The aim of the present study was to investigate minimum inhibitory concentration (MIC) distributions by broth microdilution (BMD) method and to determine the preliminary epidemiological cut-off value (ECV) of colistin by epidemiologicalcut-off (ECOFF) finder against E.coli from chickens in China.Anal swabs were collected from chicken farms in China.BMD method was used to measure MIC50 and MIC90 of colistin which were 2 and 4 μg ? mL~(-1),respectively.MIC frequency distributions for colistin were used to estimate preliminary ECV (8 μg ? mL~(-1)).High percentages of resistance to ampicillin (94.12%),nalidixic acid (94.12%),enrofloxacin (94.12%),tetracycline (94.12%),ciprofloxacin (88.24%),florfenicol (88.24%),neomycin (64.71%),gentamicin (58.82%),levofloxacin (58.82%),doxycycline (88.24%) and cefalexin (76.47%) were found.In addition,low percentages of resistance to amikacin (5.88%),spectinomycin (17.65%) and fosfomycin (41.18%) were noted.Notably,amoxicillin,sulfisoxazole and trimethoprim resulted in a 100 % resistance generation efficacy rate.Prevalence of mcr-1 in E.coli (9/17) in chromosomal DNA was higher than mcr-4 (2/17) gene,and mcr-1 (5/17) was higher than mcr-4 (3/17) in plasmid.  相似文献

This field study was conducted to compare conception and insemination efficiency responses to intrauterine polyvinylpyrrolidone-iodine (PVP-I) and antibiotic (AB) treatments in dairy cows that experienced pregnancy loss. Data were collected from lactation cows with a history of pregnancy loss 27 to 70 days post-insemination (n = 97) during the 1st to 3rd lactation (days in milk = 196 ± 28). Cows were subjected to 1 of 3 treatments: i) 50 mL saline solution intrauterine infusion (S; n = 23); ii) 2% PVP-I (n = 42); or iii) 150 mg of amoxicillin trihydrate and 40 mg/mL gentamicin sulphate (n = 32). All cows followed the progesterone-based fixed-time insemination protocol. Data were analyzed by Chi-square test and 1-way analysis of variance. The PVP-I treatment (n = 25; 59.5%) was as effective as the AB treatment (n = 19; 59.4%), compared to the S treatment (n = 5; 21.7%) to achieve a new conception. The cows in group PVP-I conceived in a shorter time than those in group AB (46.0 ± 8.7 days versus 105.0 ± 10.0 days; P < 0.05) with a more efficient insemination to conception ratio (2.32 ± 0.43 versus 4.10 ± 0.32; P < 0.05). Data suggest that intrauterine PVP-I administration is superior to intrauterine AB administration in rescheduling reproductive protocol upon late embryonic and fetal losses.  相似文献

Somatic embryogenesis (SE) in Eucalyptus spp. has been limited to germinated seeds, flowers, lignotubers or zygotic embryos. The low yield of somatic embryos from leaf explants has hampered progress, even though leaves offer a more viable source of clonal explants from superior selected genotypes. It was hypothesised that SE from leaf explants could be enhanced through pairing of synergistic exogenous plant growth regulators, such as natural auxins with natural cytokinins. Callus and embryo induction using 2,4-dichlorophenoxyacetic acid (2,4-D), α-naphthalene acetic acid (NAA), indole acetic acid (IAA), and indole butyric acid (IBA), each at either 1.0 or 3.0?mg L?1, indicated that IAA and IBA favoured significantly higher numbers of embryos compared with 2,4-D or NAA. Hence, IAA and IBA were used for subsequent experiments, combining them (at 1.0?mg L?1) with either the synthetic cytokinin, kinetin, or the natural cytokinin, trans-zeatin, both at 0.1?mg L?1. The combination of trans-zeatin and either IAA or IBA resulted in a significant increase in SE (e.g. 86 ± 17.2% and 23 ± 3.2% for IAA with trans-zeatin and kinetin, respectively), compared with kinetin, or with these auxins alone. Embryo maturation and plantlet regeneration was highest in those calli that were induced with IAA and trans-zeatin, indicating that maturation was dependent on auxin depletion, based on the stability of the analogue used for induction. For the E. grandis clone under study, the use of synergistic plant growth regulators significantly enhanced SE from leaf explants, thus presenting the opportunity to benefit from the advantages that SE offers over conventional organogenesis.  相似文献

A field trial was conducted to evaluate the role of boron (B) application at different growth stages in improving the growth,yield and net economic return of rice at farmer’s fields during summer season,2009.Boron was soil applied (1.5 kg/hm2) at the transplanting,tillering,flowering and grain formation stages of rice;foliar applied (1.5% B solution) at the tillering,flowering and grain formation stages of rice,and dipped seedling roots in 1.5% B solution before transplanting;while control plots did not apply any B.Boron application (except dipping of seedling roots in B solution,which caused toxicity and reduced the number of tillers and straw yield than control) substantially improved the rice growth and yield.However,soil application was better in improving the number of grains per panicle,1000-grain weight,grain yield,harvest index,net economic income and ratio of benefit to cost compared with the rest of treatments.Overall,for improving rice performance and maximizing the net economic returns,B might be applied as soil application at flowering.  相似文献

