OBJECTIVE: To determine the functionality of canine anti-Malassezia IgE via the passive transfer of immediate hypersensitivity localized to the skin (ie, cutaneous anaphylaxis) from atopic dogs with dermatitis attributable to overgrowth of Malassezia pachydermatis (Malassezia dermatitis [MD]) to healthy recipient dogs by use of the Prausnitz-Küstner (P-K) technique. ANIMALS: 7 clinically normal dogs, 32 atopic dogs with MD, serum from 11 atopic dogs with MD, and 3 healthy dogs without prior sensitization to M pachydermatis. PROCEDURE: Serum from atopic dogs with MD was used for P-K tests in 3 clinically normal recipient dogs. Serial dilutions of untreated, heat-inactivated, IgE-absorbed, and bovine serum albumin (BSA)-absorbed (control) aliquots of serum were injected ID in triplicate for dermal sensitization. Twenty-four, 48, and 72 hours later, a crude extract of M pachydermatis was injected ID into the sites used for sensitization injections, and immediate hypersensitivity reactions were graded on a 4-point scale. RESULTS: Untreated serum caused P-K reactivity beginning 24 hours after passive sensitization and persisting through 72 hours (titers, 1:32 to 1:64). Heat inactivation and IgE-absorption of serum eliminated P-K reactivity, whereas treatment of serum with BSA did not. CONCLUSIONS AND CLINICAL RELEVANCE: Analysis of P-K test results supports the passive transfer of cutaneous anaphylaxis by anti-Malassezia IgE and indicates it is functional in type-1 hypersensitivity reactions of atopic dogs with MD. Reduction or blockade of anti-Malassezia IgE in atopic dogs with MD may provide better clinical control of the disease.  相似文献

Results from laboratory assays for detection of animal disease are often assessed for repeatability (agreement within laboratory) and reproducibility (agreement between laboratories). This work aimed to understand the strengths and limitations of available methods for describing these quantities. Five major veterinary laboratories in Australia volunteered to participate in a designed evaluation based on repeat testing of twenty bovine sera. Sampling was stratified so that ten of the sera were negative to the virus neutralisation test (VNT) for antibody to bovine herpes virus 1 (BHV-1) and the remaining ten sera were VNT positive. Each serum was divided into 50 replicates and each laboratory assayed one replicate of each serum on a weekly basis using a commercial ELISA for BHV-1. Laboratories were blinded to the identity of sera. The data on sample to positive control ratio (S/P) for these 1000 individual assays were collated, sources of variance analysed using a random effects model, and reliability coefficients (ρ) obtained from the variance estimates as quantitative measures of within and between laboratory agreement. Coefficient of variation (CV) was calculated for combinations of sera and laboratory. CV was found to be higher for sera with the lowest mean S/P values (VNT -ve sera). For VNT -ve sera, agreement of S/P within laboratory was low to moderate (ρ: 0.01-0.27) and the agreement between all labs was low (ρ=0.02). Reliability coefficients for VNT +ve sera were very high for agreement within laboratories (ρ: 0.63-0.92) and moderate for agreement between laboratories (ρ=0.52). As well, simulation demonstrated that sero-prevalence has a dramatic affect on the reliability coefficient if sampling were to be irrespective of VNT status. We conclude that there are some limitations with the available approaches for assessing agreement within and between laboratories. Although reliability coefficients have some drawbacks they are an attractive way of reducing reliance on subjective assessment of agreement.  相似文献

A newly-formulated, high protein high fibre (HPHF) diet has recently been shown to improve satiety in dogs. The current study examined its performance during weight loss in client-owned dogs with naturally-occurring obesity. Fifteen dogs were fed the HPHF diet, whilst a matched ‘control’ group of 27 dogs, received a high protein medium fibre diet (HPMF), with an equivalent caloric density. Baseline characteristics (signalment, percentage overweight, and body fat percentage) were not significantly different between groups. However, percentage weight loss was greater (median [range] 31.8% [12.0–41.2%] vs. 20.0% [5.9–45.0%], P = 0.016) and mean rate of weight loss faster (median [range] 1.0%/week [0.3–1.6%] vs. 0.7%/week [0.3–1.5%], P = 0.028) on HPHF compared with HPMF. Percentage body fat mass decrease (measured by dual-energy X-ray absorptiometry) was also greater in dogs fed the HPHF diet (median (range] 58% [32–85%) vs. 37% [15–72%), P = 0.002). Thus, a diet formulated to include high levels of both protein and fibre, improves outcome during weight loss in obese dogs.  相似文献

