Diacetyl (DA), a natural butter flavorant, is a causative agent for the lung disease obliterative bronchiolitis. Mutagenic properties of 1,2-dicarbonyls have previously been empirically linked to their possible interaction with DNA nucleobases. This study for the first time identifies chemically the adduct of DA with 2-deoxyguanosine. Selective reactivity of DA with 5'-TTTGTTTTT-3' over 5'-TTTTTTTTT-3' indicated its propensity to modify specifically the guanosine residue. Treatment of plasmid DNA, pBR322, with DA induced changes in electrophoretic mobility that are typical of ternary structure disruption. Such DNA nucleobase interaction of DA translated into increased apoptosis in DA-treated SH-SY5Y cells in a dose-dependent manner (IC(50) = 0.114 ± 0.0421 mM). The traditional carbonyl scavengers metformin, 2-thiobarbituric acid, and d-penicillamine protected cells from DA toxicity in proportion to their rates of reaction with DA, with d-penicillamine causing a maximal increase in the IC(50) to 5.23 ± 0.0992 mM when co-incubated with DA.  相似文献

In most soil ecosystems, soil biological activity and associated processes are concentrated in the rhizosphere soil and is influenced by the external application of plant nutrients. The impacts of boron and sulfur on soil biological properties were evaluated in an Aeric Haplaquept (pH 5.7) growing rapeseed (Brassica campestris L.) as a test crop. Application of boron (B) at 2 mg kg?1 in combination with sulfur (S) at 30 mg kg?1 (B2S30) resulted in highest available Boron and sulfur of 0.239 and 15.4 mg kg?1, respectively and registered 62.5% and 71.3% increase over control (B0S0) at 60 days of crop growth compared to individual applications. The microbial populations viz. phosphate solubilizing microorganisms (PSM) and nitrogen fixing bacteria (NFB) were the highest of 52.63 and 85.87 × 105 g?1 soil, respectively, CFU in B2S30 treatment at 60 days and adjudged as the best treatment combination for enhancement of soil biological indices and seed yield.  相似文献

A study was conducted to compare the nutrient utilization, growth, and rumen enzyme profile of mithun (Bos frontalis) and Tho-tho cattle (Bos indicus) reared in the same feeding and managemental conditions. For the purpose, male mithun (n = 8) and male Tho-tho cattle (n = 8) of 1.5 years age, selected from the farm of National Research Centre on Mithun, Nagaland, India, were fed on mixed-tree-leaves-based ration as per the requirement of NRC (2001) for cattle for 12 months. Average daily gain (ADG), average dry matter intake (DMI), and feed conversion ratio (FCR) for all animals were recorded. A metabolic trial was conducted at 6 months of the experiment to assess the digestibility coefficient of different nutrients and nutritive value of ration. At 12 months of the experiment, rumen liquor was collected from all animals and analyzed for rumen enzyme profiles, viz., carboxymethylcellulase, xylanase, α-amylase, β-glucosidase, α-glucosidase, urease, and protease. It was found that ADG (507.8 g vs 392.8 g), DM intake (6.59 vs 5.85 kg/day) and DMI/W0.75 (98.75 g vs 91.00 g/day), crude protein intake (780 vs 700 g/day), and total digestible nutrient intake (3.65 vs 3.32 kg/day) were higher (p < 0.05) in mithun than cattle. The nitrogen balance was higher and FCR was better (p < 0.05) in mithun compared with cattle. The digestibility coefficient of different nutrients was similar (p > 0.05) between the species. The microbial enzyme profiles of mithun and cattle were not different (p > 0.05). The better growth performance of mithun than cattle as found in the present study clearly indicates that the mithun has higher genetic potential for growth than Tho-tho cattle of north-eastern hilly region of India.  相似文献

The independent modes of action of diuron and atrazine on the photochemical activities of chloroplasts (In vivo and in vitro) from the leaves of crop plants Pisum sativum and Pennisetum typhoides and the weeds Amaranthus viridis and Cyperus rotundus were investigated. Hill reaction activity (DCPIP photoreduction) of in vivo chloroplasts (chloroplasts isolated from herbicide-sprayed plants) was unaffected by treatment at sublethal or intermediate levels of diuron or atrazine while that of in vitro chloroplasts (chloroplasts incubated in the required herbicidal concentration) was severely inhibited. The ferricyanide catalyzed noncyclic photophosphorylation was markedly reduced in both the in vivo and in vitro chloroplast systems. N-Methyl phenozonium sulfate (PMS)-mediated cyclic photophosphorylation was inhibited in the in vivo system while a pronounced enhancement of activity was noticed in the in vitro chloroplasts. The rate of NADP+ photoreduction was severely inhibited in the in vitro chloroplasts. The unaffected in the in vivo system. The herbicidal effects on the photoreactions of isolated chloroplasts were compared with chloroplasts isolated from herbicide-sprayed plants.  相似文献

