DNA measurement by image cytometry, and a detailed immunohistochemical study using monoclonal antibodies directed against different human cytokeratin types, muscle-specific actin, vimentin and S100 protein were carried out on normal canine mammary tissue (n =4), benign canine mammary mixed tumours (n =20) and malignant canine mammary mixed tumours (n =13). The results showed that ductal and alveolar luminal cells in normal and neoplastic tissue were immunoreactive with CAM5.2 and AE1/AE3 antibodies recognizing human keratins.Basal/myoepithelial cells were clearly differentiated from ductal and alveolar epithelial cells, since the latter only expressed cytokeratins, whereas the former also expressed vimentin and muscle-specific actin. This immunohistochemical study showed that there is loss of expression of muscle-specific actin and cytokeratins in areas of myoepithelial proliferation, and enhanced expression of vimentin and S100 protein in proliferative areas with osseous and/or chondroid metaplasia. The ploidy studies revealed that 20% (4/20) of benign and 54% (7/13) of malignant mixed tumours of canine mammary gland were aneuploid and that the epithelial and myoepithelial components of the mixed tumours had identical DNA content.Our results reinforce the role of myoepithelial cells in mesenchymal metaplasia in mixed mammary tumours and suggest the possibility of a common origin of both components from a totipotential stem cell with capacity for divergent differentiation.  相似文献

The effects of squid flesh, two pelleted diets containing 41% and 53% protein, and starvation on the ammonia-N efflux rate and the biochemical composition of pink shrimp, Penaeus paulensis (Perez-Farfante), were investigated. A transient increase in the ammonia-N efflux rates of shrimp subjected to starvation was observed. Ammonia-N efflux rates of shrimp fed the diet with 41% protein were lower than in those fed the diet with 53% protein or squid flesh. Ammonia-N efflux rates were inversely related to body weight, except during starvation, which affected mainly small individuals. Increased ammonia-N excretion rates increased immediately after the ingestion of high-protein diets. Such rates were also found to be higher during light periods (cf. dark periods), possibly reflecting differences in activity levels. Dietary-related alterations in the blood, muscle and midgut gland constituents were also detected. In general terms, starvation tended to reduce the levels of such constituents, whilst pelleted diets promoted increases in lipid and carbohydrate concentrations.  相似文献

In the future, climate models predict an increase in global surface temperature and during winter a changing of precipitation from less snowfall to more raining. Without protective snow cover, freezing can be more intensive and can enter noticeably deeper into the soil with effects on C cycling and soil organic matter (SOM) dynamics. We removed the natural snow cover in a Norway spruce forest in the Fichtelgebirge Mts. during winter from late December 2005 until middle of February 2006 on three replicate plots. Hence, we induced soil frost to 15 cm depth (at a depth of 5 cm below surface up to –5°C) from January to April 2006, while the snow‐covered control plots never reached temperatures < 0°C. Quantity and quality of SOM was followed by total organic C and biomarker analysis. While soil frost did not influence total organic‐C and lignin concentrations, the decomposition of vanillyl monomers (Ac/Ad)V and the microbial‐sugar concentrations decreased at the end of the frost period, these results confirm reduced SOM mineralization under frost. Soil microbial biomass was not affected by the frost event or recovered more quickly than the accumulation of microbial residues such as microbial sugars directly after the experiment. However, in the subsequent autumn, soil microbial biomass was significantly higher at the snow‐removal (SR) treatments compared to the control despite lower CO2 respiration. In addition, the water‐stress indicator (PLFA [cy17:0 + cy19:0] / [16:1ω7c + 18:1ω7c]) increased. These results suggest that soil microbial respiration and therefore the activity was not closely related to soil microbial biomass but more strongly controlled by substrate availability and quality. The PLFA pattern indicates that fungi are more susceptible to soil frost than bacteria.  相似文献

BACKGROUND

Aedes aegypti is an important mosquito species that can transmit several arboviruses such as dengue fever, yellow fever, chikungunya and zika. Because these mosquitoes are becoming resistant to most chemical insecticides used around the world, studies with new larvicides should be prioritized. Based on the known biological profile of imidazolium salts (IS), the objective of this study was to evaluate the potential of six IS as larvicides against Ae. aegypti, as tested against Ae. aegypti larvae. Larval mortality was measured after 24 and 48 h, and residual larvicidal activity was also evaluated.

