The present study investigated genetic diversity, structure and hybridization in a collection of the endangered wild pear species Pyrus pyraster (L.) Burgd. A total of 278 putative ‘true type’ P. pyraster trees originating from seven populations in the federal state of Saxony in Germany were analyzed along with 35 pear cultivars commonly cultivated in Saxony. The genetic analysis was performed using nine nuclear microsatellite markers (ncSSR) and two paternally inherited chloroplast marker (cpDNA) amplifying in the intergenic spacer region trnQrps16 and the intron region rps16. On basis of the ncSSR dataset after STRUCTURE analysis 80 % of the wild pear individuals were assigned as ‘true type’ P. pyraster genotypes. The cpDNA analysis showed shared haplotypes in P. pyraster and P. communis but with an unequal frequency in both species. The analysis of molecular variance resulted in a moderate (ncSSR) and great (cpDNA) variation among ‘true type’ P. pyraster and the pear cultivars. The genetic diversity in the ‘true type’ P. pyraster populations was still high and the genetic structure between the populations low (ncSSR and cpDNA) indicating a genetic exchange between the populations by pollen and seeds. The clear discrimination between the P. pyraster and P. communis confirms our expectation of the existence of ‘true type’ P. pyraster individuals in the study area. The existing genetic integrity and the high genetic diversity argue for the implementation of preservation measures in P. pyraster.  相似文献

The real-time PCR methods recommended in the European Union for the quantitation of genetically modified (GM) maize events NK603, GA21, and MON 863 measure the number of copies of the GM event in relation to those of the maize-specific adh1 reference gene. The study reported here revealed that the targeted 70 base pair adh1 region exhibits a single nucleotide polymorphism (SNP839) that hampers the binding of the reverse primer used in the adh1 detection method. Partial fragments of the adh1-A and adh1-F allele were cloned. By allele-specific real-time PCR, it was shown that SNP839 corresponds to a common allelic polymorphism in maize. As a result, the quantitation of the GM maize events mentioned is positively or negatively biased, depending on the adh1 genotype of sample and calibrant. Therefore, it is proposed to revise the quantitative detection methods for NK603, GA21, and MON 863 maize.  相似文献

Earthworm activity is observed at long‐term monitoring sites as an indicator of soil function to assess changes resulting from environmental and management conditions. In order to assess changes, characteristic values of earthworm populations under different site conditions have to be known. Therefore, a classification scheme for site‐specific earthworm populations was developed for soil in agricultural use, taking interactions between earthworm populations and soil properties into account. Characteristics of sites grouped by means of a cluster analysis after principal‐component analysis served as a basis for the derivation of the classification scheme. Soil variables found to characterize site differences with respect to earthworm populations were the texture of the topsoil, the texture of the subsoil, and the soil organic‐matter (SOM) content. The textural classes of the topsoil were divided into five groups comprising sandy soils (Ss), silty sand soils (Su), slightly loamy sand soils (Sl2), medium to strongly loamy sand soils (Sl3/Sl4), and loam and clay soils. Soil organic matter was divided into grades of equal size in a range from <1%, 1%–2% up to >6%. The variables “earthworm abundance” and “earthworm species” were selected to represent earthworm populations and were divided into six groups ranging from very low to extremely high. Defined groups of earthworm populations showed a clear structure in relation to soil textural groups and the content of SOM. From this distribution, a classification scheme was derived as basis for prognostic values of site‐specific earthworm populations, thus enabling the interpretation of changes over time. For some soil textural groups, selected variables appeared to enable the derivations of expected earthworm densities and species composition outside the range of the given database, but for some soil textural groups, broader databases will be needed to specify these derivations.  相似文献

