Mammals developed an immune system able to functionally polarize into so-called type 1 or type 2 immune pathways, to resolve infections with intracellular and extracellular pathogens, respectively. In the well-studied avian immune system of the chicken, however, no evidence for polarized immunity could be found, as yet. To investigate whether these two major arms of mammalian immunity, regulated by a T helper (Th)1/Th2 cytokine balance, evolved similarly in birds, chickens were exposed to a prevalent intracellular (viral) or extracellular (helminth) infection. By using semi-quantitative RT-PCR analysis we provide evidence that polarization of Th1/Th2 type immunity extends beyond mammalian species, and, therefore, has been evolutionary conserved for more than 300 million years, when the lineages of mammalian and avian vertebrates are assumed to have segregated. 相似文献

ABSTRACT Transgenic soybean (Glycine max) plants expressing Soybean mosaic virus (SMV) helper component-protease (HC-Pro) showed altered vegetative and reproductive phenotypes and responses to SMV infection. When inoculated with SMV, transgenic plants expressing the lowest level of HC-Pro mRNA and those transformed with the vector alone initially showed mild SMV symptoms. Plants that accumulated the highest level of SMV HC-Pro mRNA showed very severe SMV symptoms initially, but after 2 weeks symptoms disappeared, and SMV titers were greatly reduced. Analysis of SMV RNA abundance over time with region-specific probes showed that the HC-Pro region of the SMV genome was degraded before the coat protein region. Transgenic soybean plants that expressed SMV HC-Pro showed dose-dependent alterations in unifoliate leaf morphologies and seed production where plants expressing the highest levels of HC-Pro had the most deformed leaves and the lowest seed production. Accumulation of microRNAs (miRNAs) and mRNAs putatively targeted by miRNAs was analyzed in leaves and flowers of healthy, HC-Pro-transgenic, and SMV-infected plants. Neither expression of SMV HC-Pro nor SMV infection produced greater than twofold changes in accumulation of six miRNAs. Leptospirosis is a zoonotic disease that occurs in humans and several animal species, including cattle, pigs, and horses, and it has a worldwide distribution. In equids, leptospirosis manifests through recurrent uveitis and abortion and other reproductive disorders. The objective of the present study was to investigate the epidemiological situation of Leptospira spp. infection in equids from the Brejo Paraibano microregion in the northeast of Brazil, that is, to assess the prevalence of and identify the major risk factors for infection. For this purpose, blood samples were taken from 257 equids (204 horses, 46 mules, and 7 donkeys), and a microscopic agglutination test (MAT) was used as serological analysis, with 24 serovars of Leptospira spp. as antigens. Antibodies were observed in 16.2% of horses, 13.0% of mules, and 28.6% of donkeys. Animals between the ages of 2.5 and 11 years had a risk of infection 0.4 times lower than those younger than 2.5 years old (odds ratio, 0.4; 95% confidence interval, 0.18-0.90). Further studies are needed to identify potential wild reservoirs and the main routes of infections. 相似文献

Although potatoes are the second most valuable crop in Kenya, poor seed potato quality has limited industry expansion. The objectives of this study were to describe the seed potato system, identify problems, determine use of high-quality seed and estimate farmer willingness to pay (WTP) for different types of seed. Data was collected from interviews, forums and surveys. A contingent valuation method (CVM) model was also developed to analyze the factors that influence farmer WTP. It was found that the seed system consists of three sub-components: Formal, Semi-Formal and Farmer. More than 96 % of Kenya seed potatoes were produced in the Farmer category, and were considered to be poor quality. Growers identified bacterial wilt, viral diseases and late blight as the most serious problems. The WTP model revealed that farmer age, distance to seed source, potato training, gender and region were statistically significant explanatory variables. The study recommends that training be conducted to increase production of “clean seed” and “positively-selected seed” in the Semi-Formal part of the seed system. 相似文献

