A simple, cost-effective and rapid viral nucleic acid release (NAR) buffer suitable for RT-PCR based diagnostic assay was developed for the detection of potato viruses. The NAR buffer and commercially available RNA isolation kit were compared for RT-PCR based assay, where an amplicon of expected size (~380 bp) targeting PVY was observed in both isolations indicating that it can be used in RT-PCR based diagnostic assays. The same was further validated for its repeatability by running across more than hundred suspected potato leaf samples collected from different sources where, it showed consistent results for the presence of PVY indicating its reliability. The NAR buffer assay was examined for its sensitivity in comparison with the kit based isolation where both the assays were able to detect even up to 10?5 dilution without affecting the sensitivity. NAR buffer was found stable up to 28 days at -20 °C and for 14 days at 4 °C without losing PCR sensitivity. The assay was also found effective to release the nucleic acid from potato leaves, thrips and aphids for PCR and RT-PCR based detection of DNA viruses like ToLCNDV-potato and other RNA viruses. The developed protocol is simple, less laborious, time-saving (10-15 min) and economical (1/100th of kit) as compared to kit based protocol. The assay can be adopted in diagnostic laboratories for detection of RNA/DNA viruses from potato plants and in thrips as well.  相似文献

Increasing global air temperatures, along with rising CO2 levels, are causing concerns about reducing available freshwater resources and altering cropping patterns. They may influence overall growth and production pattern of crop plants. These likely changes would become major limiting factors for future sustainable food production largely in the tropics and subtropics. Thus, understanding physiological responses hold the key to determining the functional relationship between the environment and crop performance. We explore here the impact of rising CO2 on the growth and yield traits of a few selected high-temperature (HT)-tolerant mungbean lines, which we earlier screened for HT tolerance using a physiological assay under managed growth conditions. The HT-tolerant lines grown under elevated CO2 levels (550 and 700 μL L?1) showed a considerable improvement in growth rates (13.5%, 67.8%, and 46.5% in plant height, leaf area, and total dry matter, respectively) and pod and seed yield (48.7% and 31.7%, respectively), compared to local checks under the same environments. Interestingly, the symptoms of accelerated pod maturity were also observed in most of these lines. The outcome of the study would undoubtedly open up opportunities for increased yield potentials of legumes under the conditions of the warming climate and elevated levels of carbon dioxide.  相似文献

Different forms of manganese (Mn) were investigated, including total, diethylenetriamine penta-acetic acid (DTPA) extractable, soil solution plus exchangeable (Mn), Mn adsorbed onto inorganic sites, Mn bound by organic sites, and Mn adsorbed onto oxide surfaces, from four soil taxonomic orders in northwestern India. The total Mn content was 200–950 mg kg?1, DTPA-extractable Mn content was 0.60–5.80 mg kg?1, soil solution plus exchangeable Mn content was 0.02–0.80 mg kg?1, Mn adsorbed onto inorganic sites was 2.46–90 mg kg?1, and Mc adsorbed onto oxide surfaces was 6.0–225.0 mg kg?1. Irrespective of the different fractions of Mn their content was generally greater in the fine-textured Alfisols and Inceptisols than in coarse-textured Entisols and Aridisols. The proportion of the Mn fractions extracted from the soil was in the order as follows: Adsorbed onto oxide surfaces > adsorbed onto inorganic site > organically bound > DTPA > soil solution + exchangeable. Based on coefficient of correlation, the soil solution plus exchangeable Mn, held onto organic site and oxide surface (amorphous) and DTPA-extractable Mn, increased with increase in organic carbon of the soil. The two forms, adsorbed onto inorganic site (crystalline) and DTPA extractable, along with organic carbon, increased with increase in clay content of the soil. DTPA-Mn and Mn adsorbed onto oxide surfaces and held on organic site decreased with increased with an increase in calcium carbonate and pH. Total Mn was strongly correlated with organic carbon and clay content of soil. Among the forms, Mn held on the organic site, water soluble + exchangeable and adsorbed onto oxide surface were positively correlated with DTPA-extractable Mn. DTPA-extractable Mn seems to be a good index of Mn availability in soils and this form is helpful for correction of Mn deficiency in the soils of the region. The uptake of Mn was greater in fine-textured Inceptisols and Alfisols than in coarse-textured Entisols and Aridisols. Among the different forms only DTPA-extractable Mn was positively correlated with total uptake of Mn. Among soil properties Mn uptake was only significantly affected by pH of the soil.  相似文献

