This study investigated the effect of acetic acid concentrations from 3 to 8% on the physicochemical and sensory quality of marinated Atlantic and Baltic herring from fresh and frozen-thawed material. The increased concentration of acetic acid was found to exert a strong impact on decreased sensory scores, weight yield, and pH value as well as on an increased brightness parameter of meat. In turn, moisture, protein hydrolysis products (PHB), total volatile bases (TVB) content, and hardness parameter were affected by the higher addition of acid to a lesser extent. Enhanced protein hydrolysis was observed only at acetic acid concentration of 5% or 5–8%, whereas there was a decrease in the content of TVB only at an acid concentration of up to 4–5%. The marinated frozen-thawed fillets, compared to the fresh ones, required a greater addition of acid in the brine, by 1.5%, to reach a similar pH value. The freezing of fish contributed also to a decrease of PHB and free hydroxyproline, along with an increased concentration of acetic acid. The marinades from frozen-thawed herring contained more TVB, had worse color, and hardness parameters, as well as less yield.  相似文献

Spermatozoa are unique cells because of their morphological and physiological characteristics. They are produced during the process called spermatogenesis. Spermatogenesis consists of three phases: spermatocytogenesis, spermiogenesis and spermiation, during which spermatozoa undergo several changes. Spermatogenesis takes place within the seminiferous tubules containing two types of cells—the germ cells and the Sertoli cells—that alongside the Leydig cells, which play an important role when it comes to normal fertility. Everything is regulated by the hypothalamic–pituitary–gonadal axis and specific hormones due to multi-hormonal feedback systems. Spermatozoa possess morphological and physiological features, which are sometimes completely different from what is observed in various somatic cells. What is more, canine spermatozoa have specific characteristics making them special compared to the spermatozoa of other mammalian species. The metabolic energy production, which is crucial for the appropriate functioning of spermatozoa, can be fuelled by different metabolic pathways utilizing different chemical substrates. Inseparable from the oxidative phosphorylation process is the production of reactive oxygen species, which are both essential and toxic to spermatozoa. Furthermore, epididymis is a very important structure, responsible for the transport and maturation of spermatozoa, which are then stored in the last segment of epididymis—the epididymal cauda. Moreover, the retrieval of spermatozoa from the epididymides is crucial for the development of assisted reproduction techniques and sperm cryopreservation methods. The information gained from the research on domestic dogs might be transferred to their wild relatives, especially those species categorized as endangered.  相似文献

The purposes of this study were to specify the occurrence and prevalence of Babesia canis, Borrelia burgdorferi sensu lato, and Anaplasma phagocytophilum in ticks removed from dogs in Warsaw, and to determine the Borrelia species occurring in Ixodes ricinus ticks. Among 590 collected ticks, 209 were identified as I. ricinus, and 381 as Dermacentor reticulatus. DNA of B. canis was detected in 11% of D. reticulatus ticks. We found that 6.2% of I. ricinus ticks harbored B. burgdorferi s.l. specific DNA and 2.9% harbored A. phagocytophilum DNA. In these samples sequencing of the detected Borrelia amplicon confirmed infection with Borrelia afzelii genospecies. New sequences were submitted to the GenBank database (accession no. EU152128, EU152127, EU152126). This work is the first detection of B. afzelii and A. phagocytophilum in ticks from Warsaw, and the first survey for the prevalence of B. canis, B. afzelii, and A. phagocytophilum in ticks in central Poland.  相似文献

