Objective To determine the specificity of a snake venom detection kit in urine samples from dogs and cats presenting to a referral centre for diseases unrelated to snake envenomation. Design Urine was collected from 50 dog and 25 cats presented for investigation and treatment of diseases unrelated to snake envenomation. Urine was collected as a voided sample, by cystocentesis or by catheterisation, and routine urinanalysis was performed. Snake venom testing was performed within 2 h of collection using a commercially available snake venom detection kit, which was observed continuously during the 10-min colour reaction phase for evidence of a visible colour indicating a positive test. Results No false-positive reactions occurred in any sample analysed. Conclusion The snake venom detection kit appears to have 100% specificity for using urine as a test sample.  相似文献

The water dynamics of cropping systems containing mixtures of Gliricidia sepium (Jacq.) Walp trees with maize (Zea mays L.) and/or pigeonpea (Cajanus cajan L.) were examined during three consecutive cropping seasons. The trees were pruned before and during each cropping season, but were left unpruned after harvesting the maize; prunings were returned to the cropping area in all agroforestry systems to provide green leaf manure. The hypothesis was that regular severe pruning of the trees would minimise competition with crops for soil moisture and enhance their growth by providing additional nutrients. Neutron probe measurements were used to determine spatial and temporal changes in soil moisture content during the 1997/98, 1998/99 and 1999/00 cropping seasons for various cropping systems. These included gliricidia intercropped with maize, with and without pigeonpea, a maize + pigeonpea intercrop, sole maize, sole pigeonpea and sole gliricidia. Soil water content was measured to a depth of 150 cm in all treatments at 4–6 week intervals during the main cropping season and less frequently at other times. Competition for water was apparently not a critical factor in determining crop performance as rainfall exceeded potential evaporation during the cropping season in all years. The distribution of water in the soil profile was generally comparable in all cropping systems, implying there was no spatial complementarity in water abstraction by tree and crop roots. However, available soil water content at the beginning of the cropping season was generally lower in the tree-based systems, suggesting that the trees continued to deplete available soil water during the dry season. The results show that, under rainfall conditions typical of southern Malawi, the soil profile contains sufficient stored water during the dry season (ca. 75–125 mm) to support the growth of gliricidia and pigeonpea, and that gliricidia trees pruned before and during the cropping season did not deleteriously compete for water with associated crops. Water use efficiency also appeared to be higher in the tree-based systems than in the sole maize and maize + pigeonpea treatments, subject to the proviso that the calculations were based on changes in soil water content rather than absolute measurements of water uptake by the trees and crops.  相似文献

The effects of ploughing and previous crop on winter wheat pests and their natural enemies under an integrated farming system was investigated in an experimental area in Slovakia. The experiment consisted of 4 small plots (50?m2 each) to which both organic and inorganic fertilisers were applied. Moreover, there was one application of herbicide and a fungicide on each plot. The ploughing was either deep or shallow, and either silage-maize or pea was used as a previous crop. The effects of the ploughing and previous crop was studied by means of multivariate redundancy analysis (RDA) and subsequent Monte-Carlo permutation test. The results showed that there was a considerable temporal variation in the abundance of insect pests and their natural enemies. Of the two husbandry treatments, only ploughing caused significant difference in the composition and abundance of both pests and natural enemies. However, while most of the pests favoured deep ploughing, the majority of natural enemies occurred more abundantly on shallowly ploughed plots. The previous crop did not affect numbers of either pests or natural enemies. Two systems, integrated and organic, which have been surveyed previously, were mutually compared with respect to the abundance of insect pests. The organic farming system turned out to be more sustainable.  相似文献

