The reactivity of different lectins with crude chromogranin A (CgA) obtained from different animals, namely, cow, horse, dog, pig, and dolphin, was examined to identify lectin(s) that would be useful as coating reagent(s) in a sandwich enzyme-linked immunosorbent assay (ELISA). Of the different lectins studied, the Amaranthus caudatus lectin (ACA), which is specific for the Thomsen-Friedenreich (T)-antigen (Galβ1-3GalNAc), was found to react with the CgA from different animals by western blotting. Purified rabbit anti-bovine CgA antibody was also found to cross-react with the crude CgA preparations. On the basis of these findings, a sandwich ELISA was developed with ACA as the coating reagent and anti-bovine CgA antibody as the probing antibody. Using this method, concentration-dependent curves ranging from 0.003 μg/mL to 25 μg/mL and from 0.02 μg/mL to 25 μg/mL were obtained for bovine CgA and canine CgA, respectively. Similarly, concentration-dependent curves were obtained for the equine, swine, and dolphin crude CgA extracts. Thus, ACA is concluded to be a valuable reagent for CgA detection in crude extracts from different animal species, and for CgA isolation/purification.  相似文献

Unreduced gamete formation is significant in the evolutionary development of complex polyploidy series found in wild strawberry, genus Fragaria (Rosaceae). Also, it is important for genetics and breeding in strawberry plants to elucidate the mechanism of unreduced gamete formation. The objective of this study was to search for ploidy anomalies resulting from artificial diploid × octoploid crosses, and examine the mechanism through which these unreduced gametes were produced. Five everbearing cultivars of Fragaria vesca L. diploid (2n = 2x = 14) were crossed with pollen from six June-bearing cultivars of Fragaria × ananassa Duch., octoploid (2n = 8x = 56). A total of 3000 mature seeds, 100 from each of the 30 parental combinations were sown at 23 °C/20 °C (day/night) under artificial lighting with a 16 h day. The seedlings were transplanted to pots and grown in a greenhouse. Reproductive and morphological observations, flow cytometry analyses, chromosome counts and DNA analyses using CAPS markers were performed to identify the genetic background of the offspring. Most of the seed (79%) did not germinate or died soon after germination. Of the seedlings produced, 7% seemed to be pure F. vesca based on morphological characteristics, flow cytometry analyses and chromosome counts; 14% were pentaploids (2n = 5x = 35), 0.1% were hexaploids (2n = 6x = 42), and 0.03% (one individual) was aneuploid (2n = 8x + 2 = 58). Electrophoresis banding patterns obtained by CAPS marker analysis were heterozygotic in the 8x pollen parent but homozygotic in the aneuploid progeny. Judging from the chromosome counts and the CAPS marker analysis, the aneuploid was the result of a homozygous unreduced pollen grain (8x) crossed with an incomplete chromosome compliment from the egg. Because of the homozygosity, the unreduced male gamete must have been derived from second division restitution (SDR) in the octoploid pollen parent.  相似文献

From the genome of a Japanese field isolate of the rice blast fungus, Magnaporthe oryzae, we newly identified Inago1 and Inago2 LTR retrotransposons. Both elements were found to be Ty3/gypsy-like elements whose copies were dispersed within the genome of Magnaporthe spp. isolates infecting rice and other monocot plants. Southern hybridization patterns of nine re-isolates derived from conidia of the strain Ina168 produced after a methyl viologen treatment were not changed, indicating that the insertion pattern of Inago elements is relatively stable.  相似文献

An important strategy to conduct intentional breeding of octoploid strawberry plants is to recognize the functions of every chromosome. To do so, a methodology must be developed to distinguish chromosomes one by one. We reported the possibility of distinguishing chromosomes using light microscopy when somatic cells of octoploid strawberry plants were stained using ordinary methods with lacto-propionic orcein (LPO). However, karyotype analysis of octoploid strawberry plants required clearer chromosome images. This study obtained clearer chromosome images of octoploid Fragaria × ananassa and F. chiloensis plants. Three staining methods were examined: 60% acetic acid (AA) alone, 1.5% LPO alone, and two-step treatments with 60% AA and 1.5% LPO. Collected root tips of the plants were placed in 2 mM 8-hydroxyquinoline solution for 1 h and were subsequently stored at 4 °C for 15 h. The samples were then fixed in a 3:1 absolute alcohol: glacial acetic acid solution for 40 min, followed by mixture with 1N HCl solutions at room temperature for 2 h and then at 60 °C for 10 min. For separate staining using 60% AA and 1.5% LPO, the root tip was expelled on the glass slide with a drop of each solution for a few minutes to stain the chromosomes. For the two-step staining method, the samples stained with 60% AA were frozen at −80 °C for at least 5 min. The cover slip was removed using a razor blade. Subsequently, the specimens were air-dried and stained with the 1.5% LPO for 3 min. Digital images of chromosomes were obtained using light microscopy. Samples of the two-step staining method produced the clearest chromosome images in both F. × ananassa and F. chiloensis. Furthermore, the greatest color difference between the chromosomes and the cytoplasm was obtained from images of the two-step staining method among the three staining methods. These results demonstrate that the two-step staining method is useful for chromosome counting and karyotype analysis in strawberry plants.  相似文献

