In order to find out a new effective accumulator of polycyclic aromatic hydrocarbons (PAHs) useful for monitoring studies on a large scale and low costs, the accumulation capacity of both biological and artificial matrixes (mosses and polyester fibers, respectively) has been tested. For this purposes, Hypnum cupressiforme and dacron® were exposed to pollution airborne in two sites located nearby an active iron industry and in center of the town of Trieste, where high PAH pollution spots, due to vehicular traffic, are usually detected. The samplers were exposed in six sampling sessions for 21 days. The results obtained were compared with data collected by active PAH samplers, usually employed for official widespread monitoring. The level of correlation between the data sets was calculated. Furthermore, a repeatability study of data was performed. Estimating the relative importance of habitat loss and fragmentation is necessary to estimate the potential benefits of specific
management actions and to ensure that limited conservation resources are used efficiently. However, estimating relative effects
is complicated because the two processes are highly correlated. Previous studies have used a wide variety of statistical methods
to separate their effects and we speculated that the published results may have been influenced by the methods used. We used
simulations to determine whether, under identical conditions, the following 7 methods generate different estimates of relative
importance for realistically correlated landscape predictors: residual regression, model or variable selection, averaged coefficients
from all supported models, summed Akaike weights, classical variance partitioning, hierarchical variance partitioning, and
a multiple regression model with no adjustments for collinearity. We found that different methods generated different rankings
of the predictors and that some metrics were strongly biased. Residual regression and variance partitioning were highly biased
by correlations among predictors and the bias depended on the direction of a predictor’s effect (positive vs. negative). Our
results suggest that many efforts to deal with the correlation between amount and fragmentation may have done more harm than
good. If confounding effects are controlled and adequate thought is given to the ecological mechanisms behind modeled predictors,
then standardized partial regression coefficients are unbiased estimates of the relative importance of amount and fragmentation,
even when predictors are highly correlated. 相似文献
The potential of bacteriophage therapy to control bacterial disease in farmed fish was tested using, as an example, furunculosis of Atlantic salmon, caused by Aeromonas salmonicida subsp. salmonicida.
In vivo testing with Atlantic salmon and rainbow trout (Oncorhynchus mykiss Walbaum) showed no adverse effects, with bacteriophage generally cleared within 96 h of administration by either intraperitoneal (i.p.) injection or oral in-feed.
Juvenile Atlantic salmon were administered a combination of bacteriophage O, R and B (1.9 × 108 pfu fish− 1) by i.p. injection, after they had been challenged with A. salmonicida subsp. salmonicida 78027, also by i.p. injection. The fish that were injected with bacteriophage immediately after challenge died at a significantly slower rate then those that were either not treated with bacteriophage, or treated 24 h post-challenge. However, the end result (100% mortality) was not affected.
In further experiments the effects of oral (1.88 × 105 pfu g− 1 fish− 1 daily for 30 days), bath (1.04 × 105 ml− 1 daily for 30 days) and i.p. (6.25 × 107 pfu fish− 1) phage treatment to control furunculosis in experimentally infected Atlantic salmon were compared with antibiotherapy (treatment with 10 mg kg− 1 bw− 1 day− 1 oxolinic acid for 10 days), using an indirect cohabitation challenge. No protection was offered by any of the bacteriophage treatments, compared to the positive challenge group, although significant protection was offered by the oxolinic acid treatment. Analysis of samples taken from the trials demonstrated that bacteriophage were correctly administered to the fish and, on occasion, were isolated from fish that had succumbed to furunculosis. It was also shown that bacteriophage resistant A. salmonicida subsp. salmonicida isolates could be recovered from mortalities in all the treatment groups.