Small-scale tree nurseries are important in fulfilling the goals of reforestation and agroforestry implementation schemes in Kenya and other developing countries. The focus in seedling production has been on quantity, instead of quality, but a change can be seen in recent tree nursery manuals. These manuals are emphasising morphological characteristics as tools for assessing potential field performance of seedlings. However, morphological criteria are debatable and their value is questioned. A survey was carried out among tree nursery operators in the Meru area, in the Eastern province of Kenya, to determine how operators perceived seedling vitality, and how they separated acceptable seedlings from those of poor vitality. Based on the survey, 3 pairs of criteria were chosen, size (tall versus small), colour (green versus yellowish), and sturdiness quotient (sturdy versus lanky). These criteria were tested on survival and growth in a field trial, a controlled bench trial, and in a root growth potential test. The results showed that the nursery operators were aware of quality differences in seedlings, but they did not cull accordingly. The results from the field trial showed that mango (Mangifera indica L.) performed poorly compared to grevillea (Grevillea robusta A. Cunn. ex. R. Br.), probably due to the high altitude. The altitudinal range for mango and grevillea are 0-1,200 m and 0-2,300 m, respectively, and the trial site was located on an altitude of 1,725 m. In grevillea, small seedlings grew better than tall in the field trial, but no differences could be found in the other trials. In mango, sturdy seedlings grew better than lanky ones in the field trial, while in the controlled trials tall seedlings grew better than small ones. The results showed that morphological characteristics as seedling quality assessment criteria could be unreliable as the effect differs with species and planting site.  相似文献

This study tested the hypothesis that incorporation of green leaf manure (GLM) from leguminous trees into agroforestry systems may provide a substitute for inorganic N fertilisers to enhance crop growth and yield. Temporal and spatial changes in soil nitrogen availability and use were monitored for various cropping systems in southern Malawi. These included Gliricidia sepium (Jacq.) Walp. trees intercropped with maize (Zea mays L.), with and without pigeonpea (Cajanus cajan L.), sole maize, sole pigeonpea, sole gliricidia and a maize + pigeonpea intercrop. Soil mineral N was determined before and during the 1997/1998, 1998/1999 and 1999/2000 cropping seasons. Total soil mineral N content (NO3 + NH4+) was greatest in the agroforestry systems (p<0.01). Pre-season soil mineral N content in the 0–20 cm horizon was greater in treatments containing trees (≤85 kg N ha−1) than in those without (<60 kg ha−1; p<0.01); however, soil mineral N content declined rapidly during the cropping season. Uptake of N was substantially greater in the agroforestry systems (200–270 kg N ha−1) than in the maize + pigeonpea and sole maize treatments (40–95 kg N ha−1; p<001). Accumulation of N by maize was greater in the agroforestry systems than in sole maize and maize + pigeonpea (p<0.01); grain accounted for 55% of N uptake by maize in the agroforestry systems, compared to 41–47% in sole maize and maize + pigeonpea. The agroforestry systems enhanced soil fertility because mineralisation of the applied GLM increased pre-season soil mineral N content. However, this could not be fully utilised as soil N declined rapidly at a time when maize was too small to act as a major sink for N. Methods for reducing losses of mineral N released from GLM are therefore required to enhance N availability during the later stages of the season when crop requirements are greatest. Soil mineral N levels and maize yields were similar in the gliricidia + maize and gliricidia + maize + pigeonpea treatments, implying that addition of pigeonpea to the tree-based system provided no additional improvement in soil fertility.  相似文献

Agroforestry, as one of integrated approaches to biodiversity conservation, has received attention. But the methods to evaluate this contribution are rare. The present study focuses on the method development for quantification of effects of agroforestry on biodiversity conservation. It includes identifying the functional groups of agroforestry components, defined as a set of species with similar impacts on a system process, and quantifying the distinctness of these groups, as related to wise use of resources and increasing well-being of local people. Combining with functional group-based Analytic Hierarchy Process (AHP) analysis, the present work gives a useful way for an integrated evaluation-planning of agroforestry management relative to biodiversity conservation. It also provides a means to improve our knowledge on the potential roles of agroforestry in biodiversity conservation of nature reserves. The analyses, using the distinctness index of functional groups, demonstrated the impacts of various land-use systems on biodiversity conservation. The analyses, using AHP, ranked the priorities of various agroforestry and plantations on the protection of nature forests, aiming at the long-term solution for the resource support. The present study suggests that agroforestry development for biodiversity conservation should take into account the impacts of functional groups in strategic planning.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献

