Aβ (beta-amyloid peptide) is an important contributor to Alzheimer's disease (AD). We modeled Aβ toxicity in yeast by directing the peptide to the secretory pathway. A genome-wide screen for toxicity modifiers identified the yeast homolog of phosphatidylinositol binding clathrin assembly protein (PICALM) and other endocytic factors connected to AD whose relationship to Aβ was previously unknown. The factors identified in yeast modified Aβ toxicity in glutamatergic neurons of Caenorhabditis elegans and in primary rat cortical neurons. In yeast, Aβ impaired the endocytic trafficking of a plasma membrane receptor, which was ameliorated by endocytic pathway factors identified in the yeast screen. Thus, links between Aβ, endocytosis, and human AD risk factors can be ascertained with yeast as a model system.  相似文献

Aluminum is considered to approach an "ideal" metal or free electron gas. The valence electrons move freely, as if unaffected by the presence of the metal ions. Therefore, the electron redistribution due to chemical bonding is subtle and has proven extremely difficult to determine. Experimental measurements and ab initio calculations have yielded substantially different results. We applied quantitative convergent-beam electron diffraction to aluminum to provide an experimental determination of the bonding electron distribution. Calculation of the electron distribution based on density functional theory is shown to be in close agreement. Our results yield an accurate quantitative correlation between the anisotropic elastic properties of aluminum and the bonding electron and electrostatic potential distributions.  相似文献

Shorebirds are declining worldwide due to loss and degradation of critical breeding and wintering habitats. Some human-modified habitats, particularly salt-pans which are used by shorebirds in many regions of the world, may help substitute for natural habitats lost for a wide range of species during migration. We studied the influence of landscape characteristics on species richness, abundance, and diversity of shorebirds at 20 sites covering most of the Inner Gulf of Thailand, a landscape with a long history of salt farming. Sites with salt-pans present held significantly higher species richness, abundance and diversity of shorebirds. Areas with larger proportions given over to aquaculture tended to have lower species richness, abundance and diversity. Generalized additive models indicated that landscapes with a larger proportion of tidal flats in conjunction with salt-pans were the best predictors of sites with higher species richness, abundance and diversity. Landscape configurations with higher richness, abundance and diversity of shorebirds also tended to be less fragmented and contained slightly larger patches. Shorebirds appeared to use ponds with exposed mud in salt-pans as both roosting sites and supplementary feeding grounds during high tide. Traditional salt-pans therefore proved to contribute significantly to maintenance of overwintering shorebird populations in this landscape and should be investigated elsewhere in Asian coastal zones. Collaboration between researchers, salt farmers and planning authorities as to how best to maintain salt-pans as potential shorebird roost sites such as in the Inner Gulf of Thailand is urgently needed in order to maintain habitat for shorebird populations in critical wintering and staging areas of this flyway.  相似文献

The fate of Amazonian forest fragments: A 32-year investigation   总被引:2,自引:0,他引:2  
We synthesize findings to date from the world’s largest and longest-running experimental study of habitat fragmentation, located in central Amazonia. Over the past 32 years, Amazonian forest fragments ranging from 1 to 100 ha have experienced a wide array of ecological changes. Edge effects have been a dominant driver of fragment dynamics, strongly affecting forest microclimate, tree mortality, carbon storage, fauna, and other aspects of fragment ecology. However, edge-effect intensity varies markedly in space and time, and is influenced by factors such as edge age, the number of nearby edges, and the adjoining matrix of modified vegetation surrounding fragments. In our study area, the matrix has changed markedly over the course of the study (evolving from large cattle pastures to mosaics of abandoned pasture and regrowth forest) and this in turn has strongly influenced fragment dynamics and faunal persistence. Rare weather events, especially windstorms and droughts, have further altered fragment ecology. In general, populations and communities of species in fragments are hyperdynamic relative to nearby intact forest. Some edge and fragment-isolation effects have declined with a partial recovery of secondary forests around fragments, but other changes, such as altered patterns of tree recruitment, are ongoing. Fragments are highly sensitive to external vicissitudes, and even small changes in local land-management practices may drive fragmented ecosystems in markedly different directions. The effects of fragmentation are likely to interact synergistically with other anthropogenic threats such as logging, hunting, and especially fire, creating an even greater peril for the Amazonian biota.  相似文献

