Juvenile rainbow trout, Oncorhynchus mykiss, were injected with estradiol-17β (E2) in order to study the source of extra calcium needed during vitellogenesis. E2-treatment increased the calcium uptake from the external medium as well as calcium mobilization from muscle and scale. Judged by the increase in plasma protein-bound calcium levels, the E2-induced increase in calcium uptake is an apparent over-mobilization of calcium, i.e., the calcium uptake of the fish is in excess of what is found bound to plasma proteins. As the calcium excretion and calcium space (calculated from free plasma calcium levels) were unaffected, the excess calcium is suggested to be incorporated into internal calcium stores. This implies that the systems regulating vitellogenesis and calcium balance are integrated on the mechanistic or endocrine level, and that E2 causes calcium mobilization of a magnitude geared to the needs of the sexually maturing female.  相似文献

A sublethal dose (0.1 pmol per honeybee, i.e., approx. 0.9 pmol/g body wt) of the synthetic pyrethroid, deltamethrin, was injected intrathoracically into the hemocoel of emerging bees and the biochemical effects were determined over a period of 3 hr. The variations in hemolymph concentrations of trehalose, glucose, phospholipid, steroid, and diacylglycerol were limited, but large variations were observed for other lipids. Mono- and triacylglycerols increased dramatically 1.5 hr after injections; then a peak of free fatty acid appeared at the same time (3 hr) as the monoacylglycerols reached their maximum and the triacylglycerols started to decrease. Atypical forms of triacylglycerols accumulated from 1.5 hr to a maximum level 2 hr after injection. Then, after 3 hr, the concentrations of regular fatty acids, monoacylglycerols, and atypical triacylglycerols returned to normal while longer-chain fatty acids and different forms of ceride simultaneously appeared. The gut alkaline phosphatase parameters (VM, K, and Hill coefficient) also showed large variations 1.5 and 2 hr after injection, indicating an initial inhibition phase followed by superimposed induction and inhibition mechanisms. These data are discussed according to the hemolymph-lipid transport scheme from the biosynthetic to the utilization sites. Hormone release associated to mitochondrial alteration could be responsible for the observed effects.  相似文献

Ivermectin is one of the most commonly used drugs in pharmacotherapy of parasitic diseases in domestic and wild animals caused by parasitic nematodes and arthropods. However, ivermectin and other avermectins very often produce side-effects in hosts. The most dominant clinical symptom of ivermectin toxicity in domestic and wild animals is CNS depression. In nematodes, the target site of ivermectin’s action is glutamate-gated chloride-channel receptor and GABA receptor. The depressive effect of ivermectin in mammals might include more than one mechanism; therefore, the anticonvulsive effect of ivermectin against convulsions caused by lidocaine and strychnine was evaluated. Ivermectin antagonized lidocaine- and strychnine-induced convulsions in rats, although these have different mechanisms. In the present study, the anticonvulsive ED50 of ivermectin for lidocaine-induced convulsions was 2.44 mg/kg (95% CL 1.67 to 3.57 mg/kg), whereas for convulsions induced by strychnine it was higher at 4.25 mg/kg (95% CL 2.32 to 3.78 mg/kg). At the same time, both anticonvulsive doses are significantly lower then the observed LD50 of ivermectin (18.20 mg/kg). Furthermore, flumazenil (0.1 and 0.2 mg/kg), an antagonist of benzodiazepine receptors, antagonizes just one part of these anticonvulsive effects of ivermectin. Our results show the significant anticonvulsive properties of ivermectin and support the findings that ivermectin in the CNS of mammals produces multiple inhibitory effects, probably through participation in the function of GABA-sensitive and GABA-insensitive chloride channels.  相似文献

