Carbohydrates are known to be important precursors in the development of roasted peanut quality. However, little is known about their genotypic variation. A better understanding of the role of carbohydrates in roasted peanut quality requires first an understanding of the genotypic variation in the soluble carbohydrate components. Ion exchange chromatography was used to isolate 20 different carbohydrate components in 52 genotypes grown in replicated trials at two locations. Inositol, glucose, fructose, sucrose, raffinose, and stachyose were quantitated, and 12 unknown peaks were evaluated on the basis of the peak height of the unknown relative to the cellobiose internal standard peak height. Peaks tentatively identified as verbascose and ajugose could not be properly integrated because of tailing. Of the 18 carbohydrates that were estimated, 9 exhibited significant variation between test environments, 5 among market types, 14 among genotypes within market types, and 11 exhibited some significant form of genotype x environment interaction. Genotypes accounted for 38-78% of the total variation for the known components, suggesting that broad-sense heritability for these components is high. The observed high genotypic variation in carbohydrate components is similar to the high genotypic variation observed for the sweetness attribute in roasted peanuts, which raises the question regarding possible interrelationships. The establishment of such interrelationships could be most beneficial to peanut breeding programs to ensure the maintenance of flavor quality in future peanut varieties. 相似文献

Distribution of microbial biomass and phospholipid fatty acids in Podzol profiles under coniferous forest 总被引:12,自引:0,他引:12
Microbial‐derived phospholipid fatty acids (PLFAs) can be used to characterize the microbial communities in soil without the need to isolate individual fungi and bacteria. They have been used to assess microbial communities of humus layers under coniferous forest, but nothing is known of their distribution in the deeper soil. To investigate the vertical distribution we sampled nine Podzol profiles on a 100‐m‐long transect in a coniferous forest and analysed for their microbial biomass and PLFA pattern to a depth of 0.4 m. The transect covered a fertility gradient from Vaccinium vitis‐idaea forest site type to Vaccinium myrtillus forest site type. The cores were divided into humus (O) and eluvial (E) layers and below that into 10‐cm sections and designated as either illuvial (B) or parent material (C), or as a combination (BC). Two measures of microbial biomass analyses were applied: substrate‐induced respiration (SIR) to determine microbial biomass C (Cmic), and the sum of the extracted microbial‐derived phospholipid fatty acids (totPLFA). The soil fertility had no effect on the results. The Cmic correlated well with totPLFA (r= 0.86). The microbial biomass decreased with increasing depth. In addition the PLFA pattern changed with increased depth as assessed with principal component analysis, indicating a change in the microbial community structure. The composition of the PLFAs in the O layer differed from that in the E layer and both differed from the upper part of the B layer and from the rest of the BC layers. The deeper parts of the B layer (BC1, BC2 and BC3) were similar to one other. The O layer had more 18:2ω6, a PLFA indicator of fungi, whereas the E layer contained relatively more of the PLFAs 16:1ω9, 18:1ω7 and cy19:0 common in gram‐negative bacteria. With increased depth the relative amount of 10Me18:0, the PLFA indicator for actinomycetes, increased. We conclude that the PLFA method is a promising discriminator between the microbial community structures of the horizons in Podzols. 相似文献

The short-term effects of N addition on CH4 oxidation were studied in two soils. Both sites are unfertilized, one has been under long-term arable rotation, the other
is a grassland that has been cut for hay for the past 125 years. The sites showed clear differences in their capacity to oxidise
CH4, the arable soil oxidised CH4 at a rate of 0.013 μg CH4 kg–1 h–1 and the grassland soil approximately an order of magnitude quicker. In both sites the addition of (NH4)2SO4 caused an immediate reduction in the rate of atmospheric CH4 oxidation approximately in inverse proportion to the amount of NH4
+ added. The addition of KNO3 caused no change in the rate of CH4 oxidation in the arable soil, but in the grassland soil after 9 days the rate of CH4 oxidation had decreased from 0.22 μg CH4 kg–1 h–1 to 0.13 μg CH4 kg–1 h–1 in soil treated with the equivalent of 192 kg N ha–1. A 15N isotopic dilution technique was used to investigate the role of nitrifiers in regulating CH4 oxidation. The arable soil showed a low rate of gross N mineralisation (0.67 mg N kg–1 day–1), but a relatively high proportion of the mineralised N was nitrified. The grassland soil had a high rate of gross N mineralisation
(18.28 mg N kg–1 day–1), but negligible nitrification activity. It is hypothesised that since there was virtually no nitrification in the grassland
soil then CH4 oxidation at this site must be methanotroph mediated.