OBJECTIVE: To investigate subjective and computerized methods of evaluation of color Doppler (CD) and power Doppler (PD) ultrasonographic images (obtained before and after administration of contrast medium) for quantitative assessment of vascularity and perfusion of various naturally occurring tumors in dogs. SAMPLE POPULATION: 34 tumors in 34 dogs. PROCEDURE: Tumors in dogs were examined via CD and PD ultrasonography before and after i.v. injection of a microbubble contrast agent (pre- and postcontrast examinations, respectively). Images were digitized for subjective assessment of vessel density and vascular pattern and computer-aided assessment of parameters of vascularity (fractional area [FA]) and perfusion (color-weighted FA [CWFA] and mean color-weighted FA [CWFA] and mean color level). RESULTS: With both analysis methods, more vessels were identified in precontrast PD ultrasonographic images than in precontrast CD ultrasonographic images. Moreover, compared with values for precontrast PD ultrasonography, FA, CWFA, and mean color level were higher for postcontrast PD ultrasonography. In postcontrast images, there was a significant association between vessel densities determined through subjective and computerized assessments. Although sample size was small, vascularity of squamous cell carcinomas was significantly greater than that of other tumor types. Ten of the 19 softer than issue that sarcomas had low vessel density with minor contrast enhancement. With increasing gross tumor volume, FA and CWFA decreased for all Doppler ultrasonographic methods. CONCLUSIONS AND CLINICAL RELEVANCE: Higher values of the ultrasonographic parameters representing vascularity and perfusion of tumors in dogs were determined via PD ultrasonography after administration of contrast medium than via PD or CD ultrasonography without administration of contrast medium.  相似文献

Headshaking is a common problem in horses. The etiology is unknown but thought to involve sensory input from branches of the trigeminal nerve, some of which are within the infraorbital canal. The objective of this retrospective cross‐sectional study was to describe the CT anatomy and variations of the infraorbital canal in horses with local disease processes and normal horses, and to examine associations between those findings and headshaking. Computed tomography scans were reviewed and morphological changes of the infraorbital canal were described. Presence of changes was then tested for association with headshaking prevalence, presence of disease processes in the region of the infraorbital canal, age, and sex. Nonparametric tests were used and a P‐value of .05 was considered significant. A total of 218 horses were included, 9% of which had headshaking and 45% had CT lesions in the region of the infraorbital canal. Morphological changes to the bone of the infraorbital canal were found in 121 horses (56%) and included the following: increased mineralization 39 (18%), decreased mineralization 89 (41%), deformed shape 51 (23%), displaced position 43 (20%), and disruption 11 (5%). All changes of the infraorbital canal significantly increased in frequency with the presence of adjacent disease. Increased mineralization and disruption of the infraorbital canal were significantly associated with headshaking in horses with adjacent disease; the latter only reached significance after exclusion of dentally immature horses. No other changes were significantly associated with the presence of headshaking. No association was found between headshaking and the age or sex of the horse.  相似文献

In this study, we investigated how co-occurrence patters of ammonia and nitrite oxidizers, which drive autotrophic nitrification, are influenced by tree species composition as well as soil pH in different forest soils. We expected that a decline of ammonia oxidizers in coniferous forests, as a result of excreted nitrification inhibitors and at acidic sites with low availability of ammonia, would reduce the abundance of nitrite-oxidizing bacteria (NOB). To detect shifts in co-occurrence patterns, the abundance of key players was measured at 50 forest plots with coniferous respectively deciduous vegetation and different soil pH levels in the region Schwäbische Alb (Germany). We found ammonia-oxidizing archaea (AOA) and Nitrospira-like NOB (NS) to be dominating in numbers over their counterparts across all forest types. AOA co-occurred mostly with NS, while bacterial ammonia oxidizers (AOB) were correlated with Nitrobacter-like NOB (NB). Co-occurrence patterns changed from tight significant relationships of all ammonia and nitrite oxidizers in deciduous forests to a significant relationship of AOB and NB in coniferous forests, where AOA abundance was reduced. Surprisingly, no co-occurrence structures between ammonia and nitrite oxidizers could be determined at acidic sites, although abundances were correlated to the respective nitrogen pools. This raises the question whether interactions with heterotrophic nitrifiers may occur, which needs to be addressed in future studies.  相似文献

