Skeletal muscle ventricles (SMVs) were constructed from canine latissimus dorsi and connected to a totally implantable mock circulation device. The SMVs, stimulated by an implantable pulse generator, pumped continuously for up to 8 weeks in free-running beagle dogs. Systolic pressures produced by the SMVs, initially of 139 +/- 7.2 mmHg and after 1 month of continuous pumping of 107 +/- 7 mmHg, were comparable to normal physiologic pressures in the adult beagles (114 +/- 21 mmHg). After 2 weeks of continuous pumping, the mean stroke work of the SMVs was 0.4 X 10(6) ergs, a performance that compares favorably with the animal's cardiac ventricles. This study shows that canine skeletal muscle which has not received prior training or electrical conditioning can perform sustained work at the high levels needed for an auxiliary cardiovascular pump. It might be possible eventually to use such muscle pumps in humans to assist the failing circulation and to provide support in children with certain types of congenital heart defects.  相似文献

Recovery of Salmonella from shell eggs.   总被引:1,自引:0,他引:1  
A preenrichment procedure and a direct selective enrichment procedure were compared for recovery of Salmonella artificially inoculated into liquid whole egg, egg yolk, and egg albumen. For liquid whole egg and egg yolk, the 2 procedures were comparable. With egg albumen, however, preenrichment in lactose broth gave significantly higher recoveries than did direct selective enrichment in either selenite cystine or tetrathlonate broths. The lactose preenrichment procedure was used to determine the survival of S. enteritidis in egg yolk and egg albumen over a period of 7 days. As shown by most probably number determinations, counts of S. enteritidis inoculated into egg albumen decreased by 3 log units, whereas those in egg yolk did not change significantly. It is recommended, therefore, that only the egg yolk be examined for this pathogen. In a comparison of 5 different preenrichment media (lactose broth, brain heart infusion broth, trypticase soy broth, buffered peptone water, and nutrient broth), lactose broth was somewhat less productive than the other 4 media for the recovery of Salmonella from egg yolks. Despite evidence that domestic sheep diseases threaten the persistence of bighorn sheep populations, the economic consequences of restricting domestic sheep grazing has polarized the debate, with some arguing that disease risk posed by domestic sheep has been exaggerated and grazing restrictions should be eased. We constructed a model to assess how different management strategies (grazing allotment closures, grazing time reductions, and reduced probability of stray domestic sheep) affect the risk of respiratory disease transmission from domestic sheep to endangered Sierra Nevada bighorn sheep, and to predict population-level impacts of an outbreak. Even when management strategies reduced risk of interspecies contact to less than 2% per year, our model predicted a 50% probability of a catastrophic respiratory disease outbreak during the next 10 bighorn sheep generations. If an outbreak occurs in the near future, the model predicts that the smallest Sierra Nevada bighorn sheep population would have a 33% probability of quasi-extinction. To eliminate all risk of contact and potential disease transmission, domestic sheep cannot be grazed on allotments that overlap with areas utilized by Sierra Nevada bighorn sheep. Where wildlife and domestic animal populations share limited habitat, and there is documented evidence of a substantial disease threat and extinction risk, stakeholders must recognize that the only way to eliminate contact and risk of disease transmission is to give priority to one species or the other. If conservation is the priority, difficult decisions will need to be made to balance trade-offs between economic livelihoods and species conservation.  相似文献

A competitive indirect enzyme-linked immunosorbent assay (CI-ELISA) was developed to quantitate the herbicide dicamba (3,6-dichloro-2-methoxybenzoic acid) in water. The CI-ELISA has a detection limit of 2.3 microg L(-1) and a linear working range of 10--10000 microg L(-1) with an IC(50) value of 195 microg L(-1). The dicamba polyclonal antisera did not cross-react with a number of other herbicides tested but did cross-react with a dicamba metabolite, 5-hydroxydicamba, and structurally related chlorobenzoic acids. The assay was used to estimate quantitatively dicamba concentrations in water samples. Water samples were analyzed directly, and no sample preparation was required. To improve detection limits, a C(18) (reversed phase) column concentration step was devised prior to analysis, and the detection limits were increased by at least by 10-fold. After the sample preconcentration, the detection limit, IC(50), and linear working range were 0.23, 19.5, and 5-200 microg L(-1), respectively. The CI-ELISA estimations in water correlated well with those from gas chromatography-mass spectrometry (GC-MS) analysis (r(2) = 0.9991). This assay contributes to reducing laboratory costs associated with the conventional GC-MS residue analysis techniques for the quantitation of dicamba in water.  相似文献

