Growth, survival, tissue ascorbate concentration and collagen content were studied in fast-growing Penaeus monodon fed two ascorbic acid (AA) derivatives. Prior to the experimental trial, shrimp (initial body weight: 0.9 ± 0.4 g) were fed on a diet lacking vitamin C for 5 weeks. During the following 13-week experiment, the shrimp (initial body weight: 8.2 ± 0.7 g) were given one of five squid-meal-based diets. The dietary treatments consisted of a diet lacking vitamin C, two diets supplemented with either 500 or 1000 mg AA kg−1 in the form of silicone-coated AA, and two diets supplemented with either 500 or 1000 mg AA kg−1 in the form of ascorbyl-2-polyphosphate. Shrimp gained 18.3 g in 13 weeks. The AA-free diet group exhibited the lowest growth rate, feed intake and survival (26.8%) after 4 weeks. Hepatopancreatic ascorbate and muscle collagen content were significantly ( P < 0.05) lower in AA-deprived shrimp. At the end of the feeding trial, growth rate and survival were not significantly ( P > 0.05) different among groups fed AA-supplemented diets. Hepatopancreatic and haemolymphatic ascorbate concentrations were significantly ( P < 0.05) higher in groups given ascorbyl-2-polyphosphate than in groups given silicone-coated AA. A supplement of 500 mg AA kg−1 diet in the form of either silicone-coated AA or ascorbyl-2-polyphosphate was adequate for optimal growth. But ascorbate concentrations in tissues remained much below saturation levels with a supplement of 1000 mg AA kg−1 diet. It is questionable whether the established vitamin C requirements for P. monodon are adequate to maintain tissue ascorbate in fast-growing juveniles.  相似文献

The phytosterol‐containing oil in the corn fiber (corn fiber oil) has potential use as a natural low‐density lipoprotein (LDL) lowering nutraceutical but its low concentration (1–3%) makes it difficult and expensive to extract. Pretreatment of corn fiber with dilute acid or glucosidases removed nonlipid components of fiber, producing oil‐enriched fractions that should be more amenable to efficient and inexpensive oil extraction. Acid, as well as enzymes, significantly increased the content of corn fiber oil and its phytosterol compounds by hydrolyzing (and removing) the starch and nonstarch (cell wall) polysaccharides from the wet‐milled corn fiber. Dual treatment of the fiber with acid and enzyme greatly increased the concentrations of corn fiber oil and its phytosterol components, compared with acid or enzyme treatments alone. Depending on the treatment, the oil concentration in the residual solids increased from 0.3 to 10.8% (21–771% increase in conc.) and the total phytosterol concentration increased from 19.8 to 1256.2 mg/g of fiber (11–710% increase in conc.) compared with untreated fiber.  相似文献

This study investigated the value of using real‐time monitoring of Phytophthora infestans airborne inoculum as a complement to decision support systems (DSS). The experiment was conducted during the 2010, 2011 and 2012 potato production seasons in two locations in New Brunswick, Canada. Airborne sporangia concentrations (ASC) of P. infestans were monitored using 16 rotating‐arm spore samplers placed 3 m above the ground. The first cases of late blight (2010 and 2011) were detected 6–7 days after the first ASC peak, and all samplers captured their first sporangia within the same week (at 3‐ and 9‐day periods). The cumulative ASC curve and the risk curves from two DSS (PLANT‐Plus and Pameseb Late Blight) had the same shape but different magnitudes. In both locations, the negative binomial distribution fitted the data better than the Poisson distribution, which is indicative of heterogeneity, and based on Taylor's power law, the heterogeneity increased with increasing ASC. Therefore, the present results suggest that spore‐sampling network devices may be a suitable approach for early detection of incoming inoculum and, when combined with DSS, represent a potential aid for targeting the optimal time to apply a disease‐control product. In this context, cumulative ASC can be a counterweight to the DSS risk estimate: a high risk combined with significant ASC will trigger fungicide spraying. Moreover, spore sampling can be used to assess the efficiency of management strategies by means of examining the area under the inoculum progress curve.  相似文献

This study investigated the dose dependency of the hemodynamic effects of IV medetomidine (MED) constant-rate infusion (CRI) during isoflurane anesthesia. Twenty-four healthy beagles randomly received one of six MED CRI regimens. A loading dose of MED was administered IV at 0.2, 0.5, 1.0, 1.7, 4.0, or 12.0 ug/kg-1 for 10 minutes, followed by a maintenance CRI providing identical dose amounts over 60 minutes. Heart rate and mean arterial blood pressure were recorded, blood gases were analyzed, and cardiac index (CI) was determined. Statistical analysis involved a repeated measures linear model. Baseline CI demonstrated a dose-dependent decrease as the MED dose increased, with decreases of 14.9% (SD, 12.7%), 21.7% (17.9%), 27.1% (13.2%), 44.2% (9.7%), 47.9% (8.1%), and 61.2% (14.1%) at doses of 0.2, 0.5, 1.0, 1.7, 4.0, and 12.0 ug/kg-1, respectively. The four lowest doses induced limited and transient changes in heart rate, mean arterial pressure, and CI. Further investigation into potential perioperative uses of MED CRI is warranted.  相似文献

