Low supply of the nutrients nitrogen (N) and phosphorus (P) limit plant growth and spreading, and increase the plant-microbial nutrient competition in subarctic and arctic regions. We investigated the mycorrhizal community structure of a polar shrub willow (Salix polaris) and the microbial turnover in its rhizosphere to explore the adaptation of a mycorrhizal plant in the subarctic tundra. The ectomycorrhizal colonisation ranged from 35 to 64% of the fine root tips and decreased with an increasing soil C/N ratio. In total, 16 ectomycorrhizal morphotypes were found under S. polaris (eight to 13 morphotypes per site, five morphotypes at all four sites). Cenococcum sp. was the most common EM fungus (32% of the ectomycorrhizal fine roots). The abundance of Cenococcum sp. increased with an increasing organic matter content and N/P ratio in the soil. Arbuscular mycorrhizal colonisation of S. polaris was absent or less than 1% of the fine root length. Microbial biomass P accounted for 21–75% of the organic soil P and 6–49% of the total soil P. Microbial biomass P, alkaline and acid phosphatase activities in the rhizosphere increased with increasing soil N concentration. We conclude that a higher N supply decreases the diversity in the mycorrhizal community on polar willows and increases the role of P turnover from the soil microbial biomass for the nutrient supply.  相似文献

The effect of 2,4-dichlorophenoxyacetic acid (2,4-D), 4-chloro-2-methylphenoxyacetic acid (MCPA), and their derivatives: phenol, 2,4-dichlorophenol (2,4-DCP), 2,4-dimethylphenol (2,4-DMP), and catechol on the activity of acetylcholinesterase (AChE, EC 3.1.1.7) in human erythrocytes was studied. Phenol, MCPA, and 2,4-DMP did not significantly change AChE activity in human erythrocytes (in vitro). Decrease of AChE activity was observed under the highest applied dose of 2,4-D—500 and 1000 ppm. Decrease of AChE activity exposed to 2,4-DCP and catechol was noted and depended on the doses of applied compounds. The relationship between activities and substrate concentrations (curves) was analyzed for reactions of acetylcholinesterase. Catalytic constants Km and Vmax were calculated from the Michaelis curve. Statistically significant decrease of Vmax and Km was observed in the activity of AChE incubated with 2,4-DCP and catechol, revealing mixed inhibition type of AChE inhibition (this compound may affect not only on enzyme but also on complex ES as well). 2,4-D decreases Vmax but do not change Km value, what reveals non-competitive type of AChE inhibition by this compounds. Non-competitive inhibition does not depend on the substrate concentrations but only on the inhibitor concentration and its Ki value, characterizes the affinity of inhibitors towards enzyme. In conclusion, changes of AChE activity upon 2,4-D, 2,4-DCP, and catechol are the consequences of direct interactions between compounds and the enzyme and indirect via membrane modification and increase of Reactive Oxygen Species.  相似文献

Broiler chicken (Gallus gallus) manure, a rich source of plant nutrients, is generated in large quantities in southeastern USA where many row crops, such as corn (Zea mays L.), are also extensively grown. However, the use of broiler manure as an economical alternative source of nutrients for corn production has not been extensively explored in this region. This study was conducted to examine the use of broiler litter as a source of nutrients for corn production, as influenced by tillage and litter rate, and any residual effects following application. In addition, the consequence of litter application to soil test nutrient levels, particularly P, Zn and Cu, was explored. The treatments consisted of two rates of broiler litter application, 11 and 22 Mg ha−1 on a wet weight basis, and one rate of chemical fertilizer applied under no-till and conventional tillage systems. Treatments were replicated three times in a randomized complete block design. Corn was grown with broiler litter and inorganic fertilizer applied to the same plots each year from 1998 to 2001. In 2002 and 2003, corn was planted no-till, but only N fertilizer was applied in order to make use of other residual litter nutrients. Soil samples were taken yearly in the spring prior to litter application and 4 years after the cessation of litter application to evaluate the status of the residual nutrients in soil. Two years out of the 4-year experiment, broiler litter application produced significantly greater corn grain yield than equivalent chemical fertilizer application and produced similar grain yield in the other 2 years. Corn grain yield was significantly greater under no-till in 1999, but significantly greater under conventional-till in 2000, and no difference between the two tillage systems were observed in 1998 and 2001. With 4 years of litter application, Mehlich-3 P increased from an initial 18 mg kg−1 to 156 mg kg−1 with 11 Mg ha−1 litter and to 257 mg kg−1 with 22 Mg ha−1 litter. For every 6 kg ha−1 of P applied in poultry litter Mehlich-3 P was increased by 1 mg kg−1. Modest increases in Mehlich-3 Cu and Zn did not result in phytotoxic levels. This study indicated that an optimum rate of broiler litter as a primary fertilizer at 11 Mg ha−1 applied in 4 consecutive years on a silt loam soil produced corn grain yields similar to chemical fertilizer under both no-till and conventional tillage systems and kept soil test P, Cu and Zn levels below values considered to be harmful to surface water quality or the crop.  相似文献