利用根际箱在红壤上研究了有机肥对水稻根际有效磷、根际微生物和土壤酶活性的影响。试验结果表明,有机肥明显地提高水稻根际和非根际土壤真菌、放线菌和细菌的数量及土壤有效磷的含量。根际土壤各类微生物的数量大于非根际土壤,表现出明显的根际效应。施用有机肥使根际效应增加,其效应为细菌 放线菌 真菌。有机肥还明显地促进水稻根际无机磷溶解菌和有机磷分解数量以及磷酸酶和脲酶的活性,同时对水稻根际土壤磷转化速率有明显的提高,从而加速了土壤养分的转化。 相似文献

Proanthocyanidin polymers, oligomers, and the structurally related monomer (+)-catechin were labeled by incorporation of radioactive precursors in shoots of willow tree (Salix caprea L.). [1-(14)C]-Acetate and [U-(14)C]-phenylalanine precursors were fed through the cut stems or petioles of leaves. Optimization of several parameters such as the nature and origin of the plant material, leaf maturity, nature, and quantity of radioactive precursor applied and the duration of metabolism led to incorporation yields of 3.2% and to specific activities of 500 microCi/g. Detailed characterization of the products (polymerization degree, procyanidin/prodelphinidin ratio, specific activities) and purification by chromatography are reported. Some sugars bound to radiolabeled proanthocyanidin polymers were removed by enzymic treatment with a mixture of glycosidases. A radioactive purity close to 100% and specific activities suitable for bioavailability studies were obtained. 相似文献

White grape seeds were subjected to sequential supercritical fluid extraction. By increasing the polarity of the supercritical fluid using methanol as a modifier of CO(2), it was possible to fractionate the extracted compounds. Two fractions were obtained; the first, which was obtained with pure CO(2), contained mainly fatty acids, aliphatic aldehydes, and sterols. The second fraction, obtained with methanol-modified CO(2), had phenolic compounds, mainly catechin, epicatechin, and gallic acid. The fractions were bioassayed. Antimicrobial activities were checked on human pathogens, and a high degree of activity was obtained with the lipophilic fraction. Agrochemical activities on phytopathogenic fungi and activities on the etiolated wheat coleoptile bioassay were also checked. The more polar fraction was active in the latter bioassay. 相似文献

Length of sample storage can become significant in sensory studies due to panel fatigue limitations and samples needed for a reasonable expectation of finding significant differences. In roasted peanut sensory studies samples are stored between -10 and -23 degrees C to prevent or retard changes. Studies of up to 13 months' duration have examined stability and slow-rate sensory changes. Sweet taste was relatively stable, whereas bitter and tongue burn attributes increased slightly. Stale taste increased, suggesting lipid oxidation was taking place even at -23 degrees C. Painty attribute did not increase until stale was >3. An increase in fruity attribute was unexpected. With increases in fruity and stale attributes a decrease in roasted peanut was expected. However, storage at -23 degrees C seems to stabilize the roasted peanut lability when compared to storage at -10 degrees C. Fruity and stale interactions with roasted peanut and lability of roasted peanut were shown to be three separate and identifiable effects on roasted peanut. 相似文献

Three groups of anions were distinguished in order of effectiveness with respect to the dispersion and flocculation of soil samples treated with Fe(III) polycations: phosphate and fulvate citrate, oxalate, silicate and tartrate and salicylate, catechol, aspartate, lactate and acetate. This was also the order of the amount of anion adsorbed by the soil. The addition of phosphate and fulvate to soil samples with a net charge of zero lowered the zero point of charge producing particles with a net negative charge. This increased the amount of dispersible clay present from 0 to 9% by weight of soil. The sorption of phosphate and fulvate by soil samples with a net positive charge reduced the zero point of charge and caused flocculation of dispersed clay. Electrophoretic and electron microscopic studies confirmed the dispersion-flocculation phenomena.Treatments which produced dispersed clay led to increased bulk densities, plastic limits and moduli of rupture but lower porosities, water holding capacities and hydraulic conductivities. Changes in soil properties and yield response in relation to tillage and residue management treatments in an irrigated groundnut (Arachis hypogea L.)—wheat (Triticum aestivum, L.) rotation on a sandy soil (1972–1974) and on a sandy loam soil (1974–1976) were evaluated. Tillage treatments (T1 and T2) did not cause significant changes in soil properties. Incorporation of crop residues (5 t residue per ha per crop, T3) caused a substantial increase in organic C, available N, and NaHCO3 -extractable P contents in the top 15 cm of both soils. Residue management treatments (T3, T4 and T5) showed no significant effect on soil bulk density and exchangeable K. Crop yields under no-tillage (T1) and conventional tillage (control, T2) were comparable on sandy soil but on the sandy loam soil, no-tillage yielded significantly lower than conventional tillage. Compared with the control, incorporation of crop residues (T3) caused higher yields on sandy soil for groundnut and wheat by 34.1 and 47.4%, respectively, and on sandy loam by 17.1 and 7.2%, respectively. Mulching with crop residues conserved soil moisture and reduced maximum soil temperature (1.5–5.3°C in groundnut and 1.5–2.9°C in wheat) but other measured soil properties were not significantly affected. Significant yield increase due to residue mulching (T4) was observed in sandy soil but it was significantly less than when total crop residue was incorporated (T3). Increased wheat root-weight density in the top 15 cm soil with residue mulching was not reflected in grain yield. Compared with the incorporation of the total amount of crop residue (T3), incorporation of half the amount and application of the other half as mulch (T5) caused lower yields — on sandy soil for groundnut and wheat by 31.5 and 15.7%, respectively, and on sandy loam by 4.8 and 3.6%, respectively. 相似文献
