Black shank, a fuagal disease caused by Phytophthora parasitica var. nicotianae, is a major hazard in the production of burley tobacco (Nicotiutta tubacum L.). Moderate levels of resistance have been bred into cultivars, but little success has been achieved in utilizing high levels of resistance found in the cigar line. Beinhart 1000-1. Beinhirt 1000-1 and seven burley cultivars were combined into a synthetic population and three cycles of selection for increased black slunk resistance were conducted. The objectives of this study were (i) t0 estimate genetic variability for agronomic traits in the original base population (Co) and die three selection cycles and (ii) to characterize the effects of selection for black shank resistance on the agronomic traits. Fifty selfed lines From the base population and each of three selection cycles were evaluated in replicated field trials at Lexington and Princeton, KY. Differences among cycle means for six agronomic traits were not statistically significant. Genetic variation for the six traits among lines within cycles was highly significant and did not change during selection. The results indicated that previously assumed associations between black shank resistance in Beinhart 1000-1 and undesirable agronomic traits were due to chromosomal linkages which were apparently broken by intercrossing. The C3 population would be a useful base population for selection to improve agronomic performance in black shank resistant genotypes.  相似文献

The use of lipoarabinomannan (LAM; obtained from Mycobacterium paratuberculosis) in and ELISA (LAM-ELISA) to test 75 sheep sera from a paratuberculosis-infected flock resulted in an approximate threefold increase in sensitivity (from 23.5% to 70.6%), compared with the use of Annau's polysaccharide in a complement fixation test (P-CFT). Even after manipulation of the LAM-ELISA cut-off value to produce a specificity of 100% to match that of the P-CFT, the sensitivity still was approximately twofold greater than that of the P-CFT. Anti-bovine monoclonal antiglobulin-enzyme conjugates matched commercially available anti-ovine polyclonal antiglobulin-enzyme conjugates with respect to sensitivity and specificity. False-positive results were found to be less frequent after combining 2 serodiagnostic tests, LAM-ELISA and D antigenagar gel immunodiffusion, resulting in an increase in specificity from 88.1% to 95.2%. The repeatability of true seropositive and seronegative results was found to be 89.5% and 91.1%, respectively, for sera obtained less than or equal to 1 month prior to slaughter and 91.7% and 95.5%, respectively, for reanalysis of sera obtained at the time of slaughter.  相似文献

Naturally occurring cutaneous fibromas affecting white-tailed deer (Odocoileus virginianus) and mule deer (O hemionus), and cutaneous fibropapillomas of domestic cattle were tested for papillomavirus using indirect immunofluorescence (IF), peroxidase-antiperoxidase (PAP), and negative-stain electron microscopic techniques. Papillomavirus was consistently detected using rabbit antiserum against papillomavirus group-specific antigen in all mule deer fibromas and bovine fibropapillomas; only 16 of 28 white-tailed deer fibromas tested by IF and 9 of 15 tested by PAP were detected. Normal skin from white-tailed deer or cattle was consistently negative for virus. Similar results were obtained by negative-stain electron microscopic examination of partially purified tumor homogenates. Using deer fibroma virus or bovine papillomavirus type 1-specific antisera, viruses were typed by IF, PAP, and immunoelectron microscopy.  相似文献

Six agglutination and two complement fixation tests were compared with respect to specificity, sensitivity and relative sensitivity for the serodiagnosis of bovine brucellosis. Based on 1051 sera from brucellosis free herds, the specificity of the tests was 98.9% for the buffered plate antigen test (BPAT), 99.2% and 99.3% for the standard tube and plate agglutination tests (STAT and SPAT), respectively, and 99.8% for the 2-mercaptoethanol test (2MET). On this small sample, the rose bengal plate test (RBPT), card test (CARD) and the complement fixation test (CFT) correctly classed all sera as negative. On a sample of 167 culture positive cattle, the sensitivities of the tests were CFT: 79.0%, BPAT: 75.4, RBPT: 74.9%, CARD: 74.3%, SPAT: 73.1%, STAT: 68.9%, and 2MET: 59.9%. All tests combined detected only 82% of these infected cattle. Analysis of the relative sensitivity of the six agglutination tests gave the following ranking: BPAT greater than RBPT greater than CARD greater than SPAT greater than STAT. The 2MET ranked between the BPAT and RBPT or between the RBPT and CARD depending on the analysis used. The use of the BPAT as a screening test is recommended provided that a test of high specificity and sensitivity such as the CFT is used to confirm screening test reactions.  相似文献

