The comparison of serological responses in a sample of adult, vaccinated and field‐infected bovines with Brucella abortus is reported. Indirect enzyme immunoassay (EIA) titration curves and Western blotting tests for smooth‐type lipopolysaccharide (S‐LPS), rough‐type LPS (R‐LPS) and lipid A were performed. In the initial screening of sera, an overall prevalence of 20.5 % was found, which corresponds to a country with a high incidence of brucellosis. End‐point EIA titres against LPS antigens from vaccinated and field‐infected cows were not significantly different. However, the absorbance values in the titration curves were significantly higher for S‐LPS as compared with the other antigens. A high correlation coefficient (r=0.933) was obtained when the titres to R‐LPS versus lipid A were compared. Western blotting reactions of vaccinated and field‐infected animals were indistinguishable. S‐LPS, R‐LPS and lipid A epitopes were recognized in a heterogeneous manner. In general, the number of bovines that reacted against LPS was higher in the field‐infected group, with a stronger binding to S‐LPS. Based on our observations, the vaccinated and field‐infected bovines are capable of producing similar antibody responses to the Brucella main outer surface antigen, LPS. It should be emphasized that the humoral response of cattle to Brucella LPS contains significant amounts of antibodies to other antigenic moieties of this important surface molecule, which may contribute to the immunity to brucellosis.  相似文献

Most studies on lactation curves only consider milk yield and describe a standard lactation curve of dairy cows, showing a peak or maximum daily yield occurring between 4 and 8 weeks after calving, followed by a daily decrease in milk yield until the cow is dried off. Wood's model is a widely used lactation curve function. Wood's model was fitted to test-day records of 95,405 lactations of parities lower than 5. Milk traits were milk yield (MY), fat percentage (F%), protein percentage (P%), fat yield (FY) and protein yield (PY), and the lactation curve was individually considered as a cluster of five linked curves. Milk trait and parity influence the goodness of fit of Wood's model. In 19.3% of the lactations, the shape of the MY, FY and PY curves follows the standard lactation curve while F% and P% have the reversed standard shape. The initial phase of lactation with the FY and PY curves contributes to the high variability of shapes.  相似文献

Dehydration, to varying degrees, will inevitably occur in horses participating in endurance trail rides. This water loss is directly related to the amount of body water lost through evaporative cooling, that in turn being related to the amount and rate of work performed, and to the environmental temperature and humidity.An electronic apparatus that can substitute for the conventional and time-consuming venous haematocrit was developed to measure the animals' hydration status. The mechanism of this electrometer is based upon the relationship between the red cell content and the electrical conductivity of the blood. The electrometer reading gave a significant correlation (p<0.001) with the venous haematocrit, thus indicating that the electrometer is a reliable apparatus for determining the animals' hydration status within seconds of venepuncture.  相似文献

The early diagnosis of bovine leukosis virus (BLV) infection, the aetiological agent in enzootic bovine leukosis, is important for the implementation of control measures. BLV infection is currently assessed by the detection of circulating antibodies against the viral envelope protein, gp51. However, this approach has shortcomings in the time taken to detect anti-BLV antibodies (three to four weeks after infection), and in the failure to detect antibodies in some animals. Clearly a technique such as the polymerase chain reaction (PCR), which directly detects the presence of viral DNA, has advantages over methods designed to measure host antibodies. The use of PCR for the detection of proviral DNA in an affected DNA sample with as little as 10(-5) micrograms of host DNA using agarose gel electrophoresis followed by ethidium bromide staining is described here. It was possible to improve the sensitivity of this assay by using hybridisation analysis with a BLV gene probe. PCR used in combination with hybridisation analysis will provide a sensitive diagnostic assay to detect BLV when antibody tests give weakly positive or equivocal results.  相似文献

