The influence of broodstock dietary lipids on egg quality and egg fatty acid composition throughout the spawning season of gilthead seabream was investigated. For this purpose, the fish were fed for 7 months either a control diet (diet C) or a diet deficient in n−3 highly unsaturated fatty acids (n−3 HUFA) but rich in both oleic (18:1n−9) and linolenic (18:3n−3) acids (diet D). Eggs spawned by both groups of fish were sampled at the beginning, middle and end of the spawning season and the fatty acid composition of their neutral (NL) and polar lipids (PL) determined. In the early season, percentages of fertilized and hatched eggs, relative proportions of NL and PL as well as their fatty acid compositions, were not affected by the lipid composition of the broodstock diet. However, the eggs spawned during the middle and late seasons showed marked differences among the two groups of fish, clearly reflecting the influence of dietary fatty acids. This influence was more evident in the neutral lipid fraction than in the polar lipids. No correlation was found between the number of buoyant eggs and eicosapentaenoic (20:5n−3, EPA), docosahexaenoic (22:6n−3, DHA) fatty acids or total n−3 HUFA contents in egg phospholipids. However, a negative correlation was detected when percentages of fertilized eggs were compared with the levels of 18:1n−9, 18:3n−3 and with the ratio 18:1n−9/n−3 HUFA present in the phospholipids. Our results indicate the importance of maintaining not only the level of n−3 HUFA in egg membrane phospholipids, but also the balance between n−3 HUFA and other fatty acids such as 18:1n−9 and 18:3n−3, in order to obtain a high spawning quality.  相似文献

ABSTRACT:   High-pressure technology is used as an alternative to heat processing because of its inactivating effect on microorganisms and enzymes. However, it can also alter the structure of other muscle proteins. The present study compares the effects of high pressure (300 MPa, 7°C, 20 min) on the proteolytic degradation and alterations in the myofibrillar proteins of sardine and blue whiting muscle. Also, muscle homogenates and enzyme extracts were pressurized in order to evaluate the high-pressure effects on unprotected proteolytic enzymes outside the whole muscle structure. Peak proteolytic activity was found to occur at 55°C in both species. The peak activity pH was pH 3 for the sardine and pH 8 for the blue whiting; the main enzyme families being aspartic proteases in the former and alkaline serine proteases in the latter. Pressurization lowered activity levels at the peak activity pH and temperature in the fish muscle (by 30.8% in the sardine and by 9.5% in the blue whiting) and also slightly in the enzyme extracts (by 16.8% in the sardine and by 19.4% in the blue whiting). The electrophoretic profiles disclosed higher protein degradation in the pressurized muscle. Overall, the observed changes in proteolytic activity can be attributed not only to the effect of high pressure on the enzymes, but also and mainly, to the effect on other muscle proteins.  相似文献

The increasing economic importance of fish parasitoses for aquaculture and fisheries has enhanced the interest in the defence mechanisms against these infections. Both innate and adaptive immune responses are mounted by fish to control parasite infections, and several mechanisms described for mammalian parasitoses have also been demonstrated in teleosts. Innate immune initiation relies on the recognition of pathogen-associated molecular patterns (PAMPs) by pathogen recognizing receptors (PRRs). A number of PRRs, mainly Toll-like receptors (TLRs), have been characterized in fish, and some molecules susceptible of functioning as PAMPs are known for some fish parasites. A lectin-carbohydrate interaction has also been described in some host fish-parasite systems, thus probably involving C-type lectin receptors. Inflammatory reactions involving cellular reactions, as phagocytosis and phagocyte activity (including oxidative mechanisms), as well as complement activity, are modulated by many fish parasites, including mainly ciliates, flagellates and myxozoans. Besides complement, a number of humoral immune factors (peroxidases, lysozyme, acute-phase proteins) are also implicated in the response to some parasites. Among adaptive responses, most data deal with the presence of B lymphocytes and the production of specific antibodies (Abs). Although an increasing number of T-cell markers have been described for teleosts, the specific characterization of those involved in their response is far from being obtained. Gene expression studies have demonstrated the involvement of other mediators of the innate and adaptive responses, i.e., cytokines [interleukins (IL-1, IL-8), tumor necrosis factor (TNF), interferon (IFN)], chemokines (CXC, CC), as well as several oxidative enzymes [inducible nitric oxide synthase (iNOS), cyclo-oxygenase 2 (COX-2)]. Information is scarcer for factors more directly linked to adaptive responses, such as major histocompatibility (MH) receptors, T cell receptors (TCRs) and IgM. Expression of some immune genes varied according to the phase of infection, and proinflammatory cytokines were mainly activated in the early stages. Gene expression was generally higher in the target tissues for some skin and gill parasites, as Ichthyophthirius multifiliis, Neoparamoeba spp. and Lepeophtheirus salmonis, thus confirming the relevance of mucosal immunity in these infections. The existence of protective responses has been demonstrated for several fish parasites, both in natural infections and in immunization studies. Most information on the mechanisms involved in protection deals with the production of specific Abs. Nevertheless, their levels are not always correlated to protection, and the precise involvement of immune mechanisms in the response is unknown in many cases. No commercial vaccine is currently available for piscine parasitoses, although experimental vaccines have been assayed against I. multifiliis, Cryptobia salmositica and scuticociliates. The known information points to the need for integrated studies of the mechanisms involved in protection, in order to choose the optimum antigen candidates, adjuvants and formulations.  相似文献

