Uapaca kirkiana (Muell. Arg.), a highly valued indigenous fruit species, is being domesticated to increase its utilization in southern Africa. Vendors, who were also the fruit gatherers, that were selling U. kirkiana fruits at roadside markets in Dedza, Malawi, were interviewed on four occasions during the marketing season (October 2003–January 2004) and fruit samples were concurrently sampled from each vendor. Information was obtained on when and how the fruits had been harvested and handled. Timing of fruit harvest was based on experience, aided by indicators such as occurrence of first heavy rains, natural abscission, size and color changes. Harvesting by knocking down unripe fruits from trees was practiced throughout the␣season, but naturally abscised fruits were also gathered in December and January. Fruits harvested when mature but unripe required incubation in soil, plain and woven plastic bags or clay-pots. The incubation method changed and the duration of incubation decreased between October (3–4 days) and January (≤2 days). Fruit samples were evaluated for damage, color and soluble solids concentration (SSC). Fruit SSC and color lightness/brightness (L*) increased from 10.1 to 16.4% (P<0.01) and from 45.7 to 50.5 units, (P<0.01), respectively. Higher SSC values were generally associated (R 2=0.68, P<0.001) with increasing pulp color intensity. Cracking, at 48%, constituted the most common form of damage in sampled fruits. On average, fruits were saleable for only 3–4 days, this being attributed to inherent fruit characteristics combined with immaturity at harvest, and damage during harvesting and storage in hot dry conditions. Lack of appropriate harvesting and handling techniques were associated with fruit darkening, drying, rotting and other postharvest problems.  相似文献

Pyrolysis behavior of levoglucosan (1,6-anhydro--d-glucopyranose), the major anhydromonosaccharide formed during cellulose pyrolysis, was studied at 250°–400°C under nitrogen. The pyrolysis products were found to change stepwise: levoglucosan MeOH-soluble fraction (lower-molecular-weight products and oligosaccharides) water-soluble fraction (polysaccharides) insoluble fraction (carbonized products). From the present experimental results, a pathway of cellulose pyrolysis via anhydromonosaccharide is proposed including polymerization to polysaccharides (a reversible reaction) as a key reaction to carbonized product formation.Part of this study was presented at the XIXth International Carbohydrate Symposium, San Diego, August 1998; and the 50th Annual Meeting of the Japan Wood Research Society, Kyoto, April 2000  相似文献

Effects of side chain hydroxyl groups on pyrolytic β-ether cleavage of phenolic model dimers were studied with various deoxygenated dimers under pyrolysis conditions of N2/400°C/1 min. Although phenolic dimer with hydroxyl groups at the C α and C γ positions was much more reactive than the corresponding nonphenolic type, deoxygenation at the C γ -position substantially reduced the reactivity up to the level of the nonphenolic type. These results are discussed with the cleavage mechanism via quinone methide intermediate formation, which is activated through intramolecular hydrogen bonds between C α and C γ hydroxyl groups.  相似文献

Fusarium wilt is the main pigeonpea production constraint in Malawi. The purpose of the study was to understand the nature and mechanism of inheritance of F. wilt resistance, yield and secondary traits in pigeonpea. 48 crosses were generated in a 12 lines × 4 testers mating scheme. Some F1 plants were selfed for segregation analysis for inheritance pattern of resistance, while others were evaluated for resistance, yield and secondary traits. There were significant variations among F1 plants for F. wilt, days to 50 % flowering, seed/pod, and number of secondary branches. Specific combining ability (SCA) effects were predominant for F. wilt, days to 50 % flowering and number of secondary branches. The general combining ability (GCA) effects, mainly due to maternal genotypes, were preponderant for yield and other secondary traits. The significance of GCA and SCA effects suggested that variations were due to additive gene action in both the testers and parental lines arising from their interactions, and the dominance effects due to interactions of the parental lines. The χ2 analysis suggested dominant patterns of inheritance for wilt in most of the F2 populations. The segregation ratios of 3:1, 15:1, and 9:7 suggested the involvement of single or two independent/complementary dominant genes in the test donors. Involvement of a few genes governing wilt resistance suggested the ease of breeding for this trait. Pedigree breeding method would be recommended for incorporating various traits in pigeonpea.  相似文献

The risk of fish pathogen transmission via eggs can be reduced by disinfection in ozonated seawater. The aim of this study was to determine the suitable conditions for ozone disinfection of the eggs of gilthead seabream Sparus aurata, sea bass Dicentrarchus labrax, red porgy Pagrus pagrus, and common dentex Dentex dentex. The eggs were disinfected with a concentration (C) of 0.5 mg of ozone/L of water at four different exposure times (T = 2, 4, 8, and 16 min). The hatching rate was determined in triplicate for each treatment. Bacterial colonies were counted on tryptic soy agar and thiosulfate-citrate-bile salts-sucrose agar. At the end of the experiment, bacterial load and hatching rate were assessed together to determine the optimal ozone treatment values, which were estimated in CT units (i.e., C [= 0.5 mg/L] x T [min]). Optimal values were CT 2-4 (T = 4-8 min; 18 degrees C) for gilthead seabream and red porgy, CT 2 (T = 4 min; 18 degrees C) for common dentex, and CT 4 (T = 8 min; 15 degrees C) for sea bass.  相似文献

