The present study identified a linear B-cell epitope in the Eastern equine encephalitis virus (EEEV) E2 glycoprotein by screening a phage-displayed random 12-mer peptide library using an EEEV E2 specific monoclonal antibody (mAb) 7C11 and defined L/F-E/R-Y-T-W-G/R-N-H/W-P as the consensus binding motif. A sequence ((321)EGLEYTWGNHPP(332)) encompassing this consensus motif was found in the EEEV E2 glycoprotein and synthesized for further epitope confirmation. Meanwhile, the corresponding epitope peptides in E2 protein of associated alphaviruses were synthesized for specificity identification. Results showed the mAb 7C11 and murine antisera all reacted strongly against the synthesized polypeptide of EEEV antigen complex, but no reaction with Western equine encephalitis virus (WEEV) and Venezuelan equine encephalitis virus (VEEV) was detected. The knowledge and reagents generated in this study may have potential applications in differential diagnosis and the development of epitope-based marker vaccines against EEEV.  相似文献

水貂绿脓杆菌(PASD03株)鞭毛蛋白的提取与分析   总被引:1,自引:0,他引:1  
本试验以水貂绿脓杆菌PASD03株为研究对象,对其鞭毛蛋白的提取方法进行研究,分别通过酸裂解、差速离心以及酸裂解结合机械振荡三种方法,用(NH4)2SO4盐析来获得鞭毛蛋白,采用垂直板聚丙烯酰胺凝胶电泳(SDS-PAGE)对提取的绿脓杆菌鞭毛蛋白进行分离纯化,并进行电镜鉴定。结果显示:成功分离出水貂绿脓杆菌的鞭毛蛋白,获得了绿脓杆菌鞭毛蛋白的初提液.绿脓杆菌鞭毛蛋白经SDS-PAGE提示纯化的鞭毛蛋白为一条相对分子质量(Mr)为53×103蛋白带,透射电镜(SEM)观察发现该鞭毛蛋白呈丝状。结论:经酸裂解法并结合机械振荡和硫酸铵沉淀法简便、耗时短、纯度高、产量高,适用于绿脓杆菌鞭毛蛋白的提取。  相似文献

  1. The effects of different fibre sources on the growth performance, carcass characteristics and gastrointestinal tract development were studied in growing Greylag geese (Anser anser).

  2. Four experimental diets were formulated with corn (maize) straw silage (CSS), steam-exploded corn (maize) straw, steam-exploded wheat straw, and steam-exploded rice straw as fibre sources. A total of 224 male Greylag geese at 28 d of age were randomly assigned to one of the 4 experimental diets.

  3. The birds fed on the CSS diets had higher average daily feed intakes than those fed on the steam-exploded straws. However, the 4 treatments had similar average daily gain, which contributed to significant differences in feed conversion ratios. The different fibre sources had no significant effects on the carcass characteristics.

  4. The CSS-fed birds had larger gizzards and lower relative length of the caeca than the other three groups. However, the relative weights and lengths of the other gut segments, the relative weights of major organs and the pH values of the gastrointestinal contents were similar between the 4 treatments.

  5. It was concluded that straw fibres with different physico-chemical properties exerted an effect on daily feed intake and gastrointestinal development, especially for the gizzard. The pretreatment of straw had a large effect on utilisation efficiency and animal performance. Steam explosion is a promising straw pretreatment for inclusion in diets for geese.