Food hoarding is one of the determinant factors of foraging behavior and benefits the hoarder. Two species of Apodemus mice, A. speciosus Temminck and A. argenteus Temminck, hoard acorns for later use. In 1988 and 1989, I conducted laboratory experiments to reveal the effects of food hoarding in the nest on the foraging behavior of these mice. I released a mouse into an indoor arena (2.8 × 2.8m) and recorded the foraging behavior of four female A. speciosus and four female A. argenteus mice in sessions with and without food hoarding in the nest box. Nest hoarding did not affect the time that a mouse allotted to excursions or the number of peanuts that a mouse ate during the night, but decreased the number of peanuts handled and the intensity of food handling (the number of handled peanuts per visit) at the food stations. The decrease in food handling outside the nest box, which seemed to be beneficial to the survival and reproduction of the hoarder in the field, was more apparent in A. speciosus than in A. argenteus. The different behavioral responses in foraging behavior to nest hoarding between two Apodemus mice may be related to their different microhabitat preferences.  相似文献

Appressorium differentiation, one of the most important steps in pathogenesis by the rice blast fungus, Pyricularia oryzae, is strongly coordinated with the cell cycle. In this study, we identified an ortholog gene of 53BP1, which encodes a signal transducer protein that participates in G2-M cell cycle checkpoint in higher eukaryotes, in the genome of P. oryzae and characterized the phenotype of deletion and overexpression mutants. Deletion mutants showed no significant deficiency in vegetative growth compared to wild-type and complemented strains, even on the media containing DNA-damaging agents. However, these null mutants had abnormal appressoria and formed more appressoria per conidium than in the wild type and were unable to penetrate the epidermis of rice leaves. eGFP-fused Mop53BP1 and qRT-PCR analyses revealed that Mop53BP1 is expressed during the first hours of appressorium formation. In addition, in overexpression mutants, Mop53BP1 localized to nuclei during all stages of appressorium maturation and penetration, and the mutants were resistant to the microtubule inhibitor benomyl, suggesting that Mop53BP1 is nuclear protein and may have some role related to microtubules.  相似文献

We estimated the amount of nitrogen (N) remobilized from 1-year-old leaves at various positions in the crowns of mature Quercus glauca Thunb. ex Murray trees and related this to the production of new shoots. Leaf N concentration on an area basis (Na) and total N (Nt= Na x lamina area of all leaves on a shoot) were related to photosynthetic photon flux (PPF) on the leaves of current-year and 1-year-old shoots. When new shoots (S02 shoots; flushed in 2002) flushed, only a portion of the leaves on the previous year's shoots (S01 shoots; flushed in 2001) were shed. After the S02 shoots flushed, S01 shoots were defined as 1-year-old shoots (S01* shoots). Both Na and Nt were positively correlated with PPF for S01 shoots, but not for S01* shoots. The fraction of remobilized N (% of the maximum Na in S01 leaves) from remaining leaves was 5-35%, with the fraction size being positively correlated with the number of S02 shoots on an S01* shoot (new shoot number). However, the mean fraction of remobilized N from fallen leaves was 45% and was unrelated to new shoot number. The total amount of N remobilized from both fallen and remaining leaves was 1-20 mg per S01* shoot. Total remobilized N was positively correlated with new shoot number. There was a statistically significant positive relationship between the light-saturated net photosynthetic rate on a leaf area basis (Amax) and Na for both S01* and S02 leaves. However, when we compared leaves with similar Na, Amax of S01* leaves was only half that of S02 leaves, indicating that 1-year-old leaves had lower instantaneous N-use efficiency (Amax per unit Na) than current-year leaves. Ratios of chlorophyll a:b and Rubisco:chlorophyll were lower in S01* leaves than in S02 leaves, indicating that 1-year-old leaves were acclimatized to lower light environments. Thus, in Q. glauca, the N allocation theory (i.e., that N is distributed according to local PPF) applied only to the current-year shoots. Although the amount of foliar N in 1-year-old shoots was not strongly affected by the PPF on 1-year-old leaves, it was affected by interactions with current-year shoots.  相似文献

A total of 29 SPF Large White prepuberal gilts (mean age 152 days at treatment) were examined for estrous and ovulatory responses after PG 600 treatment. After treatment, 85.2% of the gilts showed standing estrus within 6 days. Whereas the treatment-to-estrus interval and duration were 3.7 and 1.9 days respectively. As ovulation occurred on Day 5 to 6, appropriate timing of artificial insemination would be about 4 days after treatment. Fertility of gilts revealed to be excellent, giving rise to a high percentage of normal embryos, 85.3%. Meanwhile, development and growth of fetuses were mostly normal. Other reproductive performances recorded were: mean litter size 6.8; mean birth weight 1.26 kg; weaning-to-return estrus interval 5 to 8 days. In conclusion, PG 600 was found to be useful in inducing fertile estrus in prepuberal gilts, a result which will be of interest for commercial pig farms.  相似文献