Two-step hydrolysis of Japanese cedar (Cryptomeria japonica) was studied as treated by semi-flow hot-compressed water at 230°C/10 MPa for 15 min and 280°C/10 MPa for 30 min as the first and second stages, respectively. At the first stage, hemicelluloses and para-crystalline cellulose, whose crystalline structure is somewhat disordered, were found to be selectively hydrolyzed, as well as lignin decomposition, whereas crystalline cellulose occurred at the second stage. In all, 87.76% of Japanese cedar could be liquefied by hot-compressed water and was primarily recovered as various hydrolyzed products, dehydrated, fragmented, and isomerized compounds as well as organic acids in the water-soluble portion. The remainder, 12.24%, could not be hydrolyzed and remained as the water-insoluble residue composed entirely of lignin. Based on the distribution of various products from hemicelluloses in Japanese cedar, their decomposition pathways were proposed as independent.  相似文献

The aim of this study was to characterize the chemical composition of different granulometric fractions obtained by milling of the bark of two softwood [Norway spruce, Picea abies (L.) Karst., and Scots pine, Pinus sylvestris L.] and two hardwood species (birch, Betula pendula Roth, and eucalypt, Eucalyptus globulus Labill.), and to discriminate between them on the basis of their chemical composition content via pattern recognition techniques [principal component analysis (PCA), cluster analysis (CA), and discriminant analysis (DA)]. Bark chemical composition differed between species, and chemical variables could be used to differentiate between them. Size reduction yields granulometric fractions that are not chemically homogeneous and that can also be discriminated. Therefore, potential applications of bark in valorization programs have to carefully consider the species-specific composition and their size reduction patterns. PCA and CA were adequate tools to characterize the different bark fractions within each species. DA allowed identifying the bark samples according to species and independently from particle size. Pattern recognition statistical methods were shown to be useful tools to analyse bark fractions and chemically discriminate species and fractions.  相似文献

Parkinson's disease (PD) is a neurodegenerative disorder characterized by degeneration of dopaminergic neurons in the substantia nigra. We previously mapped a locus for a rare familial form of PD to chromosome 1p36 (PARK6). Here we show that mutations in PINK1 (PTEN-induced kinase 1) are associated with PARK6. We have identified two homozygous mutations affecting the PINK1 kinase domain in three consanguineous PARK6 families: a truncating nonsense mutation and a missense mutation at a highly conserved amino acid. Cell culture studies suggest that PINK1 is mitochondrially located and may exert a protective effect on the cell that is abrogated by the mutations, resulting in increased susceptibility to cellular stress. These data provide a direct molecular link between mitochondria and the pathogenesis of PD.  相似文献

The effect of the essential oils (EOs) of peppermint, Mentha piperita L., and tea tree, Melaleuca alternifolia (Maiden & Betche) Cheel, on the haematological, biochemical, and immunological parameters and intestinal morphology of Nile tilapia, Oreochromis niloticus L., was evaluated. Fish (58.09 ± 5.87 g) were fed 100 mg/kg and 250 mg/kg of each EO and sampled on days 7, 14, 30 and 60 after starting supplementation. The haematological and biochemical parameters were not altered by the supplementation of EOs compared to the control (p > .05). With regard to the immunological parameters, the activation of the complement system of fish fed 250 mg/kg peppermint and 100 mg/kg and 250 mg/kg tea tree EOs were significantly higher compared to the control after 60 days of feeding (p < .05). The complement system plays an essential role in innate immunity and contributes significantly to the acquired immune response; thus, its activation through supplementation with EOs is promising for the formulation of nutritional additives in aquaculture. Regarding intestinal morphology, fish fed 250 mg/kg tea tree EO presented higher villus size compared to all other groups (p < .05), which represents a healthier gut. These fish present a larger intestinal surface, which can result in better absorption and utilization of the nutrients. Based on the responses found in this study, both EOs were considered promising for the formulation of feed additives for Nile tilapia.  相似文献

There is a lack of reliable hepatotherapeutic drugs in modern medicine in the management of alcohol/drug-induced liver damage. Aloe vera extract has been used in folklore medicine for its medicinal values. This study evaluates the hepatotherapeutic activity of aqueous extract of Aloe vera gel in rats. Sprague-Dawley rats were divided into three groups; the negative control, positive control and the extract-treated groups. The negative control received only distilled water daily. The positive control received alcohol, while the extract-treated group received aqueous extract of Aloe vera and alcohol. Hepatotoxicity was induced in the positive control and extract-treated rats with alcohol. The hepatotherapeutic effect was evaluated by performing an assay of the serum total bilirubin, alkaline phosphatase, aspartate and alanine transaminases and liver histopathology. Alanine transaminase activities were comparable in all groups. Alcohol treatment alone significantly (p < 0.05) increased total serum bilirubin, alkaline phosphatase and aspartate transaminase activities. Alcohol-induced hepatic dysfunction was abrogated by Aloe vera extract. Histopathological examination revealed that alcohol induced hepatic damage. Aloe vera treatment maintained hepatic architecture similar to that seen in the control. This study shows that aqueous extract of Aloe vera gel is hepatotherapeutic and thus lends credence to the use of the plant in folklore medicine in the management of alcohol-induced hepatic dysfunction.  相似文献