诊断敏感性和特异性是诊断方法验证过程中的两个重要参数。本文通过分析和比较,详细阐述了诊断敏感性和特异性在动物检疫工作中的重要性,厘清了其与分析敏感性和特异性的区别。通过计算分析比较了不同流行率对检测结果可信度的影响,以及在同一流行率下,诊断敏感性和特异性变化对检测结果可信度的影响。结果显示,当流行率低时,诊断特异性对结果可信度的影响更大,诊断特异性的降低会导致阳性检测结果可信度显著下降。因此在进出境动物检疫工作中,不要一味强调分析特性,而要重视诊断特性;既要关注诊断敏感性,做到不漏检;也要关注诊断特异性,做到不误检。  相似文献

Interferon (IFN)-stimulated gene (ISG) 56 family (composed of ISG54, ISG56, ISG58, and ISG60) plays important roles in defense against viral infection in mammalian cells. Numerous studies have been conducted on ISG54, ISG56, and ISG60; however, little is known on ISG58. In the present study, the upstream sequence of porcine ISG58 gene was first characterized as functional promoter by luciferase reporter assay, and then two directly adjacent IFN-stimulated response elements (ISREs), one at ?206 to ?194 (ISRE-I) and a second one, directly upstream of this element at ?219 to ?207?bp (ISRE-II), were identified using the bioinformatics method. The subsequent site-directed deletion and transient transfection experiments showed the candidate ISREs are functional. ISRE-I works better than ISRE-II and synergistic cooperation exists between two ISREs. Additionally, the effect of porcine ISG58 on activation of NF-κB was analyzed using the dual-luciferase reporter assay. The results will contribute to revealing the role of ISG58 in immune response.  相似文献

The objective of this study was to determine the effect of ensiling different ratios of whole crop oat to lucerne on fermentation quality, aerobic stability and in vitro digestibility of silage on the Tibetan plateau. Four experimental treatments were produced varying in the ratio of forages on a fresh matter (FM) basis: 1) 100% oat (control, dry matter (DM) content: 317 g/kg), 2) 90% oat + 10% lucerne (OL10, DM content: 316 g/kg), 3) 80% oat+ 20% lucerne (OL20, DM content: 317 g/kg) and 4) 70% oat+ 30% lucerne (OL30, DM content: 318 g/kg). All treatments were packed into laboratory‐scale silos and ensiled for 60 days and then subjected to an aerobic stability test for 15 days. Further, the four experimental treatments were incubated in vitro with buffered rumen fluid to study the nutrient digestibility. All silages were well preserved with low pH and NH3‐N contents, and high lactic acid contents and V‐scores (evaluation of silage quality). Increasing the lucerne proportion increased (p < 0.05) crude protein (CP) content of silage, whereas neutral (NDF) and acid (ADF) detergent fibre contents were not affected. Under aerobic conditions, the control silage showed higher (p < 0.05) yeast counts (>10cfu/g FM) followed by OL10 silage, and OL10 silage improved aerobic stability for 74 h. OL20 and OL30 silages showed fewer (p < 0.05) yeasts (<105 cfu/g FM) and markedly (p < 0.05) improved the aerobic stability (>360 h). After 48‐h incubation, OL30 silage increased (p < 0.05) in vitro dry matter digestibility (IVDMD) and neutral detergent fibre digestibility (IVNDFD) compared with the control silage. These results suggest that replacing oat with lucerne had no unfavourable effects on fermentation quality of silage, but improved CP content, aerobic stability IVDMD and IVNDFD. 家蚕黑尾病主要是由曲霉菌在蚕尾部寄生引起的一种真菌病 ,从病蚕中分离到病原菌有两种 ,一种孢子表面棘多而长、黄褐色 ,孢子大 ,直径 4~ 6 μm ;一种孢子表面棘短而少、蓝绿色 ,孢子小 ,直径 2~ 3μm。黑尾病在高温多湿条件下多发 ,多发生于 4龄中期至 5龄中期。黑尾病病原菌侵染寄生家蚕的过程是 :病原菌分生孢子附着在蚕尾部背板与节间的三角形节间膜处 ,分生孢子发芽侵入蚕体产生黑色小病斑 ,随病情加重病斑加大 ,甚至在病斑处出现节间断裂 ,直至形成黑尾症状。使用药剂防治黑尾病以 5 %亚迪蚕保最佳  相似文献