Cultivar mixtures have been suggested as ways to increase crop productivity. This field study was conducted to investigate the competition among five cultivated varieties of wheat (Triticum aestivum L.) for the traits like plant height, flag leaf area, fertile tillers per plant, spike length, spikelets per spike, spike density, grain weight per spike, and seed index in relation to competition with yield per plant. It was observed that cultivar mixtures ought to be more productive than corresponding pure stands. Four binary combinations of Seher2006-Kohistan97, Farid2006-SH2002, SH2002-Shahkar95, and SH2002-Kohistan97 were superior in performance, showing land equivalent ratio (LER) values greater than one for plant height, fertile tillers per plant, spike density, grain weight per spike, and grain yield per plant. It was observed that the performance of binary mixtures was mainly due to the increased average performance in spike density, grain weight per spike, and grain yield per plant. The competitive ability of wheat varieties in mixtures was independent of each other for the respective traits, which lead to cumulative effects for the increased performance within the associates in mixtures.  相似文献

Yellow mosaic disease (YMD) caused by whitefly (Bemisia tabaci) transmitted mungbean yellow mosaic India virus (MYMIV) is an important constraint of mungbean in Pakistan. One hundred sixty-two mungbean genotypes from eight different geographic regions were evaluated for resistance to MYMIV and its whitefly vector B. tabaci. Resistance levels were assessed by visual scoring of symptoms in the field under natural conditions, in net-house by artificial transmission through whitefly and in greenhouse using graft inoculation for two consecutive years in 2008 and 2009. None of the tested genotypes was found to be disease free, while considerable variation in responses was observed among the genotypes and testing methods. All genotypes were found to be systemically infected in the field. In 2008, 32 genotypes were found to be resistant and 67 were moderately resistant with severity index (SI) values ranging from 1.0 to 1.4 and from 1.5 to 2.4, respectively. In 2009 34 genotypes were found to be resistant and 38 were moderately resistant with the same SI value ranges as in 2008, and the remaining genotypes were moderately susceptible to highly susceptible. However, only 35 genotypes were found to be moderately resistant with SI values of 1.5-2.4 and latent periods (LP) of 17-21 days when evaluated using artificial transmission through whiteflies. Similar results were obtained when these genotypes were evaluated using graft inoculation. Significant differences in adult whitefly densities among tested genotypes were also observed during both years but no correlation between the number of whiteflies and disease severity was observed. The results indicated that these genotypes did not have a high level of resistance against MYMIV, however, they may provide sources of partial resistance which can be exploited in the breeding programmes to develop mungbean genotypes resistant to MYMD or they can be used directly as varieties to manage MYMD after evaluation for acceptable agronomic characteristics, adaptation and stability in various regions.  相似文献

Response Surface Methodology (RSM) via Central Composite Design (CCD) was used to optimize the alkaline hydrolysis of paddy straw to improve ferulic acid extraction. The parameters involved, namely temperature (°C), concentration of NaOH (M) and duration of extraction (h) are screened by Full Factorial Studies (FFD). The result obtained from FFD was 0.518% (5.18 mg/g). The interactions between each parameters involved were studied using Central Composite Design (CCD). Upon optimization, the percentage of ferulic acid improved from 0.518% to 0.817% (8.17 mg/g). The optimum conditions obtained from this study are 125 °C, 3.90 M, 2.30 h for temperature, concentration of NaOH and extraction time, respectively. This result shows a significant improvement than the previous published work. Details of the experimental design, statistical analysis and interpretation are discussed below.  相似文献

Arsenic(As) contamination in soils has posed a severe threat to safe crop production. The previous studies showed the antagonism between phosphorus(P) and As in plant growth and As uptake, while the mechanisms of alleviating As toxicity by P is not completely clear. Due to the limiting P condition, it is imperative to understand how low P addition can be used to suppress arsenate As(V) uptake and the subsequent mechanisms involved. Thus in this study we investigated the effect of P addition on As uptake, anti-oxidative enzyme activity, and anti-oxidant content, and the relative expression of transport, defense, and detoxification genes using two barley genotypes differing in As toxicity tolerance. P addition significantly reduced As concentration in plant tissues, and caused the great changes in activities of catalase and superoxide dismutase, glutathione content, and the relative expression of examined genes when the plants of the two barley genotypes were exposed to 100 μmol L~(–1) As, with ZDB160(As-tolerant) being much more affected than ZDB475(As-sensitive). The current results show that P addition can alleviate As toxicity by regulating the expression of As transport, defense, and detoxification genes to a greater extent in As tolerance of barley, suggesting the possibility of controlling As uptake and toxicity by applying low amount of P fertilizers in the As-contaminated soils.  相似文献
