Carotenoid pigments are good biomarkers of grass feeding in sheep. However, grazing lambs are often stall-finished because of grass shortage. We investigated the nature of the carotenoids present in sheep blood and their persistence in this tissue. Four treatments were compared: 1) feeding a concentrate-based diet (n = 10 lambs), 2) grazing followed by a long stall-finishing period (n = 10), 3) grazing followed by a short stall-finishing period (n = 10), and 4) grazing to slaughter weight (n = 10). The concentrate supply was regulated to have similar average daily gain for all treatments. The 40 lambs were allocated to either the grazing or the stall treatments on the basis of their birth date, birth weight, and body weight. The 30 grazing lambs were further allocated to long-stall, short-stall, or grass treatment on the basis of their body weight and plasma carotenoid content. Plasma content of total carotenoids was measured by spectrophotometry during the grazing and the stall periods for all lambs and at slaughter weight for the eight heaviest lambs of each treatment. Analysis of the nature and the concentration of individual carotenoids was performed by HPLC on pasture and stall diets and on blood of grazing lambs. The carotenoid content of the stall diet was 2 to 3% that of the pasture diet. Lutein, zeaxanthin, and beta-carotene accounted for 43 to 58%, 3 to 17%, and 0 to 7% of total plasma carotenoids in grazing lambs, respectively. Two unknown polar carotenoids, expressed in lutein equivalent, accounted for 10 to 22% and 0 to 9% of total carotenoids. Plasma carotenoid content during the grazing and the finishing periods varied among animals (P < 0.001). At slaughter weight, plasma carotenoid content was higher for grass-fed than for stall-fed, long-stall finished, or short-stall finished lambs (P < 0.001), and reliably distinguished grass-fed lambs from all the others. Plasma carotenoid content decreased exponentially with the interval from starting on the stall diet (P < 0.005). The deceleration parameter of the model increased linearly with lamb average daily gain during the stall-finishing period, suggesting that the turnover of carotenoids in the blood may depend on the level of intake of the stall-finishing diet. After 4 to 13 d on the stall diet, depending on the initial plasma carotenoid concentration, plasma carotenoid concentration of previously grazed, stall-finished lambs fell to the values of lambs fed a concentrate diet without grazing. The lipopolysaccharide of Salmonella and other Gram negative pathogenic species has been implicated as a major virulence determinant and in this study we report the role of LPS of S. Enteritidis in the colonisation and persistent gastrointestinal infection of young poultry. The gene encoding the unique O-antigen ligase, waaL, was mutated by insertional inactivation in a well characterised S. Enteritidis strain, S1400/94. The waaL mutant, designated PCP, produced rough colonies on agar medium, did not agglutinate O9 antiserum, did not produce an LPS ladder on silver stained gels and was serum sensitive. PCP and a nalidixic acid marked derivative of S1400/94 (S1400/94 Nalr) were used to orally challenge young chicks, separately and together in competitive index experiments. At post-mortem examination of 1-day-old chicks challenged S1400/94 Nalr and PCP separately there were no significant differences in the numbers of S1400/94 Nalr and PCP bacteria in tissues sampled on days 1, 2, and 5. By day 42 after challenge S1400/94 Nalr bacteria were recovered in significantly higher numbers than PCP from the caecal contents (P < 0.001). In competitive index studies in the 1-day-old chick PCP colonised, invaded and persisted in lower numbers than S1400/94 Nalr. In 4-week-old chicks challenged separately, PCP bacteria were recovered from all tissues examined in significantly lower numbers than S1400/94 Nalr. In competitive index experiments in 4-week-old chicks, PCP was not detected at any site and at any time point. Therefore, the O-antigen of S. 水产动物红螯螯虾(Cherax quadricarinatus),俗称澳洲淡水龙虾,在热带和亚热带区域养殖发展十分迅速。由于饲料工业处于初级发展期,红螯螯虾营养需求与各种原料消化吸收率数据极其有限,生产饲料一般并未按照螯虾的特殊要求来单独设计配方。考虑到近期鱼粉价格上涨,使用类似鸡肉粉等替代蛋白源显得更为经济。本研究评价螯虾幼虾投喂使用鸡肉粉替代鱼粉的商业饲料生长效果。红螯螯虾幼虾(0.45±0.01) g暂养在户外圆池(容积24 600 L),密度为12.5尾/m2。养殖池水源为水库抽提,养殖期间保持流水。小龙虾投喂6种饲料,蛋白为26%,粗脂肪为7%,每个处理4个重复,在同等环境条件下生长良好。基础饲料中的鱼粉按比例被鸡肉粉替代,鱼粉含量递减为15%、12%、9%、6%、3%和0,而鸡肉粉含量递增为7.8%、10.5%、13.2%、15.8%、18.5%和21.2%,等氮等能饲料。经过56 d养殖试验,小龙虾所有试验组的存活率大于83%,增长11.3 g以上,处理组间没有显著差异。基于以上试验结果,可以得出结论,鸡肉粉适合作为小龙虾饲料中鱼粉的替代物。 相似文献

