Dimethyl sulphoxide (DMSO) at 14 mM inhibits CH4 oxidation in forest soil, but the inhibition mechanism is unknown. When soil slurries are incubated in gas tight flasks, there is a lag of about 45 h before DMSO inhibits CH4 oxidation. We tried to determine if some metabolic compound derived from DMSO, as a result of microbial activity, is responsible for the inhibition. Dimethyl sulphide (DMS) accumulated in the sealed flasks up to 5-83 μl l−1 in the headspace during a 2-week period. DMS at 1 μl l−1 in the headspace (0.64 μM in soil-water slurry) had a negligible effect on CH4 oxidation but 50 μl l−1 DMS (32 μM) inhibited CH4 oxidation completely. However, the inhibition by DMSO was already evident after 45 h, when DMS concentrations were generally non-inhibiting (0.1-0.7 μl l−1). DMSO was also shown to inhibit CH4 oxidation when the DMS produced was continuously removed. Results suggest that the production of DMS from DMSO makes a minor contribution to the inhibition of CH4 oxidation by DMSO with incubation times relevant in CH4 oxidation studies.  相似文献

Near-infrared spectroscopy and soil physicochemical determinations (pHH2O, organic matter content, total C content, NH inf4 sup+ , total N content, cation-exchange capacity, and base saturation) were used to characterize fire-or wood ash-treated humus samples. The spectroscopic and the soil physicochemical analysis data from the humus samples were used separately to explain observed variations in soil respiration and microbial biomass C by partial least-square regression. The first regression component obtained from the physicochemical and spectroscopic characterization explained 10–12% and 60–80% of the biological variation, respectively. This suggests that information on organic material collected from near-infrared spectra is very useful for explaining biological variations in forest humus.  相似文献

Sulfa antibiotics (sulfonamides) are derivatives of p-aminobenzenesulfonamide that are widely used in veterinary medicine. Foods derived from treated animals may be contaminated with these drugs. However, current immunobased sulfonamide detection methods are unfit for screening of products because they are either too insensitive or specific for a few compounds only. An immunoassay capable of detecting all sulfas in a single reaction would be ideal for screening. For development of a binder capable of binding all sulfas, a protein engineering approach was chosen and the properties of monoclonal antibody 27G3 were improved with mutagenesis followed by selection with phage display. Several different mutant antibodies were isolated. The cross-reaction profile of the best mutant antibody was significantly improved over that of the wild-type antibody: it was capable of binding 9 of the tested 13 sulfonamides within a narrow concentration range and also bound the rest of the sulfas, albeit within a wider concentration range.  相似文献

The antioxidant activity of berry phenolics (at concentrations of 1.4, 4.2, and 8.4 mug of purified extracts/mL of liposome sample) such as anthocyanins, ellagitannins, and proanthocyanidins from raspberry (Rubus idaeus), bilberry (Vaccinium myrtillus), lingonberry (Vaccinium vitis-idaea), and black currant (Ribes nigrum) was investigated in a lactalbumin-liposome system. The extent of protein oxidation was measured by determining the loss of tryptophan fluorescence and formation of protein carbonyl compounds and that of lipid oxidation by conjugated diene hydroperoxides and hexanal analyses. The antioxidant protection toward lipid oxidation was best provided by lingonberry and bilberry phenolics followed by black currant and raspberry phenolics. Bilberry and raspberry phenolics exhibited the best overall antioxidant activity toward protein oxidation. Proanthocyanidins, especially the dimeric and trimeric forms, in lingonberries were among the most active phenolic constituents toward both lipid and protein oxidation. In bilberries and black currants, anthocyanins contributed the most to the antioxidant effect by inhibiting the formation of both hexanal and protein carbonyls. In raspberries, ellagitannins were responsible for the antioxidant activity. While the antioxidant effect of berry proanthocyanidins and anthocyanins was dose-dependent, ellagitannins appeared to be equally active at all concentrations. In conclusion, berries are rich in monomeric and polymeric phenolic compounds providing protection toward both lipid and protein oxidation.  相似文献

We tested here a plant-soil system to separate recent, plant-derived and native, soil-derived carbon in soil respiration. The approach uses a perennial crop cultivated on an organic soil where upper soil layers have been removed as a result of peat extraction. There, the 14C signal from native organic matter is highly depleted compared to that in vegetation established at the site after peat extraction ceased. Radiocarbon was analyzed in carbon dioxide respired from soil over one growing season, and a two-pool isotope mixing model was applied to calculate the relative contribution of old vs. new carbon sources. The analysis showed that the approach is reliable for source partitioning with isotopes. After six years of cultivation, old peat decomposition contributed less to total soil respiration than respiration of recent plant material (30% vs. 70% on average, respectively), but the relative proportions were highly variable over the growing season. The approach offers a new possibility to follow the fate of old, native soil organic matter in highly organic soils.  相似文献

Objective To use the American College of Veterinary Internal Medicine (ACVIM) validation criteria to evaluate the performance of high definition oscillometric (HDO) and Doppler blood pressure measurement techniques against invasive blood pressure measurements in anaesthetized dogs. Study design Prospective clinical study. Animals Twenty client‐owned dogs. Materials and Methods Invasive blood pressure was measured using a catheter inserted into a pedal artery and an electronic transducer. The sites of cuff placement for the HDO measurements were the mid antebrachium or the proximal tail and, for the Doppler technique, the distal tibia. Agreement between invasive and non‐invasive blood pressure measurements was estimated by the Bland–Altman method. Results Only 10% and 34% of Doppler measurements were within 10 and 20 mmHg of invasive blood pressure values, respectively. The Doppler device failed to meet the ACVIM validation criteria for blood pressure measurement devices. The best agreement between HDO and invasive blood pressure measurement technique was observed for mean arterial blood pressure (MAP); 67% and 95% of readings were within 10 and 20 mmHg of invasive blood pressure values respectively. In addition, 52% and 87% of diastolic arterial blood pressure (DAP) measurements were within 10 and 20 mmHg of invasive readings. High definition oscillometric readings did not meet ACVIM recommended limits for SAP. Conclusion and clinical relevance The Doppler technique overestimated and the HDO device showed limited agreement with invasive blood pressure measurement in anaesthetized dogs. High definition oscillometry met most of the ACVIM requirements for MAP and DAP while the Doppler technique did not.  相似文献

Jigging with artificial lights (squid jigging) and deploying of large scale trap-net (also known as a set-net in Japan), are the major methods to capture Japanese common squid Todarodes pacificus in western Japan. Squid jigging is a highly selective fishing method. However, it consumes large amount of energy for steaming to the fishing ground and for lighting. In contrast, trap-net fishing requires substantially less energy but its capture efficiency is strongly influenced by its stationary mode of capture. The primary objective of this study was to analyze how various environmental and biological factors such as the lunar cycle, tidal condition, wind direction and squid abundance affect the capture efficiency of squid jigging and trap-net fishing. We analyzed the effect of these factors on squid catch in five Fisheries Cooperative Associations located on four islands in Nagasaki Prefecture, western Japan. Our analysis shows that squid catch in jigging and trap-net fishing is mainly influenced by the lunar cycle but also tide and wind direction play a marked role. In addition, squid abundance significantly affects the catches in trap-net fishing. Recommendations are made to improve the overall profitability of squid fishing by proper choice of the capture method, location and season.  相似文献