Tuberose cut flowers, available as loose flowers, were treated with gamma (γ) irradiation and generally regarded as safe (GRAS) preservative solutions for extension of shelf life. The flowers were packaged in low-density polyethylene bags, heat sealed and stored at 23 ± 2°C, 80% relative humidity (RH) and 4 ± 1°C, 40% RH, respectively. The flowers stored at these two temperature regimes were subjected to sensory evaluation and biochemical analyses. From these assessments, the longest shelf life of tuberose flowers was found to be 8 days at 23 ± 2°C, 80% RH (compared to 4 days for control) and 24 days at 4 ± 1°C, 40% RH (compared to 8 days for control) using combination treatment of low dose γ-irradiation (0.02 kGy) and preservative solutions (4% sucrose and 0.02% CaCl2). Ethanolic extract of tuberose flowers of the most shelf stable set (stored at 4 ± 1°C), i.e. at the end of 24 days, showed antimicrobial potency against the common skin pathogen Staphylococcus aureus (ATCC 25923 and MDR strains), suggesting utility of the senesced tuberose flowers for therapeutic applications. This preservation technique would promote export of tuberose flowers by extension of their shelf lives and allow utilization of these flowers, post senescence.  相似文献

Increasing soil carbon (C) in arable soils is an important strategy to achieve sustainable yields and mitigate climate change. We investigated changes in soil organic and inorganic carbon (SOC and SIC) under conservation agriculture (CA) in a calcareous soil of the eastern Indo-Gangetic Plains of India. The treatments were as follows: conventional-till rice and wheat (CT-CT), CT rice and zero-till wheat (CT-ZT), ZT direct seeded rice (DSR) and CT wheat (ZT-CT), ZTDSR and ZT wheat without crop residue retention (ZT-ZT), ZT-ZT with residue (ZT-ZT+R), and DSR and wheat both on permanent beds with residue (PB-PB+R). The ZT-ZT+R had the highest total SOC in both 0–15 and 15–30 cm soil layers (20% and 40% higher (p < .05) than CT-CT, respectively), whereas total SIC decreased by 11% and 15% in the respective layers under ZT-ZT+R compared with CT-CT. Non-labile SOC was the largest pool, followed by very labile, labile and less labile SOC. The benefits of ZT and residue retention were greatest for very labile SOC, which showed a significant (p < .05) increase (~50%) under ZT-ZT+R compared with CT-CT. The ZT-ZT+R sequestered ~2 Mg ha−1 total SOC in the 0–15 cm soil layer in 6 years, where CT registered significant losses. Thus, the adoption of CA should be recommended in calcareous soils, for C sequestration, and also as a reclamation technique.  相似文献

Polyvinylpyrrolidone/date palm leaf fiber (PVP/DPL) biocomposites were prepared by melt mixing fabrication technique with different weight percentage of fibers. DPL fibers were chemically modified by acrylic acid in order to have better dispersion and compatibility with PVP matrix. The interaction of DPL fibers with PVP matrix was studied by Fourier transforms infrared spectroscopy (FTIR). Field emission scanning electron microscope (FESEM) was used for the study the morphology of chemically modified DPL fibers and PVP/DPL biocomposites. Mechanical properties were improved with fiber loading due to strong interfacial adhesion between PVP and DPL fibers. The storage modulus, loss modulus and tan delta values of PVA/DPL biocomposites were measured by DMTA. The rheological properties were investigated to study the shearing storage and loss modulii along with complex viscosity of biocomposites. The thermal and conducting properties of biocomposites were measured and compared with that of virgin PVP.  相似文献

Cultivated groundnut is susceptible to late leaf spot (LLS) caused by Phaeoisariopsis personata [(Berk. & M.A. Curtis) Aex] and resistance is low to moderate in the primary gene pool of groundnut. Closely related wild species in the secondary gene pool are highly resistant to the disease. All the closely related Arachis species are diploid and cultivated groundnut is a tetraploid. Utilization of diploid Arachis species to transfer LLS resistance is time consuming and cumbersome. New sources of Arachis hypogaea (also called synthetic groundnut) were developed at ICRISAT. These are tetraploids and the present investigation has shown that they are resistant to LLS.  相似文献

A line × tester analysis was carried out in mulberry (Morus spp.) to determine the genetic interaction in the expression of various quantitative characters including leaf yield. Eight clonal varieties were selected, 3 of them were designated as lines (♀) viz. Berhampore-1, China white and MS-5 and 5 of them were called as testers (♂) viz. Mandalaya, Kosen, Assamjati, MS-1 and Kajli. Combining ability studies were conducted on these parents along with their F1 hybrids for the variables laminar index, growth rate, weight of 100 dry leaves, number of primary branches per plant, plant height, nodal distance, leaf-twig ratio, aerial biomass, moisture content, moisture retention capacity and leaf yield. Broad genetic variability was observed among the genotypes. The ratio of General Combining Ability (GCA) and Specific Combining Ability (SCA) indicated the predominance of non-additive genes in mulberry. While China white (female) and MS-1 (male) were the best general combiners among the parents, Berhampore-1 × Kajli was the best cross for leaf yield. Results suggest that selective crossing followed by proper screening may be the best approach for breeding of high yielding varieties in mulberry.