Relationship between somaclonal variation and type of culture in cucumber   总被引:3,自引:0,他引:3  
Highly inbred B line of cucumber was used to compare the effect of four types of in vitro culture on somaclonal variation. The plants were regenerated from the following types of culture: twelve- and eighteen-month-old liquid culture of meristematic clumps (LMC12(18)), ten-month-old embryogenic cytokinin-dependent suspension (CDS), eighteen-month-old embryogenic cytokinin-dependent suspension in medium with modified NH+ 4/NO3 - ratio (CDS 1.7), twelve-month-old embryogenic auxin-dependent suspension (ADS), thirty six-month-old embryogenic auxin-dependent suspension in medium with modified NH+ 4/NO3 - ratio (ADS 1.7) and recurrent leaf callus regeneration (RLC) – repeated 5 times. The differences in the incidence of the following properties were observed: the ploidy of R0 plants, the segregation of new morphological traits in R1 and the germination ability of R1 seeds. R1 families with the segregation of new phenotypes were most numerous in CDS (62.5%) and LMC18 (57.9%), next in CDS1.7 (35.7%), while the smallest number was found in LMC12 (11.1%) and RLC (3.4%).Tetraploid and mixoploid plants occurred in ADS1.7 and ADS (100%) whereas CDS and RLC were observed to contain only tetraploids, respectively 33.3% and 55.2%. There were no changes of ploidy after LMC12, LMC18 and CDS1.7. Among new phenotypes there were such that have not been described so far in cucumber: ginkgolike leaf (gll), yellow-green chlorophyll mutants (y-gc), serrate margin of corolla in male and female flowers (smc). Selenium has been recognised as a basic bioelement that determines normal development and health of animals and humans alike. Studies of many authors revealed the influence of selenium deficiency on immunity, health, reproduction and production of dairy cows. Selenium deficiency have been reported in a number of countries, for example in different regions of Poland. The aim of the study was to determine the Se levels in blood serum of dairy cows in the Lublin region. In the periods: autumn 2003, winter 2003 and autumn 2004 selenium concentrations were investigated in serum samples from 180 dairy cows. The samples were obtained from 11 farms. The selenium concentrations were estimated with the ASA method. The monitoring studies have found that Se serum levels are dependent on season of the year, physiological period, maintenance systems and methods of feeding. Mean selenium serum levels observed in the study were between 0.21-0.92 micromol/l, especially (micromol/l): lactation 0.74 +/- 0.13, drying-off period 0.67 +/- 0.15, autumn 2003 0.38 +/- 0.17, winter 2003 0.48 +/- 0.16, autumn 2004 0.69 +/- 0.18. These results suggest that selenium should be supplemented, especially in high-producing dairy cow herds.  相似文献

Key message

Quercus robur seedling mass was affected more by planting density than by taproot pruning. Root pruning enhanced stem biomass at the expense of roots in later growth stages. Alteration of biomass allocation due to nursery practices may result in greater susceptibility to injury and death of the seedlings under unfavorable environmental conditions.

Context

Plants adjust their growth and modulate the resource allocation in response to applied treatments and environmental conditions.

Aims

The aim was to examine how taproot pruning in seedlings grown at different densities affected long-term growth of Quercus robur.

Methods

Seedlings, sown as acorns at two planting densities, with or without pruned roots were harvested in the second, fourth, and fifth years of growth. The effect of root pruning on biomass allocation was determined by measuring leaf, stem, and root mass fractions; carbohydrate concentrations in the roots; and C/N ratios. Specific leaf area and root length were also determined to assess morphological adaptations to growth conditions.

Results

Total seedling mass was affected more by planting density than by taproot pruning. After 4 years of growth, root mass fractions were lower and stem mass fractions were greater in seedlings planted at a higher density. Five-year old root-pruned seedlings also had a lower root mass fraction and higher stem mass fractions than unpruned seedlings. Specific root length was not affected by root pruning or planting density.

Conclusion

Decrease of relative root biomass with simultaneous increase of stem biomass may be a long-term consequence of taproot pruning of Q. robur, and the effects may manifest years after the seedling stage.