In vineyards in Spain, tillage and semiarid Mediterranean climatic conditions accelerate organic matter loss from the soil. Cover crops are a conservation management practice that can provoke changes in soil quality which requires evaluation. Stratification ratios of soil properties such as soil organic C and labile C fractions have been proposed for the assessment of soil quality under different soil management systems. Our objective was to study the effect of different cover crop management on various soil parameters and their stratification ratios. We evaluated three different soil managements in a Typic Haploxerept from NE Spain: conventional tillage (CT); 5‐y continuous cover crop of resident vegetation (RV); and 4‐y continuous cover crop of Festuca longifolia Thuill., followed by 1‐y Bromus catharticus L. after resowing (BV). We monitored soil organic C, particulate organic C, water soluble C, potentially mineralizable N, microbial biomass C, β‐glucosidase and urease enzymatic activities, and water stable aggregates at 0–2.5, 2.5–5, 5–15, 15–25, and 25–45 cm soil depths. We calculated soil depth stratification ratios of those soil properties. Resident cover crop increased microbiological properties, labile C fractions, and aggregation with respect to conventional tillage at 0–2.5 and 2.5–5 cm soil depths. However, for Bromus cover crop the same soil properties were lower than for the resident cover crop at 0–2.5 cm depth. Stratification ratios of β‐glucosidase and urease enzymatic activities, and particulate organic C showed a higher sensitivity than other soil properties; therefore, they would be the best indicators for soil quality assessment in semiarid Mediterranean vineyards.  相似文献

The aim of this study was to evaluate the effect of body weight (BW) change during the early postpartum period and BW on day 50 post partum on the subsequent expression of estrus and fertility at first insemination before day 90 post partum. The data were derived from 1036 postpartum cows reaching 90 days in milk. The following data were recorded for each animal: lactation number, daily milk production at day 50 post partum, calving date, days in milk at estrus and at AI, insemination date, insemination number, inseminating bull, AI technician and body weight at calving, at day 50 post partum and at AI. Of the 1036 cows registered, 817 (78.9%) showed first estrus before day 90 post partum and were inseminated. Using logistic regression procedures and based on the odds ratio a one unit (kg) increase in the daily milk production at day 50 post partum and a one unit (kg) decrease in the BW between calving and day 50 post partum were related to a 1.03-fold increase and a 0.97-fold decrease in the estrus expression rate before day 90 post partum, respectively. Of the 817 cows inseminated before day 90 post partum, 437 became pregnant (53.5%). A one unit increase in the BW at day 50 post partum produced a 1.003-fold increase in the early fertility rate. Pregnancy rate before day 90 was also influenced by the season at calving, bull providing semen and technician. In conclusion, the register of BW during the postpartum period may be a useful tool for evaluating the nutritional status and its relationship with the subsequent reproductive efficiency in dairy cows.  相似文献

