Ecosystem management is a conservation strategy, but there is not a standard protocol for implementation. In theory, ecosystem management will utilize the best available science to sustain social-ecological systems in the landscape by maximizing multiple ecosystem services expected from the stakeholders’ of those systems. Llancahue is a watershed (1,270 ha) that provides fresh water to the city of Valdivia (130,000 inhabitants) and protects >700 ha of old-growth forest within a severely disturbed landscape in the lowlands of south-central Chile. The native vegetation of this landscape is the threatened Valdivian Temperate Rainforests. Management of the watershed by the Universidad Austral de Chile needs to both provide timber and work to the neighboring campesinos (small poor rural land owners) who were illegally logging the forest and improve the conservation of the old-growth forests and the quantity and quality of water provided by the watershed. This paper demonstrates how adaptive management has utilized a multi-step process to improve management of the watershed. This process has included (1) understanding stakeholders’ views towards the project, (2) developing an ecosystem management plan for the watershed that balanced multiple societal demands and ecosystem functions from the watershed, and (3) monitoring the Llancahue forest and streams to ensure activities provided desired results. This paper reports results after 4 years of implementation and provides perspectives on the ecosystem management approach.  相似文献

Abstract

AIM: To make valid recommendations on the use of serological test methods for the detection of serum antibodies in ruminants against Coxiella burnetii (Q-fever), by comparing the performance of the complement fixation test (CFT) and two ELISA, and by identifying reasons for discrepancies between the test methods.

METHODS: A total of 73 serum samples from infected cattle, 69 from infected goats, and 100 samples from non-infected cattle and 57 samples from non-infected sheep, as well as 95 samples from infected cattle herds (mix of seropositive and seronegative samples), were tested using the CFT, the IDEXX ELISA (I-ELISA) and the Pourquier ELISA (P-ELISA). A mixed panel of 12 serum samples from sheep from inter-laboratory proficiency testing (proficiency panel) was also tested using the CFT and both ELISA, and further investigated using IgG- and IgM-specific ELISA.

RESULTS: Generally, the two commercial ELISA were more sensitive than the CFT for the detection of infected ruminants. Good agreement between ELISA for positive and negative results was found for samples from the infected herd, while results for the positive panels varied between the two ELISA. For the total of the positive serum panels, the I-ELISA detected 95% of samples as positive or suspicious, while the P-ELISA detected only 81%. In the P-ELISA, more samples were considered suspicious (18%) than in the I-ELISA (14%). All sera from noninfected sheep and cattle tested negative in the serological test methods employed, except for one positive sample from a sheep in the P-ELISA. Further investigation revealed that a CFT-positive but ELISA-negative result was due to high IgM and low IgG reactivity.

CONCLUSIONS: The two commercial ELISA were more sensitive than the CFT in all panels from infected ruminants. However, they could only detect IgG. The I-ELISA should be the serological test method of choice for cattle, sheep and goats for import testing of animals into New Zealand because it was more sensitive than the P-ELISA and was equally specific to the PELISA and the CFT. For other animal species, such as deer and camelids, the CFT should still be used since none of the ELISA has been evaluated for these species. This study has shown that the two commercial ELISA will detect the majority of infected ruminants but may miss animals that have not developed an IgG response.  相似文献

A method for detection of neurectomy by electrical stimulation of the skin, distal to the neurectomy site, was tested for accuracy and feasibility. An electrical stimulation device was designed and tested to find the minimum peak voltage that would elicit a response in normal horses. Twenty seven horses were then tested at regular intervals following neurectomy and their responses recorded. Immediately following neurectomy there was an area of insensitivity with sharply defined borders extending in an arc from the back of the pastern down towards the bulbs of the heels. In the months following neurectomy, re-innervation occurred and this area gradually reduced in size, although it was still possible to detect insensitive areas 14 months after neurectomy in all horses tested. The method proved to be safe, accurate and ethically acceptable.  相似文献

ABSTRACT Pythium irregulare is a plant-pathogenic oomycete that causes significant damage to a variety of crops, including ornamentals and vegetables. Morphological as well as molecular studies have reported high levels of genetic diversity within P. irregulare sensu lato which has raised the question as to whether it is a single species or is actually a complex of morphologically similar (cryptic) species. In this study, we used amplified fragment length polymorphism (AFLP) fingerprinting and DNA sequence analysis of the internal transcribed spacer (ITS) region of the ribosomal genes (ITS region) and a portion of the mitochondrial cytochrome oxidase II gene and the spacer region between coxI and coxII to characterize 68 isolates of P. irregulare from the United States. The ITS sequence of a P. irregulare neotype at the CBS collection as well as ITS and coxII sequences for P. irregulare, P. spinosum, and P. sylvaticum from previous studies were included in our analysis. Cluster analysis identified a 19-isolate group (IR-II) that separated itself from the rest of the sample (IR-I). Population structure and sequence analyses supported the distinction of IR-I and IR-II and identified IR-II as P. irregulare sensu stricto. IR-I was designated Pythium sp. clade IR-I. Two insertion/deletion mutations and nine nucleotide substitutions in the ITS region and three in the sequence of coxII and the adjacent spacer region separated the two species. Additionally, they differed significantly (P > 0.01) in the frequency of 182 (77%) AFLP alleles. Gene flow results suggested that P. irregulare sensu stricto and Pythium sp. clade IR-I are cryptic species capable of exchanging favorable alleles (Nm = 0.72).  相似文献

