Cellular oncogenes and multistep carcinogenesis   总被引:122,自引:0,他引:122  
Two dozen cellular proto-oncogenes have been discovered to date through the study of retroviruses and the use of gene transfer. They form a structurally and functionally heterogeneous group. At least five distinct mechanisms are responsible for their conversion to active oncogenes. Recent work provides experimental strategies by which many of these oncogenes, as well as oncogenes of DNA tumor viruses, may be placed into functional categories. These procedures may lead to definition of a small number of common pathways through which the various oncogenes act to transform cells.  相似文献

  1. The Juan Fernandez Ridge, a vulnerable marine ecosystem located far off the coast of central Chile and formed by several seamounts, guyots and three islands (Robinson Crusoe, Santa Clara and Alejandro Selkirk), has recently been declared a Coastal Marine Protected Area of Multiple Uses with several National Parks embedded in it.
  2. Recent studies have highlighted the influence of remote and local oceanographic structures on the hydrographic dynamics of this ridge. However, there is still a gap in understanding how they affect the structure and dynamics of the surrounding insular planktonic communities.
  3. A hydroacoustic and oceanographic survey was conducted during the austral spring (October 2016), including hydrographic and zooplankton sampling around Robinson Crusoe Island. Oceanographic features were identified and tracked using satellite data (chlorophyll-a (Chl-a) and sea surface temperature) and modelling results.
  4. Two events of Chl-a increase relative to a threshold (>0.45 mg m−3) were forced by different physical processes, both affecting the western side of Robinson Crusoe Island. In event A during the cruise period, Chl-a subsurface maxima were associated with the arrival of a coastal meander originating on the continental shelf off Chile (remote process); the zooplankton was dominated by copepods and salps, with an evident coastal–oceanic gradient. In event B, Chl-a maxima were linked to a local upwelling forced by the intensification of a localized SSW wind. No influence of remote eddies or local Von Kármán vortices on Chl-a distribution was observed.
  5. These findings highlight the influence of remote and local physical processes on the structure of planktonic communities around Robinson Crusoe Island. Understanding the variability of these mechanisms and their effects at the base of the pelagic food web is critical in adopting an ecosystem-level approach.

Neurofibromatosis type 1 (NF1) is one of the most prevalent dominantly inherited genetic diseases of the nervous system. NF1 encodes a tumor suppressor whose functional loss results in the development of benign neurofibromas that can progress to malignancy. Neurofibromas are complex tumors composed of axonal processes, Schwann cells, fibroblasts, perineurial cells, and mast cells. Through use of a conditional (cre/lox) allele, we show that loss of NF1 in the Schwann cell lineage is sufficient to generate tumors. In addition, complete NF1-mediated tumorigenicity requires both a loss of NF1 in cells destined to become neoplastic as well as heterozygosity in non-neoplastic cells. The requirement for a permissive haploinsufficient environment to allow tumorigenesis may have therapeutic implications for NF1 and other familial cancers.  相似文献

The discovery of cytoplasmic male sterility directed scientists to focus their research on the role and influence of cytoplasmic factors on the heredity of some important agronomic traits, especially in hybrids. In this context, numerous cytolines have been created at the Agricultural Research and Development Station Turda (Romania) in order to assess if their genetic value could be due to cytoplasm diversification. Backcross was performed for ten generations in order to transfer the nucleus of the pollenator into the cytoplasm of the maternal genotype. This paper presents the results regarding yield and yield components of 100 hybrids, originated from crosses among isolines and four testers. The isolines, used as maternal genotypes, were created by transferring the nucleus of the elite inbred lines TC 209, TC 316, TC 243, TB 367 and D 105 onto the cytoplasm of T 248, TB 329, TC 177 and TC 221 inbred lines. The cytoplasms–nucleus interaction and the triple cytoplasm–nucleus–tester interaction, caused significant differentiation between the hybrids originated from cytoline maternal genotype in comparison to those from original inbred lines. Yield, thousand kernel weight, ear weight, grain weight/ear and the number of kernels/row were used for comparisons among all genotypes used into this research.  相似文献

Free and glycosidically bound volatiles obtained from the fruit pulp of Sicana odorifera by liquid-liquid extraction and by chromatography, followed by enzymatic hydrolysis with Rohapect D5L, respectively, were analyzed by capillary gas chromatography (HRGC), HRGC-mass spectrometry (HRGC-MS), and HRGC-Olfatometry (HRGC-O) analyses. A total of 37 free volatiles was detected, with the major components being 3-methyl-2-butanol, 3-hydroxy-2-butanone, ethyl 3-hydroxybutanoate, and (Z)-3-hexenol. Among the 22 detected glycosidically bound compounds, 4-hydroxybenzyl methyl ether, 4-hydroxybenzyl alcohol, and 2-phenylethanol were found to be the major constituents. Additionally, two glucoconjugates were isolated in pure form by multilayer coil countercurrent chromatography (MLCCC) of the glycosidic extrac and further purification. Their structures were elucidated by MS and NMR analyses to be the novel [4-(beta-D-glucopyranosyloxy)benzyl] 2,3-dihydroxy-3-methylbutanoate 2, and the known 4-(beta-D-glucopyranosyloxy)benzyl alcohol 1. Compounds 1 and 2 are precursors of 4-hydroxybenzyl alcohol, one of the major volatiles generated by enzymatic hydrolysis of the glycosidic fraction.  相似文献

The movement of dislocations in a crystal is the key mechanism for plastic deformation in all materials. Studies of dislocations have focused on three-dimensional materials, and there is little experimental evidence regarding the dynamics of dislocations and their impact at the atomic level on the lattice structure of graphene. We studied the dynamics of dislocation pairs in graphene, recorded with single-atom sensitivity. We examined stepwise dislocation movement along the zig-zag lattice direction mediated either by a single bond rotation or through the loss of two carbon atoms. The strain fields were determined, showing how dislocations deform graphene by elongation and compression of C-C bonds, shear, and lattice rotations.  相似文献

The chlorophyll meter (CM) has been commonly used for in-season nitrogen (N) management of corn (Zea mays L.). Nevertheless, it has limited potential for site-specific N management in large fields due to difficulties in using it to generate N status maps. The objective of this study was to determine how well CM readings can be estimated using aerial hyper-spectral and simulated multi-spectral remote sensing images at different corn growth stages. Two field experiments were conducted in Minnesota, USA during 2005 involving different N application rates and timings on a corn-soybean [Glycine max (L.) Merr.] rotation field and a corn-corn rotation field. Four flights were made during the growing season using the AISA Eagle Hyper-spectral Imager and CM readings were collected at four or five different growth stages. The results indicated that single multi-spectral and hyper-spectral band or vegetation index could explain 64–86% and 73–88% of the variability in CM readings, respectively, except at growth stage V9 in the corn-soybean rotation field where no band or vegetation index could explain more than 37% of the variability in CM readings. Multiple regression analysis demonstrated that the combination of 2–4 broad-bands or 3–8 narrow-bands could explain 41–92% or 61–94% of the variability in CM readings across the two fields and different corn growth stages investigated. It was concluded that the combination of CM readings with high spatial resolution hyper-spectral or multi-spectral remote sensing images can overcome the limitations of using them individually, thus offering a practical solution to N deficiency detection and possibly in-season site-specific N management in large continuous corn fields or at later stages in corn-soybean rotation fields.  相似文献
