Potato cyst nematodes (PCN) Globodera rostochiensis (Wollenweber, 1923) Behrens, 1975 and Globodera pallida (Stone, 1973) Behrens, 1975 are the most troublesome pests of potatoes worldwide. Since 1968 there have been no investigations of the presence of PCN in ware potato lands in Croatia although these pests were recognised as an A1 plant-health quarantine species. After the first discovery of G. rostochiensis in the locality of Belica (Medjimurska County) in 2001, intensive monitoring of the distribution of PCN in Croatia started. In 2001 G. rostochiensis was detected in 55% of soil samples taken in ware potato lands. In 2002 G. rostochiensis was detected in 4.1% of soil samples and in 0.08% of soil samples in mixed populations with G. pallida. The pathotype Ro1 was found in all tested populations and Ro1 and Pa 2/3 were detected in populations from localities Vidovec, Ivanovec and Sivice. In 2003 G. rostochiensis was detected in 1.03% of soil samples, and also in 12.8% of soil samples collected in 2004. In 2003 cysts of G. rostochiensis were detected in several seed potato fields. These results indicate the urgent need for the application of further control measures to determine the presence and distribution of PCN in ware and seed potato lands in Croatia, in order to prevent the spread of PCN to non-infested areas. Integrated management strategies, neglected until 2001, have now been implemented and will prevent the build-up of infestation and replace the previous practices of monoculture, the use of short rotations and growing susceptible cultivars.  相似文献

BACKGROUND: To check for correlation between the insecticidal properties and the specificity of lectins, a comparative study was made of the insecticidal activities of two garlic lectins with different biological activities.RESULTS: The insecticidal activity of the garlic (Allium sativum L.) leaf lectin ASAL and bulb lectin ASAII towards the tobacco aphid Myzus nicotianae Blackman was studied using bioassays with transgenic tobacco (Nicotiana tabacum L. cv. Wisconsin 38). Bioassays were started with newborn nymphs of the tobacco aphid. Although during the first 7-8 days when nymphs developed to adults there were no apparent effects, part of the nymphal population was found to develop into winged (alate) forms. Later it became clear that transgenic plants expressing ASAL and ASAII had a significant effect on the reproduction capacity of the resulting adults, with a reduction of up to 40%. Different life table parameters such as prereproductive time, intrinsic rate of natural increase, generation time and doubling time were significantly affected (P < 0.05) in aphids grown on transgenic plant material expressing ASAL and ASAII.CONCLUSION: Bioassays with tobacco plants expressing ASAL and ASAII demonstrated a significant impact on the population growth of M. nicotianae. Therefore, both lectins can be considered as valuable candidate aphid control agents.  相似文献

Double-stranded RNA and type I interferon-like activity induce an antiviral state in vertebrate cells and in several fish cell lines by increasing the expression of proteins that inhibit virus replication. We compared the protection induced by the polyinosinic:polycytidylic acid (poly I:C) or poly I:C plus transfection agents against the infectious pancreatic necrosis virus (IPNV) and the infectious hematopoietic necrosis virus (IHNV) in BF-2 cells, with that induced in RTG-2, CHSE-214, or SAF cells. In addition, we examined the reduction in the infective titers of these viruses and the correlation with Mx protein expression as IFN marker. Furthermore, the suitability of BF-2 cells for the evaluation and optimization of immune responses in an IPNV-IHNV co-infection was assessed. The results demonstrated strong anti-IPNV and anti-IHNV activity (around 90% of infected cells surviving) in BF-2 cells transfected with poly I:C, in which a loss of 1log(10) or 3log(10) of the IPNV or IHNV infective titers, respectively, was observed. No antiviral activity was evident in the cells incubated with poly I:C alone. The protection recorded in the co-infection experiments was comparable with those of the single infections. The SAF cell line exhibited the lowest antiviral capacity (45%), which was also increased after transfection with poly I:C. In addition, medium from transfected BF-2 provided protection against IPNV (1log(10) loss of infective titer) and IHNV (2log(10) loss of infective titer) in new monolayers, indicating that these cells secreted the factors that induce antiviral activity. A correlation between antiviral activity and Mx protein expression was observed in all the cells. These results indicate that poly I:C transfection could improve IFN-like production in these cell lines. However, the antiviral effectiveness of poly I:C differed between cell lines. On the basis of our findings, we conclude that the BF-2 cell line is a useful model in which to study the role of IFN-induced cytokines in resistance against single or double infections with salmonid fish viruses.  相似文献

