Tetracapsula bryosalmonae, previously referred to as PKX, causes proliferative kidney disease (PKD) in salmonids and is an economically important myxozoan pathogen in salmonid culture. A variety of molecular and immunological tools have been developed to detect the parasite. To determine the specificity of four monoclonal antibodies (MAbs) raised against T. bryosalmonae, archive material of fish infected with various myxosporean species was obtained and immunostained. Wild fish were also collected from enzootic waters and examined for T. bryosalmonae infection using immunohistochemistry and the polymerase chain reaction (PCR). Three of the MAb probes appear to be specific for T. bryosalmonae while only two of the five sets of primers tested appeared to specifically amplify T. bryosalmonae DNA. The results of the immunostaining and the PCR demonstrate that T. bryosalmonae occurs in the tubules of grayling Thymallus thymallus L., brown trout, Salmo trutta L. and Atlantic salmon, Salmo salar L. outside of the PKD season (June‐September) in the UK. This confirms the results of previous studies that these species are the preferred fish hosts for the parasite in the UK.  相似文献

 Composts made from biosolids and municipal solid wastes contain heavy metals which may be exported outside soil systems by plants, animals and surface and subsurface waters after the compost has been added to soils. Chemical distributions of Cu, Zn, Cr, Pb, Ni and Co were determined by eight sequential extractions of co-composted materials sampled on days 0, 13, 27 and 41. The concentrations of residual Zn, Cr, Cu and Pb increased by 145, 124, 73.6 and 26.3% during the composting period, respectively, whereas the concentration of residual Ni remained relatively constant and that of Co decreased by 60% over the same period. These results show that co-composting contaminated residues substantially reduces the extractability and exchangeability of four out of six heavy metals, suggesting that the risks of entering the food chain and contaminating crops, animals and water reserves would be equally reduced. Fourier-transform infrared spectra showed that heavy metals in the compost are bonded to COO- groups of the organic matter. Received: 20 March 1998  相似文献

Market demands for new sensors for food quality and safety stimulate the development of new sensing technologies that can provide an unobtrusive sensor form, battery-free operation, and minimal sensor cost. Intelligent labeling of food products to indicate and report their freshness and other conditions is one important possible application of such new sensors. This study applied passive (battery-free) radio frequency identification (RFID) sensors for the highly sensitive and selective detection of food freshness and bacterial growth. In these sensors, the electric field generated in the RFID sensor antenna extends from the plane of the RFID sensor and is affected by the ambient environment, providing the opportunity for sensing. This environment may be in the form of a food sample within the electric field of the sensing region or a sensing film deposited onto the sensor antenna. Examples of applications include monitoring of milk freshness, fish freshness, and bacterial growth in a solution. Unlike other food freshness monitoring approaches that require a thin film battery for operation of an RFID sensor and fabrication of custom-made sensors, the passive RFID sensing approach developed here combines the advantages of both battery-free and cost-effective sensor design and offers response selectivity that is impossible to achieve with other individual sensors.  相似文献

This study evaluated the effects of inhibitors on polyphenol oxidase (PPO) activity, the effect of the PPO inhibitor tropolone on noodle darkening, and the correlation of PPO activity with darkening of alkaline noodles. The PPO inhibitors tropolone and salicylhydroxamic acid (each at 1 microM) reduced kernel PPO activity by approximately 50% in three hexaploid wheat cultivars but did not inhibit PPO activity in the two very low PPO cultivars, durum Langdon, and the synthetic hexaploid-derived ID580. Tropolone (100 microg/g flour) inhibited alkaline noodle darkening (deltaL*) by 13-25% in the low PPO wheat cultivar, ID377s, and by 39-54% in the high PPO wheat cultivar, Klasic. Alkaline noodle darkening among 502 wheat samples was correlated with kernel PPO activity (r = 0.64). Results substantiate the hypothesis that PPO plays a major role in darkening of alkaline noodles. However, results also indicate that substantial darkening would occur even at zero PPO activity, as measured in the kernel PPO assay. Therefore, darkening of alkaline noodles is probably due to the cultivar-specific level of PPO activity and the presence of at least one additional darkening mechanism. Further investigation is required to identify the phenolic discoloration agent(s) and to determine the potential roles of non-PPO discoloration mechanisms, both enzymatic and nonenzymatic, in wheat products.  相似文献