Summary The fungusPhytophthora fragariae Hickman is the causal agent of red stele disease in strawberry (Fragaria × ananassa), this being a major disease in many areas with cool and moist conditions. Success of resistance breeding can be nullified by the appearence of specific races of the fungus capable of overcoming the introduced resistance. In some countries (USA, UK, Japan and Canada) races were identified by using a differential set of strawberry cultivars. The absence of an international standard differential set and the use of different test methods make a comparison of the identified races difficult or even lead to contradictory results for one variety/race combination. The aim of this study was to obtain information about the spectrum of pathogenic races in Germany as a basic contribution to research on resistance breeding against the fungus. The susceptibility of different strawberry cultivars to German isolates ofP. fragariae was evaluated. The inoculation was done by modifying a method described by Milholland et al. (1989). Rootedin vitro plants, four weeks after transferring them to the soil, were used for the investigation. Inoculation was done with a zoospore suspension of defined concentration. Up to now three German isolates, G-1, G-2, and G-3, can be separated by their ability to infect and produce oospores in the roots of the strawberry cultivars ‘Senga Sengana’, either ‘Saladin’, ‘Redgauntlet’ or ‘Climax’ andFragaria chiloensis clone ‘YaquinaB’. These results confirm the existence of pathogenic races ofP. fragariae in Germany and should be taken into consideration for resistance breeding against the fungus.  相似文献

Plant genotypes differ in their capacity to grow in soils with low manganese (Mn) availability. The physiological mechanisms underlying differential tolerance to Mn deficiency are poorly understood. To study the relationship between Mn content in soil, plant genotypes, and rhizosphere microorganisms in differential Mn efficiency, two wheat (Triticum aestivum L.) cultivars, RAC891 (tolerant to Mn deficiency) and Yanac (sensitive), were grown in a Mn‐deficient soil to which 5, 10, 20 or 40 mg Mn kg–1 were added. The shoot dry matter of both cultivars increased with increasing Mn addition to the soil. At all soil Mn fertilizer levels, the tolerant RAC891 had a greater shoot dry matter and a higher total shoot Mn uptake than the sensitive Yanac. The concentration of DTPA‐extractable Mn in the rhizosphere soil of RAC891 at Mn20 and Mn40 was slightly lower than in the rhizosphere of Yanac. The population density of culturable microorganisms in the rhizosphere soil was low (log 6.8–6.9 cfu (g soil)–1) in both cultivars and neither Mn oxidation nor reduction were observed in vitro. To assess the non‐culturable fraction of the soil microbial community, the ribosomal intergenetic spacer region of the bacterial DNA in the rhizosphere soil was amplified (RISA) and separated in agarose gels. The RISA banding patterns of the bacterial rhizosphere communities changed markedly with increasing soil Mn level, but there were no differences between the wheat cultivars. The bacterial community structure in the rhizosphere was significantly correlated with the concentration of DPTA‐extractable Mn in the rhizosphere, fertilizer Mn level, shoot dry matter, and total shoot Mn uptake. The results obtained by RISA indicate that differential tolerance to Mn deficiency in wheat may not be related to changes in the composition of the bacterial community in the rhizosphere.  相似文献