Variance components and genetic parameters for greasy fleece weights of Muzaffarnagari sheep maintained at the Central Institute for Research on Goats, Makhdoom, Mathura, India, over a period of 29 years (1976 to 2004) were estimated by restricted maximum likelihood (REML), fitting six animal models including various combinations of maternal effects. Data on body weights at 6 (W6) and 12 months (W12) of age were also included in the study. Records of 2807 lambs descended from 160 rams and 1202 ewes were used for the study. Direct heritability estimates for fleece weight at 6 (FW6) and 12 months of age (FW12), and total fleece weights up to 1 year of age (TFW) were 0.14, 0.16 and 0.25, respectively. Maternal genetic and permanent environmental effects did not significantly influence any of the traits under study. Genetic correlations among fleece weights and body weights were obtained from multivariate analyses. Direct genetic correlations of FW6 with W6 and W12 were relatively large, ranging from 0.61 to 0.67, but only moderate genetic correlations existed between FW12 and W6 (0.39) and between FW12 and W12 (0.49). The genetic correlation between FW6 and FW12 was very high (0.95), but the corresponding phenotypic correlation was much lower (0.28). Heritability estimates for all traits were at least 0.15, indicating that there is potential for their improvement by selection. The moderate to high positive genetic correlations between fleece weights and body weights at 6 and 12 months of age suggest that some of the genetic factors that influence animal growth also influence wool growth. Thus selection to improve the body weights or fleece weights at 6 months of age will also result in genetic improvement of fleece weights at subsequent stages of growth.  相似文献

Estimates of (co)variance components were obtained for weights at birth, weaning and 6, 9 and 12 months of age in Chokla sheep maintained at the Central Sheep and Wool Research Institute, Avikanagar, Rajasthan, India, over a period of 21 years (1980–2000). Records of 2030 lambs descended from 150 rams and 616 ewes were used in the study. Analyses were carried out by restricted maximum likelihood (REML) fitting an animal model and ignoring or including maternal genetic or permanent environmental effects. Six different animal models were fitted for all traits. The best model was chosen after testing the improvement of the log-likelihood values. Direct heritability estimates were inflated substantially for all traits when maternal effects were ignored. Heritability estimates for weight at birth, weaning and 6, 9 and 12 months of age were 0.20, 0.18, 0.16, 0.22 and 0.23, respectively in the best models. Additive maternal and maternal permanent environmental effects were both significant at birth, accounting for 9% and 12% of phenotypic variance, respectively, but the source of maternal effects (additive versus permanent environmental) at later ages could not be clearly identified. The estimated repeatabilities across years of ewe effects on lamb body weights were 0.26, 0.14, 0.12, 0.13, and 0.15 at birth, weaning, 6, 9 and 12 months of age, respectively. These results indicate that modest rates of genetic progress are possible for all weights.  相似文献

To evaluate terminal-sire breeds, harvest BW, prefabrication carcass measurements, and organ weights were evaluated over 3 yr for 518 crossbred wether lambs (ovis aries). Lambs were produced by single-sire matings of 22 Columbia, 22 USMARC-Composite (Composite), 21 Suffolk, and 17 Texel rams to adult Rambouillet ewes. Lambs were raised to weaning under an extensive western rangeland production system and finished in a feedlot on a high-energy finishing diet. Wethers were randomly assigned to be harvested at an average BW of 54.4, 61.2, or 68.0 kg and then transported to The Ohio State University abattoir for harvest. Wether BW was recorded before transport (off-test BW) and before harvest. Prefabrication carcass measurements and organ weights were recorded either after harvest or after an approximate 24-h chill. At comparable numbers of days on feed, Suffolk-sired lambs had heavier (P < 0.01) off-test BW, harvest BW, HCW, chilled carcass weight (CCW), and kidney weights than lambs sired by the other breeds. Suffolk-sired lambs had more (P < 0.01) kidney-pelvic fat than did Columbia-sired lambs; Composite- and Texel-sired lambs were intermediate and did not differ (P > 0.06) from the other crossbred lambs. Texel- and suffolk-sired lambs had larger lm area and greater conformation scores than Columbia-sired lambs (P < 0.03). Texel-sired lambs had greater (P < 0.01) body wall thickness, quality grades, and leg scores than Columbia-sired lambs. Composite- and Suffolk-sired lambs did not differ from each other or from lambs sired by any other breed for body wall thickness (P > 0.18) and were intermediate for quality grades and leg scores. Sire breed did not affect (P > 0.05) shipping shrink, dressing percentage, pelt weight, liver weight, and fat depth. Adjusting data to a comparable off-test BW or CCW modified sire breed rankings for some measurements. Texel-sired lambs were equal or superior to lambs sired by other breeds for HCW, CCW, dressing percentage, pelt weight, LM area, quality grade, and leg and conformation scores. However, Texel-sired lambs also had values for kidney-pelvic fat weight, fat depth, and body wall thickness that were equal to or greater than those of lambs sired by the other breeds, indicating increased fatness at comparable BW. Producers can use these results to select terminal-sire sheep breeds that will complement their production system and improve market lamb value.  相似文献

