally tolerant basidiomycete Mycena galopoda. It is suggested that the field isolates of M fijiensis can acquire resistance to Qo inhibitors due to a target site alteration with a single base pair change. Resistant isolates do not seem to contain a mixture of mutated and non‐mutated DNA, indicating a complete selection of resistant mitochondria and a maternally donated mode of resistance. © 2000 Society of Chemical Industry  相似文献

Summary This paper reports results of a 3-year evaluation of CIP advanced potato clones in a bacterial wilt-infested field (race 3) in Peru. Clones resistant or moderately resistant to wilt were selected and all tubers harvested from each clone were tested for latent infection byRalstonia solanacearum using a sensitive serological technique developed at CIP. A sampling strategy to estimate accurately the frequency of infected tubers in the clones has been evaluated. This method will allow consideration of tuber latent infection as a new selection criterion in breeding for resistance to bacterial wilt. Thirteen clones were found resistant to wilt in all three evaluations (i.e.≤6% wilt), from which five had no wilt in all trials. However, all clones harboured latent infection in tubers averaging 30%. Analysing 30 tubers/clone provides an accurate estimation of the proportion of infected tubers with a high precision level.  相似文献

We monitored the carbon isotope composition of bulk leaves and specific long-chain alkanes during a four-year litterbag experiment using 13C-labelled leaves and unlabelled reference leaves of the European beech tree (Fagus sylvatica L.). Whereas the isotope composition of alkanes from 13C-enriched leaves exhibited a marked decrease in 13C-content, the isotope composition of unlabelled reference leaves remained nearly constant. We interpreted this difference as evidence for a microbial contribution to the long-chain alkane pool of the decomposing leaves and related it to the progressive invasion of leaves by soil organisms which was revealed upon microscopic examination. These results suggest that long-chain alkanes may not provide an unaltered record of organic carbon isotope composition in soils and sediments.  相似文献
