Protected tomato is the most important horticultural crop in the Algarve (south of Portugal). However, the growing area has decreased by almost 48% since 1995, mainly as a result of the pests Bemisia tabaci and Trialeurodes vaporariorum and the epidemics of Tomato yellow leaf curl virus (TYLCV), a Begomovirus transmitted by B. tabaci. Both whiteflies are vectors of economically important viruses. Recently, Tomato chlorosis virus (ToCV), a member of the genus Crinivirus, transmitted by both B. tabaci and T. vaporariorum, was reported infecting tomato crops in Algarve. A study was carried out to evaluate the dynamics of whitefly populations on tomato crops in Algarve. Population counts of B. tabaci were high in the first months of autumn, then decreased until January, when numbers of T. vaporariorum became higher. Counts of B. tabaci then increased again.  相似文献

The whitefly Bemisia tabaci has been a serious pest in protected tomato crops since 1995 in the south of Portugal (Algarve), causing severe losses mainly resulting from Tomato yellow leaf curl virus (TYLCV), first reported in the autumn/winter season. In order to manage and control the B. tabaci/TYLCV complex, experimental field trials were carried out between 1997 and 2000. Several control methods were tested, such as the application of white screen nets to windows and doors, chemical treatments against B. tabaci and the use of cucumber as a trap crop. The results show that the percentage of plants with TYLCV symptoms mainly reflects B. tabaci infestation level in the first 6 weeks. Screen net protection was the control method that clearly provided a low incidence of plants with TYLCV symptoms in the autumn/winter season. Additionally, insecticide treatments, made weekly in the first 6 weeks, may give improved protection. In the winter/spring season, the B. tabaci/TYLCV complex is a minor problem because of the low populations of B. tabaci and the climatic conditions. Information about the work in progress and results was given to growers through visits to the experimental fields and oral presentations.  相似文献

Objective To compare serological responses in pig herds classified as low or high risk for disease caused by Actinobacillus pleuropneumoniae, using two ELISA tests based on serovar‐independent antigens. Procedure Cross‐sectional sampling was undertaken in 13 commercial herds, the clinical and slaughter histories of which indicated either freedom from (n = 5) or prior confirmed cases of A. pleuropneumoniae (n = 8). In nine herds, approximately 40 pigs each were sampled at 4, 8, 12, 16 and 20 weeks. Three of the remaining four herds were sampled between 6 and 30 weeks of age, and the last was sampled only prior to slaughter, at approximately 24 weeks. Sera were tested in ELISA based on two antigens common among A. pleuropneumoniae serovars: a 39‐kDa outer membrane protein and a recombinant ApxIVA‐N terminus protein. Results Sampling of 1 and 5 to 6‐month‐old pigs provided the most useful information on herd status. The 39‐kDa ELISA was sensitive in detecting infected herds, but had evidence of cross‐reactivity with high seroreactivity rates in older pigs in some low‐risk herds. The ApxIVA‐N ELISA was less seroreactive in high‐risk herds and had higher specificity in low‐risk herds. Conclusion ELISA based on the 39‐kDa subunit are of limited use, because of possible cross‐reactivity, but a high negative predictive value may be useful for risk assessment in suspect herds. Maternal antibody to ApxIVA‐N may be of value in detecting high‐risk herds, but 5% of 4‐week‐old pigs in low‐risk herds were also seropositive in this assay.  相似文献

Canine babesiosis is an infectious disease caused by either Babesia gibsoni or Babesia canis protozoans. The latter is also classified under three different phylogenetic groups, referred to as subspecies B. canis canis, B. canis vogeli and B. canis rossi. The objective of the present study was to validate and standardize a PCR assay to discriminate the organisms at the subspecies level. First, the reference sequences of the 18S rRNA, 5.8S rRNA and 28S rRNA genes, including the internal transcribed spacer 1 (ITS1) and 2 (ITS2) of the most common species and subspecies of the genus Babesia were retrieved from the GenBank database. Subspecies-specific primers (BAB3, BAB4 and BAB5) and one genus-specific primer were designed from the alignment of the sequences. The PCR assays were evaluated in three different combinations of primer pairs in order to assure complete specificity for each reaction. The results of the tests had demonstrated effectiveness of the novel primer pairs BAB1/BAB3, BAB1/BAB4 and BAB1/BAB5 for the amplification of the subspecies-specific target fragments of 746 bp (B. c. canis), 546 bp (B. c. vogeli) and 342 bp (B. c. rossi) by PCR. The original enzymatic amplification assays with novel primers reported in this paper were confirmed to be a reliable tool for the specific discrimination among B. canis subspecies by single-step PCR assays.  相似文献