A longitudinal observational study of 15 red deer farms was carried out in New Zealand for two years from March 1992. The deer were monitored for performance and health problems, and farm management practices were recorded. Numbers at risk were 4,683 hind-years, 2,459 stag-years and 3,202 weaner-years. The numbers of primiparous and adult hinds at risk of losing their progeny by weaning were 653 and 3,364, respectively. Where possible, postmortem examinations were carried out, and organs were sampled for histological and microbiological investigation. Rates of mortality varied with season with most stag and hind deaths in winter and weaner deaths in autumn and winter. Overall mortality rates were 1.77, 2.60 and 5.87 per 100 deer-years for hinds, stags and weaners, respectively. Malignant catarrhal fever accounted for 0.53 stag and 0.17 hind deaths per 100 deer-years at risk Misadventure, including broken bones, accounted for 15.4 per cent of weaner mortalities, or 0.53 weaner mortalities per 100 weaner-three months in autumn. Yersiniosis was confirmed in 18.6 per cent of weaner deaths, or 1.09 weaners per 100 weaner-years during the first six months after weaning, but was also suspected but unconfirmed in a further 41 per cent of weaner mortalities. Overall, 17 per cent of yearling hinds, and 9.2 per cent of adult hinds lost their progeny between pregnancy diagnosis in June and weaning in March. One outbreak of osteochondrosis was recorded. Mortality rates varied between farms and many mortalities were preventable.  相似文献

The aim of this study was to determine which cells are the source of production and target for leukotriene (LTs) action within the bovine ovary. Luteal (CL, days 14–16 of the oestrous cycle), steroidogenic cells (LSC) and endothelial cells (LEC) of the bovine corpus luteum (CL), and granulosa cells (GC) were isolated enzymatically, cultured in a monolayer and incubated with LTC4, LTB4, Azelastine (an antagonist of LTC4) or Dapsone (an antagonist of LTB4). Then cells were collected for determination of mRNA expression for LT receptors (LTRs) and 5‐lipoxygenase (5‐LO) by real time RT‐PCR, and media were collected for determination of prostaglandin (PG)E2, F, progesterone (P4; LSC only), endothelin‐1 (ET‐1; LEC only) and 17‐β oestradiol (E2; GC only). The greatest mRNA expression for LTR‐II and 5‐LO were found in LEC, whereas LTR‐I mRNA expression did not differ among cell types. The level of PGE2 increased after LTs treatment in each type of ovarian cell, excluding LTC4 treatment in LEC. The secretion of PGF was also increased by LTs, but decreased after LTB4 treatment of LSC. In GC cultures, both LTs stimulated E2 secretion; in LEC cultures, LTB4 stimulated whereas LTC4 inhibited P4 secretion; in LEC cultures, LTC4 stimulated but LTB4 inhibited ET‐1 secretion. The results show that LTs are produced locally and are involved in PGs production/secretion in all examined cells (LSC, LEC and GC) of bovine ovary. Leukotriene treatment modulate secretion of E2, by GC, P4 by LSC and ET‐1 by LEC, which indicates that LTs are involved in regulation of ovarian secretory functions.  相似文献