A biological source treatment (BST) technique using remote sensing and biogeochemistry has been developed to address acid mine drainage (AMD) at its source. The BST technique utilizes down-hole injections of microbial inoculum and substrate amendments to establish a biofilm on the surface of metal sulfides (AMD source material). The treatment results in an elevated groundwater pH (from acidic to circum-neutral levels) and prevents further oxidation of AMD source material. The first 2 years of an ongoing field study of the BST technique at a reclaimed coal mine in central Tennessee (USA) has produced successful results. For instance, the water chemistry in a monitoring well down-gradient from injection wells has improved substantially as follows: the pH increased 1.3 units from 5.7 to 7.3, the dissolved (0.45 µm-filtered) iron concentration decreased by 84% from 93 to 15 mg/l, the conductivity decreased by 379 µS/cm, and sulfate decreased by 78 mg/l. Electromagnetic induction surveys were conducted to identify AMD source material and monitor BST performance by measuring changes in subsurface resistivity throughout the site. These surveys revealed a treatment zone created between injection wells where the resistance of contaminated groundwater from up-gradient AMD sources increased as it flowed past injection wells, thus, suggesting this technique could be used to treat AMD sources directly or to intercept and neutralize sub-surface AMD.  相似文献

Cowpea is a legume widely grown in Africa, North, Central and South America, and Asia. The cowpea plant growth habits consist of erect, semi-prostrate, and prostrate types. Developing a cultivar while considering plant growth habit is essential within a breeding program since the need for a particular growth habit is region-specific, and significantly depends on the end user preference. Some cowpea growers might prefer erect types over semi-prostrate and prostrate types, while others would chose prostrate types, which provide more leaves for feed supplies to livestock. However, very little is known regarding the genetics of plant growth habit in cowpea to assist plant breeders in developing suitable cowpea cultivars having the desired growth habit plus the other required features. Therefore, the objectives of this study were to conduct an association mapping for cowpea growth habit, and to identify SNP markers associated with this trait. A total of 487 cowpea genotypes were evaluated for growth habit and a total of 1031 SNPs postulated from genotyping-by-sequencing to conduct association analysis study for cowpea growth habit. Our results showed that: (1) significant differences in cowpea growth habit were identified between countries, (2) the cowpea erect-type was prevalent, and (3) ten SNP markers, C35060651_729, C35061339_799, C35062457_1855, C35072764_1384, C35080248_2355, Scaffold2771_4351, Scaffold29522_3213, Scaffold35913_2678, Scaffold53560_188, and Scaffold58098_4297, were significantly associated with cowpea growth habit. These results could be used for enhancing marker-assisted selection (MAS) in breeding programs aimed at developing cowpea cultivars having a particular growth habit.  相似文献

Semi-purified diets containing 39% crude protein and 5% lipid were used to identify the qualitative requirement of African cattish, Clarias gariepinus (Burchell), for niacin and to characterize the pathologies associated with a deficiency of this vitamin. After 48 days of feeding, C. gariepinus supplied with the unsupplemented diet had developed severe deficiency symptoms and were subsequently withdrawn from the growth study. Niacin deficiency was characterized by feed refusal, listlessness, weight loss, poor feed utilization and high mortality. The skin overlaying the lateral line of the deficient fish became haemorrhagic and this clinical sign was accompanied by severe anaemia. After 126 days of feeding, fish fed diets containing 17.0 mg niacin kg?1 had also developed a dermopathy. but without anaemia or high mortality. The feeding of diets containing less than 33.1 mg niacin kg?1 resulted in suboptimal feed efficiency and poor protein utilisation. Allometric analysis of proximate composition indicated that carcass moisture, protein and ash were influenced by fish size, and not by dietary niacin content. However, significantly more lipid per unit of weight gain was deposited in the carcasses of fish fed the unsupplemented diet than in fish fed diets containing 17.0mgkg?1. The indicators used in the present study could not be applied to accurately determine a value for niacin requirement. However, until a more accurate assessment is performed, it is recommended that diets for C. gariepinus contain not less than 33.1 mg nicotinic acid kg?1 feed.  相似文献

Agroecosystems cover vast areas of land worldwide and are known to have a large impact on the environment, yet these highly modified landscapes are rarely considered as candidates for landscape design. While intentionally-designed agricultural landscapes could serve many different functions, few resources exist for evaluating the design of these complex landscapes, particularly at the scale of the whole-farm. The objective of this paper is to introduce an evolving framework for evaluating the design of agroecosystems based on a critical review of the literature on landscape multifunctionality and agroecology. We consider how agroecosystems might be designed to incorporate additional functions while adhering to agroecology principles for managing the landscape. The framework includes an assessment tool for evaluating farm design based on the extent of fine-scale land use features and their specific functions, to consider the present state of the farm, to plan for future conditions, or to compare alternative futures for the design of the farm. We apply this framework to two farms in Vermont that are recognized locally as successful, multifunctional landscapes. The Intervale Center, an agricultural landscape located within the city limits, serves as an incubator for new farm startups and provides unique cultural functions that benefit the local community. Butterworks Farm, a private operation producing organic yogurt and other food products, achieves important ecological functions through an integrated crop-livestock system. These farms and many others in Vermont serve as models of a framework that integrates landscape multifunctionality and agroecology in the design of the landscape. In the discussion section, we draw from the literature and our work to propose a set of important themes that might be considered for future research.  相似文献