The present study was aimed at disclosing the chemical coding of nerve structures in the porcine ciliary ganglion (CG) using immunohistochemical methods. The substances under investigation included markers of "classical" neurotransmitters, choline acetyltransferase (ChAT), vesicular acetylcholine transporter (VAChT), tyrosine hydroxylase (TH) and dopamine beta-hydroxylase (DbetaH) as well as neuropeptides, somatostatin (SOM), galanin (GAL), substance P (SP), calcitonin gene-related peptide (CGRP), vasoactive intestinal polypeptide (VIP) and neuropeptide Y (NPY). Immunoreactivity to ChAT and VAChT was found virtually in all the neuronal somata and in numerous intraganglionic, varicose nerve fibres which often formed basket-like formations around the nerve cell bodies. Many CG neurons contained immunoreactivity for SOM (46%) or GAL (29%). Interestingly, a small number (approx. 1%) of the cholinergic somata stained for TH but not for DbetaH; nevertheless, some extra- and intraganglionic nerve fibres displayed immunoreactivity for DbetaH or TH. The CG perikarya stained neither for vasoactive intestinal polypeptide (VIP) nor for neuropeptide Y (NPY), but some NPY- or VIP-positive nerve terminals were observed within nerve bundles distributed outside the ganglion. SP- and CGRP-immunoreactivity was found in some intraganglionic nerve fibres only. The present study revealed that the porcine CG consists of cholinergic neurons many of which contain SOM and GAL. Thus, it can be assumed that in the pig, these neuropeptides are involved, complementary to acetylocholine, in the parasympathetic postganglionic nerve pathway to structures of the eye including the ciliary and iris sphincter muscles.  相似文献

The oomycete Phytophthora infestans, the cause of late blight, is one of the most important potato pathogens. During infection, it secretes effector proteins that manipulate host cell function, thus contributing to pathogenicity. This study examines sequence differentiation of two P. infestans effectors from 91 isolates collected in Poland and Norway and five reference isolates. A gene encoding the Avr‐vnt1 effector, recognized by the potato Rpi‐phu1 resistance gene product, is conserved. In contrast, the second effector, AvrSmira1 recognized by Rpi‐Smira1, is highly diverse. Both effectors contain positively selected amino acids. A majority of the polymorphisms and all selected sites are located in the effector C‐terminal region, which is responsible for their function inside host cells. Hence it is concluded that they are associated with a response to diversified target protein or recognition avoidance. Diversification of the AvrSmira1 effector sequences, which existed prior to the large‐scale cultivation of plants containing the Rpi‐Smira1 gene, may reduce the predicted durability of resistance provided by this gene. Although no isolates virulent to plants with the Rpi‐phu1 gene were found, the corresponding Avr‐vnt1 effector has undergone selection, providing evidence for an ongoing ‘arms race’ between the host and pathogen. Both genes remain valuable components for resistance gene pyramiding.  相似文献

The aim of this study was to assess the influence of growth on the pharmacokinetics of sodium salicylate (SS) in male turkeys. SS was administered intravenously at a dose of 50 mg/kg. Plasma drug concentrations were assessed by high‐performance liquid chromatography, and pharmacokinetic parameters were calculated by noncompartmental analysis. As the age increased from 6 to 13 weeks (body weight increase from 2.35 to 9.43 kg), median body clearance decreased from 1.34 to 0.87 ml/min/kg. This caused a significant increase in the median mean residence time from 3.42 to 4.44 hr. Elimination phase proved to be biphasic and two elimination half‐lives (T1/2el) were distinguished. Whereas T1/2el1 was found to increase with age by 128%, T1/2el2 represented a later but faster and less age‐dependent phase of elimination (increase by 56% in the respective groups). Volume of distribution decreased with age. These effects may lead to different therapeutic response to SS in turkeys of different age and body weights.  相似文献

Birds obtained by embryo engineering are used to study embryo development and to produce transgenic birds. As this method of producing birds still generate strong emotions of the public opinion head ornaments, testes and semen characteristics of sex chimera roosters were examined to check whether they differ from chickens obtained by non‐manipulated methods. Measurements of head ornaments, testes and semen were correlated with each other. Semen quality factor (SQF) was calculated, as well as the level of fluctuating asymmetry (FA) of bilateral traits (wattles and testes). Positive correlation was found for comb width and wattle length and comb thickness and sperm concentration. Semen characteristics and FA did not exceed the level encounter in other chicken lines. Results obtained indicate that germline chimeras are similar in appearance of secondary sexual traits, and semen and testes characteristics to chickens produced in non‐manipulated way.  相似文献
