Gliadins, and primarily α-gliadins containing several sequences such as aa 31-49, aa 56-88 (33-mer), aa 57-68, and aa 69-82, are critical in the induction of immune response or toxic reaction leading to the development of celiac disease (CLD). The role of IgA anti-gliadin antibodies (IgA AGA) is unknown. To this end, we prepared several humanized monoclonal IgA AGA using transgenic α1KI mice. Employing Pepscan with overlapping decapeptides of α-gliadin we observed a robust similarity between the specificity of humanized mouse monoclonal IgA AGA and IgA AGA from patients with florid CLD. The common immunodominant region included several sequential epitopes localized in the N-terminal part of α-gliadin (QFQGQQQPFPPQQPYPQPQPFP, aa 29-50, and QPFPSQQPYLQL, aa 47-58). Notably, IgA AGA produced by clones 8D12, 15B9, 9D12, and 18E2 had significant reactivity against sequences localized in the 33-mer, LQLQPFPQPQ (aa 56-65) and PQLPYPQPQPFL (aa 69-80). Humanized mouse monoclonal IgA AGA that have a known specificity are suitable as standard in ELISAs to detect serum IgA AGA of CLD patients and for studying the AGA pathogenic role in CLD, especially for analyzing the translocation of complex of specific IgA antibodies and individual gliadin peptides through enterocyte barrier.  相似文献

Several strains of porcine bocaviruses have been reported worldwide since their first detection in Sweden in 2009. Subsequently, the virus has been reported to be associated with gastrointestinal and respiratory signs in weaner and grower pigs. Although Malaysia is host to a self-sufficient swine livestock industry, there is no study that describes porcine bocavirus in the country. This report is the first to describe porcine bocavirus (PBoV) in Malaysian swine herds. PBoV was identified in various tissues from sick and runt pigs using the conventional PCR method with primers targeting conserved regions encoding for the nonstructural protein (NS1) gene. Out of 103 samples tested from 17 pigs, 32 samples from 15 pigs were positive for porcine bocavirus. In addition, a higher detection rate was identified from mesenteric lymph nodes (52.9%), followed by tonsil (37.0%), and lungs (33.3%). Pairwise comparison and phylogenetic analyses based on a 658-bp fragment of NS1 gene revealed that the Malaysian PBoV strains are highly similar to PBoV3 isolated in Minnesota, USA. The presence of porcine bocavirus in Malaysia and their phylogenetic bond was marked for the first time by this study. Further studies will establish the molecular epidemiology of PBoV in Malaysia and clarify pathogenicity of the local isolates.  相似文献

An attempt was made to understand the physicochemical attributes that are the basis of physical differences between alkaline and salted noodle doughs. Flour and dough properties of one soft and three hard‐grained wheat cultivars were observed. Doughs were made with either sodium chloride or sodium carbonate. Each formulation variant was tested at both high and low water additions. Samples for glutenin macropolymer (GMP) isolation were taken at selected noodle dough processing stages. When a 1.67% w/v Na2CO3 solution was used for mixograph testing, dough characteristics were radically altered and differences between cultivars were masked. In lubricated squeezing flow (LSF) testing, hard wheat noodle doughs had significantly (P < 0.01) longer relaxation times and higher % residual force values than soft wheat doughs in both the salted and alkaline variants. LSF maximum force and biaxial viscosity were significantly higher in alkaline doughs than salted. GMP extracted from alkaline doughs was gummy and sticky, and was more opaque than GMP from salted doughs. GMP weight decreased sequentially when extracted from samples taken in the active phase (mix, compound, sheet) of noodle dough processing and decreased more in alkaline doughs. GMP weight increased more after 24 hr of dough rest in salted doughs. GMP gel strength was noticeably higher in GMP extracted from alkaline doughs. After dough resting, alkaline GMP gel strength significantly increased, whereas it decreased in GMP from salted doughs, suggesting a role for GMP in the increased stiffness of alkaline noodle doughs.  相似文献