This work was carried out to predict the combining abilities, both general and specific, for performance traits and bodily yields of Serrasalmidae. Ninety‐six 30‐day‐old juveniles were purchased from two commercial fish farms, 12 of each of the following eight genetic groups: pacu, pirapitinga, tambaqui, tambacu, tambatinga, patinga, paqui and piraqui. Six fish from each genetic group were grown in 500‐L fibreglass tanks (two tanks per genetic group) until they were 495 days old. At the end of the growth period fish were weighed, subjected to morphometric analysis and processed to obtain their bodily yields. Two nuclear markers and one mitochondrial marker were used to confirm the identity of the animals. Combining abilities were obtained using the method proposed by Griffing in 1956 (Australian Journal of Biological Science, 4, 463–493) adapted to a mixed models analysis, environmental effects were estimated by the empirical best linear unbiased estimator method and genetic effects (general and specific combining abilities) were estimated with the empirical best linear unbiased predictor. Predictions of the combining abilities of advanced hybrids were obtained by the mixed models mixture method with normal distributions. Tambaqui showed higher general and specific combining abilities than the other groups for most of the variables, making it the most important genetic group. General combining ability makes a greater contribution to phenotypic variance than specific combining ability for most variables, indicating a predominance of genes with an additive effect in the control of evaluated traits.  相似文献

Allelopathy is the biological phenomenon of chemical interactions between living organisms in the ecosystem, and must be taken into account in addressing pest and weed problems in future sustainable agriculture. Allelopathy is a multidisciplinary science, but in some cases, aspects of its chemistry are overlooked, despite the need for a deep knowledge of the chemical structural characteristics of allelochemicals to facilitate the design of new herbicides. This review is focused on the most important advances in allelopathy, paying particular attention to the design and development of phenolic compounds, terpenoids and alkaloids as herbicides. The isolation of allelochemicals is mainly addressed, but other aspects such as the analysis and activities of derivatives or analogs are also covered. Furthermore, the use of allelopathy in the fight against parasitic plants is included. The past 12 years have been a prolific period for publications on allelopathy. This critical review discusses future research areas in this field and the state of the art is analyzed from the chemist's perspective. © 2019 Society of Chemical Industry  相似文献

Background: This study addresses the antitumoral properties of Penicillium purpurogenum isolated from a polluted lagoon in Northeastern Brazil. Methods: Ethyl Acetate Extracellular Extract (EAE) was used. The metabolites were studied using direct infusion mass spectrometry. The solid Ehrlich tumor model was used for antitumor activity. Female Swiss mice were divided into groups (n = 10/group) as follows: The negative control (CTL−), treated with a phosphate buffered solution; the positive control (CTL+), treated with cyclophosphamide (25 mg/kg); extract treatments at doses of 4, 20, and 100 mg/kg; animals without tumors or treatments (Sham); and animals without tumors treated with an intermediate dose (EAE20). All treatments were performed intraperitoneally, daily, for 15 days. Subsequently, the animals were euthanized, and the tumor, lymphoid organs, and serum were used for immunological, histological, and biochemical parameter evaluations. Results: The extract was rich in meroterpenoids. All doses significantly reduced tumor size, and the 20 and 100 mg/kg doses reduced tumor-associated inflammation and tumor necrosis. The extract also reduced the cellular infiltration of lymphoid organs and circulating TNF-α levels. The extract did not induce weight loss or renal and hepatic toxic changes. Conclusions: These results indicate that P. purpurogenum exhibits immunomodulatory and antitumor properties in vivo. Thus, fungal fermentation is a valid biotechnological approach to the production of antitumor agents.  相似文献

We examined how post‐larval blue crab (Callinectes sapidus) dispersal occurs within Pamlico Sound, NC, USA, a predominantly wind‐driven system. We sampled during multiple 24‐h periods over 2 years (2000–01) to relate the spatial distribution of post‐larvae in the water column with circulation patterns. A hydrodynamic model of the region was used to recreate dispersal trajectories and to assess potential transport mechanisms and pathways that link near‐inlet and across‐Sound nursery habitats. Most post‐larval blue crabs were collected in surface waters at night, and were consistently distributed within the north‐western region of Pamlico Sound. Particle‐tracking simulations suggested that dispersal from the inlets to across‐Sound nursery habitats only resulted from the combination of tidal and wind‐driven currents. Our simulation results further indicated that the northernmost inlet (Oregon Inlet) was the primary supplier of post‐larval blue crabs throughout the northern basin of Pamlico Sound, as crabs ingressing through Hatteras Inlet to the south were not retained within our study area. A dispersal pathway connecting Oregon Inlet and across‐Sound settlement habitats was evident from field observations. Collectively, our results indicate how multiple forcing agents, coupled with post‐larval vertical positioning within the water column, drive estuarine dispersal and connect spatially separated nursery habitats.  相似文献
