The 4-(dimethylamino)cinnamaldehyde (DMAC) assay is currently used to quantify proanthocyanidin (PAC) content in cranberry products. However, this method suffers from issues of accuracy and precision in the analysis and comparison of PAC levels across a broad range of cranberry products. Current use of procyanidin A2 as a standard leads to an underestimation of PACs content in certain cranberry products, especially those containing higher molecular weight PACs. To begin to address the issue of accuracy, a method for the production of a cranberry PAC standard, derived from an extraction of cranberry (c-PAC) press cake, was developed and evaluated. Use of the c-PAC standard to quantify PAC content in cranberry samples resulted in values that were 2.2 times higher than those determined by procyanidin A2. Increased accuracy is critical for estimating PAC content in relationship to research on authenticity, efficacy, and bioactivity, especially in designing clinical trials for determination of putative health benefits.  相似文献

Taylorella equigenitalis is a gram-negative coccobacillus and the causative agent of a transmissible venereal disease in horses known as contagious equine metritis. Outbreaks of contagious equine metritis have been documented in various countries since 1977, with the most recent discovery in the United States in December 2008. During disease occurrences, culturing semen samples for T equigenitalis before breeding may help to prevent transmission of this disease; however, little is known about the antimicrobial activity of equine semen extenders against the organism. The purpose of this study was to investigate the infectivity levels of T equigenitalis in three equine semen extenders inoculated with known concentrations of the organism. The semen extenders used for this study included INRA 96, E-Z Mixin BF, and VMDZ. In addition, Timentin was added to INRA 96 at three different concentrations (0.5, 1.0, and 1.5 mg/mL) to investigate possible synergistic effects of antibiotic supplementation of extenders. Results were based on the visual counting of the colonies on chocolate Eugon agar plates. Both INRA 96 (with added Timentin) and VMDZ (as supplied by the manufacturer) significantly reduced the numbers of T equigenitalis isolated from semen extenders as compared with INRA 96 (as supplied by the manufacturer) or the antibiotic free E-Z Mixin BF. Our findings indicate that INRA 96 (with added Timentin) or VMDZ may significantly decrease the growth of T equigenitalis in extended semen; however, it is also important to consider the possible effects of antibiotic supplemented extenders on sperm longevity and fertility in addition to eliminating specific pathogens in semen.  相似文献

To investigate the influence of maternal Se supply and plane of nutrition on lamb morbidity, mortality, and passive transfer of IgG, pregnant ewe lambs were used in 2 experiments with 2 × 3 factorial treatment arrangements. Supplementation of Se began at breeding and was either adequate Se (ASe, 9.5 μg/kg of BW) or high Se (HSe, 81.8 μg/kg of BW) in Exp. 1 or ASe (11.5 μg/kg of BW) or HSe (77.0 μg/kg of BW) in Exp. 2. On d 50 or 40 of gestation for Exp. 1 or 2, respectively, ewes were assigned randomly to 1 of 3 nutritional planes: 60% (RES), 100% (control, CON), or 140% (HI) of NRC requirements. This resulted in the following treatments: ASe-RES, ASe-CON, ASe-HI, HSe-RES, HSe-CON, and HSe-HI. Upon parturition, lambs were separated from their dams and serum samples obtained. Lambs were fed artificial colostrum for the first 20 h and then placed on milk replacer and grain pellets until completion of the study (Exp. 1, 57 d; Exp. 2, 21 d). Twenty-four hours after parturition, lamb serum samples were collected for IgG analysis. All lambs were reared similarly and morbidity and mortality assessed. Main effects were considered significant when P ≤ 0.05. In Exp. 1, there was a Se × plane of nutrition interaction (P ≤ 0.01) for lamb morbidity from birth to weaning and for 24-h IgG concentration. Lambs from ASe-RES and HSe-HI ewes were treated more frequently (P < 0.01) for respiratory and gastrointestinal disease, and lambs from HSe-HI ewes had the smallest (P < 0.01) 24-h serum IgG concentration. In Exp. 1, lambs from HI ewes also had the greatest (P < 0.01) mortality rates from birth to weaning compared with lambs from CON and RES ewes. In Exp. 2, there was an effect (P < 0.01) of maternal plane of nutrition with lambs from RES ewes having increased 24-h IgG compared with lambs from CON and HI ewes. There was no effect of maternal Se supplementation on lamb 24-h IgG in Exp. 2; however, there was a Se × plane of nutrition interaction (P < 0.01) for morbidity. From birth to 21 d of age, lambs from ASe-CON ewes had fewer (P < 0.01) treatment days compared with lambs from any of the other treatment groups. There also tended (P = 0.08) to be an effect of maternal Se supplementation on lamb mortality with increased mortality observed in lambs from HSe ewes. Results from the studies show a restricted maternal plane of nutrition can increase lamb serum IgG concentration. Selenium results were not consistent between the 2 experiments and may be due to differences in maternal Se.  相似文献