Denitrification has the potential to remove excess nitrogen from groundwater passing through riparian buffers, thus improving water quality downstream. In regions with markedly seasonal precipitation, transient stream flow events may be important in saturating adjacent floodplain soils and intermittently providing the anaerobic conditions necessary for denitrification to occur. In two experiments we characterised the denitrification potential of soils from two contrasting floodplains that experience intermittent saturation. We quantified under controlled laboratory conditions: 1) potential rates of denitrification in these soils with depth and over time, for a typical period of saturation; and 2) the influences on rates of nitrate and organic carbon. Treatments differed between experiments, but in each case soil-water slurries were incubated anaerobically with differing amendments of organic carbon and nitrate; denitrification rates were measured at selected time intervals by the acetylene-block technique; and slurry filtrates were analysed for various chemical constituents. In the first experiment (ephemeral tributary), denitrification was evident in soils from both depths (0-0.3 m; 0.3-1.1 m) within hours of saturation. Before Day 2, mean denitrification rates at each depth were generally comparable, irrespective of added substrates; mean rates (Days 0 and 1) were 5.2 ± 0.3 mg N kg dry soil−1 day−1 (0-0.3 m) and 1.6 ± 0.2 mg N kg dry soil−1 day−1 (0.3-1.1 m). Rates generally peaked on Days 2 or 3. The availability of labile organic carbon was a major constraint on denitrification in these soils. Acetate addition greatly increased rates, reaching a maximum in ephemeral floodplain soils of 17.4 ± 1.8 mg N kg dry soil−1 day−1 on Day 2: in one deep-soil treatment (low nitrate) this overcame differences in rates observed with depth when acetate was not added, although the rate increase in the other deep-soil treatment (high nitrate) was significantly less (P ≤ 0.01). Without acetate, peak denitrification rates in this experiment were 6.9 ± 0.4 and 2.8 ± 0.2 mg N kg dry soil−1 day−1 in surface and deep soils, respectively. Differences in rates were observed with depth on all occasions, despite similar initial concentrations of dissolved organic carbon (DOC) at both depths. Levels of substrate addition in the second experiment (perennial stream) more closely reflected natural conditions at the site. Mean denitrification rates were consistently much higher in surface soil (P ≤ 0.001), while the source of water used in the slurries (surface water or groundwater from the site) had little effect on rates at any depth. Mean rates when all treatments retained nitrate were: 4.5 ± 0.3 mg N kg dry soil−1 day−1 (0-0.3 m depth); 0.8 ± 0.3 mg N kg dry soil−1 day−1 (0.3-1.0 m); and 0.6 ± 0.1 mg N kg dry soil−1 day−1 (1.8-3.5 m). For comparable treatments and soil depths, denitrification potentials at both sites were similar, apart from higher initial rates in the ephemeral floodplain soils, probably associated with their higher DOC content and possibly also their history of more frequent saturation. The rapid onset of denitrification and the rates measured in these soils suggest there may be considerable potential for nitrate removal from groundwater in these floodplain environments during relatively short periods of saturation.  相似文献

The N extracted after chloroform (CHCl3) fumigation was determined as a possible index of soil N supply to plants. The relationships between extractable N following fumigation and reference indices such as total N, alkali-hydrolyzable N, N released by the Stanford short-term incubation method, and the N extracted by KCl and by CaCl2, were measured in nine soils of differing soil N supply capacity. A highly significant correlation was achieved between the extractable N released by fumigation and the N released by the Stanford method, i.e. a short-term aerobic incubation (r = 0.87). Similarly, the correlation between extractable N by fumigation and the N uptake by ryegrass was highly statistically significant (r = 0.93). Using the N extracted following fumigation has the advantage that laboratory results are available in two days and are both reproducible and of high precision. Therefore, the N extracted following fumigation is a valid, timesaving and precise index of soil N supply capacity.  相似文献

