A radiolabelling method is generally used to determine the foliar uptake of xenobiotics. This technique cannot provide any information on the localization of chemicals inside leaf tissues. The influence of an alcohol ethoxylate surfactant on the uptake of three fluorescent dyes, difluorofluorescein (hydrophilic), rhodamine B (moderately lipophilic) and a naphthalimide dye (lipophilic), into the leaves of three contrasting species, bean (Vicia faba), wheat (Triticum aestivum) and cabbage (Brassica oleracea), at 16 h after treatment was measured using a conventional wash‐off method and also visualized in vivo by confocal laser scanning microscopy (CLSM). Whereas the lipophilic dyes showed greater intrinsic uptake than the hydrophilic one, the enhancing effect of the surfactant on uptake was more pronounced for the latter. CLSM revealed that the presence of the surfactant increased the transport of difluorofluorescein into the epidermal cells of bean and wheat leaves, but not cabbage leaves. Rhodamine B showed greatest transcuticular diffusion in all three species, but most of the dye moved into the vacuole of the epidermal cells. The naphthalimide dye was strongly retained by the wax–cuticle layer of all species, even in the presence of the surfactant. CLSM has proven to be an attractive tool for studying xenobiotic diffusion. The results obtained using fluorescent dyes are believed to be applicable to the foliar uptake of herbicides. 相似文献
In April 2003, persistent scouring and ill-thrift that was reported in calves form an intensive beef rearing operation in central Argentina despite treatments with benzimidazole and ivermectin. In order to conduct a controlled faecal egg count reduction test on this herd, 40 calves 5-8-months-old were selected on the basis that they had a nematode eggs per gram (epg) of faeces count greater than 150. Animals were divided into four groups (1-4) of 10 calves. Calves of groups 1-3 were treated, respectively, with subcutaneous injection of ivermectin (200 mcg/kg), ricobendazole (4 mg/kg) and levamisole (7.5 mg/kg), while calves of group 4 remained as untreated controls. The egg count reductions carried out 10 days later were lower than 15% in calves treated with ivermectin and ricobendazole, but 100% in animals receiving levamisole. Pooled post-treatment faecal cultures showed larval percentages of 92 and 95 for Haemonchus and 8 and 5 for Cooperia in the faeces of calves treated with ivermectin and ricobendazole, respectively. This is the first reported case of Haemonchus parasiting cattle showing simultaneous resistance to avermectins and benzimidazole type anthelmintics. 相似文献
Sainfoin (Onobrychis viciaefolia Scop.) is adapted to the calcareous soils of the southern Great Plains and can provide early season forage that does not induce bloating; however, little is known about performance by ruminants grazing sainfoin. Our objective was to determine the effect of plant growth stage and grazing pressures on potential animal production from sainfoin as predicted from energy intake as a multiple of maintenance. Nitrogen-fertilized (100 kg of N/ha) Renumex sainfoin was grown under irrigation on a Pullman clay loam (fine, mixed, thermic Torretic Paleustoll) near Lubbock, TX. Light (L), medium (M), and heavy (H) grazing pressures were applied with steers grazing sainfoin that was at the bud (B), flower (F), and seed shatter (S) stages of growth. The L, M, and H pressures were grazed to remove 50, 75, and 90% of the standing plant height. Across growth stages, L, M, and H grazing pressures averaged 52, 69, and 87% removal of pregrazed herbage mass. Dry matter intake as a percentage of BW of steers averaged 3.9, 2.8, and 1.7 for L, M, and H grazing pressures. Across growth stages, predicted live weight gain for L, M, and H grazing pressures averaged .86, .67, and .03 kg/d. Our findings indicate that the multiple of maintenance method may be useful for evaluating treatments from small-plot grazing experiments. 相似文献
Efforts at mitigating global biodiversity loss have often focused on preserving large, intact natural habitats. However, preserving biodiversity should also be an important goal in the urban environment, especially in highly urbanized areas where little natural habitat remains. Increasingly, research at the city/county scale as well as at the landscape scale reveals that urban areas can contain relatively high levels of biodiversity. Important percentages of species found in the surrounding natural habitat, including endangered species, have been found in the urban forest.
This contribution concisely highlights some examples of urban biodiversity research from various areas of the world. Key issues involved in understanding the patterns and processes that affect urban biodiversity, such as the urban–rural gradient and biotic homogenization, are addressed. The potential for urban areas to harbor considerable amounts of biodiversity needs to be recognized by city planners and urban foresters so that management practices that preserve and promote that diversity can be pursued. Management options should focus on increasing biodiversity in all aspects of the urban forest, from street trees to urban parks and woodlots. 相似文献
Besides infection in humans, Salmonella enteritidis can also cause serious illness in young chickens. However, the genetic and immunological parameters important for the disease in chickens are not well characterized. In this study, processes in the chicken intestine in response to a Salmonella infection were investigated in two different chicken lines. One-day-old chickens were orally infected with Salmonella. T-cell subpopulations, phagocytic properties of intestinal mononuclear cells and RNA expression levels of the jejunum were investigated. The two chicken lines differed in the amount of cfu in the liver and growth retardation after the infection. Differences in phagocytic activity of intestinal mononuclear cells were found between control and Salmonella infected chickens. The number of CD4+ T-cells of the intestine decreased after the Salmonella infection in one chicken line, while the number of CD8+ T-cells increased in both chicken lines, but the time post infection of this increase differed between the lines. In one chicken line the expression levels of the genes carboxypeptidase M and similar to ORF2 decreased after the Salmonella infection, which might be related to a decrease in the amount of macrophages. With the microarray, ten genes were found that were regulated in only one of the chicken lines, while we found six genes regulated in response to the infection in both chicken lines. So differences in genetic background of the chickens influence the intestinal host response of the Salmonella infection as observed by phagocytic activity, gene expression and changes in the number of T-cell subpopulations and macrophages. 相似文献