To identify high-valued coproducts from commercially processed corn germ, it was necessary to determine the effect of processing conditions on corn germ proteins. We found that significantly less protein was extracted from commercial wet-milled as compared to dry-milled corn germ using Tris, sodium dodecyl sulfate (SDS) buffer containing 14 mM 2-mercaptoethanol at 100 degrees C for 10 min. SDS-polyacrylamide gel electrophoresis (PAGE) revealed a number of proteins with molecular masses ranging from approximately 10 to 66 kDa for the dry-milled corn germ as compared to only a few significant protein bands centered around 23 kDa in the wet-milled corn germ. The protein content of the wet- and dry-milled corn germ was approximately the same; however, nonprotein nitrogen values were significantly greater for the wet-milled than for the dry-milled germ. The distribution of fractionated germ protein freshly excised from the embryo of yellow dent corn kernels was more similar to that of dry-milled than wet-milled corn. SDS-PAGE of laboratory preparations of wet-milled corn germ more closely resembled commercial dry- than wet-milled corn germ, which could be attributed to limited microbial growth during steeping in the laboratory preparations.  相似文献

The Global Oscillation Network Group (GONG) project estimates the frequencies, amplitudes, and linewidths of more than 250,000 acoustic resonances of the sun from data sets lasting 36 days. The frequency resolution of a single data set is 0.321 microhertz. For frequencies averaged over the azimuthal order m, the median formal error is 0.044 microhertz, and the associated median fractional error is 1.6 x 10(-5). For a 3-year data set, the fractional error is expected to be 3 x 10(-6). The GONG m-averaged frequency measurements differ from other helioseismic data sets by 0.03 to 0.08 microhertz. The differences arise from a combination of systematic errors, random errors, and possible changes in solar structure.  相似文献

We recently reported that corn fiber oil contains high levels of three potential cholesterol-lowering phytosterol components: ferulate-phytosterol esters (FPE) (3–6 wt%), free phytosterols (1–2 wt%), and phytosterol-fatty acyl esters (7–9 wt%). A previous study also indicated that corn bran oil contained less phytosterol components than corn fiber oil. The current study was undertaken to attempt to confirm this preliminary observation using more defined conditions. Accordingly, oil was extracted from corn fiber and corn bran prepared under controlled laboratory conditions, using the same sample of corn hybrid kernels for each, and using recognized bench-scale wet-milling, and dry-milling procedures, respectively. After extraction, the chemical composition of the phytosterol components in the oil were measured. This study confirmed our previous observation—that FPE levels were higher in corn fiber oil than in corn bran oil. During industrial wet-milling, almost all of the FPE are recovered in the fiber fraction (which contains both fine and coarse fiber). During laboratory-scale wet-milling, ≈60–70% of the FPE are recovered in the coarse fiber (pericarp) and 30–40% are recovered in the fine fiber. During laboratory-scale dry-milling, <20% of the FPE are recovered in the bran (pericarp), and the rest in the grits. The recoveries of the other two phytosterol components (free phytosterols and phytosterol-fatty acyl esters) revealed a more complex distribution, with significant levels found in several of the dry- and wet-milled products.  相似文献

Five diets were formulated to provide an isoproteic daily dietary intake of 15 g kg?1 day?1, while maintaining daily starch intake ranging from 0 to 40 g kg?1 day?1. The 4‐week experiments started with initial mean weights of 4.7 and 4.4 g for Pangasius bocourti and P. hypophthalmus, respectively. The results clearly show the protein sparing action of starch in both species. Best protein retention was obtained with starch intake of 20 and 10 g kg?1 day?1 for P. bocourti and P. hypophthalmus, respectively, which was equivalent to 40 and 20% starch in the feed. Pangasius bocourti and P. hypophthalmus fingerlings attained maximal growth with starch intake of 30 and 10 g kg?1 day?1, equivalent to 60 and 20% starch in the feed. It was noted that body lipid accumulation was much higher in P. bocourti than in P. hypophthalmus, and that excess dietary starch intake depressed diet digestibility and growth.  相似文献

Background  

Dense genetic maps, together with the efficiency and accuracy of their construction, are integral to genetic studies and marker assisted selection for plant breeding. High-throughput multiplex markers that are robust and reproducible can contribute to both efficiency and accuracy. Multiplex markers are often dominant and so have low information content, this coupled with the pressure to find alternatives to radio-labelling, has led us to adapt the SSAP (sequence specific amplified polymorphism) marker method from a 33P labelling procedure to fluorescently tagged markers analysed from an automated ABI 3730 xl platform. This method is illustrated for multiplexed SSAP markers based on retrotransposon insertions of pea and is applicable for the rapid and efficient generation of markers from genomes where repetitive element sequence information is available for primer design. We cross-reference SSAP markers previously generated using the 33P manual PAGE system to fluorescent peaks, and use these high-throughput fluorescent SSAP markers for further genetic studies in Pisum.  相似文献