Mass media images offer audiences models for how to perform the social roles they depict. Opinions and other attributes of credible media models may likewise be embraced by audience members seeking to identify with those models. Thus farm magazine narratives about “successful” farmers may encourage readers to model or aspire to featured farmers' production and management techniques and ascribe legitimacy to models' responses to current agricultural issues. However, production of agrarian images in the mass media — including images of farms, farmers, and farmers' values — are inevitably biased such that media representations of successful farmers selectively present objective characteristics in terms of the media's own ideological frameworks, which in turn reflect the dominant ideology of the social relations in which the media are engaged. As a first step in identifying farm magazines' role in creating social models for farmers, this study analyzes articles featuring “successful,” “leading,” or “innovative” farmers in leading agricultural magazines. The featured farmers are categorized according to enterprise characteristics and characterizations of them and their management philosophies. Findings show that farmers in farm magazines have larger than average operations and are portrayed in a way that blends a “farming as business” orientation with more conventional agrarian values but that generally omits non-business aspects of farm life.  相似文献

Despite the growing popularity of ecosystem‐based management (EBM) in national legislation and in research and institutional literature, there is often an implementation gap ‘on the ground’, impeding widespread adoption in fisheries. This gap reflects in part the differing understandings of EBM held by fishermen and by management institutions. To explore and seek to close this gap, the underlying principles of EBM considered priorities by fishermen were systematically compared with the priorities identified in the published literature. The fishermen's priorities were determined by asking Atlantic Canadian fishermen to identify the EBM principles they consider most important. Four priority principles were identified: Sustainability, Stakeholder Involvement, Develop Long‐Term Objectives and Use of All Forms of Knowledge. The latter two were not frequently noted as priorities in the literature, while some literature priorities were less commonly chosen by fishermen, indicating a significant difference in perspectives on EBM. The rationale for fishermen's choice of priorities was explored by analysing the fishery management issues they raised – many directly connected to the above four priorities. In addition, another principle, Commit to Principles of Equity, often arose as an implicit priority among fishermen. We suggest that success in implementation of EBM may depend on reconciling differing priorities among its underlying principles, and combining knowledge and expertise from fishermen with research and institutional sources. The comparative methodology used here, which could be replicated elsewhere, should lead to better recognition of local challenges in EBM implementation and encourage support for EBM, to further its contribution to sustainable fisheries.  相似文献

Dispersion during the larval phase is of central importance in the dynamics of marine fish and invertebrate populations. Rapid transport or dispersion of larvae may contribute to connectivity and mixing, whereas spatial persistence (retention) is hypothesized to favour stock complexity and local subpopulations. Larval retention, while rarely quantified, may be defined in species with protracted spawning by the spatial co‐occurrence of larvae of different sizes or ages. The spatial distributions of larval Atlantic herring (Clupea harengus) were examined from 22 annual autumn surveys (1975–1998) and 9 spring surveys (1975–1984) from the Bay of Fundy, a region with large tides and residual flow. Larvae of all sizes (3–27 mm in length, from hatch to nearly 4 months post‐hatch) were observed each year in two major aggregations; one off southwestern Nova Scotia, and the other in the mid‐inner Bay of Fundy off the northwestern shore of Nova Scotia. Two similar aggregations were evident over 5 months later from 9 spring surveys (1975–1984), despite the residual flow that would have swept the larvae from the region within 1 month. Larval retention was apparent from overlapping centres of mass of different size (=age) classes of larvae, and tested using a size diversity index, based on the co‐occurrence of 1‐mm‐size categories, derived from protracted spawning of several weeks. Geospatial ‘hot spots’ (Gi* statistic) of four size (age) classes were evident at specific stations in the 50–100 m bathymetric zone and not elsewhere. These metrics provide quantitative measures of retention that may be applied to many ichthyoplankton data sets. One of the three main spawning areas collapsed during the study period after a period of intense fishing and failed to rebuild, but there was no substantial change in the location of larval hotspots in subsequent years. While larval retention does not directly relate to each spawning location, larval retention in the Bay of Fundy contributes to the complex ‘metapopulation’ structure of herring stocks in the western Atlantic.  相似文献

Recent approaches to the planning of marine protected area (MPA) networks for biodiversity conservation often stress the need for a representative coverage of habitat types while aiming to minimize impacts on resource users. As typified by planning for the Australian South‐east Marine Region, this strategy can be manipulated by political processes, with consequent biased siting of MPAs. Networks thus created frequently possess relatively low value for biodiversity conservation, despite significant costs in establishment and maintenance.
  • 2. Such biases can be minimized through application of the data‐driven and species‐based concept of key biodiversity areas (KBAs).
  • 3. By mapping locations of threatened species and populations that are highly aggregated in time or space, the KBA process allows marine sites of global biodiversity significance to be systematically identified as priority conservation targets. Here, the value of KBAs for marine conservation planning is outlined, and guidelines and provisional criteria for their application provided.