Experiments were conducted in irrigation runoff containment basins to assess the effects of bait species ( Camellia japonica , Ilex crenata or Rhododendron catawbiense ), bait type (whole leaf vs. leaf disc), baiting duration (1, 2, 7 or 14 days), baiting depth and growth media (modified PARP-V8 or PARPH-V8) on the recovery of Phytophthora species. A two-rope, flexible bait-deployment system was compared with a one-rope fixed system for bait stability at designated locations and depths. A total of 907 Phytophthora isolates were subjected to PCR-based single-strand conformation polymorphism (PCR-SSCP) analysis to identify to species level. Seven distinct SSCP patterns representing six morphospecies: P. citricola (Cil I), P. citrophthora (Cip I), P. hydropathica (Hyd), P. insolita (Ins), P. megasperma (Meg I & II) and an unidentified Phytophthora species were identified. Irrespective of culture medium, 7 days of baiting with rhododendron leaves consistently resulted in the recovery of the greatest diversity and populations of Phytophthora species with minimum interference from Pythium species. The flexible bait-deployment system was superior to the fixed system, minimizing the risk of bait loss and dislocation of baiting units and allowing baits to remain at designated depths from the surface under inclement weather.  相似文献

OBJECTIVE: To compare a recently developed recombinant MSP-5 competitive inhibition ELISA with a card agglutination test for detection of antibodies to Anaplasma marginale and Anaplasma centrale in Australian cattle. MATERIALS AND METHODS: The ELISA was compared with the card agglutination test using 208 sera from cattle in Anaplasma-free herds, 86 sera from cattle experimentally infected with A marginale or A centrale and 757 sera from cattle in areas endemic for A marginale. RESULTS: The specificity of the ELISA, based on testing 208 sera from cattle in Anaplasma-free areas, was 99.5%, and the sensitivities for detection of antibodies to A marginale and A centrale in sera from the experimentally infected cattle were 98.0% and 100%, respectively. For the same sets of sera, the specificity of the card agglutination test was 98.6% and the sensitivities for detection of antibodies to A marginale and A centrale were 98.0% and 100%, respectively. For the 757 sera collected from cattle in areas endemic for A marginale, the agreement between the ELISA and the card agglutination test depended on the positive threshold selected for the ELISA. The maximum achievable agreement was 91.5% (kappa = 0.73; 95% confidence interval 0.66, 0.79). CONCLUSION: We conclude that the competitive inhibition ELISA is a useful alternative to the card agglutination test for detection of A marginale or A centrale infection in cattle. The assay should be particularly useful for epidemiological applications such as prevalence studies and control programs.  相似文献

Containment basins (CB) are an integral part of recycling irrigation systems that foster agricultural sustainability through water resource conservation. However, little is known regarding this aquatic ecosystem and the lack of water quality data has become an increasingly serious liability in crop health management. Here we report on four distinct seasonal and two diurnal patterns of change in water quality in the CBs. The four seasonal patterns are (a) periodic fluctuation in chlorophyll a, pH, and dissolved oxygen (DO), (b) oxidation–reduction potential (ORP) rises with decreasing DO, (c) tendency for increase in electrical conductivity, salinity, and total dissolved solids, and (d) weather-dependent changes in turbidity and temperature. The two diurnal patterns are (1) chlorophyll a, pH, DO, and temperature consistently peak between 16:00 and 17:00 hours and bottom out around 08:00 hours, and (2) ORP peaks in the morning and bottoms in the evening. Eight of the nine parameters excluding temperature were correlated; and algal blooms appear to be a major driving force for changes in the other seven parameters. These results underscore the importance of water quality monitoring in irrigation management and provide a framework for better understanding of pathogen aquatic ecology and how changes in water quality might be employed in a manner that suppresses plant pathogens and improves crop quality and productivity.  相似文献

Microbial biomass, size and community structure along with an estimate of microbial activity and soil chemical parameters were determined at three depths in two soils (e.g. sandy loam Ultic Hapludalf and silt loam Mollic Hapludalf) replicated three times under one winter and summer season. Microbial biomass and community structure were estimated from phospholipid-PO4 content and fatty acid methyl ester (FAME) measurements. Microbial activity and assimilative capacity were estimated using a 3H-acetate incorporation into phospholipids and by incubating the soil samples at the average winter and summer temperatures, 3 and 20 °C, respectively. We found that the size of the microbial biomass in both the surface and the subsurface soils was not significantly affected by the seasonal variation but activity increased by as much as 83% at the summer temperatures in the surface soil. We demonstrated using FAME analysis that for both soils seasonal changes in the subsurface microbial community occurred. These findings suggest that winter conditions will shift the population activity level in both the surface and subsurface systems and the biochemical structure of the community in the subsurface. In all cases, the inorganic chemical properties of the soil, as a function of season, remained constant. The greatly increased activity of microbial population at the higher temperature will favor the capacity of the system to utilize nutrients or organic materials that may enter soil. During low temperature seasons the capacity of either surface or subsurface soils to assimilate materials is generally diminished but the reduction reflects changes in metabolism and not a reduced biomass size.  相似文献
