The in vitro performance of mango shoot culture is delimited by several factors, of which explant necrosis and media browning are considered as major constraints for successful exploitation of such an important commercial crop. Our results showed that source of explants had a significant effect on the performance of in vitro cultures of mango. The variations in survival of explants collected from glasshouse grown seedlings and field-grown stock plants indicated towards differences between their physiological/ontogenic age. Though, chronologically, both are young, the glasshouse grown shoots are ontogenetically younger and; therefore, responded better over field-grown shoots. Improved performance of explants harvested from mycorrhizal plants suggests integration of mycorrhizal biotechnology with tissue culture biotechnology in order to achieve better results as mycorrhiza being a well-known stress alleviator may help explants mitigate in vitro stresses. Furthermore, shoot tip explants of field-grown ‘Amrapali’ mango was assayed for their browning propensity at pre- and post-culture stages. Status of different bio-chemicals such as in vivo phenol, in vitro phenol exudation, phenylalanine ammonia lyase, peroxidase and polyphenol oxidase as affected by various pre-treatments were estimated to establish the relationship of these bio-chemicals with necrosis of mango shoot cultures. The different pre-treatments comprised etiolation of newly emerged shoots (5–7 days old) of stock plants using black polythene for 7 days, etiolation treatment + agitation of explants in antioxidant solution (antioxidants: ascorbic acid at 100 mg l−1 + citric acid at 50 mg l−1), etiolation treatment + agitation of explants in polyvinylpyrrolidone solution (PVP) at 0.2% and non-etiolated control. Of these, explants treated with PVP proved to be superior to other treatments with regards to explant necrosis percentage as such explants exhibited least activities of phenols and oxidative enzymes. Correlation studies indicated existence of high positive correlation between explant necrosis and these bio-chemicals.  相似文献

Every year, multiple outbreaks of salmonellosis in humans are linked to contact with mail‐order chicks and ducks. The objective of this study was to describe the temporal changes in the prevalence of serovars, genotypes and antimicrobial resistance (AMR) phenotypes of non‐typhoidal Salmonella (NTS) recovered from shipped boxes of mail‐order hatchling poultry in the United States during 2013 to 2015. In each year, a sample of feed stores belonging to a single national chain participated in the study. The store employees submitted swabs or hatchling pads from hatchling boxes and shipment tracking information of the arriving boxes to the investigators. NTS was cultured from the samples and isolates were sent to the National Veterinary Services Laboratories (Ames, IA) for serotyping, pulsed‐field gel electrophoresis (PFGE) and AMR phenotyping. The PFGE patterns of Salmonella serovars isolated from hatchling boxes were compared with those from human outbreaks of salmonellosis linked to live poultry contact. The box‐level prevalence of NTS was significantly higher in 2015 compared to 2014. Also, the population of Salmonella serovars recovered in 2015 was more diverse and substantially different from those recovered in the previous two years. Of PFGE patterns recovered from hatchling boxes, seven distinct patterns in 2015, three in 2014 and four in 2013 were indistinguishable from the PFGE patterns of human outbreaks‐associated strains in the respective years. Importantly, a significant positive correlation was found between the box‐level prevalence of PFGE patterns and the number of human illnesses associated with the same patterns. Also, the proportion of multidrug‐resistant isolates was higher in 2014 and 2015 compared to that in 2013. The results demonstrate that shipments of mail‐order hatchling poultry are frequently contaminated with Salmonella genotypes indistinguishable from human outbreaks‐associated strains each year, and control efforts at hatchery level are likely to have an important public health impact.  相似文献