The effects of landscape patchiness on the diversity of birds of the Georgia Piedmont were investigated during 1993. Birds were sampled along line transects within relatively large (10–13.25 ha) and small (less than 3.25 ha) forest patches located within nonforest agricultural landscapes. Patterns of habitat use in these patches were compared to those in contiguous forest patches larger than 13.25 ha. Analysis of variance revealed significant differences in diversity between large and small woodlots and between contiguous and fragmented landscapes, especially in terms of the numbers of edge and interior species and winter-resident, summer-resident, and year-round birds observed. 相似文献

Ecological patterns and processes operate at a variety of spatial scales. Those which are regional in nature may not be effectively captured through the combination of conservation plans derived at the local level, where land use planning frequently takes place. Conversely, regional conservation plans may not identify resources important for conservation of intraregional ecological variation. We compare modeled conservation networks derived at regional and local scales from the same area in order to analyze the impact of scale effects on conservation planning. Using the MARXAN reserve selection algorithm and least cost corridor analysis we identified a potential regional conservation network for the Central Valley ecoregion of California, USA, from which we extracted those portions found within five individual counties. We then conducted the same analysis for each of the five counties. An overlay of the results from the two scales shows a general pattern of large differences in the identified networks. Especially noteworthy are the trade-offs and omissions evident at both scales of analysis and the disparateness of the identified corridors that connect core reserves. The results suggest that planning efforts limited to one scale will neglect biodiversity patterns and ecological processes that are important at other scales. An intersection of results from the two scales can potentially be used to prioritize areas for conservation found to be important at several spatial scales. 相似文献

Tephritid fruit flies are serious economic pests worldwide. As larvae, they feed and develop within the pulp of host fruits, making infestation difficult to detect by visual inspection. At U.S. ports of entry, incoming produce shipments are checked for infestation by manually cutting open a small sample of fruit and searching for tephritid larvae. Consequently, there is a need for more sensitive, high-throughput screening methods. This study evaluated gas chromatography (GC) as a potential technology for improved detection of hidden infestation. Grapefruits (Citrus × paradisi Macfad.) infested with immature stages of the Caribbean fruit fly Anastrepha suspensa (Loew) (Diptera: Tephritidae) were examined to determine if infested fruit emitted a chemical profile distinct from that of non-infested fruit. Peaks identified by GC analysis were grouped into three classes. Chemicals detected in similar quantities in all samples, or slightly elevated in infested samples, were regarded as non-diagnostic background volatiles. Chemicals highly elevated after oviposition, during the last instar exit stage, and in experimentally-pierced fruit were interpreted to be indicators of citrus peel injury, and included d-limonene and β-ocimene. Chemicals elevated exclusively in the larval infestation stages were considered indicators of feeding damage and potentially diagnostic of infestation, and included hexyl butanoate and an unidentified compound. The peaks associated with injury and feeding were also detectable with a portable ultra-fast GC analyzer that required less than 80 s per sample. Further studies will investigate the potential application of these results for development of a rapid, non-destructive screening method for detection of tephritid infestation. 相似文献

Plant landraces have long been recognized as potential gene pools for biotic and abiotic stress-related genes. This research used spring wheat landrace accessions to identify new sources of resistance to the wheat stem sawfly (WSS) (Cephus cinctus Norton), an important insect pest of wheat in the northern Great Plains of North America. Screening efforts targeted 1409 accessions from six geographical areas of the world where other species of grain sawflies are endemic or where a high frequency of accessions possesses the resistance characteristic of solid stems. Resistance was observed in approximately 14% of accessions. Half of the lines displayed both antixenosis and antibiosis types of resistance. Among the resistant accessions, 41% had solid or semi-solid stems. Molecular genetic screening for haplotypes at the solid stem QTL, Qss.msub.3BL, showed that 15% of lines shared the haplotype derived from ‘S-615’, the original donor of the solid stem trait to North American germplasm. Other haplotypes associated with solid stems were also observed. Haplotype diversity was greater in the center of origin of wheat. Evaluation of a representative set of resistant landrace accessions in replicated field trials at four locations over a three year period identified accessions with potential genes for reduced WSS infestation, increased WSS mortality, and increased indirect defense via parasitoids. Exploitation of distinct types of plant defense will expand the genetic diversity for WSS resistance currently present in elite breeding lines. 相似文献
