Home Plate is a layered plateau in Gusev crater on Mars. It is composed of clastic rocks of moderately altered alkali basalt composition, enriched in some highly volatile elements. A coarsegrained lower unit lies under a finer-grained upper unit. Textural observations indicate that the lower strata were emplaced in an explosive event, and geochemical considerations favor an explosive volcanic origin over an impact origin. The lower unit likely represents accumulation of pyroclastic materials, whereas the upper unit may represent eolian reworking of the same pyroclastic materials.  相似文献

Midbrain dopamine neurons regulate many important behavioral processes, and their dysfunctions are associated with several human neuropsychiatric disorders such as attention deficit hyperactivity disorder (ADHD) and schizophrenia. Here, we report that these neurons in mice selectively express guanylyl cyclase-C (GC-C), a membrane receptor previously thought to be expressed mainly in the intestine. GC-C activation potentiates the excitatory responses mediated by glutamate and acetylcholine receptors via the activity of guanosine 3',5'-monophosphate-dependent protein kinase (PKG). Mice in which GC-C has been knocked out exhibit hyperactivity and attention deficits. Moreover, their behavioral phenotypes are reversed by ADHD therapeutics and a PKG activator. These results indicate important behavioral and physiological functions for the GC-C/PKG signaling pathway within the brain and suggest new therapeutic targets for neuropsychiatric disorders related to the malfunctions of midbrain dopamine neurons.  相似文献

A molecule or larger body is chiral if it cannot be superimposed on its mirror image (enantiomer). Electromagnetic fields may be chiral, too, with circularly polarized light (CPL) as the paradigmatic example. A recently introduced measure of the local degree of chiral dissymmetry in electromagnetic fields suggested the existence of optical modes more selective than circularly polarized plane waves in preferentially exciting single enantiomers in certain regions of space. By probing induced fluorescence intensity, we demonstrated experimentally an 11-fold enhancement over CPL in discrimination of the enantiomers of a biperylene derivative by precisely sculpted electromagnetic fields. This result, which agrees to within 15% with theoretical predictions, establishes that optical chirality is a fundamental and tunable property of light, with possible applications ranging from plasmonic sensors to absolute asymmetric synthesis.  相似文献

Epinephrine and norepinephrine condensed with acetaldehyde or with formaldehyde in dilute aqueous solution at neutral pH and room temperature to form 1,2,3,4-tetrahydroiso-quinoline alkaloids. Similar condensation reactions occurred in cow adrenal glands perfused with dilute aldehyde solutions at 37 degrees C. Biosynthesis of these alkaloids in vivo could play a role in altering an individual's behavior during and after the ingestion of alcohol.  相似文献

BACKGROUND: Pseudoperonospora cubensis, the causal oomycete agent of cucurbit downy mildew, is responsible for enormous crop losses in many species of Cucurbitaceae, particularly in cucumber and melon. Disease control is mainly achieved by combinations of host resistance and fungicide applications. However, since 2004, resistance to downy mildew in cucumber has been overcome by the pathogen, thus driving farmers to rely only on fungicide spray applications, including carboxylic acid amide (CAA) fungicides. Recently, CAA‐resistant isolates of P. cubensis were recovered, but the underlying mechanism of resistance was not revealed. The purpose of the present study was to identify the molecular mechanism controlling resistance to CAAs in P. cubensis. RESULTS: The four CesA (cellulose synthase) genes responsible for cellulose biosynthesis in P. cubensis were characterised. Resistant strains showed a mutation in the CesA3 gene, at position 1105, leading to an amino acid exchange from glycine to valine or tryptophan. Cross‐resistance tests with different CAAs indicated that these mutations lead to resistance against all tested CAAs. CONCLUSION: Point mutations in the CesA3 gene of P. cubensis lead to CAA resistance. Accurate monitoring of these mutations among P. cubensis populations may improve/facilitate adequate recommendation/deployment of fungicides in the field. A liquid chromatographic (LC) method is proposed for the determination of aflatoxin M1 in milk. The method was successfully applied to both liquid whole and skim milk and also whole and skim milk powder. The samples are initially extracted with acetonitrile-water followed by purification using a silica gel cartridge and a C18 cartridge. Final analysis by LC was achieved using a radial compression module equipped with a 5 micron C18 column and a fluorescence detector. The method was successfully applied to samples at levels of 10 to 0.08 ppb added aflatoxin M1 with recoveries in the range of 70-98%.  相似文献