Abstract

The simultaneous incorporation of heavy metals into the soil is still a matter of great concern. Interaction (competitive sorption) between these metals and the soil solid phase may result in a deterioration of soil quality which relies basically on amounts of alkaline cations saturating soils sorptive complex. Results of this study indicate that Pb, Cu, C d, and Zn have induced solution pH decreases which were more intensive at highest metal loading rates. Partition parameters (Kd)-based sequences showed that Pb and Cu were more competitive than Cd and Zn and the overall selectivity sequence followed: Pb > Cu > Cd > Zn. Metal loadings and their competitive sorption have led to a strengthened displacement of alkaline cations (i.e. Ca2+, Mg2+, K+, Na+), especially of Ca2+ as a factor “stabilizing” soil sorptive complex. Such metals impact jointly with soils acidification are of great environmental concern since tremendous amounts of alkaline cations (especially Ca2+) may be potentially leached out, irrespective of the degree of soil contamination, as evidenced in the current study. High and positive ΔG values implied that the studied soils were characterized by generally low concentrations of exchangeable potassium which required high energy to get displaced (desorbed). Further studies on heavy metal uncontaminated or contaminated areas should be undertaken to provide with data which should be used for predictions on changes related to soil buffering capacity as impacted by heavy metal inputs.  相似文献

The aim of this investigation was to compare the liver and aorta changes in rats fed cholesterol-containing diets and the possible improvement when diets would be supplemented with frequently used raw vegetables. The phenolic compounds of three vegetables in methanol-water (1:1) fraction were characterized using electrospray ionization mass spectra (ESI-MS). Results showed that the content of polyphenols, flavonoids, quercetin, flavanols, tannins, and ascorbic acid varied for garlic and white and red onions ranging from 6.68 to 18.08 mg GAE/g DW, 490.4-701.0 μg CE/g DW, 281.2-1100.0 μg, 32.40-41.30 μg CE/g DW, 2.88-3.12 mg CE/g DW, 1.87-2.33 mg AA/g DW, 1388.2-1442.3 μg CGE/g DW, respectively. The radical scavenging capacities (μM TE/g DW) for the same investigated vegetables for ABTS, FRAP, CUPRAC, and DPPH assays ranged from 48.78 to 92.42, 9.41-28.56, 3.06-10.41, and 6.49-23.42, respectively. Good correlations were observed between the phenolic contents and the radical scavenging capacities of the vegetables. The interaction between BSA and quercetin, BSA and garlic and onions extracts was measured by 3-dimensional fluorescence (3D-FL) and Fourier transform infrared (FT-IR) spectroscopy. The highest polyphenol content was found in methanol/water fraction of onions and garlic; therefore, for the investigation of in vitro interactions with BSA only polyphenols of this fraction were used. For in vivo studies, 30 male Wistar rats were divided into 5 groups each of 6 and named Control, Chol, Chol/Garlic, Chol/OnionRed, and Chol/OnionWhite. During 6 weeks, the rats of all 5 groups were fed a basal diet (BD). The rats of the Control group were fed the BD only. The BD of the Chol group was supplemented with 10 g/kg of nonoxidized cholesterol (NOC). Each of the other three groups was supplemented with 10 g/kg of NOC and 500 mg of raw fresh garlic, 500 mg of raw fresh red onion, and 500 mg of raw fresh white onion on 1 kg of body weight for Chol/Garlic, Chol/OnionRed, and Chol/OnionWhite diet groups, respectively. In order to detect the changes in the liver and aorta, a histological procedure was applied, and the liver enzymes were determined and compared. It was found that the main changes vs the Control group were in the liver of rats fed the cholesterol-containing diet without vegetable supplementation. Significantly less histological changes in the liver and lower level of liver enzymes vs those of the Chol group were detected in rats of the Chol/Garlic group (P < 0.05). The interaction between the polyphenol extract of garlic and BSA in vitro showed its strong ability comparable with that of quercetin to quench the intrinsic fluorescence of BSA. In conclusion, all studied vegetables showed protective effects, but raw garlic supplemented with cholesterol-containing diets significantly prevented the aorta and liver damages of rats.  相似文献

Tuber yield, together with tuber number and size are the basic agronomic and breeding traits in Jerusalem artichoke and can be significantly affected by environmental factors. We report the results of a long term trial on the performance of 20 Jerusalem artichoke cultivars. The random model for means with restricted maximum likelihood (REML) procedure was used to estimate the overall effects of the genotype, environment and genotype by environment interaction on traits. The partial least square regression (PLSR) model was used for modeling genotype by environment interaction variance components with a set of available correlated environmental variables. The REML variance component estimates model revealed that tuber number and yield are more dependent on GE interaction which allowed identification of best genotypes for specific environments. The PLSR model revealed that the most important climatic variables for optimal emergence, canopy development, high tuber number and yield are adequate soil and air temperatures in April. For larger tuber mass, precipitation variables and even distribution of rainfall were the most important factor, together with soil and air temperature in June when tuber growth is initiated. The knowledge obtained in this study is valuable for the identification and understanding of key environmental factors that contribute to the performance of Jerusalem artichoke.  相似文献