The objectives of the present study were to quantify the effects of a biological chronic stressor (lameness) on the duration and frequency of different oestrous behaviours in parallel with milk hormone profiles. Dairy cows 51.8 ± 1.4 days postpartum (n = 59), including 18 non‐lame control cows, were scored for lameness and closely observed for signs of oestrus having had their follicular phases synchronized by administration of gonadotrophin‐releasing‐hormone (GnRH) followed by prostaglandin F (PG) 7 days later. Lameness shortened the period when herd‐mates attempted to mount the lame cows (1.83 ± 0.69 h vs 5.20 ± 1.53 h; p = 0.042) but did not affect the overall duration of total behaviours (lame 12.3 ± 1.3 h vs non‐lame 15.2 ± 1.3 h). Lameness also lowered the intensity of oestrus [1417 ± 206 points (n = 18) vs 2260 ± 307 points (n = 15); p = 0.029]. Throughout the synchronized oestrous period, lame cows mounted the rear of herd‐mates less frequently (p = 0.020) and tended to chin rest less (p = 0.075). Around the period of maximum oestrous intensity, lameness also diminished the proportion of cows mounting the rear of another cow and chin resting (p = 0.048, p = 0.037, respectively). Furthermore, lame cows had lower progesterone values during the 6 days before oestrous (p ≤ 0.05). Fewer lame cows were observed in oestrus following PG (non‐lame 83%, lame 53%; p = 0.030); however, if prior progesterone concentrations were elevated, lame cows were just as likely to be observed in oestrus. In conclusion, following endogenous progesterone exposure, lameness shortens the period when herd‐mates attempt to mount lame cows but does not affect the incidence of oestrous. However, lame cows are mounted less frequently and express oestrus of lower intensity. This is associated with lower progesterone prior to oestrus but not with abnormal oestradiol or cortisol profiles in daily milk samples.  相似文献

AIMS: To determine the effects of liveweight (LW) and condition score (CS) of ewes at mating on both mating performance and scanning percentage, and to determine if effects of CS were independent of LW. METHODS: Two groups of ewes, comprising 1,780 mixed-aged Romney (Rom MA) and 692 composite two-tooth (1/8 East Friesian 1/16 Finn 13/16 Romney, Comp TT) ewes were grazed separately with harnessed rams for two reproductive cycles. Romney rams were used for the first 17 days of mating and then replaced on the morning of Day 18 by Suffolk rams. Ewes were weighed and CS assessed on a scale of 0-5 before the introduction of the rams. Ewes were identified as having been mated during the first cycle only, during both cycles, during the second cycle only, or not mated. All mated ewes were scanned using ultrasound, 50 days after the end of mating, and identified as being either non pregnant, single-, twin- or triplet-bearing. RESULTS: Linear relationships (R2=0.99) between mean LW and CS at mating were evident for both Rom MA and Comp TT ewes and the mean increase in LW per unit increase in CS was greater in the Rom MA ewes (7.88 kg) than the Comp TT (4.78 kg) ewes (p<0.05). Ewes mated in the second cycle only were significantly (p<0.05) lighter than those mated in the first cycle only. In Rom MA ewes, a greater proportion were mated in the first cycle only if CS was >/=2.0 than if CS was <2.0, and the proportions of ewes mated in the first cycle only or in both cycles did not differ if CS was >/=2.5. In Comp TT ewes, there was no difference in the proportions mated in the first cycle or both cycles, if CS was >/=2.0. Triplet-bearing ewes were heavier than twin-bearing ewes, which were in turn heavier than both singleton-bearing and non pregnant ewes. Twin-bearing ewes had higher CS than both single-bearing and non pregnant ewes (p<0.05) but differences were not significant if LW was included as a covariate. In both Rom MA and Comp TT ewes, greater proportions were non pregnant and lower proportions had twins if CS was <2.0 than if CS was >/=2.0. In Rom MA ewes only, a greater proportion again had twins and a lower proportion had singletons if CS was >/=3.0 than if CS was <3.0. In Comp TT ewes, the proportion that were non pregnant or twin-bearing did not vary with CS if CS was >/=2.0. Reproductive performance was maximal for Rom MA and Comp TT ewes that had CS at mating >/=3.0 and >/=2.0, respectively, equating to LW of 62.6 and 48.5 kg, respectively. CONCLUSIONS: In both groups of ewes, CS and LW at mating positively affected reproductive performance measured at the time of mating and at scanning, however, no reproductive advantage was evident above a minimum CS of 3.0 and 2.0, equating to LW of 62.6 and 48.5 kg, for Rom MA and Comp TT ewes, respectively. Sheep farmers should direct management to ensure that no ewes are below these minimum targets rather than ensuring high average LW and/or CS of a mob.  相似文献