Five crossbred wethers (58 kg) fitted with cannulas in the rumen, duodenum, and ileum were used in a 5 x 5 Latin square design to study effects of feeding combinations of alkaline hydrogen peroxide-treated wheat straw (AHP-WS) and alfalfa hay at restricted intakes on site and extent of nutrient digestion. Additionally, flows and disappearance of N and amino acids (AA) in the small intestine were regressed on alfalfa nitrogen intake (ANI) to estimate alfalfa's contribution to postruminal N and AA supplies. Diets consisted of 80:20 forage:concentrate mixtures; diet designations were 80:0, 80% AHP-WS and no alfalfa; 60:20, 60% AHP-WS and 20% alfalfa; 40:40, 40% AHP-WS and 40% alfalfa; 20:60, 20% AHP-WS and 60% alfalfa; and 0:80, no AHP-WS and 80% alfalfa. A modest positive quadratic (P less than .05) response was noted for total tract digestibility (TTD) of OM; values were 76.5% for diets 80:0 and 0:80 vs 78% for diet 40:40. Ruminal digestibility (percentage of intake) of NDF and ADF increased in a quadratic manner from 43 and 30%, respectively, for diet 0:80 to 71 and 70%, respectively, for diet 80:0. Ruminal digestibility of fiber may have been enhanced due to linear (P less than .05) decreases in liquid and particulate dilution rates, resulting in increased ruminal residence time of fiber as alfalfa hay replaced AHP-WS. Liquid and particulate dilution rates decreased linearly from 6.4 and 5.2%/h, respectively, for diet 80:0 to 5.4 and 3.4%/h, respectively, for diet 0:80. Regression analysis of N data indicated that alfalfa N had a ruminal escape value of 26%.  相似文献

Because certain inflammatory processes are dependent on the fatty acid composition of the cellular membrane, dietary manipulations that replace omega-6 fatty acids with omega-3 fatty acids may modify inflammatory responses. We investigated the effect of supplemental dietary linseed oil, containing the omega-3 fatty acid, alpha-linolenic acid, on in vivo responses of horses to endotoxin. One group of horses (n = 6) was fed a control pelleted ration (0% linseed oil), and another group of horses (n = 6) was fed an 8% linseed oil pelleted ration. After 8 weeks of consuming these rations, all horses were given 0.03 micrograms of Escherichia coli 055:B5 endotoxin/kg of body weight, infused over 30 minutes. Horses were monitored over 24 hours. Compared with baseline values within each ration group, endotoxin infusion caused significant (P less than 0.05) increase in rectal temperature, heart rate, and plasma concentration of thromboxane B2, 6-keto-prostaglandin F1 alpha, and fibrinogen and significant (P less than 0.05) decrease in total WBC count. Compared with baseline values within each ration group, endotoxin infusion failed to cause significant changes in prothrombin, activated partial thromboplastin, thrombin, or whole blood recalcification times, serum concentration of fibrin degradation products, PCV, or plasma total protein concentration. Before and after endotoxin infusion, horses given the linseed oil ration had longer mean whole blood recalcification time and activated partial thromboplastin time than did horses fed the control ration.  相似文献

The medical records of 31 dogs diagnosed with prostatic carcinoma at the teaching hospital between January 1970 and October 1987 were reviewed to determine whether gender status had an effect on the clinical features or biologic behavior of the disease. The influence of temperature (10 degrees C and 20 degrees C) on pharmacokinetics and metabolism of sulphadimidine (SDM) in carp and trout was studied. At 20 degrees C a significantly lower level of distribution (Vdarea) and a significantly shorter elimination half-life (T(1/2)beta) was achieved in both species compared to the 10 degrees C level. In carp the body clearance parameter (ClB(SDM)) was significantly higher at 20 degrees C compared to the value at 10 degrees C, whereas for trout this parameter was in the same order of magnitude for both temperatures. N4-acetylsulphadimidine (N4-SDM) was the main metabolite of SDM in both species at the two temperature levels. The relative N4-SDM plasma percentage in carp was significantly higher at 20 degrees C than at 10 degrees C, whereas there was in trout no significant difference. In neither species was the peak plasma concentration of N4-SDM (Cmax(N4-SDM)) significantly different at two temperatures. The corresponding peak time of this metabolite (Tmax(N4-SDM)) was significantly shorter at 20 degrees C compared to 10 degrees C in both carp and trout. In carp at both temperatures, acetylation occurs to a greater extent than hydroxylation. Only the 6-hydroxymethyl-metabolite (SCH2OH) was detected in carp, at a significant different level at the two temperatures. Concentrations of hydroxy metabolites in trout were at the detection level of the HPLC-method (0.02-micrograms/ml). The glucuronide metabolite (SOH-gluc.) was not detected in either species at the two temperatures.  相似文献

A double L-shaped intestinal cannula was developed in an attempt to overcome problems observed previously with simple T-type cannulas. The cannula was constructed from cyclopolyvinyl chloride water pipe fittings. Construction materials were fairly rigid, but by connecting the split cannula pieces with elastic castration bands the cannula had some flexibility. Placing a short cone over the exposed cannula barrel reduced mechanical damage to the intestine. The double L cannula required a much smaller incision in the intestine during surgical insertion than a T-type cannula; it also simplified replacement. Construction is described; use and performance of the cannula has been satisfactory.  相似文献
