ABSTRACT

1. Chicken-associated Campylobacter spp. are the cause of most food poisoning cases in Europe. In order to study the host–pathogen interactions, a reliable and reproducible method of colonising chickens with the bacteria is required.

2. This study aimed to identify a more appropriate and less invasive method of colonisation (cf. gavaging) by seeding bedding material (litter) that commercial chickens are kept on with a mixture of Campylobacter spp., broth and faeces.

3. The first phase of the study tested the longevity of Campylobacter spp. recovery in seeded litter over 24 h: significantly more Campylobacter spp. was recovered at 0 or 3 h post-seeding than at 6 and 24 h post-seeding, indicating that the pathogen can survive to detectable levels for at least 3 h in this environment.

4. In the second phase, three groups of 10 broiler chickens (negative for Campylobacter spp. prior to exposure) were exposed at 21 days of age to one of three different Campylobacter jejuni and C. coli mixes (A, B, C), using the method above. At 28 days of age, birds were euthanised by overdose of barbiturate or cervical dislocation, and livers and caeca removed for Campylobacter spp. assessment.

5. All liver and 28/30 caeca samples tested positive for Campylobacter spp., with mix A and C giving higher counts in the caeca than mix B. The method of euthanasia did not affect Campylobacter spp. counts.

6. In conclusion, a successful method for reliably colonising broiler chickens with Campylobacter spp. has been developed which negates the need for gavaging and is more representative of how contamination occurs in the field.  相似文献

External iliac artery atherosclerotic disease and aneurism occur in man. For treatment, imaging is required to facilitate minimally invasive introduction and advancement of stents within the intended vessels. Sheep are commonly used to test and improve stents. However, little information is published regarding the angiographic anatomy of the iliac arteries in the ovine species. The objective of this study was to describe the angiographic anatomy of the iliac arteries in the sheep. Computed tomography (CT) angiography and gross anatomical dissection were performed in, respectively, 10 and 43 adult ewes. Diameters and lengths of the arteries were measured. In comparison with man, salient anatomical differences were identified in the sheep: (1) the absence of common iliac arteries, (2) the common trunk at the origin of internal iliac arteries and (3) the location of the bifurcation of the external iliac arteries into femoral arteries in the pelvis (not in the limb). External iliac arteries in this series of sheep were 86 mm long in average and had a mean diameter of 7.5 mm. Lengths of arteries are only slightly different between man and sheep, while diameters are rather similar. Therefore, the sheep model appears to be sufficiently similar to man to test stent properties. This study provides useful reference images and measures of lengths and diameters of relevant arteries that could be applied to research with ovine models.  相似文献

对牛体细胞核移植技术中核供体细胞周期同期化处理,电融合条件及去核方法等进行了研究。结果表明,血清-饥饿组(细胞周期同期化处理组)的移核重构胚细胞融合率(83.3%),卵裂率(70%)和囊胚发育率(33.8%)与血清-选择组对应指标间无显著性差异(82.3%,69.2%,32.4%,P>0.05);但上述2组分别极显著高于血清-随机组(64.8%,33.8%,4.35%,P<0.01),试验确立了场强1.025kV/cm,脉冲宽度50μs,连续2次脉冲为最佳电融合条件。摸索出的点击去核法,对卵母细胞进行去核,去核成功率达90%,极显著高地吸引法的68.3%(P<0.01);其囊胚发育率为34.1%,显著高于吸引法的18.0%(P<0.05),与挤压法(20.9%)相比差异不显著(P>0.05),移植重构胚胎移植后,产2头成活的体细胞克隆牛。  相似文献

A new serotype of calicivirus was isolated from California sea lions (Zalophus californianus) with severe vesicular disease. Neutralizing antibodies were found in 27 of 82 (32.9%) serum samples from California sea lions and in 15 of 146 (10.3%) serum samples from Steller sea lions (Eumetopias jubatus) tested. The seropositive animals were widely dispersed along the margins of the eastern Pacific basin, from the Bering Sea to the Santa Barbara Channel. Seropositive samples were found from as early as 1976 through the present time. This new calicivirus serotype, San Miguel sea lion virus type 13, was inoculated into weaned pigs, resulting in induction of severe vesicular disease, which spread to all pigs, including uninoculated pen contacts. Virus was continually shed by most of the pigs throughout the 2-week duration of the experiment.  相似文献

Eleven cows in late lactation were exposed to butyric acid for three weeks and were compared to five controls. Two intraruminal doses were daily applied, that is 1.0 g/kg B.W. of butyric acid to six animals and 1.0 g/kg B.W. of sodium butyrate to another five. Decline in milk yield was clinically recorded in response to butyric acid, while muscle tremor and diarrhoea resulted additionally from sodium butyrate. Behaviours of the clinico-chemical parameters of beta-OH-butyrate, glucose, free fatty acids, bilirubin, ASAT, gamma-GT, AP, and cholesterol were comparable to those in fattening bulls. Liver damage was not safely established. Some of the clinico-chemical alterations were more strongly pronounced after administration of sodium butyrate. One cow fell ill with ketosis under butyric acid load.  相似文献