Most research on the extraction of high-priced compounds from vineyard/wine byproducts has traditionally been focused on grape seeds and skins as raw materials. Vine-shoots can represent an additional source to those materials, the characteristics of which could depend on the cultivar. A comparative study of hydroalcoholic extracts from 18 different vineyard cultivars obtained by superheated liquid extraction (SHLE), microwave-assisted extraction (MAE), and ultrasound-assisted extraction (USAE) is here presented. The optimal working conditions for each type of extraction have been investigated by using multivariate experimental designs to maximize the yield of total phenolic compounds, measured by the Folin-Ciocalteu method, and control hydroxymethylfurfural because of the organoleptic properties of furanic derivatives and toxicity at given levels. The best values found for the influential variables on each extraction method were 80% (v/v) aqueous ethanol at pH 3, 180 °C, and 60 min for SHLE; 140 W and 5 min microwave irradiation for MAE; and 280 W, 50% duty cycle, and 7.5 min extraction for USAE. SHLE reported better extraction efficiencies as compared to the other two approaches, supporting the utility of SHLE for scaling-up the process. The extracts were dried in a rotary evaporator, reconstituted in 5 mL of methanol, and finally subjected to liquid-liquid extraction with n-hexane to remove nonpolar compounds that could complicate chromatographic separation. The methanolic fractions were analyzed by both LC-DAD and LC-TOF/MS, and the differences in composition according to the extraction conditions were studied. Compounds usually present in commercial wood extracts (mainly benzoic and hydroxycinnamic acids and aldehydes) were detected in vine-shoot extracts.  相似文献

Gains and losses of soil carbon (C), have been reported when tropical forests are converted to pastures. Regional studies are crucial for setting regional baselines and explaining each particular trend, in order to solve this controversy. Tropical deciduous forest (TDF) is under high deforestation pressure, mainly for conversion to pastures. The present study compared soil organic C (SOC) and nitrogen (SON) in the surface layer (0–5 cm) of forest and pasture soils in a TDF of western Mexico. SOC and SON concentrations were 18 and 60% lower in pasture soils than in forest soils, and C:N ratio increased in pasture soils. Furthermore, pasture soils had lower labile C and available inorganic nitrogen (N) than forest soils. These results can be explained as a reduction in C inputs to pasture soils and management-induced disruption of soil aggregates. In forest soils, macroaggregates (> 250 μm) were predominant (85%), whereas in pasture soils they were reduced to 35% of dry sand-free soil mass. The estimated SOC and SON losses from the top 5 cm of soil were 3 Mg C ha−1 and 0.9 Mg N ha−1, respectively.  相似文献

Thermal stabilities of main enzymes involved in the biosynthesis of virgin olive oil (VOO) aroma through the lipoxygenase (LOX) pathway were studied in crude enzymatic preparations. Kinetic parameters of thermal inactivation for LOX were determined graphically and were shown to be compatible with the presence of two LOX isoenzymes (LOXlab and LOXres) having different thermal stabilities and displaying relative activities of 88 and 12% each. Data on hydroperoxide lyase (HPL) suggest the existence of just one HPL isoform. Thermal stabilities of LOX and HPL enzymatic activities in crude preparations seem to explain the observed decrease of volatile contents in VOO aroma as a consequence of heat treatments of olive fruit. Moreover, differences in thermal stability of LOXlab and LOXres would justify the distinct pattern of reduction of C6 and C5 compound contents observed in the aroma of these oils.  相似文献

Controlled atmosphere storage technology to extend the shelf life of "Reina de los Valles" wild strawberry fruit (Fragaria vesca L.) was studied. Fruits were stored at 3 degrees C for three weeks in different atmosphere compositions: 0.05% CO2/21% O2 (air), 3% CO2/18% O2, 6% CO2/15% O2, 10% CO2/11% O2, and 15% CO2/6% O2. The effect of gas composition on soluble solids content, titrable acidity, pH, off-flavor, aroma volatiles, and consumer preference was monitored. The result showed that the 10% CO2/11% O2 combination can efficiently prolong the shelf life of wild strawberries by maintaining the quality parameters within acceptable values, through inhibiting the development of Botrytis cinerea, without significantly modifying consumer acceptance.  相似文献
