The pharmacokinetic properties of marbofoxacin, a third generation fluoroquinolone, were investigated in 12 healthy adult cats after single subcutaneous (SC) administration of 2 mg/kg BW (Part I, n=8 cats) and 4 mg/kg BW (Part II, n=4 cats). In each part of the study blood and urine samples were collected before treatment and thereafter for 5 days. The plasma and urine concentrations of marbofloxacin were determined by HPLC with UV detection. Pharmacokinetic calculations were performed for each treated animal using an open one-compartment-model with first-order elimination after SC dosing. Marbofloxacin in plasma (means): Maximum concentrations (Cmax) of about 1.2 and 3.0 microg/ml were measured 2.3 and 4 hours (tmax) after dosing of 2 and 4 mg/kg BW, respectively. Elimination from the body was low with a total clearance (Cl/F) of approximately 0.1 l/h/kg for both dosages. The half-life (t 1/2) for this process was calculated with 8-10 hours. AUC increased almost proportional when doubling the dose, i.e., 19.77 +/- 6.25 microg * h/ml (2 mg/kg BW) and 51.26 +/- 11.83 microg * h/ml (4 mg/kg BW). Plasma kinetics measured were in accordance with data from literature. Marbofloxacin in urine (means): Maximum drug concentrations were detected 4 and 8 hours after dosing with 70 microg/ml (2 mg/kg BW) and 160 microg/ml (4 mg/kg BW), respectively. Inhibitory effects of the urinary matrix on the antimicrobial activity of the drug were taken into account when performing PK/PD calculations. However, a concentration-dependent bactericidal activity (Cmax/MIC > 8-10) which is claimed for fluoroquinolones was sufficiently met with focus on Escherichia (E.) coli (MIC90 0.5 microg/ml). In the same matrix a threshold value of 1.0 microg/ml was undercut 82 and 116 hours after SC dosing, respectively. Hence, a time-dependent bacteria killing kinetic (T > MIC) which may be of relevance for some Gram-positive germs like Staphylococcus spp. (MIC90 1.0 microg/ml) should be covered, too.  相似文献

Contents: Experimental investigation on the regulative capacity of sheep embryos in vivo and in vitro: Embryos aged five or six days were obtained surgically or after slaughter. They were divided in two equal halves by micromcinipulation. Both halves of the embryos diweloped into blastocysts during cultivation over a period of 3 to 48 hours if conditions were fauourable. 2 out of 1.3 ewes lambed into which hulves of embryos had been transplanted that were obtained after slaughter. 14 out of 24 ewes into which halves of embryos had been implanted which were obtained surgically did not come into estrus again. 7 out of the 14 ewes lambed so far. In potato plants induced for systemic resistance by infiltration with Pseudomonas syringae pv. maculicola, 12-oxo-phytodienoic acid (OPDA) accumulated in infiltrated leaves as well as in non-treated leaves of infected plants. In contrast, jasmonic acid (JA) levels increased only in infiltrated leaves, suggesting that the biosynthetic precursor of JA, OPDA, might play a role in systemic acquired resistance.  相似文献

There is a dearth of knowledge on the available sources of botanical products for smallholder farmers in Africa and their bioactivity against stored-product pests. Many locally available plants across Africa exhibit both promising insecticidal, medicinal activity and are nutrient rich yet remain underutilized. Laboratory studies on toxicity of powders of Senna siamea and Piper guineense alone or combined with diatomaceous earth (DE) to adult Sitophilus granarius, Tribolium castaneum and Acanthoscelides obtectus were conducted in stored grains. Adult mortality was observed up to 7 d while progeny production was recorded 6–10 weeks. Adult A. obtectus were most susceptible (LD50 0.0084% and 0.0696% wt/wt for S. siamea and P. guineense, respectively), followed by S. granarius. T. castaneum was most tolerant (LD50 0.6589% and 0.6829% wt/wt for S. siamea and P. guineense, respectively). Combination of plant powders and DE controlled the beetles faster compared to the plant powders alone. LT50 ranged from 20 h to 101 h for T. castaneum exposed to 1.0% S. siamea and 1.0% DE, and 0.5% P. guineense and 1.0% DE, respectively. Botanicals caused significant reduction of F1 adults compared to the control. The potential of using botanicals combined with DE to control stored-product insects is discussed.  相似文献

The study applies structural indices using the example of an oak (Quercus petraea (Matt.) Liebl.) — chequer tree (Sorbus torminalis (L.) Crantz) stand in order to derive recommendations for the silvicultural treatment of Chequer trees. The investigated stand, located in the northern part of Bavaria, comprises eight tree species and four shrub species. Various indices were used to analyse the stand structure and the crown coverage frequency. It was shown that chequer trees, which are presently of high economic interest, are strongly oppressed in the upper layer and almost completely missing in the lower layers of the stand. The possible reasons for this finding and alternatives for the further management of the stand are discussed. Persistent and repeated thinnings in order to ensure sufficient crown development of the chequer trees seem to be essential for their survival.  相似文献

The calculation of critical loads and their exceedance is one method to describe the vulnerability of forests to environmental stress caused by anthropogenic impact. Exceedance of critical loads for acidifying inputs and nitrogen was compared to different indicators of the soil and forest conditions in the German part of the extensive forest monitoring (ICP Forests/EU Level I), including more than 1,800 plots. In addition, an empirical relationship between the C/N ratio of the forest floor humus layer (C/N Humus) and the estimated nitrogen output for ten plots of the intensive monitoring (ICP Forests/EU Level II) was established in order to estimate the potential nitrogen output on Level I plots dominated by Norway spruce. Regarding all tree species assessed, the exceedance of critical loads for nitrogen and sulphur is negatively correlated with pH and base saturation up to 30 cm soil depth. The sulphur deposition and the exceedance of critical loads are highly correlated with the sulphur content of leaves and needles, whereas the respective relations for nitrogen were lower. The crown condition was weakly positively related to the sulphur content in tree leaves and needles. For Norway spruce sites, high exceedance of critical loads for nitrogen and nitrogen deposition corresponded well with low C/N Humus. In regions with high nitrogen load and low C/N ratios in the humus layer, the calculated nitrogen output was high. The results support the concept of critical thresholds in that way that their exceedance can impair forest ecosystem functions like nitrogen retention.  相似文献