Estimation of the developmental age of the bovine fetus and newborn calf   总被引:1,自引:0,他引:1  
Jersey cows from several herds provided 97 fetuses and 24 calves at fixed gestational intervals between 80 and 290 days after conception. The fetuses and calves were killed, weighed and measured and, after dissection, the sizes and weights of a range of skeletal and soft tissues were recorded. Six morphological measurements emerged as most suitable for the determination of developmental age in the normal fetus. By plotting their mean values and 95 per cent tolerance limits, the rates of growth and the variability of each measurement were defined. Long bone length was the most useful single measurement for predicting the developmental age of the fetus. Brain weight, bodyweight, crown-anus length and long bone length showed curvilinear growth patterns; age prediction equations derived from these measurements are complicated to use and additional simplified formulae have been derived. The number of appendicular ossification centres also had predictive value, but it could not be used to determine fetal developmental age between 100 and 160 days gestation.  相似文献

Amyloid angiopathy with cerebral hemorrhage and senile plaques was found in the brain of aged dogs. In all 9 cases examined, 13 to 19 years old, 6 males and 3 females, amyloid deposits were observed mostly in the wall of cerebral arterioles and capillaries showed hyaline degeneration. The accumulation of amyloid fibrils measuring about 10 nm in diameter was seen in the cerebral vessel wall by electron microscopy. The cerebral hemorrhage was observed in 6 of 9 dogs and 2 of them showed massive hemorrhage. The hemorrhagic foci were sometimes closely contact with the vessels involved in amyloid angiopathy. In addition, senile plaques being classified into 2 types were found in the cerebral cortex of 3 dogs. The first type was characterized by the accumulation of degenerative neurites and often contained granular argyrophilic material. The second type had a well-defined amyloid core with neuritic halo. Amyloid deposits were also found in or around intestinal vessel walls of 3 dogs. The amyloid deposited in the cerebral vessels, senile plaques and intestinal vessels showed characteristic green birefringence under the polarized light even after potassium permanganate treatment.  相似文献

The relative antiandrogen-induced prostate involution activity of the newly synthesized hydroxyflutamide pro-drug was compared with that of flutamide in 25 Beagles. Secondary antiandrogen activity of both drugs on the testes and mammary tissue was investigated. Daily oral administration of both compounds at 2 dosages (ie, 2.5 and 5.0 mg/kg of body weight) during a 7-week period was monitored by transabdominal ultrasonography of the prostate twice a week. Cross-sectional area estimates of the prostate gland calculated from oblique dorsoventral, and transverse sonographic measurements were diminished significantly in some of the treated dogs as early as day 14 of drug administration. All treated dogs had significant differences in reduction by day 47. Involution was related directly to dose (P less than 0.05), but no difference was observed between test compounds. Differences in secondary antiandrogen activity were not remarkable. Flutamide was not found to have any activity advantage in vivo over hydroxyflutamide. It was concluded that ultrasonography can be a highly effective means of monitoring prostate size, and of monitoring drug-induced involution over time.  相似文献

An experiment was conducted to develop a system useful for measuring methionine requirements of growing steers. Seven ruminally cannulated steers (312 kg, gaining .91 kg/d) were fed a diet based on ammoniated corn cobs, corn starch, molasses and urea. Quantities of N and sulfur-containing amino acids disappearing from the small intestine were 96.0 and 14.8 g/d, respectively. Postruminal infusions of Na-caseinate (CAS) resulted in linear (P less than .01) increases in N retention with values increasing from 30.1 g/d with no postruminal CAS infusion to 39.3, 50.8 and 59.2 g/d (averaged across methionine supplementation) when 100, 200 and 300 g/d CAS were infused. Postruminal infusions of a mixture of crystalline L-amino acids (simulating the nonsulfur-containing essential amino acid pattern of casein; SIM) at levels of 100, 200 and 300 g/d also led to linear increases (P less than .01) in N retention with steers retaining 30.9, 38.9 and 50.5 g N/d (averaged across methionine supplementation), respectively. Postruminal infusion of 12 g/d L-methionine across CAS and SIM infusions improved (P less than .01) N retention by 7.6 g/d but infusion of SIM, which is devoid of sulfur amino acids, also increased N retention. Responses to methionine supplementation was greatest when 200 or 300 g/d SIM were abomasally infused. The data are interpreted to demonstrate that, for steers fed a diet containing little true protein, postruminal supplementation with nonsulfur-containing amino acids tended to increase the ability of growing steers to respond to methionine supplementation.  相似文献
