Bacterial blight of grapevine is caused by a slow‐growing bacterium Xylophilus ampelinus. It has been suspected to occur in Slovenia on the basis of visual observation of characteristic symptoms in the 1960s. In the present study, symptoms were recorded in an infected vineyard during three consecutive years (2002/2004). Samples from this vineyard were tested by nested‐PCR and isolation of bacteria on media was attempted. In the first year, angular lesions on leaves were highly expressed and an isolate morphologically similar to X. ampelinus was obtained from one sample. It was purified and identified as X. ampelinus using biochemical and nutritional tests, fatty acid analysis, immuno‐fluorescence, nested PCR and partial sequencing of the 16S rRNA gene. The 16S rDNA sequence showed 99–100% homology to known sequences of X. ampelinus strains, including the type strain. Pathogenicity of the isolate was confirmed in tissue‐cultured and potted grapevine plants. In the following two years, symptoms of bacterial blight were only faintly expressed. Using isolation on media and nested‐PCR, 23 and 17 extracts prepared from 10 and 8 grapevines, respectively, were analysed. Four simple and rapid field tests for the serodiagnosis of brucellosis in cattle, goat, sheep and swine were developed. The performance of the assays was investigated using serum samples collected in Portugal from animals originating from herds with a defined sanitary status with respect to the presence of brucellosis. The sensitivity calculated for the bovine, caprine, ovine and swine Brucella lateral flow assays based on results obtained for samples collected from animals with culture confirmed brucellosis was 90%, 100%, 90% and 73%, respectively. None of the samples from animals from herds free of brucellosis reacted in the flow assays indicating a high specificity. However, as expected, some degree of reactivity was observed when testing selected serum samples that reacted non-specific in reference tests for brucellosis. 相似文献

Although ordinary chondrite (OC) meteorites dominate observed falls, the identification of near-Earth and main-belt asteroid sources has remained elusive. Telescopic measurements of 35 near-Earth asteroids ( approximately3 kilometers in diameter) revealed six that have visible wavelength spectra similar to laboratory spectra of OC meteorites. Near-Earth asteroids were found to have spectral properties that span the range between the previously separated domains of OC meteorites and the most common (S class) asteroids, suggesting a link. This range of spectral properties could arise through a diversity of mineralogies and regolith particle sizes, as well as through a time-dependent surface weathering process. 相似文献

Floodplains along large European rivers are diffusely polluted with heavy metals due to emissions in the past. Because of low mobility of heavy metals in floodplain soils and improvements of water quality, these pollutants will remain in place, and can gradually become covered with less contaminated sediments. Bioturbators, especially earthworms, can play an important role in the mixing and surfacing of contaminated substrate. Surfaced substrate can be redistributed by recurrent flooding events, even to areas outside the floodplain. The question remained to what extent bioturbation by small mammals contributes to the redistribution of heavy metals from river sediments in floodplains. Extensive fieldwork on bioturbators such as voles, moles and earthworms and their distribution patterns, as well as on sediment deposition and bioturbation, was conducted at the ‘Afferdensche en Deestsche Waarden’ floodplain over the years 2001–2003. Field data were combined with data of experiments in field enclosures and substrate columns to calculate the amounts of sediment and heavy metals (Zn, Cu, Pb and Cd) redistributed during the floods as well as on an annual basis. Moles and voles surfaced considerable amounts of substrate and heavy metals, but not as much as earthworms which contribute a substantial proportion of the total deposition and redistribution during floods. Although the impact of moles and voles on the redistribution during floods was only locally important, on an annual basis the bioturbation activity of especially moles in floodplains cannot be neglected. The annual amounts of substrate and heavy metals surfaced by all investigated bioturbators were even larger than the total amounts of substrate and heavy metals deposited during floods. 相似文献

Brucella melitensis biovar 1 was isolated from bovine milk samples from a herd in central Kenya, and Brucella abortus biovar 3 was isolated from aborted fetus materials and vaginal discharge fluids from cattle in central and eastern provinces
of Kenya. All infections including those with B. melitensis were in cattle with reproductive problems kept in mixed herds indicating that cross infection occurs from small ruminants.
Multiple-locus variable-number tandem repeat analysis genotyping revealed a close molecular homology of the B. melitensis isolates with an isolate from Israel and a close homology of the B. abortus isolates with an isolate from Uganda indicating that these genotypes have a wide geographic distribution. Infection of cattle

Bacterial clearance, L-selectin adhesion receptor expression and neutrophil recruitment during experimentally induced Escherichia coli mastitis were investigated. Acute mastitis was induced by injection of 10(4) cfu E. coli into both left quarters of 12 clinically healthy lactating cows, 2-6 weeks after parturition. Half the cows were treated intravenously 10 h after infection, and subcutaneously 30 h after infection with enrofloxacin. In both groups, bacterial count, milk serum albumin, milk somatic cell count, circulating leukocyte count and L-selectin expression on neutrophils were determined. Both groups responded to challenge with udder inflammation and fever. Treatment with enrofloxacin affected the elimination rate of bacteria at hours +14, +18, +24, +48, and +72 after challenge, but not the bacteriological cure rate (five out of six for treated and three out of six for nontreated cows). The drop in L-selectin expression found following challenge did not differ between groups throughout the study. No effect of treatment was found on SCC. Based on these findings, it may be concluded that when treatment is administered 10 h after infection, the number of bacteria in milk is not correlated with L-selectin expression on circulating PMN during experimentally induced E. coli mastitis. The initial bacterial load probably dictates the extent of the decrease in L-selectin expression and milk somatic cells. 相似文献

Cows affected with bovine spongiform encephalopathy (BSE) display chronic neurological signs consisting of behavioural changes, abnormalities of posture and movement, and/or hyperaesthesia. At present, there are no laboratory test available to diagnose BSE in the live animal. In this article, we describe the post-mortem diagnostic examination of brains from BSE-suspected cattle as currently performed at ID-Lelystad. The routine laboratory diagnosis of BSE consists of histopathological examination of the brain and detection of the modified prion protein, PrP(BSE), in brain tissue. These tests, however, have the disadvantage of being laborious and time consuming, so that results are available only after several days. Recently, at ID-Lelystad a new post-mortem test has been developed that enables screening of larger volumes of brain samples for PrP(BSE) within 1 day. This BSE test is especially suited for slaughterline monitoring. A preliminary validation study has shown that both sensitivity and specificity are 100% compared to the gold diagnostic standard of histopathology. 相似文献

The ‘developmental origins of health and disease’ hypothesis proposes not only that we are what we eat, but also that we could be what our parents ate. Here, we aimed to improve health and performance of young piglets via maternal diets based on the hypothesis that maternal nutritional interventions change metabolic programming in piglets, reflected by differential gene expression early in life. Therefore, sows were fed either a regular diet, based on barley, wheat and wheat by‐products, sugar beet pulp, palm oil and oilseed meal, or a high‐fat (HF) diet consisting of the regular diet supplemented with an additional amount of 3.5% soybean oil and 1% fish oil at the expense of palm oil and wheat. Performance results, physiological parameters and gene expression in liver of piglets and blood of piglets and sows at day 7 after farrowing from both diet groups were compared. The HF diet tended to enhance growth rate of the offspring in the first week of life. No significant differences in gene expression in liver tissue and blood could be detected between the two groups, neither with whole‐genome microarray analysis, nor with gene specific qPCR analysis. In this study, the feeding of a high‐fat diet with increased amounts of polyunsaturated fatty acid (PUFA) to gestating sows under practical farm settings did not induce significant changes in gene expression in sows and offspring. 相似文献