The aim of this study was to determine the prevalence of Tritrichomonas foetus infection and associated clinical signs in purebred cats in Germany, to investigate the role of co-infection, and identify determinants of infection. Faecal specimens accompanied by epidemiological questionnaires were scored and collected from 230 purebred cats. Faeces were examined for trichomonads and other enteroparasites. The prevalence of T foetus was 15.7% among cats and 18.5% among catteries. An abnormal faecal score and history of diarrhoea were observed in 64% and 61% of T foetus-positive cats, respectively, and correlated significantly with infection. Co-infection, observed in 36% of T foetus-infected cats, was not associated with diarrhoea. Norwegian Forest cats were infected significantly more often than other breeds. No association was found with any environmental factors. This study demonstrated a high prevalence of symptomatic T foetus infections in purebred cats in Germany. Co-infection with other enteroparasites did not worsen clinical signs of trichomonosis.  相似文献

A six-state, 5 year field project was initiated in 2006 to study weed management methods that foster the sustainability of genetically engineered (GE) glyphosate-resistant (GR) crop systems. The benchmark study field-scale experiments were initiated following a survey, conducted in the winter of 2005-2006, of farmer opinions on weed management practices and their views on GR weeds and management tactics. The main survey findings supported the premise that growers were generally less aware of the significance of evolved herbicide resistance and did not have a high recognition of the strong selection pressure from herbicides on the evolution of herbicide-resistant (HR) weeds. The results of the benchmark study survey indicated that there are educational challenges to implement sustainable GR-based crop systems and helped guide the development of the field-scale benchmark study. Paramount is the need to develop consistent and clearly articulated science-based management recommendations that enable farmers to reduce the potential for HR weeds. This paper provides background perspectives about the use of GR crops, the impact of these crops and an overview of different opinions about the use of GR crops on agriculture and society, as well as defining how the benchmark study will address these issues.  相似文献

All transgenic cultivars of potatoes registered in Canada and the United States have been modified to express a synthetic cry3A gene as a means of conferring resistance against the Colorado potato beetle, an important economic pest of potatoes. A PCR method was developed to amplify a 499 bp region of the synthetic cry3A gene. Using this method, synthetic cry3A could be detected in six different transgenic cultivars. Positive results could be confirmed with PvuII restriction digestion of the PCR-generated amplicon, which resulted in two fragments that were 283 and 216 bp in size. Of the 52 tuber extracts tested with this method, no false positive or false negative results were obtained, suggesting the method could be used with a high degree of accuracy. The absolute limit of detection was the number of cry3A copies present in one or perhaps two haploid copies of the potato genome. The practical limit of detection in tubers on a fresh weight basis was 0.02% for the NL 10-SUP and 0.01% for the remaining cultivars. Synthetic cry3A could also be detected in processed food products such as potato chips, shoestring potatoes, and frozen French fries. The method was suitable for screening potato tuber lots and some processed foods for the presence of synthetic cry3A.  相似文献

Arbuscular mycorrhizal fungi (AMF) are key components of ecosystems through their influence on plant communities and ecosystem processes. A major source of information regarding the importance of AMF species richness on process rates are mesocosm experiments using different levels of diversity of AMF as provided by single-species cultures of AMF. Since AMF inocula are generally made available in the form of non-sterile pot culture material, it is possible that AMF symbiosis-associated microbiota are at least partially responsible for some effects hitherto directly attributed to the AMF mycelium. Here, we provide evidence that microbiota associated with single-species cultures of AMF (after long-term pot culture enrichment of 7–8 years) can strongly affect the ecosystem process of soil aggregation. This effect occurred in an AMF isolate specific manner, but in the absence of live and active AMF mycelium. We additionally documented large differences in microbiota communities associated with the different AMF inocula (using PLFA analyses), suggesting that these differences were at least partly responsible for the observed changes in soil aggregation. This result points to AMF–microbiota interactions as a largely unexplored mechanism underlying soil aggregation (and potentially other ecosystem processes). We suggest that a reinterpretation of previous experiments using greenhouse-derived AMF cultures may be necessary, and the need to consider AMF symbiosis-associated microbiota in mechanistic studies of AMF and mycorrhizae in general is emphasized.  相似文献
