Bending resistance of repaired column members and shear resistance of opening frames with repaired columns of conventional Japanese wooden houses

When it is necessary to repair conventional Japanese wooden houses, the decayed lower parts of columns should be replaced with new wood material. The bending resistance of columns repaired by four methods and the shear resistance of opening frames with those repaired columns were investigated in this study. Bending tests of the repaired columns showed differences in initial bending stiffness and maximum bending moment related to the repair methods and loading direction. Racking tests were conducted on door opening frames with conventional door head members or upper partial walls sheathed with 12-mmthick plywood. The conventional frame specimens broke at door head-column joints with no obvious bending deformation of the columns, resulting in little difference in load-shear deformation curves among the repair methods. The columns of plywood-sheathed specimens, on the other hand, clearly were bent after the nails at the plywood-to-wood frame joints started to pull off. The load-shear deformation curves of the plywood-sheathed specimens did not vary regardless of the repair methods when shear deformations were small but were affected by repair methods as shear deformation increased.     

Plasma Membrane Lipids Are the Powerful Components for Early Stage Aluminum Tolerance in Triticale

The order of aluminum (Al) tolerance in triticale lines (ST2>ST22) after re-elongation in an Al-free 0.2 mM calcium (Ca) solution for 9 h (Ca period) following 1 h pretreatment with 20 μM Al (Al period) agreed with that after 24 h of Al treatment. Permeability of the plasma membrane (PM) of root-tip cells after the Ca period was significantly increased in Al-sensitive ST22. Al was accumulated more heavily in the root-tip portion of ST22 than in that of ST2, although similar amounts of malic and citric acid anions were released from both triticale lines. We established a new system examining lipid permeability using synthesized nylon-2,8 ultrathin and porous capsules trapped previously with 0.1% (w/v) methylene blue solution and coated thereafter with PM lipid isolated from root tips by a newly developed technique. Permeability of the PM lipid measured with time in 0.2 mM Ca with or without 50 μM Al photometrically ( A ₆₈₀) was significantly greater in Al-sensitive ST22 after 5 min of Al treatment. This is the first report to directly show the primary and early role of PM lipid in Al tolerance in triticale.     

Expression of cell adhesion molecules in canine choroid plexus tumors

Choroid plexus tumor (CPT) is a primary intracranial neoplasm of the choroid plexus epithelium in the central nervous system. In the current World Health Organization classification, CPT is classified into two categories; choroid plexus papilloma (CPP) and carcinoma (CPC). In the present study, we investigated immunohistochemical expressions of N-cadherin, E-cadherin and β-catenin in 5 canine CPT cases (1 disseminated CPC, 2 CPCs and 2 CPPs). One CPP case was positive for N-cadherin and β-catenin, but negative for E-cadherin. The disseminated CPC case was positive for E-cadherin and β-catenin, but negative for N-cadherin. The other cases were positive for the three molecules examined. These results suggest that loss of the N-cadherin expression might associate with the spreading of CPC cells.

Experimental transmission of Bovine leukemia virus in cattle via rectal palpation

We examined whether Bovine leukemia virus (BLV) was transmitted by rectal palpation using a common sleeve between a BLV-infected cow and BLV-negative steers. Three of four steers developed antibodies against BLV as determined by agar-gel immunodiffusion (AGID) test between 7 to 10 weeks after the first rectal palpation using common sleeves from BLV-infected cow. In the steers, BLV proviral DNA were detected by PCR 1 to 5 weeks earlier than detection of the antibodies by the AGID test. Our experiments demonstrated that rectal palpation is a potential cause of BLV spread in herds and that detection of BLV proviral DNA in cattle by PCR is useful screening test for early diagnosis of BLV infection.     

Comparative assessment of Th1 and Th2 cytokines of swamp type buffalo and other bubaline breeds by molecular cloning, sequencing and phylogenetics

Comparative assessment of Th1 and Th2 cytokines of three bubaline breeds namely swamp buffalo, its crossbreed with riverine buffalo (CB), and the improved breed of Bulgarian Murrah buffalo (BMB), was done by molecular cloning, sequencing and phylogenetic analysis. The Th1 cytokines analyzed included IL-2, IL-12p35, IL-12p40, and IFN-gamma while Th2 cytokines included IL-4 and IL-10. Both groups showed strict conservation in the putative secondary structures and amino acid residues within the tribe Bovini, which indicated functional cross-reactivity. Nucleotide sequence homology ranged from 98.6 to 100.0% and was lowest for IL-12p35. With regard to amino acid sequence, the lowest homology was observed in IL-4 with 97.8%. This substitution was mainly due to differences in mRNA splicing. The phylogenetic relationship of the buffalo breeds was analyzed and showed them as a cluster comprised mainly of species belonging to the order Artiodactyla, including cattle and pigs. A deeper knowledge of these cytokine structures will favor understanding of water buffalo immunology and how much it differs from its closest subspecies and other animals.     

Spontaneous lymphoma in a prairie dog (Cynomys ludovicianus)

A 4-year-old, female, black-tailed prairie dog (Cynomys ludovicianus) was examined for acute dyspnea and two skin masses. Tentative diagnosis of lymphoma was made based on cytology of pleural effusion and the skin masses. Administration of prednisolone was initially effective in improving the condition; however, the clinical signs deteriorated after radiation therapy and administration of cyclophosphamide. Postmortem examination revealed that neoplastic lymphocytes had infiltrated most of the organs, and the histopathological diagnosis was multi-centric lymphoma.