Background, Aim and Scope   One of the first occurrences of pharmaceutically active compounds in groundwater was reported from the sewage irrigation farms south of Berlin. At these sewage irrigation farms treated sewage effluent passed the soil and unsaturated zone before reaching the aquifer. Clofibric acid was detected in pore water from soils of those sewage irrigation farms in concentrations between 65 ng/L and 1430 ng/L. The aim of this study was to investigate the transport behavior of regularly detected clofibric acid, diclofenac, ibuprofen, and propyphenazone under conditions comparable to those at the sewage irrigation farms in a multiple compound sand column laboratory experiment. Materials and Methods   Sediment column experiments were conducted to study the transport of pharmaceuticals in the unsaturated zone. The migration was measured in fine to medium grained sand and leaching solution containing 1 mg/L of pharmaceutically active compounds and 61 mg/L of the tracer lithium chloride (LiCl). For the analysis of the pharmaceutical compounds the water samples were adjusted to a pH value of 2 and then extracted by solid-phase extraction (SPE). Before extraction, the samples were spiked with a surrogate standard for analytical quality control. The sample extracts were analyzed by capillary gas chromatography-mass spectrometry (GC-MS) with selected ion monitoring (SIM). Depending on the sample volume (100 to 200 mL) and the matrix, the limits of detection were between 1 and 10 ng/L, and the limits of quantitation were between 5 and 25 ng/L. Analysis for calcium, magnesium and lithium were carried out using a 'Trace Scan' ICP-AES from Thermo Jarrel Ash. Sodium, potassium, iron and manganese were analyzed using a Philips PU 9400 flame AAS. Analysis of anions was performed on a Dionex ion chromatograph DX 120. Results   At the sewage irrigation farms the average concentrations of clofibric acid in the unsaturated zone declined from higher values near ground surface (480 ng/L) to lower values near the groundwater table (65 ng/L). From the pharmaceuticals analyzed only clofibric acid, primidone and propyphenazone could be analyzed in the first (upper) aquifer at the sewage irrigation farms. All other pharmaceuticals could neither be detected in the first aquifer nor in the deeper aquifers. Breakthrough curves from soil column experiments revealed no transformation and no retardation for clofibric acid, whereas transformation of diclofenac was so high (79%) that no retardation factor could be calculated. Ibuprofen was significantly transformed (37%), transformation of propyphenazone (17%) was quite low and retardation (Rf = 2.05) was in the range of previously conducted column experiments. Discussion   The results confirm previously conducted experiments with clofibric acid where this compound was identified as highly mobile and persistent. The results that diclofenac and ibuprofen are significantly transformed where unexpected as other studies exhibited much lower transformation under saturated conditions at least for diclofenac. However, lower pH values and higher oxygen contents in the unsaturated zone compared to the aquifer may explain this observed high transformation of these compounds at the column experiments. Conclusions   We conclude that irrigation with sewage effluent containing the compounds used in our experiments will lead to an input into groundwater of clofibric acid, whereas diclofenac and ibuprofen will most likely be transformed during the passage. Propyphenazone will be retarded but will most likely occur in groundwater. These results from the column experiments coincide very well with the occurrence of the pharmaceuticals clofibric acid, primidone, and propyphenazone in the first aquifer. Recommendations and Perspective  : The results underline the need to study the sorption of pharmaceuticals on various materials. e.g. organic matter, surfaces at pH values occurring in the unsaturated zone. Future field studies will also include the investigation of desorption behavior in the unsaturated zone.  相似文献

Bioavailability of dietary folate might be impaired by the polyglutamate chain to which approximately 70% of dietary folates are bound. This chain must be removed enzymatically in the intestine before folate is absorbed as a monoglutamate. To increase formation of monoglutamate folate in vegetables, the vegetables were subjected to various processing treatments. Treatments included freezing (-18 degrees C, 16 h) and thawing (4 degrees C, 24 h) and hydrostatic high-pressure treatment (200 MPa, 5 min). Both freezing/thawing and high-pressure treatment increased the proportion of folate in the monoglutamate form in leeks, cauliflower, and green beans 2-3-fold. However, loss of total folate after these treatments was >55%. It is concluded that conversion of folate polyglutamate to the monoglutamate form in vegetables is possible by certain processing treatments. Potentially this could lead to vegetables with higher folate bioavailability. However, to prevent folate loss into processing water, processing in a closed system should be applied.  相似文献

Summary The occurrence of chromosomal interchanges was studied in F1 hyrids in two diallel schemes, including nine accessions of Triticum aestivum subsp. spelta, originating from Iran, Europe and North Africa, and the common wheat variety Chinese Spring as a reference. Seven accessions of subsp. spelta differed in one translocation from Chinese Spring. The European accession Grey differed in one or perhaps two interchanges from the reference variety, and Iran 417a showed a difference of three interchanges with Chinese Spring. In total six or perhaps seven interchanges were observed. Thus the number of translocation events in subsp. spelta appeared to be low. One translocation was unique and occurred only in the accession from North Africa. Also one accession from Europe, the landrace Schwarzwald, possibly carried a unique translocation. In both diallels an interchange with a relatively low coefficient of multivalent realisation at first meiotic metaphase was observed in most accessions. The two extra translocations in Iran 417a are considered to be the result of more recent translocation events, whereas the possible second translocation in accession Grey showed such a low coefficient of multivalent realisation that its existence could be doubted.  相似文献