Anthocyanins occur in potato tuber skin and flesh, sprouts, leaves, stems and flowers. The goal of this study was to identify genomic regions and candidate gene alleles key for accumulation of anthocyanins in potato corolla in various quantities. QTL analyses were performed in two mapping populations segregating for flower colour intensity and candidate genes were identified on the basis of function and location (chalcone isomerase, chi; chalcone synthase, chs) or location (RNA-dependent RNA polymerase 1, RDR1). We detected three and four QTL affecting the violet flower colour intensity using the two mapping populations, respectively. In both populations a locus F, necessary for violet flower colour, segregated and we used different approaches to differentiate the qualitative effect of this locus and to detect the genetic factors affecting the quantitative flower colour intensity. The strongest QTL and the only one common for the two mapping populations was located on chromosome V. The role of all three candidate genes, chi, chs and RDR1, in control of flower colour intensity is supported to different extents by the performed genetic analyses. The most important QTL on chromosome V is most likely in the same position as the QTL for anthocyanin tuber flesh coloration described previously, which indicates that the natural variation in some biosynthetic and/or regulatory genes may influence anthocyanin levels in multiple tissues.  相似文献

The study was conducted in a small (7.4 ha) peatland system with a humic lake concentrically surrounded by Sphagnum mat (Caricetum limosae) and Sphagnum bog (Ledo-Sphagnetum) in the Mazurian Lakeland in North-Eastern Poland. The peatland was situated in a forested catchment with a total area of 16.25 ha, and was influenced by surface drainage, artificially designed for forestry purposes. The content of zinc (Zn), cadmium (Cd), lead (Pb), copper (Cu) and hydrogen (H) ions was analysed in waters from precipitation, humic lake, mat, bog and surface outflow. The yearly budget of these ions, expressed as difference between atmospheric inflow and surface outflow, for the whole catchment was presented. The drainage of the peatland system over three years was accompanied by an increase in pH of lake, mat and bog waters. The whole catchment retention of H+ and trace metals studied decreased yearly in absolute values under these conditions. The peatland system began to leach Pb into surface waters and lost its ability to intercept Cd. These effects could be caused by intensity of outflow of water and sulphates and lowering of water storage. However, the potential to retain Zn and Cu ions was preserved.  相似文献

The characteristically clustered occurrence and low level of mobility of Heterodera schachtii and Rhizoctonia solani in the soil and the induction of stress symptoms in the sugar beet canopy make them ideal targets for site-specific arrangements with precision agriculture tools. A field site infested with H. schachtii and R. solani was investigated in 2009 with near-range and aerial hyperspectral sensors during the growing season. At 31 sample points ground truth data for incidence and severity of the two organisms were collected and geo-referenced. Spectral vegetation indices computed from reflectance measurements obtained from two flight campaigns (AISA, 17th of June; HyMap, 28th of August) and the near-range spectroradiometers were significantly correlated (P < 0.01) with symptoms caused by the nematode or Rhizoctonia crown and root rot. A supervised classification with Spectral Angle Mapper of leaf symptoms induced by the organisms resulted in a classification accuracy of 72 and 64% for the AISA and HyMap data, respectively. The results demonstrated that remote sensing in combination with geographic information system technologies can be used effectively for the detection and mapping of symptoms caused by beet cyst nematode and Rhizoctonia crown and root rot.  相似文献

The aim was to evaluate the effect of sourdough and dietary fiber addition, as well as partially baked frozen (PBF) technology, on biological value of proteins, lipid profile, and some minerals content in selected tissues of rats.In all breads, the limited amino acid was lysine. Proteins of PBF conventional and PBF breads with an addition of dietary fiber (PBF DF breads) had significantly higher Chemical score and Essential amino acid index in comparison to proteins of Direct bread (P < 0.05). Protein digestibility corrected amino acids score was significantly lower in Direct DF and PBF improved breads (P < 0.05).Addition of fiber to Direct and PBF bread decreased significantly total digestibility of protein compared to all groups (P < 0.05).Concentration of calcium was significantly lower in liver of rats fed with Direct sourdough and PBF improved breads in comparison to rodents fed with Direct bread. The concentration of iron was significantly higher in liver of animals fed with Direct bread. The highest level of calcium in femoral bones of rats was shown in groups fed with PBF conventional bread. The lowest level of iron was measured in femoral bone of rats fed with PBF sourdough bread in comparison to the rest of the experimental groups.  相似文献