The results suggest that, although there were no safety problems associated with the approach, furunculosis in Atlantic salmon is not readily controllable by application of bacteriophage. 相似文献
The cryopreservation of testicular tissue is a potential method for preserving male fertility. However, the effect of cryopreservation on bovine calf testicular tissue is scarce. This study investigated the effect of different cryoprotectants on bovine calf testicular tissue at the molecular level. Testicular tissue from ten immature bovine calves (6 months) was collected after slaughter and cryopreserved in an extender containing different concentrations of the following five cryopreservation solutions (CP): bovine serum albumin (BSA) with 5% dimethyl sulfoxide (DMSO), trehalose with 5% DMSO, DMSO and glycerol and ethylene glycol (EG). After 7‐day cryopreservation, the expression levels of three spermatogonial stem cell (SSC)‐related genes, octamer‐4 (OCT4), KIT ligand (MGF/SCF) and kit oncogene (C‐KIT), were investigated by quantitative PCR (qPCR). The cell viability was highest for the tissues preserved with 30 mg/ml BSA (77.82% ± 1.22) and 40 mg/ml trehalose (74.23% ± 1.16) compared with other groups (p < 0.05), and the level of expression of the three genes was highest with 30 mg/ml BSA (p < 0.05). Compared with other CPs, the 30 mg/ml BSA and 40 mg/ml trehalose have the better cryopreserve protection. The 30 mg/ml BSA is the most viable media for the cryopreservation of testicular tissue from cattle. 相似文献
The effect of three annually consecutive additions of pig slurry at two rates (90 and 150 m3 x ha(-1) x year(-1) on soils and soil humic acids (HAs) was investigated in a field experiment under semiarid conditions. Soils and pig slurries were analyzed by standard methods. The HAs were isolated from soils and pig slurry by a conventional procedure based on alkaline extraction, acidic precipitation to pH 1, purification by repeated alkaline dissolutions and acidic precipitations, water washing, dialysis, and final freeze-drying. The HAs obtained were analyzed for elemental (C, H, N, S, and O) and acidic functional group (carboxylic and phenolic) composition, and by UV-vis, FT-IR, fluorescence, and ESR spectroscopies. With respect to the control soil, the pig slurry amended soils had greater pH and electrical conductivity, slightly larger total N content, and smaller values of C/N ratio. A decrease of total organic C was observed only in soils amended for 2 and 3 years at the higher slurry rate. With respect to control soil HA, pig slurry HA was characterized by larger contents of S- and N-containing groups, smaller acidic functional group and organic free radical contents, a prevalent aliphatic character, extended molecular heterogeneity, and smaller aromatic polycondensation and humification degrees. Amendment with pig slurry HA determines a number of modifications in soil HAs, including increase of C, S, and COOH contents, C/N ratios, and aliphaticity and decrease of extraction yields and N, O, phenolic OH, and organic free radical contents. These effects are generally more evident after the first year of slurry application and tend to disappear with increasing number of treatments. Most probably, over the years the slightly humified slurry HA is mineralized through extended microbial oxidation, whereas only the most recalcitrant components, such as S-containing, phenolic, and aliphatic structures, are partially accumulated by incorporation into soil HA. 相似文献

Upscaling of agro-ecological indicators applied in regional analyses is sensitive to scale issues of the input data. This study develops a methodology to quantify this sensitivity for an indicator of soil organic carbon (SOC) dynamics at the farming system level. A reference case consists of seven fully described farms in northern Italy. Both upscaling in complexity by substituting measured input with estimated input and upscaling in space by extending the methods to farms not included in the reference case are addressed. The indicator increased with 3-107% at four farms after substituting measured management input with that estimated by an expert, whereas it remained unchanged or decreased at the other three farms. Taking the modal value from a cluster of pedological input did not lead to additional uncertainty in most cases, and only slightly increased it in others. We evaluated spatial upscaling by including 733 farms divided in 18 clusters that were described with less information as compared to the reference farms. Within each cluster, we observed relevant variability of the indicator (coefficients of variation of 12-43%), as a consequence of the heterogeneity of farms comprised in each cluster. In each cluster we calculated the indicator for one virtual farm, defined by using modal values for basic farm inputs. In this case the indicator was highly correlated (R2 = 0.98) with the average of the values obtained using measured basic farm inputs. We conclude that upscaling in complexity and space introduces uncertainty in the values of the indicator compared to the reference case. The extent of such differences depends on the variability of the systems under analysis and on indicator sensitivity. 相似文献