RESULTS

Promising results were obtained with aqueous solutions of two IS: 1‐n‐octadecyl‐3‐methylimidazolium chloride ( C 18 MImCl ) and 1‐n‐hexadecyl‐3‐methylimidazolium methanesulfonate ( C 16 MImMeS ), showing up to 90% larval mortality after 48 h exposure. C 18 MImCl was more effective than C 16 mIMeS , causing mortality until day 15 after exposure. An application of C 18 MImCl left to dry under ambient conditions for at least 2 months and then dissolved in water showed a more pronounced residual effect (36 days with 95% mortality and 80% mortality up to 78 days).

CONCLUSION

This is the first study to show the potential of IS in the control of Ae. aegypti. Further studies are needed to understand the mode of action of these compounds in the biological development of this mosquito species. © 2017 Society of Chemical Industry

The frequency of fish diseases diagnosed at the Fish Disease Laboratory at the University of Berne from 1979 to 1988 is presented. Parasitic infections have been found to be the most frequent diagnosis. Among noninfectious diseases poisonings were predominant. Six infectious diseases have been considered to be economically relevant. These diseases (according to their frequency: Gyrodactylosis, Costiosis, Ichthyophthiriosis, Viral Hemorrhagic Septicemia (VHS), Saprolegniosis and Furunculosis) were further analysed. Their incidence varies with season, species, origin and length of fish. Possible explanations for these findings are discussed.  相似文献

Vesicular stomatitis is an infrequent yet important vesicular disease of cattle, horses, and swine. Periodic outbreaks of this disease in the United States have caused economic losses in cattle herds because of decreased production, movement restrictions, and trade embargoes. Vesicular stomatitis causes clinical signs indistinguishable from those of foot-and-mouth disease. It is of utmost importance that appropriate samples are collected from clinical cases of vesicular disease in cattle and swine so a rapid laboratory diagnosis can be made.  相似文献

Recently, feline Langerhans cells (LC) were immunophenotypically characterized as CD1a+, CD4+, CD18+, CD53+ and MHC II+ cells. In mice, these cells are known to internalize antigens and to migrate to the lymph nodes (LN). In the cat, we have investigated the migration of LC from the skin and vaginal mucosa to regional LN in response to chemical exposure (fluorescein isothiocyanate). Three days after the administration of a FITC solution on the posterior limb of two male cats and in the vagina of one female, a biopsy was carried out on the draining LN of the sensitized zones. Immunostaining with monoclonal antibodies anti-CD79, anti-CD8, and antibodies recognizing LC was performed on cytospins and frozen sections of LN and showed that a majority of FITC+ cells displayed a LC immunophenotype and were localized in T-cell areas, but not in follicular areas. These results are the first evidence of migration of feline LC from skin and vaginal mucosa to the regional LN.  相似文献

As part of the National Pesticide Monitoring Program, the U.S. Fish and Wildlife Service analyzed organochlorine contaminant residues in fish samples collected from about 100 stations each year from 1970 to 1974. During this period, mean residues of DDT and its metabolites declined nationally but remained widespread, and high concentrations continued to be present in areas where DDT use was extensive. Results of interlaboratory crosscheck analyses supported these conclusions, despite interpretation problems posed by intercompound analytical interferences in 1970 and 1971. Temporal trends in PCB residues were less obvious. Highest PCB residues were found in the industrialized areas of the Northwest and Midwest, and traces were present at most stations. Dieldrin and endrin residues remained essentially unchanged during this period; dieldrin residues were widespread and were highest in Hawaii and in areas of the Midwest where aldrin was used extensively. Toxaphene occurrence increased; it was formerly found only in fish from streams draining cotton-farming regions, but residues were detected in 1974 samples from other areas. According to the recommendation of the National Academy of Sciences' Water Quality Criteria, organochlorine residues in freshwater fish may have represented a hazard to piscivorous fish and wildlife at 71 percent of the stations sampled in 1970 and 66 percent in 1974.  相似文献