Malus sylvestris (L.) Mill. is the only indigenous wild apple species in Europe and recognized as rare and endangered species. An important prerequisite for the preservation of M. sylvestris is the identification of ‘true type’ M. sylvestris individuals because hybrids with cultivated apple could be also occurred. In this study variations among 625 putative M. sylvestris trees originated in the East Ore Mountains were evaluated with 20 selected morphological traits. The importance of these traits concerning the differentiation of ‘true type’ and hybrids of M. sylvestris were determined by statistical analysis. The results demonstrated a strong correlation of individual traits and those contributing mostly to variation of M. sylvestris were detected by the Principal Component Analysis. The relationship of 284 M. sylvestris, 18 M.?×?domestica, M. floribunda 821 and M. robusta 5 individuals were calculated based on the morphological data and presented in a dendrogram. The individuals grouped in two main clusters and 68 hybrids of M. sylvestris were identified. The main result of the morphological data of these hybrids indicates that the grouping is mainly based on the flower and leaf pubescence and less on the fruit size.  相似文献

Abandonment of mountain grassland often changes vegetation composition and litter quantity and quality, but related effects on labile soil organic matter (SOM) are largely unknown. The aim of this study was to investigate the impacts of grassland management and abandonment on soil carbon distribution in light (< 1.6 g cm–3) particulate organic matter (POM) and aggregation along a gradient of management intensity including hay meadows, pastures, and abandoned grasslands. The reduction of management intensity is an interregional phenomenon throughout the European Alps. We therefore selected sites from two typical climate regions, namely at Stubai Valley, Austria (MAT: 3°C, MAP: 1097 mm) and Matsch Valley, Italy (MAT: 6.6°C, MAP: 527 mm), to evaluate effects of land‐use change in relation to climate. Free water‐floatable and free POM (wPOM, fPOM), and an occluded POM fraction (oPOM), were isolated from three water‐stable aggregate size classes (2–6.3 mm, 0.25–2 mm, < 0.25 mm) using density fractionation. Aggregate mean weight diameter slightly decreased with decreasing management intensity. In contrast to absolute POM‐C, fPOM‐C increased in aggregates at both sites with abandonment. Because the oPOM‐C was less affected by abandonment, the ratio of oPOM‐C : fPOM‐C shifted from > 1 to < 1 from meadow to abandoned grassland in aggregates at both sites and thus independent of climate. This suggests that in differently managed mountain grasslands free and occluded POM are functionally different SOM fractions. In bulk soil, the oPOM‐C : fPOM‐C ratio is better suited as an indicator for the response of SOM to management reduction in subalpine grasslands than the total soil C, absolute or relative POM‐C content.  相似文献

Biochar (BC) application as a soil amendment has aroused much interest and was found to considerably improve soil nutrient status and crop yields on poor, tropical soils. However, information on the effect of BC on temperate soils is still insufficient, with effects expected to differ from tropical soils. We investigated the effects of BC on soil nutrient dynamics, crop yield, and quality in a greenhouse pot experiment. We compared three agricultural soils (Planosol, Cambisol, Chernozem), and BCs of three different feedstocks (wheat straw [WS], mixed woodchips [WC], vineyard pruning [VP]) slowly pyrolyzed at 525°C, of which the latter was also pyrolyzed at 400°C. The BCs were applied at two rates (1% and 3%, which would correspond to 30 and 90 t ha–1 in the field). Three crops, namely mustard (Sinapis alba L.), barley (Hordeum vulgare L.), and red clover (Trifolium pretense L.) were grown successively within one year. The investigated soil properties included pH, electrical conductivity (EC), cation‐exchange capacity (CEC), calcium‐acetate‐lactate (CAL)–extractable P (PCAL) and K (KCAL), C, N, and nitrogen‐supplying potential (NSP). The results show a pH increase in all soils. The CEC increased only on the Planosol. The C : N ratio increased at 3% application rate. Despite improving the soil nutrient status partly, yields of the first crop (mustard) and to a lesser extent of the second crop (barley) were significantly depressed through BC application (by up to 68%); the yield of clover as third crop was not affected. Only the BC from WS maintained yields in the range of the control and even increased barley yield by 6%. The initial yield reduction was accompanied by notable decreases (Cu, Fe, Mn, Zn) and increases (Mo) in micronutrient concentrations of plant tissues while nitrogen concentrations were hardly affected. The results of the pot experiment show that despite additional mineral fertilization, short‐term growth inhibition may occur when applying BC without further treatment to temperate soils.  相似文献