Single nucleotide polymorphisms (SNPs) were discovered in 44 cDNA clones from leaves by comparison of the commercial cultivar ‘Mona Lisa‘ with a wild population of Anemone coronaria L. One hundred and fifty five SNPs were discovered with an average frequency of one SNP per 167 bp. Forty nine percent of the SNPs are transitions, 43 are transversions, 26 are heterozygotes and 9% are InDels. Eighty two (68%) of the SNPs located in the ORF, resulted in a change of amino acid while 39 (32%) did not affect the amino acid. Thirty eight SNPs (46%), which change the amino acid, resulted in similar amino acid, while 44 SNPs (54%) resulted in a non-similar amino acid. Nine polymorphic sites were genotyped by matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) using genomic DNA. Phylogenetic analysis of 50 individuals from six wild populations and the commercial cultivar revealed high polymorphism within and low polymorphism between A. coronaria L. wild populations. The phylogenetic tree revealed only one clear cluster of the ‘Mona Lisa’ cultivar. All the other individuals from the various wild populations are distributed on different nodes indicating that the genetic variation within the wild populations is high while the commercial cultivar is quite homogenous and genetically different from all other populations.  相似文献

Recombinant proteins containing tetracysteine tags can be successively labeled in living cells with different colors of biarsenical fluorophores so that older and younger protein molecules can be sharply distinguished by both fluorescence and electron microscopy. Here we used this approach to show that newly synthesized connexin43 was transported predominantly in 100- to 150-nanometer vesicles to the plasma membrane and incorporated at the periphery of existing gap junctions, whereas older connexins were removed from the center of the plaques into pleiomorphic vesicles of widely varying sizes. Selective imaging by correlated optical and electron microscopy of protein molecules of known ages will clarify fundamental processes of protein trafficking in situ.  相似文献

A model for optimal operation of water supply/irrigation systems of various water quality sources, with treatment plants, multiple water quality conservative factors, and dilution junctions is presented. The objective function includes water cost at the sources, water conveyance costs which account for the hydraulics of the network indirectly, water treatment cost, and yield reduction costs of irrigated crops due to irrigation with poor quality water. The model can be used for systems with supply by canals as well as pipes, which serve both drinking water demands of urban/rural consumers and field irrigation requirements. The general nonlinear optimization problem has been simplified by decomposing it to a problem with linear constraints and nonlinear objective function. This problem is solved using the projected gradient method. The method is demonstrated for a regional water supply system in southern Israel that contains 39 pipes, 37 nodes, 11 sources, 10 agricultural consumers, and 4 domestic consumers. The optimal operation solution is described by discharge and salinity values for all pipes of the network. Sensitivity of the optimal solution to changes in the parameters is examined. The solution was found to be sensitive to the upper limit on drinking water quality, with total cost being reduced by 5% as the upper limit increases from 260 to 600 mg Cl l–1. The effect of income from unit crop yield is more pronounced. An increase of income by a factor of 20 results in an increase of the total cost by a factor of 3, thus encouraging more use of fresh water as long as the marginal cost of water supply is smaller than the marginal decrease in yield loss. The effect of conveyance cost becomes more pronounced as its cost increases. An increase by a factor of 100 results in an increase of the total cost by about 14%. The network studied has a long pipe that connects two distinct parts of the network and permits the supply of fresh water from one part to the other. Increasing the maximum permitted discharge in this pipe from 0 to 200 m3 h–1 reduces the total cost by 11%. Increasing the maximum discharge at one of the sources from 90 to 300 m3 h–1 reduces the total cost by about 8%.  相似文献