The objective of this study was to document the expression and localization of angiopoietin (ANGPT) family members comprising of angiopoietin (ANGPT1 and ANGPT2), and their receptors (Tie1 and Tie2) in buffalo corpus luteum (CL) obtained from different stages of the oestrous cycle, and the modulatory role of ANGPT1 and ANGPT2 alone or in combinations on progesterone (P4) secretion and mRNA expression of phosphotidylinositide‐3kinase‐protein kinase B (PI3K‐AKT), phosphoinositide‐dependent kinase (PDK), protein kinase B (AKT), Bcl2 associated death promoter (BAD), caspase 3 and von willebrand factor (vWF) in luteal cells obtained from midluteal phase (MLP) of oestrous cycle in buffalo. Real‐time RT‐PCR (qPCR), Western blot and immunohistochemistry were applied to investigate mRNA expression, protein expression and localization of examined factors whereas, the P4 secretion was assessed by RIA. The mRNA and protein expression of ANGPT1 and Tie2 was maximum (p < .05) in mid luteal phase (MLP) of oestrous cycle. The ANGPT2 mRNA and protein expression was maximum (p < .05) in early luteal phase, decreased in MLP and again increased in late luteal phase of oestrous cycle. ANGPT family members were localized in luteal cells and endothelial cells with a stage specific immunoreactivity. P4 secretion was highest (p < .05) with 100 ng/ml at 72 hr when luteal cells were treated with either protein alone. The mRNA expression of PDK, AKT and vWF was highest (p < .05) and BAD along with caspase 3 were lowest (p < .05) at 100 ng/ml at 72 hr of incubation period, when cultured luteal cells were treated with either protein alone or in combination. To conclude, our study explores the steroidogenic potential of angiopoietins to promote P4 secretion, luteal cell survival and angiogenesis through an autocrine and paracrine actions in buffalo CL.  相似文献

The wheat-rye translocation (IBL-IRS) that carries the tightly linked genes Lr26/Sr31/Yr9, has been widely exploited in the development of wheat cultivars worldwide. This resistance, however, has become ineffective owing to the evolution of new pathotypes of Puccinia recondita that neutralize the resistance of Lr26. Inheritance studies on ‘Federation4′/‘Kavkaz’ revealed complementary genes derived separately from ‘Federation’ and ‘Kavkaz’ for adult plant resistance. This previously undescribed source of resistance appears to be widely effective and could therefore be used to broaden the genetic base for resistance in India. Its effectiveness in other geographical areas is unknown.  相似文献

Thirteen monogenic morphological marker traits of lentil were analysed for their joint inheritance in 25 crosses. Only three traits showed linkage: Ert (for spreading-erect growth habit), Gs (for brown-green stem) and Bl (for brown-green leaf). The recombination frequency between Bl and Gs was 14%, between Bl and Ert 34%, and between Gs and Ert 38%. Thus, the gene Bl is located between Ert and Gs.  相似文献

In-gel hybridization patterns were studied in a set of nine diverse bread wheat ( Triticum aestivum L. em. Thell) genotypes using 23 simple sequence repeat (SSR) probes in combination with 14 different restriction enzymes. Multilocus fingerprints due to SSR probes, shown earlier to be characteristic of a majority of plant genomes, were not obtained and only a very low level of polymorphism was detected when using as many as 142 probe-enzyme combinations. The hybridization of a prominent solitary high molecular weight fragment (> 23 kb) with a number of SSR probes suggested the presence of these SSRs (microsatellites) within the long stretches of repeated DNA sequences. This indicates that the genome of bread wheat differs from that of other plants in the organization and distribution of SSRs and that SSR probes detect very little polymorphism.  相似文献

Abstract. A field experiment was conducted over two years in one of the mountain Alfisols of the Western Himalayas to study the effects of phosphorus and mulching on phosphorus use efficiency (PUE) and productivity of wheat (cv. S-308) at difference growth stages. The source of phosphorus was single superphosphate at 0, 26, 52 and 78 kg P/ha whereas the sources of mulching materials were pine needles ( Pinus longifolia ), lantana weed ( Lantana camara ) at 8 t/ha and transparent polyethylene sheet compared with no mulch.
The phosphorus use efficiency (PUE) at tillering, flowering and harvesting was greatest at the lowest dose of P and decreased as the P levels were increased. However, the increased levels of P up to 78 kg/ha, significantly improved the dry matter yield at tillering stage (30%) and flowering stage (93%) and also the grain yield (139%) and straw yield (148%) at the harvest stage. The application of mulching materials in general, and polyethylene in particular, significantly increased the phosphorus use efficiency and the productivity of wheat by 27% at tillering stage; 17% at flowering stage, and by 98 and 110% in the case of grain and straw yield at harvest stage.
Although the polyethylene mulch effect was physically superior to the other materials, the latter gave greater financial returns. Consequently, the economic optimum application of P in association with 8 t/ha of lantana mulch was found to be 53 kg/ha over two years, whereas, in the absence of mulching, it was 58 kg/ha.  相似文献