An ELISA measuring IgM rheumatoid factor (RF) in dog serum is presented. Dog sera and a human IgM RF standard, calibrated against the international World Health Organisation (WHO) standard, are compared. It is concluded that the human IgM RF standard may be used as reference serum in the canine assay, which makes it possible to compare results from different veterinary laboratories.  相似文献

The purpose of the present study was to calculate the critical difference between 2 analytical results for the red blood cell count (RBC), the white blood cell count (WBC), the haemoglobin concentration (Hb), and the haematocrit (PCV) in blood from Red Danish Dairy cows. The critical difference can help to judge whether the difference between 2 consecutive analytical results from the same animal may be safely ascribed to natural variation or not. To calculate the critical differences, blood samples from 20 clinically healthy lactating cows were collected once daily for 5 consecutive days. The total variance of the analytical results was divided into the component of variance between cows (S2Inter), the component of variance for days within cows (S2Intra), and the component of variance for measurements (S2Anal) using nested analysis of variance. The critical difference was then calculated from S2Intra and S2Anal as 0.61 x 10(12)/l for RBC, 2.2 x 10(9)/l for WBC, 0.79 mmol/l for Hb, and 0.07 for PCV. The critical differences may be used as guidelines to indicate potentially important changes in the parameters. However, the analytical results should not be assessed by the critical differences alone, but should also be compared to the corresponding reference intervals.  相似文献

Context

The application of regional-level airborne lidar (light detection and ranging) data to characterize habitat patches and model habitat connectivity over large landscapes has not been well explored. Maintaining a connected network of habitat in the presence of anthropogenic disturbances is essential for regional-level conservation planning and the maintenance of biodiversity values.

Objectives

We quantified variation in connectivity following simulated changes in land cover and contrasted outcomes when different conservation priorities were emphasized.

Methods

First, we defined habitat patches using vegetation structural attributes identified via lidar. Second, habitat networks were constructed for different forest types and assessed using network connectivity metrics. And finally, land cover change scenarios were simulated using a series of habitat patch removals, representing the impact of implementing different spatial prioritization schemes.

Results

Networks for different forest structure types produced very different patch distributions. Conservation scenarios based on different schemes led to contrasting changes during land cover change simulations: the scheme prioritizing only habitat area resulted in immediate near-term losses in connectivity, whereas the scheme considering both habitat area and their spatial configurations maintained the overall connectivity most effectively. Adding climate constraints did not diminish or improve overall connectivity.

Conclusions

Both habitat area and habitat configuration should be considered in dynamic modeling of habitat connectivity under changing landscapes. This research provides a framework for integrating forest structure and cover attributes obtained from remote sensing data into network connectivity modeling, and may serve as a prototype for multi-criteria forest management and conservation planning.

After years of decline and neglect, low woodland types based on coppice management experience renewed interest. Substantial research has demonstrated the potentials of coppice for biomass production and for nature conservation, and coppices are increasingly being suggested for urban situations. Yet, our understanding of the more social aspects of coppice woodlands in modern urban situations is limited. Against this background, this contribution classifies coppice management systems as a basis for identification of social aspects of coppices and their potential use in contemporary urban forestry. Based on this classification, the social perspectives and potential niches for urban coppices are discussed, while lines of research are suggested which will support the development of a thorough and up-to-date knowledge base, against which the social merits of urban coppice woodlands can be critically evaluated.  相似文献