Farm animal genetic diversity is required to meet current production needs in various environments, to allow sustained genetic improvement, and to facilitate rapid adaptation to changing breeding objectives. Production efficiency in pastoral species is closely tied to the use of diverse genetic types, but greater genetic uniformity has evolved in intensively raised species. In poultry, breeding decisions are directed by a few multinational companies and involve intense selection, the use of distinct production lines, and very large populations. In dairy cattle, the Holstein breed dominates production. Intensive sire selection is leading to relatively rapid inbreeding rates and raises questions about long-term effects of genetic drift. Key questions in management of farm animal genetic diversity involve the distribution of potentially useful quantitative trait locus alleles among global livestock breeds. Experiments with tomato, maize, and mice suggest that favorable alleles can exist in otherwise lowly productive stocks; this cryptic variation may potentially contribute to future selection response. Genetic improvement under relatively intense unidirectional selection may involve both increases in the frequency of favorable additive alleles as well as the progressive breakdown of homeostatic regulatory mechanisms established under the stabilizing selection that is characteristic of natural populations. Recombination among closely linked regulatory loci and new, potentially favorable mutations are possible sources of long-term genetic variation. A greater understanding of the potential that these alternative mechanisms have for supporting long-term genetic improvement and of genetic relationships among global livestock populations are priorities for managing farm animal genetic diversity.  相似文献

Using a geographic information system (GIS), by analysis of the relationship between the spatial distribution of cattle density and the risk factors temperature, altitude and rainfall, we defined geographical habitats enabling optimal development and competence of Culicoides spp. to transmit Bluetongue-Virus (BTV): Risk zones (low, high, highest risk) were identified mainly in Baden-Württemberg, Hessen and Rheinland-Pfalz if persistently infected ruminants are imported into these zones in summer (June to August mainly), based on the current climatic conditions, BTD outbreaks are considered a real possibility. Overwintering of the virus seems unlikely. However, global warming will lead to a steady increase of the size of the risk zones. In addition, the possibility of primary outbreaks increases. The reason for this is not only the expected northern shift of Culicoides imicola, but in addition an increasing vector competence of domestic Culicoides species. We therefore recommend the storage of vaccines as well as conducting ecological studies analysing the presence of Culicoides vectors. Using the data from these studies, it will be possible to produce updated quantitative risk assessment via GIS.  相似文献

This study was designed to evaluate breed differences in resistance to Haemonchus contortus in lambs. A total of 181 ewe lambs representing crossbred Dorsets (DO) and Dorpers (DP; out of 1/2-Dorset, 1/4-Rambouillet, 1/4-Finnsheep ewes) and straight-bred Katahdins (KT) were evaluated over 3 yr. An additional 144 DO, DP, KT, and Barbados Blackbelly x St. Croix (HH) wethers were evaluated over 2 yr. Lambs were weaned at 60 to 90 d of age. After deworming at about 4 mo of age, ewe lambs received approximately 10,000 infective larvae and were evaluated for parasite resistance in drylot, whereas wethers were evaluated on pasture under conditions of natural infection. Each sex was analyzed separately. Egg counts per gram of feces (FEC), log-transformed FEC (LFEC), packed cell volumes (%), and body weights (kg) measured at 3, 4, 5, and 6 wk after deworming and reinfection were analyzed. Breed influenced all traits (P < 0.05) except BW in ewe lambs and PCV in wethers. Year and week influenced (P < 0.05) all traits. At most times, DP had the highest FEC, DO had the lowest PCV, and KT and HH had lower FEC and higher PCV than either DO or DP. Clearly, Dorper sheep were not more resistant to parasites than DO, but they were able to cope with infection better by maintaining similar or higher (P < 0.05) PCV and similar BW. Katahdin and HH were more resistant, with lower FEC (P < 0.05) than DO or DP. Breed differences were more apparent when infection levels were higher; DO and DP were less affected when infection levels were low and when animals were on a better plane of nutrition. Caribbean hair sheep originated in hot, humid regions of West Africa. They and the derivative Katahdin breed seem relatively resistant to parasitism. The Dorper, in contrast, was developed in more arid regions and exhibited little parasite resistance.  相似文献