Summary Three double-cut red clover (Trifolium pratense L.) varieties: Prosper I, Florex, and Tristan each with two different seedlots were space-planted in a field. The total number of plants established for each seedlot was about 900. In the seedling year, plants were classified into five growth types according to rosette development and flower production. In the fall of the third year 14% of the rosette-forming, non-flowering Type 1 plants were alive; 8–11% of the intermediate types survived; and only 2% of the nonrosette-forming, profusely flowering Type 5 plants remained growing. The non-flowering class (Types 1 and 2) persisted better than the flowering class (Types 3, 4, and 5). This difference in persistence was similar for all three varieties regardless of seed source. The two flowering classes, in the production years, had the same proportion of the most vigorous plants. The most vigorous plants of the non-flowering class had the same frequency distribution of seed yield as those of the non-flowering class. In conclusion, the flowering response can be used as a selection criterion for persistent plants; and plants not flowering in the seedling year can attain a forage and seed yield as high as flowering plants in the production years. The paper describes a tree/crop interface (TCI) experiment designed to investigate the effects of row orientation using Leucaena leucocephala Lam. Each TCI plot consisted of a regularly pruned Leucaena hedge in the middle and 12 crop rows on either side. Eight such plots were arranged at 45° around a sole Leucaena plot with rows oriented in four compass directions viz., North-South, East-West, Northeast-Southwest and Northwest-Southeast.Results of four years from 1984 to 1987 did not show any effect of row orientation, and similarly, no effect was seen on crop rows due to their location on the windward or leeward side of the hedge. The TCI effect was positive on the first crop row in the first year because Leucaena grew slowly, but depressed the yield of the first 4 to 6 crop rows(1.8 to 2.7 m from hedge) in subsequent years. The negative effect of Leucaena was noted more on sunflower in a relatively dry year than on sorghum in other years.Results from the TCI plots were used to estimate the yield of five hedgerow intercropping (HI) systems with varying alley widths (2.7 to 9.9 m). Comparison with sole stands of Leucaena and crops indicated that HI was more productive particularly at close alley widths. For example, hedges spaced at 2.7 m and 3.6 m averaged 37% and 25% higher productivity than the respective sole stands; but this advantage may be an overestimation of the real potential.The relevance of TCI experiments for studying agroforestry systems, their merits and limitations, especially of the design employed in this study are discussed.  相似文献

A simple tree-crop interaction equation is re-interpreted in terms of resource capture. Benefits in physical yields from agroforestry are to be expected only when there is complementarity of resource capture by trees and crops. Novel approaches involving a combination of sap flow measurements of transpiration and allometric estimates of biomass production were used to determine seasonal water use by trees and crops in agroforestry systems. The results were used to test the hypothesis that agroforestry may improve productivity by capturing a greater proportion of annual rainfall than annual crops. Grevillea robusta A. Cunn., which is reputed to have a deep rooting habit, was grown in semi-arid Kenya either as sole stands or in combination with maize (Zea mays L.). Water use by individual trees and maize plants was determined using constant temperature heat balance gauges and scaled to provide stand-level estimates of transpiration based on linear relationships (r2>0.70) between sap flow and leaf area across a range of tree ages and environmental conditions. Maximum stand-level transpiration rates for grevillea ranged from 2.6 to 4.0 mm per day, consistent with previous studies in similar environments. Biomass production by grevillea was closely correlated with stand-level transpiration (r2>0.69–0.74), suggesting that non-destructive estimates of biomass increments can be used to provide reliable estimates of seasonal transpiration. Cumulative water use by grevillea over the 4.5-year observation period was comparable in the sole tree and agroforestry treatments, reaching a maximum utilisation of annual rainfall of 64–68% 3–4 years after planting. Approximately 25% of the water transpired by the trees was used during the dry season, indicating that they were able to utilise off-season rainfall, comprising 16% of the total annual rainfall, and residual water remaining in the soil profile after the cropping period. During the 1995 long rains, when 221 mm of rain was received, transpiration by sole maize was <50% of precipitation, compared to ca. 85% by the trees in the sole grevillea and agroforestry treatments. These results confirm that agroforestry systems may greatly increase rainfall utilisation compared to annual cropping systems. However, careful consideration of the tradeoffs between the loss of crop production and the additional value provided by tree products is essential.  相似文献

The rate and extent to which biophysical resources are captured and utilized by the components of an agroforestry system are determined by the nature and intensity of interactions between the components. The net effect of these interactions is often determined by the influence of the tree component on the other component(s) and/or on the overall system, and is expressed in terms of such quantifiable responses as soil fertility changes, microclimate modification, resource (water, nutrients, and light) availability and utilization, pest and disease incidence, and allelopathy. The paper reviews such manifestations of biophysical interactions in major simultaneous (e.g., hedgerow intercropping and trees on croplands) and sequential (e.g., planted tree fallows) agroforestry systems. In hedgerow intercropping (HI), the hedge/crop interactions are dominated by soil fertility improvement and competition for growth resources. Higher crop yields in HI than in sole cropping are noted mostly in inherently fertile soils in humid and subhumid tropics, and are caused by large fertility improvement relative to the effects of competition. But, yield increases are rare in semiarid tropics and infertile acid soils because fertility improvement does not offset the large competitive effect of hedgerows with crops for water and/or nutrients. Whereas improved soil fertility and microclimate positively influence crop yields underneath the canopies of scattered trees in semiarid climates, intense shading caused by large, evergreen trees negatively affects the yields. Trees in boundary plantings compete with crops for above- and belowground resources, with belowground competition of trees often extending beyond their crown areas. The major biophysical interactions in improved planted fallows are improvement of soil nitrogen status and reduction of weeds in the fallow phase, and increased crop yields in the subsequent cropping phase. In such systems, the negative effects of competition and micro-climate modification are avoided in the absence of direct tree–crop interactions. Future research on biophysical interactions should concentrate on (1) exploiting the diversity that exists within and between species of trees, (2) determining interactions between systems at different spatial (farm and landscape) and temporal scales, (3) improving understanding of belowground interactions, (4) assessing the environmental implications of agroforestry, particularly in the humid tropics, and (5) devising management schedules for agroforestry components in order to maximize benefits.