The secretion of acetylcholinesterase (AChE) by female and male Heligmosomoides polygyrus was studied in different in vitro culture media. AChE secretion was increased in the presence of fetal calf serum or bovine serum albumin (BSA). In the absence of crowding effects, specific AChE activity in excretion/secretion products was higher for male (2.41 +/- 0.07 mumol min-1 l-1 mg-1) than for female (0.56 +/- 0.04 mumol min-1 mg-1) worms but on a per nematode basis both sexes showed comparable rates of secretion. Acetylthiocholine iodide was the favoured substrate of the enzyme. When the nematodes were incubated in vitro with albendazole (ABZ), ricobendazole (RBZ), mebendazole (MBZ), levamisole (LVM), morantel (MRT) or ivermectin (IVM), at concentrations from 1 mM to 10 nM, in RPMI medium for 2 or 6 h and then transferred to a drug-free medium (RPMI medium supplemented with 0.5% BSA) for 24 h or continuously exposed to the drugs in supplement-free medium (24 h), the concentration- and time-dependent inhibitory effects on AChE secretion were observed. The continued exposure to the drugs for all incubation periods (with a single exception for LVM 1 mM) produced the highest levels of inhibition. Under these conditions, the concentrations inhibiting the secretion of AChE by 50% (IC50) relative to drug-free controls were estimated. The IC50 values ranged from 0.012 microM (IVM) to 2.96 microM (MRT). Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) was first isolated in swine alveolar macrophages (SAMs) and has subsequently been reported to replicate in other cell lines. Entry of the virus inside the cell takes place by receptor-mediated endocytosis. Following the entry of the virus into the cell, several not completely understood changes take place. PRRSV has been reported to be an apoptotic-inductor virus both in vivo and in vitro. Interestingly, it has been suggested that PRRSV-induced apoptosis occurs in cells other than those in which PRRSV replicates by a bystander mechanism. A study was conducted to determine the influence of sodium salicylate on the behaviour and the food and water consumption of broiler chickens after lipopolysaccharide (LPS) injection. An oral dose of 100 mg/kg sodium salicylate was given and an acute phase reaction in broiler chickens was provoked through the intravenous injection of Escherichia coli LPS. Water intake was higher in the LPS and salicylate-treated group than in the positive control group. The salicylate treatment, however, did not restore the food intake, or influence the behaviour of the chickens. These data show that sodium salicylate has a positive effect on the water intake after intravenous injection of LPS in chickens and suggests that there is a difference in mechanism of action of food and water consumption after LPS injection in chickens. 相似文献

A 6-year-old Simmental cow infected with Theileria buffeli had a clinical disease characterized by theilerial parasitemia, macrocytic normochromic anemia with acanthocytosis and spherocytosis, lymphoid hyperplasia (lymphocytosis, edematous lymphadenomegaly), dysproteinemia, evidence of liver disease, and a low serum antibody titer against T. buffeli. The cow was in a herd in which all cattle originated in Missouri; 22/75 (29%) of cattle had a theilerial parasitemia and 26/75 (35%) had titers to T. buffeli of > or =1:160. Classification of the Missouri bovine organism as T. buffeli was based on DNA sequencing and comparison to sequences for T. buffeli and Theileria sp. type A obtained from GenBank. Intraerythrocytic veils and piroplasms were seen during transmission electron microscopy. The organism was successfully transmitted to two splenectomized calves, which developed mild anemias while parasitemic. Blood from the second calf was used as the source of T. buffeli antigen for an indirect immunofluorescence antibody test. Theilerial isolates from a Missouri white-tailed deer were also sequenced and resembled Theileria sp. types F and G and were not consistent with the bovine organism. 相似文献