The hypothesis that temporal separation of resource use between trees and crops minimises competition for wa ter in agroforestry systems during the cropping period and increases utilisation of annual rainfall was tested at Machakos in semi-arid Kenya. Four popular tree species were chosen to provide a range of leafing phenologies. These included Melia volkensii, which sheds its leaves twice a year, Senna spectabilis and Gliricidia sepium, which shed their leaves during the long dry season, and the evergreen Croton megalocarpus. All four species retained their foliage during the long rains, offering little scope for temporal separation of resource use. Maize (Zea mays) yields were reduced by 50–70% in the agroforestry treatments. Reductions in crop yield were strongly correlated with tree growth (r 2 =0.94) and available soil moisture (r 2 =0.88). G. sepium remained leafless for much of the short rains despite the presence of available soil water, and was least competitive with the bean crops (Phaseolus vulgaris) grown at this time. Reductions in crop yield in the agroforestry treatments were closely correlated with tree growth (r 2 =0.99) and available moisture (r 2 =0.79) during the 1996/97 short rains (158 mm), but not during the much wetter 1997/98 season (608 mm). Shading by trees or shade nets reduced crop yield, in contrast to previous studies in the semi-arid tropics. Low off-season rainfall during the study period (9% of annual rainfall compared to the long-term average of 20%) limited the potential for temporal separation of growing periods. Where the prospects for temporal or spatial separation in resource use are limited, shoot and/or root pruning may be necessary to manage competition between trees and crops. Being one of the most abundant and species‐rich groups of natural enemies occurring in all agroecosystems, spiders are variably affected by pesticide applications. Here, a review is given of research on spider ecotoxicology. More than 40 species of spiders and almost 130 pesticides (acaricides, insecticides, fungicides and herbicides) have been tested so far in the field or under laboratory conditions. Field studies show that the degree of population reduction following pesticide application is a function of a number of factors inherent to pesticides, crops and spider species (guilds). These studies also revealed indirect effects via habitat and prey disruption. Among laboratory studies, a number of papers have investigated only the direct lethal effect. A meta‐analysis of these data reveals that spiders are mainly affected by acaricides and insecticides, particularly neurotoxic substances. Currently, ecotoxicological research on spiders is focused more on direct sublethal effects on a variety of behavioural traits (locomotion, predation, web‐building, reproduction, development) and physiology. Yet a standardised approach to the evaluation of sublethal effects is lacking. A few studies have provided some evidence for hormesis in spiders. Future research should be more concentrated on sublethal effects and the estimation of long‐term changes in spider populations as a result of pesticide treatment. Apoptosis is an important determinant of the normal development of pre-implantation embryos in vitro. Recently, endoplasmic reticulum (ER) stress-mediated apoptosis has been extensively investigated in a wide variety of diseases. Efficient functioning of the ER is essential for most cellular activities and survival. Tauroursodeoxycholic acid (TUDCA), an endogenous bile acid, has been reported to attenuate ER stress-mediated cell death by interrupting the classic pathways of apoptosis. Therefore, in this study, the anti-apoptotic effect of TUDCA on ER stress-induced apoptosis was examined in pre-implantation pig embryos. Also, tunicamycin was used to investigate the effects of ER stress on pig embryo development. After in vitro maturation and fertilization, presumptive pig embryos were cultured in NCSU-23 medium supplemented with TUDCA or TM for 6 days at 39 °C, 5% CO(2) in air. All data were analysed using one-way anova and Duncan's multiple range test in the statistical analysis system (SAS). In addition, we also determined the optimal TM and TUDCA concentrations. Samples were treated with TM at concentrations of 0, 1, 2 or 5 μm and with TUDCA at concentrations of 0, 100, 200 or 300 μm. When TM was used during in vitro culture, only 8.2% (8/97) of the embryos developed to the blastocyst stage when the treatment concentration was 1 μm compared with 27.4% (28/102) of the embryos in the control group (p < 0.05). In contrast, the frequency of blastocyst formation and the number of cells were higher when treated with 200 μm TUDCA compared with the control group (32.8% and 39.5 vs 22.2% and 35.6, p < 0.05). Moreover, the developmental rate to the blastocyst stage embryo in the group treated with TM and TUDCA was not significantly different from that of the control group (17.8%, 26/142 vs 24.9%, 36/145). Furthermore, the blastocyst cell number was enhanced (31.9 vs 36.9) and apoptosis reduced (TUNEL-positive nuclei number, 6.0 vs 3.2) by TUDCA treatment in pig embryos. In the real-time quantitative RT-PCR analysis, the expression of anti-apoptotic Bcl-XL gene was shown to be increased in the blastocyst stage because of TUDCA treatment, whereas expression of pro-apoptotic Bax was decreased. In addition, we also found that TUDCA decreased the rate of TM-induced apoptosis in the pre-implantation stage. Taken together, our results indicate that TUDCA improves the developmental competence of pig embryos by modulating ER stress-induced apoptosis during the pre-implantation stage.  相似文献