Vertisol soils of central India are heavy in texture, with high clay content and low organic matter. These soils are prone to degradation and the soil loss is due to poor management practices including excessive tillage. Based on a long-term study conducted for improving the quality of these soils, it was found that management practice such as low tillage (LT) + 4 t ha?1 compost + herbicide (Hb) recorded significantly higher organic carbon (OC) (6.22 g kg?1) and available N (188.5 kg ha?1) compared to conventional tillage (CT) + recommended fertilizer (RF) + off-season tillage (OT) + hand weeding (HW) (OC: 4.71 g kg?1, available nitrogen (N) (159.3 kg ha?1). Among the physical soil quality parameters, mean weight diameter (MWD) was significantly higher under LT + 4 t ha?1 straw + Hb (0.59 mm). The practice of LT + 4 t ha?1 straw + HW recorded significantly higher microbial biomass carbon (MBC) (388.8 μg g?1). The order of key indicators and their contribution towards soil quality was as follows: OC (29%) >, MBC (27%) > available zinc (Zn) (22%) > MWD (9%) > available boron (B) (8%), > dehydrogenase activity (DHA) (5%). The order of the best treatment which maintained soil quality index (SQI) values reasonably good (>1.5) was as follows: LT + 4t ha?1 compost + HW (1.65) > LT + 4 t ha?1 compost +Hb (1.60) > LT + 4t ha?1 straw + HW (1.50). Hence, these treatments could be recommended to the farmers for maintaining higher soil quality in Vertisols under soybean system. Correlation studies revealed stronger relationship between key indicators like OC (R2 = 0.627), MBC (R2 = 0.884), available Zn (R2 = 0.739) and DHA (R2 = 0.604) with Relative Soil Quality Index (RSQI). The results of the present study would be highly useful to the researchers, farmers and land managers.  相似文献

Black gram (Vigna mungo L.) var. Shyam plants were raised in refined sand at four levels of molybdenum (Mo), i.e., low (0.002 µM) to excess (2 µM) for 70 days. The molybdenum deficiency symptoms appeared as interveinal chlorosis of young and middle leaves. Compared to control (0.2 µM Mo), total dry matter, seed yield and seed protein decreased at low and excess Mo. The concentration of Mo in leaves and seed as well as activity of nitrate reductase increased with an increase in Mo supply. Low and excess Mo deteriorated the quality of seeds by lowering the content of starch, sugars, protein, and nitrogen and increasing electrical conductivity of seed leachate. Molybdenum deficiency and excess both resulted in production of lightweight immature seeds, poor in vigor and germination potential. The values of sufficiency and threshold of toxicity in leaves were 0.078 and 2.15 µg Mo g?1 dry matter of black gram.  相似文献

Thirty field experiments were conducted in North Dakota during 2011 and 2012 to compare two ground-based active-optical sensors for their relationship between sensor readings and INSEY (in-season estimate of yield). The experimental design at each site was a randomized complete block with four replications and six nitrogen (N) rate treatments: control, 45, 90, 134, 179, and 224 kg ha?1 applied pre-plant as ammonium nitrate within five days of planting. The two sensors, GreenSeeker® (Trimble, Sunnydale, CA, USA) (GS) and Crop Circle ACS 470® sensor (Holland Scientific, Lincoln, NE, USA) (CC) were used to scan over the top of the corn at V6 and V12 growth stages. The GS INSEY and the CC INSEY were similarly related to corn yield at V6. The CC using the red-edge lens option improved the INSEY relationship to corn yield compared to the GS or the CC using the red lens option at V12.  相似文献