Extensive lignite mining in the Indian (Thar) Desert commenced within the past decade. Accompanying extraction of this valuable resource there have been visible, important environmental impacts. The resultant land degradation has prompted concern from both public and regulatory bodies. This research assesses the success of rehabilitation plans implemented to revegetate a lignite mine‐disturbed area, near the village of Giral in western Rajasthan State. Rehabilitation success was achieved within the environmental constraints of this northwest Indian hot‐desert ecosystem using a combination of: (1) backfilling (abandoned pits) with minespoil and of covering the backfilled‐surfaces with fresh topsoil to a thickness of about 0·30 m; (2) use of micro‐catchment rainwater harvesting (MCWH) technique; (3) soil profile modification approaches; (4) plant establishment methodologies; and (5) the selection of appropriate germplasm material (trees, shrubs and grasses). Preliminary results indicate that the resulting vegetative cover will be capable of self‐perpetuation under natural conditions while at the same time meeting the land‐use requirements of the local people. The minespoil is alkaline in nature and has high electrical conductance. The average content of organic carbon, N, P and K is lower than in the regional topsoil. However, the concentration of Ca, Mg, Na and total S in the minespoil is much higher than in the topsoil. Further, the spoil material has no biological activity. Enhanced plant growth was achieved in MCWH plots, compared to control plots, where minespoil moisture storage was improved by 18–43 per cent. The rehabilitation protocol used at the site appears to have been successful because, in addition to the planted species, desirable native invasive species have become established. This study developed methods for the rehabilitation of lignite mine‐disturbed areas and has also resulted in an understanding of rehabilitation processes in arid regions with an emphasis on the long‐term monitoring of rehabilitation success. The variation in the seed shape, colour and yield, and content, yield and fatty acid composition of seed oil of 109 accessions of opium poppy Papaver somniferum, (majority of them Indian land races), was investigated. The seeds were white, pale yellow or light brown in colour, reniform or round in shape and varied in size up to three fold. The oil content, seed and the oil yield varied between 26 to 52%, 1.0 to 7.4 g/plant and 0.4 to 2.7 g/plant, respectively. The % content of palmitic, oleic and linoleic acid in the seed oil ranged between 9.3 to 40.0%, 7.5 to 58.4% and 0.7 to 72.7%, respectively. On average basis, the levels of major fatty acids in the seed oil were: oleic (37.1%) > palmitic (27.3%) > linoleic acid (17.2%). The palmitoleic, stearic and linolenic acids were present in the oils of only some of the accessions. Two of the accessions yielded linoleic acid rich seed oil of about the same quality as soybean and maize oils, and in four accessions, the proportion of palmitic, oleic and linoleic acids was roughly equal. The palmitic acid was relatively less and linoleic acid more in the seed oil from accessions rich in oil content. The oil that contained higher amount of oleic acid also contained higher amount of palmitic acid and relatively lower amount of linoleic acid. The correlation analyses revealed a strong positive relationship between seed yield and oil yield (r = +0.81), oil yield and oil content (r = +0.54) and oleic acid and palmitic acid content in the seed oil (r = +0.49), and a weak positive relationship between oil content and linoleic acid content of oil (r = +0.24), and a negative correlation was observed between oil content and palmitic acid content (r = –0.32), palmitic acid and linoleic acid (r = –0.55) and oleic acid and linoleic acid contents of oil (r = –0.68). The observations have permitted selection of accessions that are high seed and oil yielding and/or rich in linoleic, palmitic and oleic acids or containing palmitic, oleic and linoleic acids in about equal amounts.  相似文献

Crop consumptive water use and productivity are key elements to understand basin water management performance. This article presents a simplified approach to map rice (Oryza sativa L.) water consumption, yield, and water productivity (WP) in the Indo-Gangetic Basin (IGB) by combining remotely sensed imagery, national census and meteorological data. The statistical rice cropped area and production data were synthesized to calculate district-level land productivity, which is then further extrapolated to pixel-level values using MODIS NDVI product based on a crop dominance map. The water consumption by actual evapotranspiration is estimated with Simplified Surface Energy Balance (SSEB) model taking meteorological data and MODIS land surface temperature products as inputs. WP maps are then generated by dividing the rice productivity map with the seasonal actual evapotranspiration (ET) map. The average rice yields for Pakistan, India, Nepal and Bangladesh in the basin are 2.60, 2.53, 3.54 and 2.75 tons/ha, respectively. The average rice ET is 416 mm, accounting for only 68.2% of potential ET. The average WP of rice is 0.74 kg/m3. The WP generally varies with the trends of yield variation. A comparative analysis of ET, yield, rainfall and WP maps indicates greater scope for improvement of the downstream areas of the Ganges basin. The method proposed is simple, with satisfactory accuracy, and can be easily applied elsewhere.  相似文献