High fractional concentrations of inspired oxygen (FiO(2)) delivered over prolonged periods produce characteristic histological changes in the lungs and airway of exposed animals. Modern medical anaesthetic machines are adapted to deliver medical air (FiO(2)=0.21) for the purpose of reducing FiO(2); anaesthetic machines designed for the veterinary market have not been so adapted. Two inexpensive modifications that allow medical air to be added to the gas flow from veterinary anaesthetic machines are described. We previously described successful treatment, including surgical drainage, of a Streptococcus equi subspecies equi brain abscess that caused severe neurological deficits in a 7-year-old Quarter Horse mare. This report details the long-term successful outcome of the case, findings of a magnetic resonance imaging (MRI) study performed 14 years after surgery and necropsy findings 18 years after initial treatment. Despite persistent cerebral and midbrain lesions detected by MRI and at necropsy, the mare returned to serviceable function within a year of initial treatment and had a successful performance career for over 10 years until carpal arthritis prompted retirement. This case demonstrates that brain abscess in horses can be successfully managed by combined medical and surgical treatment.  相似文献

Various glues and adhesives were tested for their ability to remove the unabsorbed residue from drops of foliar applied radioactive nutrients to Ficus benjamina L. and Glycine max Merr. Most glues/adhesives adequately removed the unabsorbed residue from nutrients that dried forming a salt deposit on the leaf surface, but only 1.6–3.2% agar, Rhoplex 33, and Elmers Glue removed greater than 95% of the residue from nutrients that formed a moist hygroscopic residue on the leaf surface. Adding 25 mM KH2PO4 to 3.2% agar, followed by swabbing the treated area with lN HCl‐1% Liquinox detergent removed all unabsorbed residue from monovalent and divalent cationic an anionic nutrients including heavy metals. Methods to assay foliar absorption were compared by applying “Rb phosphate to leaves of intact plants, excised leaves and leaf discs. Excised leaves with their petiole or stem bases in water behaved similar to intact plants, whereas excised leaves or discs maintained in in a moist atmosphere behaved atypically due to slower and incomplete drying compared to intact plants.  相似文献

Extraction of Fe from fresh leaves with 0.1 N HCl proved to be a better indicator of the Fe status of a variety of ornamental tropical foliage and flowering plants compared to total Fe, 0.1 N HCl‐ether, and IN HCl extraction. It consistently gave higher correlations (r = 0.73 to 0.95 depending on the species) with chlorophyll concentration than the other methods tested. However, even 0.1 N HCl extraction did not distinguish levels of Fe deficiency accurately when compared between species.  相似文献

In a series of experiments, plants of Salvia splendens ’St. John's Fire’ were sprayed to run‐off with NH4NO3 solutions that varied from 0.10–1.87 M (0.8–15.0% NH4NO3). Damage to young fully expanded leaves was determined by measuring percent visually estimated necrosis, leakage of UV (264 nm) absorbing constituents, K, Ca and phosphate, and post‐treatment dry weight:fresh weight ratio. The average correlation coefficient of percent visually estimated necrosis was 0.992 with leakage of UV absorbing constituents, 0.956 with leakage of phosphate, 0.951 with dry weightfresh weight ratio, 0.919 with leakage of Ca and 0.741 with leakage of K. Leakage of UV absorbing constituents consistently provided the best correlation with the visual degree of leaf burn.  相似文献

For dicots, bicarbonate (HCO3‐) is regarded as a main factor in the induction of iron (Fe) chlorosis in calcareous soils, and sand and solution culture. In sand culture experiments, peach [Prunus persica (Batsch) L.] rootstock developed chlorosis only when HCO3‐ levels were equal to or higher than 6 mM. Above this level, chlorosis increaeed as HCO3‐ level was increased. In spite of the lack of chlorosis at to or below 6 mM of HCO3‐, large growth reductions (40–60% reduction in fresh shoot weight) were seen in all rootstocks, although the tolerant rootstock had less reduction than the more susceptible rootstocks. Shoot growth was affected by HCO3‐ more than was root growth.  相似文献
