Chloroplast division involves plastid-dividing, dynamin, and FtsZ (PDF) rings. We isolated intact supertwisted (or spiral) and circular PDF machineries from chloroplasts of the red alga Cyanidioschyzon merolae. After individual intact PDF machineries were stretched to four times their original lengths with optical tweezers, they spontaneously returned to their original sizes. Dynamin-released PDF machineries did not retain the spiral structure and could not be stretched. Thus, dynamin may generate the motive force for contraction by filament sliding in dividing chloroplasts, in addition to pinching-off the membranes. 相似文献

Five compounds oxidizing canine erythrocytes were isolated from an aqueous ethanol garlic extract by silica gel column chromatography and preparative thin-layer chromatography. On the basis of nuclear magnetic resonance, infrared spectroscopy, and mass spectrometry, they were identified as three known compounds: bis-2-propenyl trisulfide (1), bis-2-propenyl tetrasulfide (2), and bis-2-propenyl pentasulfide (3) as well as two novel compounds, bis-2-propenyl thiosulfonate (4) and trans-sulfuric acid allyl ester 3-allylsulfanyl-allyl ester (5). A mixture of compounds 1-3 and compounds 4 and 5 induced methemoglobin formation in canine erythrocyte suspension in vitro resulting in the oxidation of canine erythrocytes. These groups of characteristic organosulfur compounds contained in garlic probably contribute to oxidations in blood. The constituents of garlic have the potential to oxidize erythrocytes and hemoglobin, suggesting that foods containing quantities of garlic should be avoided for feeding dogs. 相似文献

Recent progress in studies concerning behavior of, and management for, grazing cattle are reviewed. Since 1950, much study has been conducted on ‘How’, ‘When’, ‘Where’ and ‘How long’ regarding grazing. After the 1980s, grazing ecology introduced the concept of hierarchy at different spatial and temporal scales, and since then grazing behavior has been investigated on the foraging hierarchy of large grazing herbivores: bite, feeding station (FS), patch, feeding site, camp and home range. From the sequence of activities, FS is grouped within a feeding patch, and movement of grazing cattle has been studied between FSs, feeding patches, feeding sites and between camps. Grazing behavior and production relates closley with defoliation, and grazing management should control both grazing behavior and vegetation according to three rules: planning, operational and adaptation rules. Planning rules relate the stocking rate of cattle; operational rules relate to defoliation; and adaptation rules vary with regional situations. Recent studies on grazing have been carried out in the fields of animal diversity and welfare. Future studies in this field should be conducted on the ecology, neurophysiology and psychology of grazing. Nonlinear analysis will also be significant in this field. Grazing cattle production should also utilize supplementation by roughage and/or grains. 相似文献

Vacuolar degeneration of the thyroid follicular epithelium was observed in two untreated female cynomolgus monkeys assigned to control groups. In light microscopy, large vacuoles containing a homogenous substance occupied the basal region of the epithelium, and the nuclei had shifted toward the apical region. The vacuoles showed negative reactions to PAS and thyroglobulin. Electron microscopic observation revealed dilatation of the rough endoplasmic reticulum corresponding to the vacuoles. The plasma TSH, T3 and T4 levels determined for the samples kept frozen were within the normal ranges, suggesting that the thyroid function was kept intact. 相似文献

Spina bifida aperta (SBA) is a congenital malformation of the spinal cord with complications such as spinal ataxia and bowel and bladder dysfunction. We have developed a chick model with surgery-induced SBA that shows spinal ataxia after hatching. In the present study, motor neurons in the early stages in chicks with and without SBA were observed by immunohistochemical staining with a monoclonal antibody against Islet-1, a motor neuron marker. Delay in migration and maturation of motor neurons was observed in SBA. Although the final numbers of Islet-1-positive neurons in these two groups were not different, a defect in the production and elimination of excess motor neurons in the early developmental stages in the SBA group may be involved in the pathological mechanism of the motor complications of this disease. 相似文献

In the past 25 years, there has been only one case of Japanese encephalitis in horses in Japan. We determined the full genome sequence of the Japanese encephalitis virus (JEV) strain JEV/eq/Tottori/2003 isolated from an afflicted horse and also analyzed its virulence in mice. The sequence analysis showed that the genome of JEV/eq/Tottori/2003 is similar to that of genotype I, a dominant genotype of JEV presently circulating in Japan. Its neurovirulence, but not neuroinvasiveness, was still as high as it was for genotype III, thus indicating the necessity for continuation of a vaccination program of horses against JEV. 相似文献

A genomic library of Xanthomonas oryzae pv. oryzae (X. o. pv. oryzae) T7174 was screened for 4-methylumbelliferyl β-D-glucoside-hydrolyzing (MUGase) activity. In subcloning of one of the MUGase-positive clones, an approximately 4.2-kb SacI-SphI fragment conferred not only MUGase activity but also 4-methylumbelliferyl β-D-cellobioside-hydrolyzing (MUCase) activity. Sequence analysis showed that the fragment contained an ORF of 2951 bp. The conceptual
ORF product was significantly homologous with 1,4-β-D-glucan glucohydrolase D (CELD) from Pseudomonas fluorescens subsp. cellulosa, and was named CELDXo. Cell fractionation experiments suggested that CELDXo is localized in the cell-envelope fraction. We constructed a CELDXo-deficient mutant (74ΔCELD) from X. o. pv. oryzae. Little MUCase activity was detected in the cell-envelope fraction prepared from the mutant. The mutant 74ΔCELD did not grow
in synthetic medium containing cellobiose as the sole sugar source. On the other hand, growth in rice leaves and pathogenicity
of the mutant and the parental strain did not differ. These results suggested that CELDXo is involved in cellobiose utilization of X. o. pv. oryzae but that the gene is not required for bacterial growth in rice leaves.

We studied in vivo fiber digestibility and fermentation parameters such as volatile fatty acid concentration and in vitro degradation in each segment of hindgut of horses fed timothy hay or silage made from the same sward. Six Thoroughbred horses were fed timothy hay or silage in equal amounts (1.6% of bodyweight per day, bodyweight mean 572 kg) every 3 h per day, then slaughtered. There were no differences between hay and silage in the concentration of the total volatile fatty acids, the apparent digestibility of dry matter, organic matter, and fiber, and in vitro neutral detergent fiber (NDF) disappearance rate in each segment. The total volatile fatty acid concentration and the apparent digestibility of dry and organic matter and fiber differed ( P < 0.01) depending on the digestive segments. Both for hay and silage, the concentration of total volatile fatty acids in the digesta liquid phase largely increased ( P < 0.01) from the cecum to the right ventral colon, and kept a constant value from the right ventral colon to the right dorsal colon, and then decreased from the right dorsal colon to the small colon. For dry and organic matter and fiber components, the apparent digestibility changed in the same manner from former to hinder segments of the hindgut. Regardless of diet, dry and organic matter and fiber components showed lower values in the cecum and the right ventral colon and increased ( P < 0.01) largely from the right ventral colon to the left dorsal colon, then kept constant values in segments to the rear of left dorsal colon. There were no differences in in vitro NDF disappearance among cecum, ventral colon and dorsal colon. 相似文献
